Abstract:AIM:To investigate the changes of oxidative stress in the stomach tissues and their roles in gastric motility and interstitial cells of Cajal(ICC) in diabetic rats.METHODS: Thirty-eight SD rats(8-week-old,male) were intraperitoneally injected with streptozotocin(STZ).Diabetes was successfully induced in 36 of them.The diabetes rats were randomly divided into untreated diabetes group and treated diabetes group.Eighteen healthy SD rats(8-week-old,male) served as controls.The body weight and the levels of blood glucose and glycosylated hemoglobin were measured.At the end of week 1 and week 10,9 rats were sacrificed in each group.The gastric emptying rate and the levels of malondialdehyde(MDA),superoxide dismutase(SOD),tumor necrosis factor α(TNF-α),tyrosine kinase receptor c-Kit and stem cell factor(SCF) in gastric smooth muscle were analyzed.The apoptosis of ICC in gastric tissues was detected by the methods of immunocytochemistry and TUNEL.RESULTS: Compared with control group,gastric motility and SOD activity in untreated diabetes group were significantly weakened,the levels of MDA and TNF-α increased,the levels of c-Kit and SCF decreased,and apoptosis of ICC enhanced.In treated diabetes group,the oxidative stress level was attenuated,antioxidant capacity was enhanced,the levels of c-Kit and SCF were significantly increased,and the ICC apoptosis was reduced.Gastric motility was significantly improved after antioxidant therapy.CONCLUSION: Hyperglycemia affects the expression of antioxidant enzymes in the stomachs of diabetic rats.Oxidative stress is caused by hyperglycemia and is an important factor in the etiology of gastric motility dysfunction in diabetic rats,which may be correlated with the augmentation of ICC apoptosis resulting from oxidative stress-induced c-Kit/SCF damage.

Abstract:To explore the mechanism of impairment and defense of oxidative stress in cavernous mitochondria of diabetic rats.Adult male SD rats(n=42)were randomly divided into normal control group(n=10) and experimental group(n=32). The diabetic model rats induced by streptozotocin were randomly divided into diabetes group(n=13) and therapeutic group with reduced glutathione (GSH) treatment(n=12). Eight weeks later, erectile function was assessed by measuring the rise in intracavernous pressure (ICP) of the rats following cavernous nerve eletrostimulation before the rats were sacrificed. The levels of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD) in cavernous tissue were detected. The masson staining was used to show the structure of the rat penis. Mitochondrial transmembrane potential was detected.A significant decrease in ICP was recorded in the diabetic rats [(50.80 ± 9.80)vs.(90.42 ± 7.02) mmHg,P<0.05], with improvement measured in the rats receiving GSH [(74.20 ± 5.69)vs.(50.80 ± 9.80)mmHg, P<0.05]. The levels of MDA increased remarkably [(6.15 ± 1.07)vs.(3.52 ± 0.94)mmol/g protein, P<0.01] and the activities of SOD decreased significantly[(73.34 ± 6.56)vs.(114.22 ± 6.34)U/mg protein, P<0.05)] in cavernous tissue of the diabetes group. Mitochondria transmembrane potential was decreased [(727.98 ± 68.33)vs.(1223.15 ± 222.92),P<0.01]. A remarkable decrease in MDA [(3.90 ± 0.96)vs.(6.15 ± 1.07)mmol/g protein, P<0.05] and increase in SOD[(95.74 ± 4.65)vs.(73.34 ± 6.56)U/mg protein,P<0.05] were observed in GSH treatment group. Meanwhile, the morphology changes of cavernous tissue and the decrease of mitochondria transmembrane potential were inhibited[(930.30 ± 48.36) vs.727.98 ± 68.33),P<0.05], in diabetic rats with GSH treatment.Hyperglycemia could cause oxidative stress in the cavernous tissue of diabetic rats and this impairment could contribute to diabetic erectile dysfunction; Oxidant treatment could attenuate oxidative stress by improving the function of mitochondria in cavernous tissue. Oxidative stress plays an important role in diabetic erectile dysfunction (DED) and our study might provide a new insight into the prevention and treatment of DED.

