Abstract:Objective To study myogenic differentiation of rat bone mesenchymal stem cells (BMSCs) in vitro for obtaining the available seed cells to treat erectile dysfunction (ED). Methods Rat BMSCs were isolated and cultured from the femur and tibia of Sprague Dawley (SD) rat. The expressions of CD49d, CD73, CD90, CD105, CD106 and CD31, CD34 and CD45 in the isolated BMSCs were analyzed by flow cytometry. The fourth passage of BMSCs were further induced and differentiated into myogenic cells by conditioned medium. Results Mesenchymal stem cell surface markers CD49d, CD73, CD90, CD105 and CD106, were detectable in the isolated cells but no detectable for the expression of CD31, CD34 and CD45. Myogenic differentiation cells were identified by the expression of alpha-smooth muscle actin (α-SMA) and Desmin. Conclusion Rat BMSCs were successfully isolated and direct differetiated into myogenic cells in vitro. They could be used as autogenous BMSCs and gene modified BMSCs for ED therapy.
All-Trans Retinoic Acid Promotes Smooth Muscle Cell Differentiation of Rabbit Bone Marrow-Derived Mesenchymal Stem Cells

Abstract:Bone marrow-derived mesenchymal stem cells are multipotent stem cells, an attractive resource for regenerative medicine. Accumulating evidence suggests that all-trans retinoic acid plays a key role in the development and differentiation of smooth muscle cells. In the present study, we demonstrate, for the first time, that rabbit bone marrow-derived mesenchymal stem cells differentiate into smooth muscle cells upon the treatment with all-trans retinoic acid. All-trans retinoic acid increased the expression of myocardin, caldesmon, 22-kDa smooth muscle cellspecific protein (SM22 alpha), and SM-myosin heavy chains in rabbit bone marrow-derived mesenchymal stem cells, as detected by reverse transcription polymerase chain reaction (PCR). Immunostaining of SM22 alpha and SM-myosin heavy chains using monoclonal antibodies also indicated smooth muscle cell differentiation of rabbit bone marrowderived mesenchymal stem cells following the treatment with all-trans retinoic acid. In addition, more than 47% of bone marrow-derived mesenchymal stem cells demonstrated the contractile phenotype of smooth muscle cells. Western blot results showed that SM-1 and SM-2 were highly expressed in the differentiated cells. These results suggest that all-trans retinoic acid may serve as a potent agent for functional smooth muscle cell differentiation in tissue engineering.

Abstract:Bone marrow mesenchymal stem cells (BMMSCs) are a kind of adult stem cells with multiple potential functions in cell proliferation and differentiation and will be wildly used for cell therapy and tissue engineer- ing in the future. Many factors are reviewed in this paper which include donors’ age, isolation methods, cell culture density, surface materials and their effects on cell proliferation. It is emphasized that the development of different substitution, such as growth factors and human autologous serum, for fetal bovine serum (FBS) to avoid the risk from some viruses. The BMMSCs introduced by the plasmid with telomerase gene have shown stronger prolifera- tion ability and multiple differentiation potential. The development of perfusion culture systems will make the prepa- ration of BMMSCs more suitable for clinical application.

