Abstract:Humic acid (HA)is a principal component of dissolved organic matters in all aquatic ecosystems. It comprises of a variety of molecular structures and functional groups and can bind pollutants in the water, thereby affecting the pollutant toxicity to aquatic organisms. In this study, a triazine herbicide prometryn, commonly present in the surface and ground water, was chosen as the target pollutant and a model organism zebrafish(Danio rerio)as the target organism. The aim of this study was to investigate the acute toxicity of prometryn to Danio rerio at 5 and 15 mg·L-1 HA concentrations. Under single prometryn exposure, the 24 h, 48 h, 72 h, and 96 h median lethal concen-trations(LC50)of prometryn to Danio rerio were 9.143, 8.432, 7.645, and 7.448 mg·L-1 and 95%confidence intervals were 8.635~9.861, 7.408~9.934, 7.041~8.310, and 7.032~7.971 mg·L-1, respectively. Based on these acute toxic experiment results, prometryn had the high toxicity to Danio rerio. In addition, 96 h cumulative mortality rate of Danio rerio did not change obviously with 5 mg·L-1 HA when the con-centrations of prometryn were 6.5, 7.5, and 8.5 mg·L-1. In contrast, the same toxicological parameter became 40%higher at 15 mg·L-1 HA than HA-free when prometryn concentration was 8.5 mg·L-1. It was shown that HA can increase the acute toxicity under higher prometryn concentrations. This might be explained by a hypothesis that the association of prometryn with DOM reducing its toxicity might be overshad-owed by other mechanisms actually enhancing its toxicity in water.

Abstract:Humic acid (HA) is a major component of dissolved organic matter, is ubiquitous in the aquatic environment and influences the biological toxicity of organic pollutants. In this study, we investigated the cytochrome P450 1A (CYP 1A) mRNA expression and ethoxyresorufin-O-deethylase (EROD) activity in the gills and liver of zebrafish following exposure to the s-triazine herbicide prometryn with or without HA present. Prometryn induced both CYP 1A mRNA expression and EROD activity. The CYP 1A mRNA expression of zebrafish that were exposed to a combination of prometryn and HA was increased compared to those exposed to prometryn alone. A likely cause for CYP 1A induction is the impact of special components of HA, functioning as aryl hydrocarbon receptor (AHR) agonists. In combination with HA, these increase prometryn levels in tissues. Similar results for EROD activity were evident. In our time course study, CYP 1A mRNA expression reached maximum values during 24h. This revealed CYP 1A mRNA transcription as a comparatively sensitive toxicity index. In a recovery experiment, we found a faster decrease of CYP 1A mRNA expression to control levels (CK) in gills compared to liver tissue. Following exposure to HA, CYP 1A mRNA expression in liver tissue displayed a faster decrease to CK levels. HA induced enhanced metabolic rates for prometryn. In contrast, recovery regularity of CYP 1A expression in gills was independent of the presence of HA. This result indicates different detoxification mechanisms for HA in liver and gills.
Effect of HA on Ecotoxicity of NanoZnO in Carassius auratus

Abstract:To investigate the effect of humic acid(HA) with different concentrations(0,1,1.5 and 2.5 mg L-1) on toxicity of ZnO nanoparticle(ZnO-NP) in the fish(Carassius auratus),the experimental animals were exposed to HA and ZnO-NP mixtures for seven days.Then the distribution of zinc,and the changes in the ROS content and antioxidative system in the brain and liver of C.auratus were recorded.The results indicated that with the increasing of HA,the concentration of zinc increased in the gills and muscles and reduced in the eyes,brain and liver of C.auratus.Becasue ZnO-NP induced the production of OH free radicals,and the increase of HA promoted the activity of SOD in the brain and liver of C.auratus,the concentrations of MDA and GSHA increased.It was also found that the stress of ZnO-NP caused the oxidative damage of the liver and brain,which could be further induced by HA.Therefore,the results showed that HA changed biological effects of ZnO-NP,and suggested that enviornment factors could not be disregarded when considering the ecological safety of nanomaterials in the aquatic system.
The influence of humic acid on pyrene biotoxicity in liver of Carassius auratus