Abstract:Diabetic retinopathy (DR) is one of the most common microvascular complications of diabetes and is the leading cause of blindness in young adults. Oxidative stress has been implicated as a critical cause of DR. Metabolic abnormalities induced by high-glucose levels are involved in the development of DR and appear to be influenced by oxidative stress. The imbalance between reactive oxygen species (ROS) production and the antioxidant defense system activates several oxidative stress-related mechanisms that promote the pathogenesis of DR. The damage caused by oxidative stress persists for a considerable time, even after the blood glucose concentration has returned to a normal level. Animal experiments have proved that the use of antioxidants is a beneficial therapeutic strategy for the treatment of DR, but more data are required from clinical trials. The aims of this review are to highlight the improvements to our understanding of the oxidative stress-related mechanisms underlying the development of DR and provide a summary of the main antioxidant therapy strategies used to treat the disease.
The effect of reduced GSH on oxidative stress in corpus cavernosum tissue of diabetes rats

Abstract:Objective To investigate the mechanism of oxidative damnification during the developement of diabetic erection dysfunction(DMED) and explore the effect of reduced glutathione on oxidative damnification. Methods Fifty male Sptague-Dawley rats were randomly divided into 4 groups including group A(control group, n=10),group B(no intervention diabetic group,n=13),C group(insulin intervention diabetic group,n=13) and D group(diabetic insulin and GSH intervention group,n=14).The diabetic model was established by the intraperitoneal injection of streptozotoc.Homogenates of corpus cavernosum tissue of rats was ready for next analysis.The thiobarbaturic acid(TBA) method was used to determine the lipid peroxidation,SOD activity was measured using the xanthine-oxidase cytochrome c method.The expression of SOD_(EX) gene was detected by RT-PCR and level of CuZnSOD protein by Western-blot.Results There was almost no lipid peroxidation product detected in the corpus cavernosum tissue of the rats in group A.Compare with that of group A,SOD activity,the expression of SOD_(EX) gene and CuZnSOD protein significantly decreased in B,C,D groups(P<0.05) whereas MDA level significantly increased(P<0.05).Compare with that of group B,SOD activity,the expression of SOD_(EX) gene and CuZnSOD protein in C,D groups significantly increased(P<0.05) whereas MDA level significantly decreased(P<0.05).Compare with that of C group,the expression of SOD_(EX) gene of D groups, and CuZnSOD protein significantly increased(P<0.05).Conclusion The metabolism was distinctly disturbed in the corpus cavernosum tissue of the diabetic male rats such as increase of the lipid peroxidation product and decrease of the free radical eliminating.Reduced GSH could reduced the oxidative damnification partly,which indicated that oxidative damnification might be involved in the development of DMED.Our study might provide a new insight for prevention and treatment of DMED.

Abstract:AbstractDiabetic peripheral neuropathy (DPN) is a major complication of diabetes mellitus with a high incidence. Oxidative stress, which is a crucial pathophysiological pathway of DPN, has attracted much attention. The distortion in the redox balance due to the overproduction of reactive oxygen species (ROS) and the deregulation of antioxidant defense systems promotes oxidative damage in DPN. Therefore, we have focused on the role of oxidative stress in the pathogenesis of DPN and elucidated its interaction with other physiological pathways, such as the glycolytic pathway, polyol pathway, advanced glycosylation end products, protein kinase C pathway, inflammation, and non-coding RNAs. These interactions provide novel therapeutic options targeting oxidative stress for DPN. Furthermore, our review addresses the latest therapeutic strategies targeting oxidative stress for the rehabilitation of DPN. Antioxidant supplements and exercise have been proposed as fundamental therapeutic strategies for diabetic patients through ROS-mediated mechanisms. In addition, several novel drug delivery systems can improve the bioavailability of antioxidants and the efficacy of DPN.

Abstract:Pathological and physiological changes in dermal tissue in a rat model of diabetes mellitus (DM) were investigated. Sixteen male 8-week-old Sprague-Dawley rats were randomized into two groups of eight, the DM group (Group DM) and the normal control group (Group (NC) normal control). Group DM rats were injected with streptozotocin (STZ) intraperitoneally at a dose of 65 mg/kg body weight. Group NC rats were injected with the same volume of citric acid buffer. All rats were sacrificed 12 weeks later. The impact of exposure to (AGE) advanced glycation end products-modified human serum albumin (AGE-HSA) on epidermal cells and ECV304 cells was evaluated in cell culture experiments. The diabetic rats exhibited changes in skin tissue, including a decrease in thickness, disappearance of the multilayer epithelium structure, degeneration of collagen fibres and an increase in the infiltration of inflammatory cells, in addition to a significant increase in skin glucose and AGEs. Moreover, diabetic rats had increased plasma glycosylated protein (GSP) and malondialdehyde (MDA) and decreased plasma glutathione (GSH). The percentage of epidermal cells in S phase was similar between the two group rats; however, there was a marked decrease in the G2/M phase in Group DM. Additionally, exposure of ECV304 cells to AGE-HSA led to a time-dependent and dose-dependent increase in apoptosis. Therefore, the high glucose in the skin tissue, coupled with the accumulation of toxic substances such as AGEs, promote the dysfunction of dermal cells and/or the matrix. This may be a significant mechanism of diabetes-induced early-stage endogenous skin damage.
Study on the Role of Oxidative Stress in the Kidneys of Diabetic Rats

