Spectrofluorimetric Determination of Proteins Based on Fluorescence Quenching of N-Amino-4-(N-methylpiperazine)-1,8-naphthlimide

Abstract:A new spectrofluorimetric method for determining proteins has been developed based on fluorescence quenching of N-amino-4-(N-methylpiperazine)-1,8-naphthlimide(AMN)by bovine serum albumin(BSA).In the medium of phosphate buffer(pH=3.5),addition of BSA could quench the fluorescence of AMN at 533nm,and the fluorescence intensity change was linear to BSA concentration in the range of 0.1-12.0μg·mL-1.The detection limit was 1.05×10-8g·mL-1.The proposed method has advantages of simplicity,rapidity,good selectivity and sensitivity.It has been applied to the determination of total proteins in serum samples with recoveries of 98.7%-103.2%(RSD≤3.63%).