Abstract:In the current study,whether the different concentrations of humic acid(HA)may influence bioaccumulation and oxidative stress of pyrene in liver of Carassius auratus was investigated.The fish(C.auratus)was exposed to HA(0,0.05,0.1,0.5,1.0 and 2.5 mg/L)and pyrene(0.05 mg/L)mixtures for 10 days.Then the pyrene bioaccumulation as well as their effects on the reactive oxygen species,Malondialdehyde(MDA)content and antioxidative system in the liver of C.auratus were analyzed.With the HA concentration increase,bioaccumulation of pyrene in liver of C.auratus reduced.In the meantime,0.05 mg/L and 0.1 mg/L HA could induce the production of reactive oxygen species in the liver of C.auratus,0.5 mg/L and 1.0 mg/L HA could decrease the MDA contents.The superoxide dismutase,catalase,glutathione-s-transferase activities and glutathione contents decreased after an initial increase,under HA concentration increase.It was pyrene stress which caused the oxidative damage of the liver and brain,which could be further induced by HA.Overall,our present work illustrated that the presence of HA could reduce pyrene concentration in fish liver and change the pyrene ' s biological effects,which should not be disregarded when considering the ecological safety of pyrene in the aquatic system.
Influence of humic acid on arsenic bioaccumulation and biotransformation to zebrafish: A comparative study between As(III) and As(V) exposure.

Abstract:Previous studies have indicated that natural organic matter in the aquatic environment could affect arsenic bioaccumulation and biotransformation to aquatic organisms. However, the differences between the effects of arsenite and arsenate exposure have not been studied and compared in fish exposure models. In this study, adult zebrafish (Danio rerio) were exposed to 5 mg/L inorganic As solutions, in the presence of a range of humic acid (HA) concentrations (1, 2.5, 5, 10, 20 mg/L) in 96 h waterborne exposure. Results showed that in the presence of HA, total As bioaccumulation was significantly reduced in zebrafish following arsenite exposure, while this reduction was not observed during arsenate exposure. The reduction in total arsenic bioaccumulation for arsenite exposure can be explained by the fact that HA forming a surface coating on the cell surface, hindering transport and internalization. However, this reduction in total As was not observed due to differences in uptake pathways for arsenate exposure. Results also showed that Arsenobetaine (AsB) was the main biotransformation product in zebrafish following inorganic As exposure, accounting for 44.8%-64.7% of extracted arsenic species in all exposure groups. The addition of HA caused levels of MMA and As(III) to decrease, while the distribution of AsB significantly increased in arsenite exposure groups. The increase in AsB could be because the As(III)-HA complex was formed, affecting the methylation of As(III). In contrast, the addition of HA to arsenate exposure groups, did not affect the reduction of As(V) to As(III) and therefore, an increase in the distribution of AsB was not observed in arsenate exposure groups. This study provides useful information on the mechanisms of toxicity, for improved risk assessment of As in natural aquatic environments.

Abstract:The studies on how humic acid (HA) influences the oxidative stress of arsenic in aquatic organism is limited. Using Danio rerio as case study, we explored the oxidative stress effects in aquatic organism after 96 h exposure to the HA and arsenic. Results revealed the co-exposure of HA and arsenite elevated the superoxide dismutase activities and downgraded the malondialdehyde. Thus, we speculate that HA may alleviate the oxidative stress induced by arsenite, which may be caused by the HA’s coating in combination with the complexation of arsenite and HA. In addition, HA acted as the reactive oxygen species scavenger, promising to eliminate the oxygen free radicals. Contrastingly, HA may impact little on the arsenate exposure. This study can help better understand oxidative stress mechanism of co-exposure of arsenic and HA in aquatic environment.
Influences of Humic Acid on the Bioavailability of Phenanthrene and Alkyl Phenanthrenes to Early Life Stages of Marine Medaka (oryzias Melastigma).