Abstract:Background As commonly used scaffold material in tissue engineering, chitosan has many advantages, such as strong biodegradability, low antigenicity, good biocompatibility and no pyrogen reaction. This study aims to isolate, cultivate and purify Sprague-Dawley (SD) rat bone marrow-derived mesenchymal stem cells (BMSCs), and to observe the growth of BMSCs when co-cultured with self-made chitosan porous scaffold in vitro and to test the biocompatibility of this tissue engineering scaffold, so as to lay the foundation for promoting nerve regeneration of transplant treatment. Methods Three-week-old healthy male SD rats were used in this study, and BMSCs were isolated and purified through bone marrow adherent culture method. The surface markers of BMSCs at Passage 3 were detected and identified by flow cytometry (FCM) and the BMSCs were three?dimensionally cultured in vitro on chitosan porous scaffolds produced by freeze-drying method. Ethanol alternative method was used to detect the chitosan scaffold porosity. Scanning electron microscope was used to explore the internal structure of the scaffold, measure the size of its aperture, and observe the morphology and development of the cells within the scaffold. Methyl thiazolyl tetrazolium (MTT) method was used to determine the cells' proliferation. Results The cultured BMSCs were uniform and similiar to fibrous arrangement, and mixed cells reduced obviously. The identification result of FCM showed the CD29 positive rate was 98.49% and CD45RA positive rate was only 0.85%. The chitosan scaffold had an interlinked, uniform similar three-dimensional porous structure and its aperture porosity was 90%. Some cells stretched out pseudopod and infiltrated into the porous structure of scaffold, even fusing with them. The BMSCs were seeded in the scaffold successfully. The chitosan scaffold had no obvious effect on BMSCs' proliferation. Conclusions Chitosan porous scaffolds have good structural character and biocompatibility, and can be used as an alternative cell-carrier for later cell replace treatment of central nervous system diseases. doi: 10.3969/j.issn.1672-6731.2014.11.015

Abstract:Background: Autograft and allograft transplantation are used to prompt the regeneration of axons after nerve injury. However, the poor self-regeneration caused by the glial scar and growth inhibitory factors after neuronal necrosis limit the efficacy of these methods. The purpose of this study was to develop a new chitosan porous scaffold for cell seeding. Material/Methods: The bone marrow mesenchymal stem cells (BMSCs) and tissue-engineered biomaterial scaffold compound were constructed and co-cultured in vitro with the differentiated BMSCs of Wistar rats and chitosan scaffold in a 3D environment. The purity of the third-generation BMSCs culture was identified using flow cytometry and assessment of induced neuronal differentiation. The scaffolds were prepared by the freeze-drying method. The internal structure of scaffolds and the change of cells' growth and morphology were observed under a scanning electron microscope. The proliferation of cells was detected with the MTT method. Results: On day 5 there was a significant difference in the absorbance value of the experimental group (0.549 +/- 0.0256) and the control group (0.487 +/- 0.0357) (P>0.05); but on day 7 there was no significant difference in the proliferation of the experimental group (0.751 +/- 0.011) and the control group and (0.78 +/- 0.017) (P>0.05). Conclusions: Tissue engineering technology can provide a carrier for cells seeding and is expected to become an effective method for the regeneration and repair of nerve cells. Our study showed that chitosan porous scaffolds can be used for such purposes.

Abstract:Objective:To explore the feasibility of myoblast-derived exosomes (MB-Exos) in inducing myogenic differentiation of bone marrow mesenchymal stem cells (BM-MSC) in vitro. Methods:Rat skeletal muscle satellite cells were differentiated into myoblasts. And MB-Exos were extracted and co-incubated with BM-MSC for inducing myogenic differentiation. The expressions of myogenin, desmin and myosin were measured and the level of growth factors after exosome inducing was detected.Results:There was a positive expression of Pax7 and myogenic differentiation was detected. The purification rate of myoblast cells was 94. 5% under fluorescence-activated cell sorting (FACS) . Vesicle-like morphology of exosomes was visualized via transmission electron microscope (TEM) after an extraction of gradient ultracentrifugation. With a typical cup shape, the diameter was 30 to 200 nm with a negative stain of phosphotungstic acid. Western blot revealed that exosome markers of CD9, CD63, CD81 and TSG101 were expressed in MB-Exos. GFP-labeled MB-Exos were ingested by BM-MSC. Under the stimulation of MB-Exos, myogenic differentiation of BM-MSC was verified as an expression of myogenin in 3 days and an expression of desmin and myosin in 6 days. The levels of IGF-1, HGF and FGF2 in supernatant of cultured BM-MSC spiked markedly according to enzyme-linked immunosorbent assay (ELISA) .Conclusions:MB-Exos induces myogenic differentiation of BM-MSC in vitro and promotes the secretion of various growth factors. It may be one of the mechanisms of exosomes promoting cellular proliferation and differentiation.