Abstract:AIM: To investigate the changes of oxidative stress in the kidneys and their roles in nephropathy in diabetic rats. METHODS: Diabetic rats were induced by streptozotocin (STZ). 36 rats were divided into three groups randomly: (1) NC group, normal control rats; (2) DM group, diabetic rats received protamine zinc insulin (PZI) 2U-4U/2 d; (3) DT group, diabetic rats received PZI 9-12 U/kg body weigh/day. 12 weeks later, rats were killed, blood glucose, blood cholesterol, serum creatinine, blood urea nitrogen, HbA1c, urinary creatinine, and urinary protein for 24 h were measured. The activities of antioxidant enzymes in renal cortex, including total superoxide dismutase (TSOD), Cu-Zn superoxide dismutase (Cu-Zn SOD), Mn superoxide dismutase (Mn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), and maleic dialdehyde (MDA) were measured by chromatometry. RT-PCR was performed to detect the expression of different antioxidant enzymes mRNA. RESULTS: For all the targets we measured, there was no significant difference between NC and DT groups. Compared with the other two groups, the levels of blood glucose, cholesterol, trigalloyl glycerol, HbA1c in DM group increased significantly. The activities of TSOD, Cu-Zn SOD and CAT decreased significantly. The activity of GSH-Px increased significantly. There was no significant difference among the activities of Mn SOD in all three groups. The level of MDA in DM group was much higher than that in NC or DT group. The relative expression levels of GSH-Px and Cu-Zn SOD mRNA in DM group were higher than those in other two groups, while the relative expression level of CAT decreased. Mn SOD mRNA was expressed without significant difference in all groups. Compared with NC or DT group, urinary protein in DM group increased significantly, while creatinine clearance rate decreased. CONCLUSIONS: Hyperglycemia affected the expression of antioxidant enzymes. Oxidative stress was caused by hyperglycemia in diabetic rats and may be an important factor in the etiology of diabetic nephropathy.

Abstract:Objective To explore the effects of melatonin on oxidative stress in cavernous tissues of diabetic rats and to investigate the mechanism of oxidative stress during the development of diabetic erection dysfunction.Methods The diabetic model rats induced by streptozotocin were randomly divided into the diabetic group and melatonin treatment group.The normal rats were raised as normal control group.Eight weeks later,the erectile function was assessed by measuring the rise in intracavernous pressure(ICP)of the rats following cavernous nerve electrostimulation before the rats were sacrificed.The levels of malondialdehyde(MDA)and the activities of superoxide dismutase(SOD)in cavernous tissues were detected.The content of endothelium and smooth muscle in penis were analyzed with immunohistochemical assay.Results A significant decrease in ICP was recorded in the diabetic rats(P<0.01).The levels of MDA increased remarkably(P<0.01)and the activities of SOD decreased significantly(P<0.05)in cavernous tissues of the diabetic group.Compared with the diabetic group,the levels of MDA decreased significantly(P<0.05)and the activities of SOD increased remarkably(P<0.05)in cavernous tissues of the melatonin treatment group.Meanwhile,melatonin increased the concentration of endothelium and smooth muscle in penis of diabetic rats.Conclusions Supplements of melatonin could attenuate oxidative stress and improve erectile function.Oxidative stress plays an important role in diabetic erectile dysfunction and our study might provide a new insight for the prevention and treatment for it.

Abstract:In recent decades, localized tissue oxidative stress has been implicated as a key component in the development of diabetic retinopathy (DR). Increasing evidence shows that oxidative stress caused by diabetes-induced metabolic abnormalities is the most common mechanism associated with the pathogenesis of DR for both type 1 and type 2 diabetes. Increase in intracellular reactive oxygen species (ROS) concentrations results in the activation of several mechanisms involved in the pathogenesis of DR. In particular, damage or dysfunction caused by oxidative stress still persists even after glycemia has been normalized. Despite considerable evidence showing the beneficial effects of antioxidants in preventing the development of retinopathy, results from large-scale clinical trials on classic antioxidants are somewhat ambiguous. Scavenging reactive radicals may not be the most ideal antioxidant strategy in DR. Advances in understanding the function of ROS in the development of DR can lead to the development of new therapeutic strategies based on the mechanisms of ROS generation and scavenging. Increasing amounts of data have demonstrated the promising prospect of antioxidant therapy and its beneficial effects in vision protection. Therefore, new strategies that utilize antioxidants as additive therapy should be implemented in the treatment of DR.