Abstract:The influences of humic acid (HA) on the environmental behavior and bioavailability of parent polycyclic aromatic hydrocarbons (PAHs) and alkyl PAHs were investigated and compared using the early life stages of marine medaka (Oryzias melastigma, O. melastigma). It was demonstrated that the binding affinity of parent phenanthrene (PHE) with HA was smaller than that of 3-methyl phenanthrene (3-MP) and 9-ethyl phenanthrene (9-EP). Furthermore, the bioaccumulation of the three PAHs and the levels of lipid peroxidation (LPO) were calculated to study the changes in bioavailability of PAHs in presence of HA. The results indicated that the addition of HA significantly decreased the bioaccumulation and toxicity of PAHs by decreasing free PAHs concentrations. The bioavailable fractions of HA-bound PAHs in bioaccumulation (α) and toxicity (β) were evaluated, indicating that the HA-bound 3-MP and 9-EP show higher bioavailability in bioaccumulation and lower bioavailability in toxicity relative to those of PHE. The β/α values were less than 1 for all PAH treatment groups containing HA, suggesting that the fraction of HA-bound PAHs contributing to bioaccumulation was higher than that of HA-bound PAHs inducing toxic effect. In addition, we proposed that the free PAHs generated by desorption from HA in the cell were toxic by showing that the β/α ratio values are correlated with the log KOW values (p = 0.007 and R(2) = 0.8355). Thus, oil spill risk assessments should consider both alkyl PAHs and the factors that influence the bioavailability and toxicity of PAHs in the natural aquatic environments.
Double-dose responses of Scenedesmus capricornus microalgae exposed to humic acid

Abstract:Dissolved organic matter (DOM) has been found to attenuate the ecotoxicity of various environmental pollutants, but research on its own toxic effects in aquatic ecosystems has been very limited. Herein, the toxic effects of humic acid (HA), a represent DOM typically found in natural waters, on the freshwater alga Scenedesmus capricornus were investigated. As result, HA exerted a double-dose effect on the growth of Scenedesmus capricornus. At HA concentrations below 2.0 mgC/L, the growth of Scenedesmus capricornus was slightly promoted, as was the synthesis of chlorophyll and macromolecules in the algae. Moreover, S. capricornus can maintain its growth by secreting fulvic acid as a nutrient carbon source. However, the growth of Scenedesmus capricornus was significantly inhibited when HA was beyond 2.0 mgC/L. The main mechanisms of humic acid's toxicity were membrane damage and oxidative stress. Particularly, when the oxidative stress exceeds the algae's carrying capacity, the synthesis of EPS is greatly inhibited and HA damage results. Taken together, DOM may have both positive and negative effects on aquatic ecosystems.

Effects of Prometryn Exposure on Hepatopancreas Oxidative Stress and Intestinal Flora in Eriocheir sinensis (Crustacea: Decapoda)

Abstract:There is growing evidence that long-term exposure to prometryn (a widely used herbicide) can induce toxicity in bony fish and shrimp. Our previous study demonstrated its 96 h acute toxicity on the crab Eriocheir sinensis. However, studies on whether longer exposure to prometryn with a lower dose induces toxicity in E. sinensis are scarce. Therefore, we conducted a 20 d exposure experiment to investigate its effects on the hepatopancreas and intestine of E. sinensi. Prometryn reduce the activities of antioxidant enzymes, increase the level of lipid peroxidation and cause oxidative stress. Moreover, long-term exposure resulted in immune and detoxification fatigue, while short-term exposure to prometryn could upregulate the expression of genes related to immunity, inflammation and detoxification. Prometryn altered the morphological structure of the hepatopancreas (swollen lumen) and intestine (shorter intestinal villi, thinner muscle layer and thicker peritrophic membrane). In addition, prometryn changed the species composition of the intestinal flora. In particular, Bacteroidota and Proteobacteria showed a dose-dependent decrease accompanied by a dose-dependent increase in Firmicutes at the phylum level. At the genus level, all exposure groups significantly increased the abundance of Zoogloea and a Firmicutes bacterium ZOR0006, but decreased Shewanella abundance. Interestingly, Pearson correlation analysis indicated a potential association between differential flora and hepatopancreatic disorder. Phenotypic abundance analysis indicated that changes in the gut flora decreased the intestinal organ's resistance to stress and increased the potential for opportunistic infection. In summary, our research provides new insights into the prevention and defense strategies in response to external adverse environments and contributes to the sustainable development of E. sinensis culture.
Dissolved Organic Matter Affects Both Bioconcentration Kinetics and Steady-State Concentrations of Polycyclic Aromatic Hydrocarbons in Zebrafish (danio Rerio).