Abstract:Objective: To establish an isolation and culture system of rat bone marrow stem cells(MSCs), osteogenic differentiation and compound titanium meshes in vitro in order to verify their osteogenesis. Methods: Density gradient centrifugation was used to isolate bone MSCs from SD rats and their morphological characteristics were examined under microscope. The bone MSCs underwent osteogenic induction and cytochemical and immunocytochemical staining were performed to verify their multipotential. Cells of osteogenic differentiation that compounded titanium meshes were detected by scanning electron microscope. Results: High purity of bone MSCs was successfully obtained and they showed good proliferation ability. Both ALP activity and mineraliztion node staining were positive in bone MSCs after osteogenic differentiation. Cells of osteogenic differentiation that compounded titanium meshes were detected by scanning electron microscope, it was deposited calcified matrix. Conclusion: The isolation and culture system of rat bone MSCs was successfully established and the bone MSCs sustained their osteogenic differentiation potential in vitro. The cells of osteogenic differentiation that compounded titanium meshes was deposited calcified matrix.

Transdifferentiation of rat fetal brain stem cells into penile smooth muscle cells

Abstract:Objectives: To evaluate whether rat fetal brain stem cells can be induced to acquire cell fates outside the nervous system, hypothesising that cell-based replacement therapy with stem cells can aid in the regeneration of penile smooth musculature and might help to attenuate organic erectile dysfunction (ED), as the degeneration of penile smooth muscle cells leading to subsequent impairment of function is important in organic ED. Materials and methods: Fetal brain stem cells (FBSCs) from embryonal 12-day Fisher 344 rats were isolated and characterized. For in vitro studies, undifferentiated FBSCs were cultured for 21 days in either N2 media (control) or N2 media conditioned in rat penile smooth muscle cell culture. These were then subjected to immunocytochemistry for specific markers of neural stem cells (nestin) and penile smooth muscle cells, i.e. alpha-smooth muscle actin (alphaSMA), penis-specific myosin light chain (MLC) desmin, calponin, vimentin, phosphodiesterase-5 (PDE5) and connexin. For in vivo studies, male adult Fisher 344 rats had an intracavernous injection with saline (five rats, control) or FBSCs that were labelled genetically by an expression construct for green fluorescent protein (GFP, nine rats, experimental) and maintained for 6 weeks. The rats were then killed and penile tissue was harvested and subjected to immunocytochemistry for markers of neural stem cells, smooth muscle cells, and sinusoidal endothelium (vascular endothelial growth factor, VEGF). Results: Undifferentiated cells exposed to N2 media continued to maintain the characteristic morphological and protein marker features of FBSCs, while the cells exposed to the conditioned media acquired the morphological features of smooth muscle cells. In addition, the differentiated cells (30-40%) expressed smooth muscle markers. Rats implanted with FBSCs had cells that showed double-labelling for GFP/alphaSMA, GFP/calponin and GFP/VEGF. The control group had no evidence of such double-labelling. Conclusions: These results suggest the transdifferentiation of FBSCs into penile smooth muscle cells. Such transdifferentiated cells showed long-term survival when injected into the cavernous tissue, thus raising the possibility of a novel therapeutic option for organic ED.

Abstract:Differentiation of Leydig cells plays a key role in male reproductive function. Bone marrow mesenchymal stem cells (BMSCs) have emerged as a potential cell source for generating Leydig-like cells due to their multipotent differentiation capacity and accessibility. This study aimed to investigate the morphological and genetic expression changes of BMSCs during differentiation into Leydig-like cells. Testicular extract liquid, which simulates the microenvironment in vivo, induced the third passage BMSCs differentiated into Leydig-like cells. Changes in cell morphology were observed by microscopy, the formation of lipid droplets of androgen precursor was identified by Oil Red Staining, and the expression of testicular specific genes 3β-HSD and SF-1 in testicular stromal cells was detected by RT-qPCR. BMSCs isolated from the bone marrow of Sprague-Dawley (SD) rats were cultured for 3 generations and identified as qualified BMSCs in terms of morphology and cell surface markers. After 14 days of induction with testicular tissue lysate, lipid droplets appeared in the cytoplasm of P3 BMSCs by Oil Red O staining. RT-qPCR detection was performed on BMSCs on the 3 rd , 7 th , 14 th , and 21 st day after induction. Relative expression levels of 3β-HSD mRNA significantly increased after 14 days of induction, while the relative expression of SF-1 mRNA increased after 14 days of induction but was not significant. BMSCs can differentiate into testicular interstitial cells with reserve androgen precursor lipid droplets after induction by testicular tissue lysate. The differentiation ability of BMSCs provides the potential to reconstruct the testicular microenvironment and is expected to fundamentally improve testicular function and provide new treatment options for abnormal spermatogenesis diseases.

