Abstract:In this work, we propose a MgF2/Au/MgF2/Analyte based, angle-interrogation surface plasmon resonance sensor. This symmetrical structure results in higher surface electric field strengths, and longer surface propagation lengths and depths. A refractive index resolution of 8.1×10−8RIU in fluid protocol and 3.5×10−7RIU in atmosphere protocol is acquired by a simple SPR imaging system. In order to demonstrate the biomedical applications of this system, we detect Escherichia coli with different concentrations using dopamine film on the sensor surface to immobilize antibodies. A detection limit of 103cfu/mL of E. coli bacteria is acquired from this system.

Abstract:The refractive index resolution of a surface plasmon resonance (SPR) sensor has been significantly improved these years; however, higher sensing performance is always desired. In this work, we propose a line-monitoring, long-range SPR sensor whose resolution is much better than conventional SPR sensors. Also, in contrast to mono-channel detection, multichannel detection, using line-monitoring technique, can detect multiple channels concurrently. In this way, this system achieves a refractive index resolution of 4.0 × 10− 7 refractive index units and can monitor multiple molecular interactions simultaneously. Finally, a model experiment detecting the Escherichia coli bacteria has demonstrated the potential for biomedical applications of this system.
Design of a Surface Plasmon Resonance Sensor Based on Grating Connection

Abstract:Nowdays, the study of measurement of the biological field focuses on the research of improving surface plasmon resonance (SPR) in the fields of integration and detection sensitivity. We designed a kind of grating connected surface plasmon resonance sensor. Theoretically, we analyzed the wave vector and the effective refractive index relations with the diffraction grating structure. Then we obtained the nanoparticles enhancement SPR structure with a resolution 10 times higher than that of traditional SPR sensors. Also, we used the finite-difference time-domain (FDTD) analysis and simulation which showed that it was obvious with coupling effect by the nanoparticles enhancement SPR structure that the reflectance spectral bandwidth results validated the structure significantly which improved the sensitivity. Experimental results showed that the dynamic response of the designed sensor reached 10 −6 RIU (refractive index unit). This study has the certain significance to long-distance and special sensing applications.
Study and Numerical Simulation of a Surface Plasmon Resonance Sensor Using Bi-Beam Differential Measurement Approach

Abstract:In this article, a novel measurement approach of Surface Plasmon Resonance (SPR) with bi-beam was presented in order to improve the sensitivity and anti-disturbance capability of intensity-modulated SPR sensor. The relation curves between reflected light intensity and sample refractive index were established at different incident angles and gold film thickness, based on theoretical calculation and numerical simulation of this new method. Compared with the traditional intensity-modulated technology, studies indicates that bi-beam approach has better sensitivity and broader measuring range when the incident angle difference of the two beams is equal to 5 degree and gold film thickness is in the range of 40 to 50 nm. The noiseproof feature and stability of the sensor system are greatly improved with the application of light intensity difference technology.

Abstract:Surface plasmon resonance (SPR) sensing technology is widely used in the field of biosensors due to its non-marking, high-sensitivity, and non-invasive characteristics. However, SPR technology is still limited to sensing analysis in two-dimensional plane, axial detection, as the key of SPR application in three-dimensional medium spatial detection, has not been well studied and solved. In an angle-interrogation SPR sensing system, the spatial characteristics of evanescent wave-dielectric interaction at multiple wavelengths are studied, and the factors affecting the spatial distribution of surface plasmon resonance are also analyzed. An axial spatial resolution method based on the particle swarm optimization (PSO) algorithm with multi-wavelength angle-interrogation structure is proposed, the refractive index distribution in axial space is determined by analyzing the characteristic SPR signal. In addition, the calculation and analysis of the applicable range of wavelengths are carried out. In the reliable spectral range of the incident light wavelength of 600-900 nm, the average error of the axial refractive index spatial resolution increases from 10(-5) RIU to 10(-4) RIU as the number of axial layers increases. The proposed multi-wavelength angle modulation structure analysis method based on PSO algorithm extends the SPR detection range from two-dimensional plane to three-dimensional space, which provides a new and promising analysis model for molecular biology.

