Involvement of NAME in Synaptic Plasticity Following Behavioral Trainings in Rats: an Ultrastructural Study

Abstract:One trial passive avoidance and Morris water maze trainings combined with electron microscopic examination on contical and hippocampal synapses were used to investigate the possible effects of intracerebroventricular administration of N ω nitro L arginine (NAME) on the behavioral and structural modifications associated with processes of learning and memory. The results indicated that: ① The behavioral learning and memory following trainings was not accompanied by alterations in some gross structural parameters, such as the size of synapses and vesicles, and the length and curvature of active zone; meanwhile, significant changes were induced in some subtle structural parameters following trainings, including the increase of the lengh of postsynaptic density, the decrease of the width of synaptic cleft, and also increase in some stereometric parameters of synaptic vesicles; and ② NAME administration impaired the behavioral memory of trainings and also blocked the subtle ultrastructural changes stated above induced by trainings. It is proposed that many subtle ultrastructural modifications in cortical and hippocampal synapses might contribute to the formation of learning and memory, while NO might play important roles both in the formation of behavioral memory and in the induction of some subtle ultrastructural changes underlying memory formation.