Abstract:ABSTRACT:Oxidative stress due to aberrant metabolism is considered as a crucial contributor to diabetes and its complications. Hyperglycemia and hyperlipemia boost excessive reactive oxygen species generation by elevated mitochondrial respiration, increased nicotinamide adenine dinucleotide phosphate oxidase activity, and enhanced pro-oxidative processes, including protein kinase C pathways, hexosamine, polyol, and advanced glycation endproducts, which exacerbate oxidative stress. Oxidative stress plays a significant role in the onset of diabetes and its associated complications by impairing insulin production, increasing insulin resistance, maintaining hyperglycemic memory, and inducing systemic inflammation. A more profound comprehension of the molecular processes that link oxidative stress to diabetes is crucial to new preventive and therapeutic strategies. Therefore, this review discusses the mechanisms underlying how oxidative stress contributes to diabetes mellitus and its complications. We also summarize the current approaches for prevention and treatment by targeting the oxidative stress pathways in diabetes.
Reseach on epidermal keratinocyte function impairing in rats with diabetes mellitus

Abstract:Objective To explore the effect of diabetes mellitus (DM) on biological behavior of epidermal keratinocyte in rats. Methods A total of 40 Sprague-Dawley rats were randomized into control group and streptozotocin (STZ) -induced diabetes group. Of each group, 10 rats were implemented with deep partial-thickness scalding. The re-epithelialization rate was observed at the 3rd, 7th, 14th and 21th post-burn day. Histological characteristics and thickness of epidermal tissue in both groups were observed. The adhesion rate, cell cycles, apoptosis rate and migration ability of keratinocyte were measured. The accumulation of advanced glycosylation end products (AGEs) of epidermal tissue was observed. Results The percentages of re-epithelialized area at the 7th, 14th and 21th post-burn day were much lower in DM group than in control group (P<0.05). In DM group, the epidermal thickness was reduced obviously with obscure multilayered epithelium and less amount of prickle cells; The adhesion rates of 12, 24 h after culturing keratinocyte and the percentage of G2/M phase cells were lower in DM group than in control group (P<0.05). However, apoptosis rate of keratinocyte was higher, the amount of migration cell was significantly less in DM group than in control group (both P<0.05). There were lots of AGEs accumulated in epidermal tissue in DM group, while there were hardly AGEs in control group. Conclusions Re-epithelization blocked exists on non-healing wound in DM rats, which is related with the impaired keratinocyte biological behavior. A large of AGEs accumulate in the epidermal tissue of DM rats, which may be a important reason to inhibit keratinocyte function in diabetic environment.

Abstract:Diabetes mellitus is one of the major public health problems worldwide. Considerable recent evidence suggests that the cellular reduction-oxidation (redox) imbalance leads to oxidative stress and subsequent occurrence and development of diabetes and related complications by regulating certain signaling pathways involved in β-cell dysfunction and insulin resistance. Reactive oxide species (ROS) can also directly oxidize certain proteins (defined as redox modification) involved in the diabetes process. There are a number of potential problems in the clinical application of antioxidant therapies including poor solubility, storage instability and nonselectivity of antioxidants. Novel antioxidant delivery systems may overcome pharmacokinetic and stability problem and improve the selectivity of scavenging ROS. We have therefore focused on the role of oxidative stress and antioxidative therapies in the pathogenesis of diabetes mellitus. Precise therapeutic interventions against ROS and downstream targets are now possible and provide important new insights into the treatment of diabetes.
Study on the amount of superoxide anion produced in coronary arterioles of diabetic rats

Abstract:Objective To study the amount of superoxide anion produced in coronary arterioles of diabetic rats under high glucose and observe the effect of high glucose on producing of superoxide anion. Methods Diabetes model of rats was established with streptozotocin. Coronary arterioles were dissected from rats utilizing surgical microscope and micrurgy instruments, followed by the preparation of frozen section. The amount of superoxide anion produced in coronary arterioles of diabetic rats and normal rats was determined using immunofluorescent technique. Results The amount of superoxide anion in coronary arterioles of diabetic rats increased obviously comparing with that in normal rats arteries and the diabetic rats added with free radical scavenger in advance. The results of fluorescence staining of coronary arterioles frozen transection indicate that the amount of superoxide anion produced increased obviously in both vascular smooth muscle and endothelium of coronary arterioles under high glucose. Conclusions High glucose can stimulate vascular smooth muscle and endothelium of coronary arterioles to produce excess superoxide anion, thus participates oxidative stress under diabetic state, which is possibly in close relation with diabetic capillary vessel complication.