Abstract:The spatial distribution detection and characterization of multi-adsorption layers, biomembranes, and cells are important techniques to study biomolecular properties and mechanisms. Using the surface plasmon resonance (SPR) technology, we investigated the spatial characteristics, penetration mechanism, and detection depth of the interaction between evanescent waves and a complex medium. In addition, parameters correlated with the axial spatial distribution were analyzed. We found that the spatial refractive-index distribution of an axial layered model has a unique correlation with the following three characteristic parameters: resonance angle at different wavelengths, first-derivative extreme-point of the angular spectrum, and effective refractive index. A new layer-analysis, based on wavelength-scanning angle interrogation (WSAI), was introduced to enable refractive-index measurements in an axial spatial medium. This new method extends the detection capabilities of SPR sensors and provides a more accurate analysis method for interaction events within an evanescent field.

Abstract:Surface plasmon resonance (SPR) has become an indispensable tool for label-free detection and quantification of molecular binding. Traditionally, the principle of SPR biosensors is described with a stratified medium model, in which discrete molecules are approximated with a uniform thin film. With the recent technical advances, SPR can now detect extremely low coverage of molecules, which raises the question of the validity of the traditional model. Here, we present combined theoretical, numerical and experimental analysis of SPR detection principle by considering the discrete nature of the molecules (particles). Our results show that the stratified medium model can provide reasonable description of SPR biosensors for relatively high coverage and weakly scattering samples. However, interference between the SPR images of individual particles needs to be considered for high spatial resolution images and for strong scattering samples at certain incident angles of light.

Abstract:Surface plasmon resonance (SPR) sensors are tending toward miniaturization, and easy-to-use, low-cost sensors are in demand. Here, to meet this demand, we propose a capillary SPR sensor consisting of a capillary with a gold film plated onto the inner wall. A simple ideal model is constructed, cylindrical metal–dielectric waveguide theory is discussed, analytic equations are established, and the SPR curve of the capillary containing water was obtained by calculating the reflectivity. Optical simulations were also carried out in TracePro, and the simulation results are shown to be consistent with the theoretical calculations. Both ideal and Gaussian light sources are studied and small differences in the spectra are compared. Sensor performances and the CCD placement were also discussed. A method of s-polarized light as reference light to eliminating aberration was introduced. The results demonstrate the feasibility of the proposed capillary SPR sensor.
Study on the Flow Field Distribution in Microfluidic Cells for Surface Plasmon Resonance Array Detection

Abstract:This research is dedicated to optimizing the design of microfluidic cells to minimize mass transfer effects and ensure a uniform flow field distribution, which is essential for accurate SPR array detection. Employing finite element simulations, this study methodically explored the internal flow dynamics within various microfluidic cell designs to assess the impact of different contact angles on flow uniformity. The cells, constructed from Polydimethylsiloxane (PDMS), were subjected to micro-particle image velocimetry to measure flow velocities in targeted sections. The results demonstrate that a contact angle of 135° achieves the most uniform flow distribution, significantly enhancing the capability for high-throughput array detection. While the experimental results generally corroborated the simulations, minor deviations were observed, likely due to fabrication inaccuracies. The microfluidic cells, evaluated using a custom-built SPR system, showed consistent repeatability.

Abstract:Surface plasmon resonance microscopy (SPRM) is an excellent platform for in situ studying cell-substrate interactions. However, SPRM suffers from poor spatial resolution and small field of view. Herein, we demonstrate plasmonic scattering microscopy (PSM) by adding a dry objective on a popular prism-coupled surface plasmon resonance (SPR) system. PSM not only retains SPRM's high sensitivity and real-time analysis capability, but also provides approximate to 7 times higher spatial resolution and approximate to 70 times larger field of view than the typical SPRM, thus providing more details about membrane protein response to ligand binding on over 100 cells simultaneously. In addition, PSM allows quantifying the target movements in the axial direction with a high spatial resolution, thus allowing mapping adhesion spring constants for quantitatively describing the mechanical properties of the cell-substrate contacts. This work may offer a powerful and cost-effective strategy for upgrading current SPR products.