Abstract:Non-obstructive azoospermia is the most challenging type of male infertility. Stem cell based therapy provides the potential to enhance the recovery of spermatogenesis following cancer therapy. Bone marrow-derived mesenchymal stem cells (BMSCs) possess the potential to differentiate or trans-differentiate into multi-lineage cells, secrete paracrine factors to recruit the resident stem cells to participate in tissue regeneration, or fuse with the local cells in the affected region. In this study, we tested whether spermatogenically-induced BMSCs can restore spermatogenesis after administration of an anticancer drug. Allogeneic BMSCs were co-cultured in conditioned media derived from cultured testicular Sertoli cells in vitro, and then induced stem cells were transplanted into the seminiferous tubules of a busulfan-induced azoospermatic rat model for 8 weeks. The in vitro induced BMSCs exhibited specific spermatogonic gene and protein markers, and after implantation the donor cells survived and located at the basement membranes of the recipient seminiferous tubules, in accordance with what are considered the unique biological characteristics of spermatogenic stem cells. Molecular markers of spermatogonial stem cells and spermatogonia (Vasa, Stella, SMAD1, Dazl, GCNF, HSP90α, integrinβ1, and c-kit) were expressed in the recipient testis tissue. No tumor mass, immune response, or inflammatory reaction developed. In conclusion, BMSCs might provide the potential to trans-differentiate into spermatogenic-like-cells, enhancing endogenous fertility recovery. The present study indicates that BMSCs might offer alternative treatment for the patients with azoospermatic infertility after cancer chemotherapy.

Abstract:Introduction. Stem cells (SCs) show significant benefits in the treatment of postprostatectomy erectile dysfunction (ED). However, the low retention rate of the traditional single-cell strategy at the injection sites limits its therapeutic potential. Aim. This study aims to investigate the feasibility and mechanism of adipose-derived stem cells (ADSCs)-based micro-tissues (MTs) in the treatment of ED in a rat model of bilateral cavernous nerves (CNs) injury. Methods. ADSCs labeled with 5-ethynyl-2-deoxyuridine (EdU) were used to generate MTs with hanging drop method. 10 Sprague-Dawley (SD) rats underwent sham surgery and intracavernous (IC) injection of phosphate buffer solution (PBS) (the sham group). Another 70 rats underwent bilateral CN crush and were then treated with PBS (n = 10, the crush group), dissociated ADSCs (n = 30, the ADSCs group), and MTs (n = 30, the MTs group), respectively. At day 1, 3, 7, 14 (n = 5), and 28 (n = 10) postsurgery, specimens were harvested for histology. At day 28, 10 rats in each group were examined for erectile function before tissue harvest. Main Outcome Measures. Light microscopy of the dynamic aggregation of the MT, immunohistologic examination of the MTs, the retention and distribution of EdU + ADSCs in the corpus cavernosum (CC), and the penis histological analyses of collagen content, Western blot of functional proteins in MTs, intracavernous pressure recording on CN electrostimulation. Results. Three-day-old MTs became stable and expressed nerve growth factor, vascular endothelial growth factor, C-X-C chemokine receptor type 4, Wnt5a, and collagen IV. More EdU + ADSCs retained in the CC in the MTs group than that in the ADSCs group. IC injection of MTs resulted in significant restoration of the erectile function and histopathological changes compared with the ADSCs group. Conclusion. IC-injected MTs resulted in a better restoration of erectile function than traditional single-cell strategy. The underlying mechanisms of recovery appear to involve enhanced cellular retention in the penis and upregulation of some paracrine factors. Abstract:Here we present an efficient method for isolating and culturing mandibular bone marrow mesenchymal stem cells (mBMSCs) in vitro to rapidly obtain numerous high-quality cells for experimental requirements. mBMSCs could be widely used in therapeutic applications as tissue engineering cells in case of craniofacial diseases and cranio-maxillofacial regeneration in the future due to the excellent self-renewal ability and multi-lineage differentiation potential. Therefore, it is important to obtain mBMSCs in large numbers. In this study, bone marrow was flushed from the mandible and primary mBMSCs were isolated through whole bone marrow adherent cultivation. Furthermore, CD29+CD90+CD45- mBMSCs were purified through fluorescent cell sorting. The second generation of purified mBMSCs were used for further study and displayed potential in differentiating into osteoblasts, adipocytes, and chondrocytes. Utilizing this in vitro model, one can obtain a high number of proliferative mBMSCs, which may facilitate the study of the biological characteristics, the subsequent reaction to the microenvironment, and other applications of mBMSCs.
Study of Inducing Differentiation of Bone Marrow Mesenchymal Stem Cells of Rat into Cardiac Myocytes in Vitro