Abstract:Diabetic retinopathy is one of the most serious microvascular complications induced by hyperglycemia via five major pathways, including polyol, hexosamine, protein kinase C, and angiotensin II pathways and the accumulation of advanced glycation end products. The hyperglycemia-induced overproduction of reactive oxygen species (ROS) induces local inflammation, mitochondrial dysfunction, microvascular dysfunction, and cell apoptosis. The accumulation of ROS, local inflammation, and cell death are tightly linked and considerably affect all phases of diabetic retinopathy pathogenesis. Furthermore, microvascular dysfunction induces ischemia and local inflammation, leading to neovascularization, macular edema, and neurodysfunction, ultimately leading to long-term blindness. Therefore, it is crucial to understand and elucidate the detailed mechanisms underlying the development of diabetic retinopathy. In this review, we summarized the existing knowledge about the pathogenesis and current strategies for the treatment of diabetic retinopathy, and we believe this systematization will help and support further research in this area.

Abstract:Under physiological conditions,radicals on cellular activities play an important role.When the body is subjected to noxious stimuli lead to oxidative stress,the increased production or decreased removal of radicals result in oxidative damage to cells and tissues.Diabetic cataract is one of the complications of diabetes,and an important class of blinding eye disease.Currently its pathological mechanisms were considered including polyol pathway activation,oxidative stress and non-enzymatic glycation of lens proteins.Oxidative stress action as a central part is attractive,which is closely related to the generation of diabetic cataract.The development of nzymatic and non-enzymatic antioxidant defense systems provides a strategy for anti-cataract,and provides a new idea for mitigation and prevention of diabetic cataract studies.
Effect of oxidative stress on wound surface healing in diabetic rats with scald

Abstract:Objective To study the effect of oxidative stress on wound surface healing in diabetic rats with scald.Methods Scald models were induced by STZ in diabetic rats.Levels of H2O2 and MDA in serum and wound surface healing were observed.The effect of H2O2 on fibroblast apoptosis in rats was detected by flow cytometry.Results Compared with non-diabetic controls,wound healing rates were delayed while the levels of MDA and H2O2 in serum increased obviously in diabetic rats.And H2O2 could promote apoptosis of fibroblast cell.Conclusion Oxidative stress might be one of the positive candidates for diabetic impaired wound healing.

Increased Oxidative Stress Caused by Impaired Mitophagy Aggravated Liver Ischemia and Reperfusion Injury in Diabetic Mice.

Abstract:Aims/Introduction Emerging evidence has suggested the detrimental role of oxidative stress in aggravating ischemia and reperfusion (IR) injury in diabetic livers. Interplay between oxidative stress and mitophagy has been shown. However, the role and mechanism of mitophagy in regulating oxidative stress and IR injury in diabetic livers remain unclear. Materials and Methods Wild-type and db/db (DB) mice were subjected to a partial warm liver IR model. Liver injury, oxidative stress, mitophagy and related molecular pathways were analyzed. Results Here, we found that increased liver IR injury was observed in DB mice, as evidenced by higher levels of serum alanine aminotransferase and serum aspartate, worsened liver architecture damage and more hepatocellular death. DB mice also showed increased mitochondrial oxidative stress. Mitochondrial reactive oxygen species scavenge alleviated liver IR injury in DB mice. Mechanistic analysis showed that 5 ' adenosine monophosphate-activated protein kinase-mediated mitophagy was suppressed in DB mice post-IR. Pharmacological activation of 5 ' adenosine monophosphate-activated protein kinase by its agonist effectively restored mitophagy activation, leading to decreased mitochondrial oxidative stress and attenuated liver IR injury in DB mice. Conclusions Our findings showed that diabetes increased oxidative stress to exacerbate liver IR injury by impairing 5 ' adenosine monophosphate-activated protein kinase-mediated mitophagy. Strategies targeting oxidative stress and mitophagy might provide a promising approach to ameliorate liver IR injury in diabetes patients.