Abstract:In the present work we present a simple method for analyzing the surface plasmon resonance (SPR) spectra numerically. According to our calculation, we found that the shift of the resonance angle is proportional to the surface density of the adsorbed molecular layer and the value difference between the refractive index of the protein layer and that of the buffer. The variation of the thickness of the gold film may change the resonance angle but does not affect the linear correlation of the shift of the resonance angle with the surface density of the adsorbed protein. In addition, we found that the variation of the thickness of the cover slide, which is stuck to the base of the prism and which is covered by a thin gold film, is approximately not influenced by the resonance angle of SPR. Abstract:Objective-based surface plasmon resonance microscopy (SPRM) is a novel optical imaging technique that can map the spatial distribution of a local refractive index based on propagating surface plasmon polaritons (SPPs). Different from some other optical microscopy that shows a dot-like point spread function (PSF), a nanosized object appears as a wave-like pattern containing parabolic tails in SPRM. The geometrical complexity of the wave-like pattern hampered the quantitative interpretation of the PSF of SPRM. Previous studies have shown that two adjacent rings were obtained in the frequency domain by applying a two-dimensional Fourier transform to such patterns. In the present work, a ring-fitting method was developed to extract geometrical features out of the dual rings and to connect these features with several experimental parameters. It was found that the radius of ring equaled to the wavevector of SPPs. Its orientation revealed the propagation direction of SPPs. The coordinate distance of the center of ring gave the parallel component of the wavevector of the incident light, which was regulated by the incident angle. The ring-broadening factor reflected the propagation length of SPPs in a reciprocal relationship. Systematical and quantitative interpretations in the frequency domain not only advanced the basic understanding on the PSF of SPRM but also opened up the possibility to utilize these frequency-domain features for detection and sensing purposes in future.

Abstract:A D-shaped optical-fiber refractive index sensor based on surface plasmon resonance is reported. Rigorous coupled-wave analysis was used to simulate the numerical modeling. The influences of different metal grating parameters on the coupling efficiency were studied and an optimal phase match was obtained. The results show that wavelength sensitivity of the proposed optical fiber sensor is as high as 0.925 μm/RIU for refractive index values from 1.35 to 1.43 with a high linearity value of 0.997. The reported structure may promote the development of high-performance biochemical sensing devices.

Abstract:Spectral surface plasmon resonance (SPR) sensors with absorbance measurement were prepared. Resonant wavelengths (lambda) versus effective refractive indexes of the SPR mode were measured with different media in contact with the gold layer. An investigation into the refractive-index sensitivity of the sensor at a fixed angle reveals a linear dependence of lambda(R) on the refractive index of the solution (n(c)), with Dlambda /Dn(c) = 3553.6 nm in a small range of 1.333< or = n(c) < or =1.347. It was observed that the effective refractive index slowly decreases with increasing n(c), attributable to wavelength-induced modulation of optical dielectric constant for the gold layer. Adsorption of bromothymol blue (BTB) on the gold layer leads to a redshift of Dlambda(R) = 3.7 nm, larger than Dlambda(R) = 2.5 nm induced by myoglobin (Mb) adsorption. On the basis of Fresnel equations, calculations with d approximately 1 nm and n=1.69 for BTB and d approximately 3 nm and n = 1.40 for Mb also demonstrate that the SPR band shift induced by full-monolayer adsorption of BTB is larger than that for full-monolayer Mb adsorption. The combination of both measured and calculated results suggests that the contribution of the adlayer index of refraction to the sensitivity of the sensor is greater than that of the adlayer thickness.
Numerical simulations of surface plasmon resonance system for monitoring DNA hybridization and detecting protein-lipid film interactions

Abstract:This paper presents a simple method to extract information about thin organic films from surface plasmon resonance (SPR) spectra. From numerical simulations it was found that a shift (Δ θ SPR ) of an absorption peak in the SPR spectrum was directly proportional to the product of the thin organic film thickness and the refractive index difference between the thin organic film and a buffer soaking the sample. It was also found that Δ θ SPR was not sensitive to the thin organic film support of a gold film and a glass cover slip. Relationships between Δ θ SPR and distributions of macromolecule structures, in the thin organic films were theoretically established. Formulae were derived for a homemade SPR system to calculate length, transverse area, density and surface concentration of macromolecules in the thin organic film. The validity of these treatments was checked by precisely measuring the size of a single distearoylphosphatidylcholine molecule on a gold-supported phospholipid film; by quantitatively monitoring hybridization of synthesized oligonucleotides strands based on a biotin/avidin system; and by quantitatively detecting the steric hindrance of rabbit C-reactive protein specifically bound to phospholipid monolayers composed of synthesized lipids.
A Curved Top-Wall Flow Cell for Improvement of Response Consistency in Surface Plasmon Resonance Array Detection

