Abstract:Objective: We investigated a previously undefined effect of n-3 PUFA on BBB injury post-H/I in neonates. Background:The permeability of blood brain barrier (BBB) is known to determine the vulnerability of immature brain to hypoxic/ischemic (H/I) insults. Omega-3 polyunsaturated fatty acids (n-3 PUFA) has been shown to protect brain against H/I by attenuating neuronal death and reducing neuroinflammation. Methods:Female Sprague-Dawley rats were maintained on a diet with or without n-3 PUFA enrichment from the second day of pregnancy to 14d parturition. H/I was introduced in P7 offspring by carotid artery ligation followed by 2.5h hypoxia. We observed a BBB leakage to cadaverine (640Da) 4h post-H/I and a relatively delayed leakage to dextrans (3KD-40KD) 24h after injury. Results:Interestingly, the neonatal BBB is impermeable to Evans blue up to 48h post-H/I, while IgG extravasation was obvious in both ipsilateral and contralateral brain. n-3 PUFA significantly ameliorated H/I-induced BBB damage, as revealed by less IgG extravasation, reduced tracer efflux, preserved BBB ultrastructure and enhanced tight junction protein expression in n-3 PUFA-supplemented rats. Furthermore, n-3 PUFA inhibited the otherwise elevated MMP activity in the brain and blood after H/I.