Abstract:Dissolved organic matter (DOM) is ubiquitous in natural aquatic ecosystems. The association of hydrophobic organic compounds (HOCs), such as polycyclic aromatic hydrocarbons (PAHs), with DOM may have a large impact on HOC fractions in water and their bioconcentration in fish. However, the effects of DOM on HOC bioconcentration in fish are not well understood, especially whether DOM will affect the bioconcentration steady-state concentrations of HOCs in fish is still confusing. Thus, this study investigated the effects of three DOM including gallic acid (GA), tannic acid (TA), and humic acid (HA) with molecular weights ranging from 170 Da to about 10 kDa at different concentrations (1, 5, and 15 mgOC L-1) on the bioconcentration of PAHs including phenanthrene, anthracene, fluoranthene, and pyrene in zebrafish (Danio rerio), with the PAH freely dissolved concentrations maintained constant by passive dosing systems. The results revealed that the presence of DOM generally increased the bioconcentration steady-state concentrations of the PAHs in zebrafish (Cb-ss), with the increase ranging from 28.1% to 204.0%, and the increase of Cb-ss promoted by TA with middle molecular weight (1700Da) was the highest among the studied DOM. Moreover, the Cb-ss increased with the concentrations of GA with low molecular weight and TA with middle molecular weight in water, whereas decreased with increasing concentrations of HA with high molecular weight. The uptake rate constants of the PAHs in zebrafish with DOM were higher than that without DOM. Ingestion of DOM and direct accumulation of PAHs associated with DOM might be primary influencing mechanisms of DOM on the Cb-ss, and whether the facilitated diffusive mass transfer by DOM will affect the Cb-ss needs to be further studied. This study suggested that DOM-associated HOCs should be considered in future HOC risk assessment in addition to the freely dissolved HOCs. (c) 2018 Elsevier B.V. All rights reserved.
Regulation mechanisms of humic acid on Pb stress in tea plant (<i>Camellia sinensis</i> L.)

Abstract:Though the interaction between humic acid (HA) and heavy metals has been widely reported, the effects of HA on the toxicity of heavy metals to plants are still in debate. In this study, the regulation mechanisms of HA on Pb stress in tea plant (Camellia sinensis L.) was investigated through hydroponic experiments, and the experimental results were explained by using transmission electron microscope (TEM), scanning transmission X-ray microscopes (STXM) and isobaric tags for relative and absolute quantitation (iTRAQ) differential proteomics. Significant alleviation of Pb stress was found with HA coexistence. TEM results showed that HA greatly mitigated the damage of cells caused by Pb stress. Compared with sole Pb treatment, the addition of HA increased the contents of pectin and pectic acid in the cell wall by 10.5% and 30.5%, while arabinose (Ara) and galactose (Gal) decreased by 20.5% and 15.9%, respectively, which were beneficial for increasing Pb adsorption capacity of the cell wall and promoting cell elongation. Moreover, iTRAQ differential proteomics analysis proved that HA strengthened the antioxidant system, promoted the synthesis of cell wall, and stabilized protein and sulfur-containing substance metabolism in molecular level. Notably, the concentration of calcium (Ca) in the cell wall of HA coexistence treatment was 47.4% higher than Pb treatment. STXM results also indicated that the distribution of Ca in the cell wall was restored with the presence of HA. This might promote the formation of the egg-box model, thus alleviating Pb stress in cells. Our results reveal the regulation mechanisms of HA on Pb detoxification in plants and provide useful information for improving the safety of agricultural products. (c) 2020 Elsevier Ltd. Abstract:Extensive use of triazophos for the chemical control of pests in agriculture or aquaculture might strongly disturb the aquatic environment due to residue accumulation through various routes like surface run-off, spray-drift and effluent from factories, which have potential negative effects to non-target aquatic organisms. Previous studies have documented the antioxidative effects of triazophos to mammals, however, the oxidative toxicity of triazophos to fish has not been adequately studied to date. Thus, an acute exposure (96 h) to triazophos at different concentrations of 0.06, 0.3 and 1.5 mg/L (corresponding to 1/50th, 1/10th and 1/2th of 96 h-LC50, respectively), was conducted to investigate the triazophos-induced oxidative stress in adult zebrafish (Danio rerio). The results showed that the time- and dose-dependent induction of oxidative stress except for the ROS level in liver after 24 and 48 h exposure, as indicated by increased reactive oxygen species (ROS), malondialdehyde (MDA) level and a compromised antioxidant defense system, including increased superoxide dismutase (SOD) activity, catalase (CAT) activity, glutathione (GSH) content as well as the increased at first and decreased afterwards genes expression (Sod1, Sod2, Cat and Gpx) in brain. Simultaneously, ROS and MDA showed an increased trend, SOD activity, CAT activity and GSH content showed a trend of increasing at 24 h and decreasing at 48 h, and then increasing at 96 h and Sod1, Sod2, Cat and Gpx gene showed decreasing at first and then increasing in liver tissue. The present study concluded that the damage of the antioxidant system by triazophos induced oxidative stress in the brain and liver of zebrafish with concomitant lipid peroxidation, which is an important mechanism underlying the triazophos-induced acute toxicity.