Abstract:High spatially consistent responses are desired for the array-based biomolecular interaction analysis. The response consistency is dependent on the uniformity of mass transport rate. Based on theoretical analysis, we established a universal mathematical relation between the mass transport rate and the flow cell height. The optimal flow cell geometry for analyte distribution uniformity, H(l) = 1/3.l(-1/2), was derived from the mathematical relation. Using Poly(dimethylsiloxane) elastomer replica molding methods, the flow cell was fabricated. The actual flow regimes within the flow cell were confirmed by micro-Ply experiments and results agreed with expectations. To demonstrate the capability of this flow cell in improving spatial consistency of responses, goat-anti-rabbit IgG and rabbit IgG interaction experiments were performed on a home-built SPR imaging system. Results showed that the curved top-wall flow cell significantly reduced the coefficient of variance, of association rate across the sensing surface. (C) 2014 Elsevier B.V. All rights reserved.

Abstract:A novel open-loop photonic crystal fiber (PCF) refractive index sensor based on the surface plasmon resonance effect is proposed. The gold film is applied to the surface of the air-core PCF to induce the SPR effect. The finite element method is employed to examine the influence of open-angle on the sensing performance. Simulation results show that the sensor is suitable for refractive index measurements ranging from 1.26 to 1.32 when the open-angle ranges from 90 to 130°, demonstrating an average wavelength sensitivity of 1738 nm/RIU. As the angle varies from 140 to 180°, the sensor becomes more appropriate for refractive index measurements ranging from 1.32 to 1.38, displaying an average wavelength sensitivity of 1667 nm/RIU. The sensing range of the sensor is influenced by open-angle, enabling the selection of an appropriate sensor structure according to the desired measurement range.

Abstract:Surface plasmon resonance (SPR) biosensor based on phase detection possesses high sensitivity. Fully understanding the relations among the parameters and the phase detection is instructive for the optimal design. A five-layer mathematical model of an SPR biosensor based on phase detection is presented. We systematically analyzed the effects of the gold film, the linker and the sample on the phase detection by numerical simulation with Matlab. The numerical results show that the phase detection has the obvious selectivity, which narrows the linear detection scope. For the perfect detection resolution, gold film thickness should be from 30 to 45nm, the linker layer should be 1nm and the sample thickness should be greater than 20nm. At the same time, we compared the phase detection with the angular interrogation and found that they have the similar correlation factor, but the difference in resolution can be one order.

Abstract:Surface plasmon resonance (SPR) has been used extensively in biosensing implementation as a high-sensitivity and label-free sensing technique. For SPR sensors based on spectral interrogation, their performance in terms of refractive index (RI) resolution differs substantially when operating at different resonant wavelengths. This paper presents a feasible scheme of enhancing the sensor resolution by simulating measured SPR curves using an experiment-assisted simulation model to effectively explore the optimal resonant wavelength for the sensing system. It shows that the variation of experimentally obtained RI resolution with the resonant wavelength is in good accordance with the simulation results. In addition, the RI resolution at the derived optimal resonant wavelength is at least 2 times better than that at the commonly used resonant wavelength region both experimentally and theoretically. The methods of the proposed scheme potentially facilitate optimization of various sensor instrumentation for high-resolution SPR sensing.

Abstract:Wide-field surface plasmon resonance microscopy (WF-SPRM), based on Kretschmann’s configuration, is a powerful technique for detecting nano-objects in solution. In this study, key parameters for building a highly sensitive WF-SPRM are optimized, including laser wavelength, prism refractive index, numerical aperture of the objective, and gold layer thickness. A medium model describes the surface plasmon resonance (SPR) sensor principle, treating molecules bound on the sensor surface as an effective medium with an effective refractive index and thickness. On the other hand, the SPR discrete particle model is introduced to describe the detection principle for discrete particles. Theoretical, numerical, and experimental analyses are conducted, and the calculated intensity profiles of single and double nanoparticles from the discrete particle model describe the experimental profiles well. Furthermore, the influence of coating the gold layer with a dielectric layer on the SPR sensitivity is investigated for varying refractive indices.