Abstract:OBJECTIVE:To investigate the role of cardiac myocytes in the differentiation of bone marrow mesenchymal stem cells (MSCs) to cardiac myocytes and the biological properties in the course.METHODS:The bone marrow of the extremities of the rats was flushed, and bone marrow MSCs were obtained by method of density gradient centrifugation. They were cultured. The second passage of cultured MSCs were labelled with bromodeoxyuridine (BrdU). The cardiac myocytes were obtained from the apex of rat heart with trypsin digestion method, and they were cocultured with labeled MSCs. The developmental changes of bone marrow MSCs were observed under light microscope with immunohistochemical staining for BrdU and alpha-sarcomeric actin on the 3rd day, and electron microscopic examination on the 5th day.RESULTS:MSCs proliferated fast in primary culture and subculture, positive rate was (90.34+/-2.31)%, and there was statistical difference when it compared with control group [(4.07+/-1.35)%, P<0.01]. The morphology of MSCs changed significantly after coculture. There were new cells nuclei of which were positively stained for BrdU and its cytoplasm positive for actin. Under transmissive electron microscope, sarcomeres like structure and abnormal Z line were observed in cytoplasm.CONCLUSION:Cardiac myocytes can effectively induce bone marrow MSCs to differentiate into cardiac myocytes by coculturing.
Intracavernous Transplantation of Bone Marrow-Derived Mesenchymal Stem Cells Restores Erectile Function of Streptozocin-Induced Diabetic Rats

Abstract:Introduction. Erectile dysfunction (ED) is a frequent complication of diabetes mellitus. The efficacy of common ED therapies is low for diabetes-associated ED. Aim. To explore the effects of transplantation of bone marrow-derived mesenchymal stem cells (BM-MSCs) on improving erectile function of streptozocin (STZ)-induced diabetic rats. Methods. Male Sprague Dawley rats were injected either with STZ to induce diabetes or with citrate buffer as controls. Rat BM-MSCs were harvested and labeled with CM-DiI (Chloromethylbenzamido derivatives of 1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate), and then transplanted into corporal cavernosum of STZ-induced diabetic rats. Four weeks after transplantation, all rats were analyzed for erectile function and penile histology. Main Outcome Measures. Erectile function was evaluated by the ratio between intracavernous pressure (ICP) and mean arterial pressure (MAP) during electrostimulation of cavernous nerve. Fate of transplanted BM-MSCs was identified using immunofluorescence staining. Smooth muscle and endothelium in corpora cavernosum were assessed using immunohistochemistry. Results. After BM-MSCs transplantation, the ICP/MAP ratio was increased significantly compared with diabetic controls. Content of smooth muscle and endothelium in corporal cavernosa of BM-MSCs transplanted rats was significantly increased compared to diabetic controls. Immunofluorescence analysis demonstrated that CM-DiI-labeled BM-MSCs could stay in corporal cavernosa for at least 4 weeks and some of them expressed von Willebrand Factor, CD31, calponin, or alpha-smooth muscle actin, cells markers for endothelial cells or smooth muscle cells, respectively. Conclusion. Intracavernous transplantation of BM-MSCs had beneficial effects on erectile function of diabetic rats and increased the content of endothelium and smooth muscle in corporal cavernosum. Biological activity of the cultured rabbit bone marrow-derived mesenchymal stem cells