Abstract:To investigate the toxic and environmental effects of prometryn, a laboratory experiment was performed on coexistent fish and algae. The body weight and length of Carassius carassius, Microcystis aeruginosa (M. aeruginosa) densities and water quality were measured continuously for 92 days. It was observed that fish growth was significantly inhibited by prometryn. This can be partly attributed to the adverse effects of prometryn on the antioxidant system of fish; the activities of superoxide dismutase (SOD) and catalase (CAT) in crucian carp were significantly inhibited by prometryn. The growth of M. aeruginosa was greatly inhibited by prometryn (p < 0.05), and the adverse effects of prometryn on M. aeruginosa indirectly impacted water qualities. The body weight and length of Carassius carassius first increased and then tended to be stable with increasing electrical conductivity (EC) values; their quantified relationship was established based on the Gompertz and Logistic equations (R2 = 0.920–0.989). Based on the above results, we concluded that the toxic effects of prometryn can impact the antioxidant system of fish and algae which in turn affects their growth performance, and have an indirect impact on water quality. The application of equations made it realizable to obtain a more detailed interpretation of the processes involved in these biological–abiotic interactions.
Effect of humic acid on copper uptake and metallothionein induction in Daphnia magna

Abstract:Daphnia magna was used to study the influence of humic acid (HA) on the biological effect of copper (Cu) in water. Cu species in reconstituted water with different HA concentrations was calculated using MINTEQA2. The lethal concentration 50 (LC50) was predicted by the biotic ligand model (BLM). Mortality, Cu bioaccumulation, and metallothionein (MT) in D. magna exposed to Cu and HA for 3 d were measured. All indicators were proven using LC50. As the HA concentration increased from 0 mg/L to 6 mg/L, the content of free Cu2+ decreased and the content of Cu and HA complexes (Cu-HA) increased. Mortality, Cu bioaccumulation, and MT decreased significantly, indicating that the existence of HA in the water reduced the uptake and toxicity of Cu. The uptake of Cu-HA complexes was significantly lower than that of free Cu ions. Their mechanisms of bioavailability were different because the Cu-HA complexes were not involved in the synthesis of MT in vivo.

