Homocysteine Measurement and Its Clinical Significances

Abstract:Objective: Homocysteine has been identified as an independent risk factor for atherosclerosis(As) and birth defective disease. Objective: To establish a method for determining total plasma homocysteine (Hcy), to inspect the changes of Hcy level for healthy individuals, pateints with coronary atherosclerosis, and kernel families with congenital heart disease(CHD). Methods: Using the method of precolumn derivatization HPLC were measured the total plasma Hcy for 49 healthy individuals, 74 patients with coronary atherosclerosis (CAs), and 7 families having a child with congenital heart disease ( n =19). Results: The method is sensitive (detection limit 0.125 μmol·L -1 ), specific, and has a high reproducibility ( CV 2.86%). Its recovery is 101.89%.In the groups of healthy individuals, the mean level of plasma Hcy were significantly higher in men than that in women and children, which were (9.2±1.0) ( ±s , the same below); (5.8±0.6), and (5.9±0.4) μmol·L -1 respectively ( P 0.05). In the pateines with CAs and the kernel families with CHD the Hcy level were (16.9±1.7) and (28.4±1.2) μmol·L -1 separately, which were significantly higher than the control ( P 0.001). ±s , the same below); (5.8±0.6), and (5.9±0.4) μmol·L -1 respectively ( P 0.05). Conclusion: homocysteine is a risk factor for As and CHD. the total level of plasma homocysteine may be a subsidiary indicator for the diagnosis of atherosclerosis and congenital heart disease.