Abstract:Objective To observe the effect of the in vitro isolation,culture and identification of rabbit bone marrow-derived mesenchymal stem cells(BMSCs) and explore the differentiation potentials of BMSCs.Methods BMSCs were isolated and purified by density gradient centrifugation combined with attachment culture method.The biological characteristics of BMSCs were observed under an inverted microscope.The BMSCs were identified with HE staining and CD34/CD44 immunohistochemical staining.The biological activity of BMSCs was detected by osteogenic induction and steatogenic induction.Results BMSCs were isolated successfully in vitro and had good homogenicity.Immunohistochemical staining of CD34/CD44 showed strong positive expression of CD44 and negative expression of CD34.Fortis osteogenic ability and positive staining of ALP could be detected by osteogenic induction,and fortis steatogenic ability and positive staining of axungia could be detected by steatogenic induction.Conclusion BMSCs have the capability of multi-directional differentiation in vitro.

Abstract:Background: Cavernous nerve injury (CNI) is the leading cause of erectile dysfunction (ED) after radical prostatectomy and pelvic fracture. Transplantation of human adipose-derived stem cells (ASCs) has been widely used to restore erectile function in CNI-ED rats and patients. Umbilical cord blood-derived MSCs (CBMSCs) are similarly low immunogenic but much primitive compared to ASCs and more promising in large-scale commercial applications due to the extensive establishment of cord blood banks. However, whether CBMSCs and ASCs have differential therapeutic efficacy on CNI-ED and the underlying mechanisms are still not clear. Materials and methods: A bilateral cavernous nerve injury (BCNI) rat model was established by crushing the bilateral cavernous nerves. After crushing, ASCs and CBMSCs were intracavernously injected immediately. Erectile function, Masson staining, and immunofluorescence analyses of penile tissues were assessed at 4 and 12 weeks. PKH-26-labeled ASCs or CBMSCs were intracavernously injected to determine the presence and differentiation of ASCs or CBMSCs in the penis 3 days after injection. In vitro experiments including intracellular ROS detection, mitochondrial membrane potential assay, EdU cell proliferation staining, cell apoptosis assay, and protein chip assay were conducted to explore the underlying mechanism of CBMSC treatment compared with ASC treatment. Results: CBMSC injection significantly restored erectile function, rescued the loss of cavernous corporal smooth muscles, and increased the ratio of smooth muscle to collagen. PKH-26-labeled CBMSCs or ASCs did not colocalize with endothelial cells or smooth muscle cells in the corpus cavernosum. Moreover, the conditioned medium (CM) of CBMSCs could significantly inhibit the oxidative stress and elevate the mitochondria membrane potential and proliferation of Schwann cells. Better therapeutic effects were observed in the CBMSC group than the ASC group both in vivo and in vitro . In addition, the content of neurotrophic factors and matrix metalloproteinases in CBMSC-CM, especially NT4, VEGF, MMP1, and MMP3 was significantly higher than that of ASC-CM. Conclusion: Intracavernous injection of CBMSCs exhibited a better erectile function restoration than that of ASCs in CNI-ED rats owing to richer secretory factors, which can promote nerve regeneration and reduce extracellular matrix deposition. CBMSC transplantation would be a promising therapeutic strategy for CNI-ED regeneration in the future.