Abstract:Tetrabromobisphenol A (TBBPA) and Perfluorooctane sulfonate (PFOS) are emerging contaminants which coexist in marine environments, posing significant risks to ecosystems and human health. The behavior of these contaminants in the presence of dissolved organic matter (DOM), specifically the co-contamination of TBBPA and PFOS, is not well understood. The bioaccumulation, distribution, elimination, and toxic effects of TBBPA and PFOS on thick-shell mussels (Mytilus unguiculatus V.), with the absence and presence of humic acid (HA), a typical DOM, were studied. The results showed that the uptake of TBBPA decreased and the uptake of PFOS increased when exposed to 1 mg/L HA. However, at higher concentrations of HA (5 and 25 mg/L), the opposite effect was observed. Combined exposure to HA, TBBPA, and PFOS resulted in oxidative stress in the digestive gland, with the severity of stress dependent on exposure time and HA dose. Histological analysis revealed a positive correlation between HA concentration and tissue damage caused by TBBPA and PFOS. This study provides insights into the influence of HA on the bioaccumulation-elimination patterns and toxicity of TBBPA and PFOS in marine bivalves, offering valuable data for ecological and health risk assessments of combined pollutants in aquatic environments rich in DOM.
Effects of Humic Acid on Bioaccumulation and Oxidative Stress Induced by Tetrabromobisphenol A in Coontail Ceratophyllum Demersum L.

Abstract:Under simulated static conditions in laboratory, the effects of humic acid(HA)on bioaccumulation of tetrabromobisphenol A(TBBPA)and oxidative stress induced by TBBPA in Coontail Ceratophyllum demersum L.were studied.Eight gram fresh weight of C.demersum were exposed to 0.5 mg·L-1 TBBPA in glass beakers with 3 L 10% Hoagland′s solution containing 0, 0.04, 0.20, 0.40, 0.80 and 2.0 mg·L-(1DOC)HA, respectively, for 14 d under laboratory conditions.Controls were beakers containing only 10% Hoagland′s solution without TBBPA and HA addition.The results showed that the presence of HA reduced the bioaccumulation of TBBPA in C.demersum, by 25.2% at 2.0 mg·L-1 HA.In the absence of HA, 0.5 mg·L-1 TBBPA increased signal intensity of free radicals and activity of superoxide dismutase(SOD)and decreased Glutathione(GSH)content in C.demersum.At low concentrations of HA and TBBPA, the signal intensity of free radicals, GSH content, and SOD activity resumed to the control level.At higher concentrations of HA and TBBPA, the signal intensity of free radicals and SOD activity increased significantly, whereas the GSH content decreased significantly, as compared to the control.The oxidized glutathione(GSSG)contents were not significantly different at the various concentrations of HA.
Different Life-Stage Exposure to Hexafluoropropylene Oxide Trimer Acid Induces Reproductive Toxicity in Adult Zebrafish (danio Rerio).

Abstract:As a novel alternative to perfluorooctanoic acid (PFOA), hexafluoropropylene oxide trimer acid (HFPO-TA) has been widely used and has caused ubiquitous water pollution. However, its adverse effects on aquatic organisms are still not well known. In the present study, zebrafish at different life stages were exposed to 0, 5, 50, and 100 mu g/L of HFPO-TA for 21 days to investigate reproductive toxicity in zebrafish. The results showed that HFPO-TA exposure significantly inhibited growth and induced reproductive toxicity in zebrafish, including a decrease of the condition factor, gonadosomatic index, and the average number of eggs. Histological section observation revealed that percentages of mature oocytes and spermatozoa were reduced, while those of primary oocytes and spermatocytes increased. In addition, exposure to HFPO-TA at three stages induced a significant decrease in the hatching rate, while the heart rate and normal growth rate of F1 offspring were only significantly inhibited for the exposure from fertilization to 21 days postfertilization (dpf). Compared with the exposure from 42 to 63 dpf, the reproductive toxicity induced by HFPO-TA was more significant for the exposure from fertilization to 21 dpf and from 21 to 42 dpf. Expression of the genes for cytochrome P450 A1A, vitellogenin 1, estrogen receptor alpha, and estrogen receptor 2b was significantly up-regulated in most cases after exposure to HFPO-TA, suggesting that HFPO-TA exhibited an estrogen effect similar to PFOA. Therefore, HFPO-TA might disturb the balance of sex steroid hormones and consequently induce reproductive toxicity in zebrafish. Taken together, the results demonstrate that exposure to HFPO-TA at different life stages could induce reproductive toxicity in zebrafish. Abstract:This study explored the effects of humic acid (HA) on the toxicity of ZnO nanoparticles (nano-ZnO) and Zn(2+) to Anabaena sp. Typical chlorophyll fluorescence parameters, including effective quantum yield, photosynthetic efficiency and maximal electron transport rate, were measured by a pulse-amplitude modulated fluorometer. Results showed that nano-ZnO and Zn(2+) could inhibit Anabaena sp. growth with the EC50 (concentration for 50% of maximal effect) of 0.74 ± 0.01 and 0.3 ± 0.01 mg/L, respectively. In the presence of 3.0 mg/L of HA, EC50 of nano-ZnO increased to 1.15 ± 0.04 mg/L and EC50 of Zn(2+) was still 0.3 ± 0.01 mg/L. Scanning electron microscopy observation revealed that HA prevented the adhesion of nano-ZnO on the algae cells due to the increased electrostatic repulsion. The generation of intracellular reactive oxygen species and cellular lipid peroxidation were significantly limited by HA. Nano-ZnO had more damage to the cell membrane than Zn(2+) did, which could be proven by the malondialdehyde content in Anabaena sp. cells. © 2014 Wiley Periodicals, Inc. Visualization and Quantification of the Impact of Humic Acid on Zinc Accumulation in Aquatic Plants Using a Low-Molecular-Weight Fluorescent Probe

Abstract:The main aims of this study were to investigate the impact of humic acid (HA) on zinc (Zn) accumulation in aquatic plants and to study a low-molecular-weight Zn2+-selective fluorescent probe to visualize and quantify the tissue-level Zn concentrations. Ceratophyllum demersum and Aldrovanda vesiculosa were exposed to solutions containing Zn (1 and 3 mg/L), HA (0.5 mg/L), and Zn with HA for nine days. The Zn accumulation (mg/g) in the plants was measured by ICP-OES and we applied a Zn2+-selective fluorescent probe with a low molecular weight to the analysis of Zn in C. demersum plant cells using fluorescence microscopy and ImageJ software. The application of HA reduced the Zn accumulation significantly (p < 0.05) and increased the chlorophyll concentration slightly less significantly (p > 0.05) in both plants. Results obtained from ImageJ revealed a strong positive correlation between fluorescence intensity and the Zn accumulation in C. demersum (r = 0.988). We showed that the application of HA reduced the Zn accumulation in both plants, and successfully visualized and quantified that a Zn2+-selective fluorescent probe with a low molecular weight can be applied to the diagnosis of Zn osmosis into a cell or tissue on the basis of fluorescence intensity.

Abstract:The effects of humic acid (HA) on interactions between ZnO nanoparticles (ZnO NPs) and Pseudomonas putida KT2440 biofilms at different maturity stages were investigated. Three stages of biofilm development were identified according to bacterial adenosine triphosphate (ATP) activity associated with biofilm development process. In the initial biofilm stage 1, the ATP content of bacteria was reduced by more than 90% when biofilms were exposed to ZnO NPs. However, in the mature biofilm stages 2 and 3, the ATP content was only slightly decreased. Biofilms at stage 3 exhibited less susceptibility to ZnO NPs than biofilms at stage 2. These results suggest that more mature biofilms have a significantly higher tolerance to ZnO NPs compared to young biofilms. In addition, biofilms with intact extracellular polymeric substances (EPS) showed higher tolerance to ZnO NPs than those without EPS, indicating that EPS play a key role in alleviating the toxic effects of ZnO NPs. In both pure ZnO NPs and ZnO-HA mixtures, dissolved Zn2+ originating from the NPs significantly contributed to the overall toxicity. The presence of HA dramatically decreased the toxicity of ZnO NPs due to the binding of Zn2+ on HA. The combined results from this work suggest that the biofilm maturity stages and environmental constituents (such as humic acid) are important factors to consider when evaluating potential risks of NPs to ecological systems.
