Abstract:A method using high-performance liquid chromatography (HPLC) with electrospray ionization mass spectrometry (ESI-MS) in the negative mode is presented for the quantification and characterization of different soyasaponins using six authentic soyasaponin standards. This method was successfully applied to the rapid separation of diverse soyasaponins, more than 50, including soyasaponins A in different degrees of acetylation, and soyasaponins B in both their 2,3-dihydro-2,5-dihydroxy-6-methyl-4H-pyran-4-one (DDMP)-conjugated and non-conjugated forms in different samples in one single run for only 30min. Standard calibration curve was linear over the concentration range of 0.010–1.0mg/L for each soyasaponin. Within-day and day-to-day relative standard deviations were less than 9.2 and 13.1%, respectively.

Abstract:The purpose of this study was to fabricate a water-soluble phytosterol ester (PE) nanodispersion using emulsification-evaporation combined high-energy approaches (ultrasonic and microfluidization treatment). The effects of different surfactants, fabricating condition on the characterization of PE nanodispersions were investigated. Our results indicated that soybean protein isolate (SPI) was more suitable for the fabrication of PE nanodispersion than konjac gum, rice protein and sodium caseinate (NaCas). The average particle size, zeta-potential, encapsulation efficiency and loading amount of PE nanodispersion were 97.2 +/- 5.0 nm, 38.9 +/- 3.2 mV, 95.4 +/- 0.4% and 19.1 +/- 0.8%, respectively. Hydrophobic interaction played a significant role in the formation of nanodispersion. Microfluidization and high-speed shearing affected the secondary structure of SPI by breaking hydrogen bonds. Ultrasonic treatment could facilitate hydrogen bond formation, and increase hydrophobic interaction between SPI and PE. Emulsification-evaporation combined with ultrasonic method was gentler than microfluidization in the preparation of PE nanodispersion. The present method can be used to increase the water-solubility of PE and may be extended for other hydrophobic compounds.
Carboxyl-functionalized Magnetic Amide-Linked Covalent Organic Framework for Efficient Extraction of Multi-Class Mycotoxins from Soybeans Prior to HPLC-MS/MS Analysis

Abstract:Mycotoxins co -contamination in soybean poses great risks to human health, however, due to their diverse structures, it is difficult to simultaneously extract and detect these multi-mycotoxins. By incorporating modification of hydrophilic carboxyl groups with the hydrophobic properties of covalent organic frameworks, a stable cubic carboxyl-functionalized magnetic amide -linked covalent organic framework were fabricated, characterized and utilized as adsorbents for magnetic solid phase extraction (MSPE) of eleven frequently detected mycotoxins from soybeans followed by HPLC-MS/MS analysis. The key MSPE parameters were screened using the PlackettBurman design and optimized with the Box-Behnken design for response surface methodologies. Under the optimal experimental conditions, the linear range was 0.2-50 mu g/kg, the limits of detection were 0.1-1.4 mu g/kg. The established method was applied for the soybean analysis with satisfactory recoveries (70.8-108.3%) and precision (intra- and inter -relative standard deviations of 2.1-9.2% and 1.6-11.1%, respectively). With proper modification, this method may have great application potential for simultaneously analyzing and monitoring the multi-mycotoxin contamination in foods to ensure consumer safety and public health.

Abstract:A new analytical method was developed for the separation and determination of basic orange II, acid orange II and auramine O in soybean products. The technique was focused on ionic liquid reverse micelle microextraction (IL-RMME) followed by analysis and determination by ultra-high performance liquid chromatography (UHPLC) with photodiode array detector of three chemical dyes. In this method, IL-RMME solution consisting of ionic liquid [Omim]BF6 and surfactant GenapolX-080 was used as extractant. Important parameters affecting IL-RMME efficiency, such as extraction solvent type and volume, sample solution pH, salt effect, centrifugation speed and time were investigated. Under the optimal condition, the linearity of the method was in the range of 0.1-10 ng mL−1with correlation coefficient above 0.9994 and the limits of detection below 0.03 ng mL−1. At the same time, relative standard deviations of the developed procedure for intra- (n=5) and inter-day (n=5) precision were in the range of 5.04-8.50%. The results demonstrated that a simple fast environmentally friendly efficient method was successfully applied in the separation and determination of three chemical dyes in soybean products.

An Ultra-Fast and Green LC-MS Method for Quantitative Analysis of Aesculin and Aesculetin in Cortex Fraxini

Abstract:This study aims to develop a fast and eco-friendly liquid chromatography–mass spectrometry (LC-MS) method for the determination of aesculin and aesculetin in Cortex Fraxini. Ultrapure water was used as the solvent during the microwave-assisted extraction process to prepare the Cortex Fraxini sample. This extraction method reduces the cost of the harmful solvent (only ultrapure water was used) and microwave extraction time (1 min). The LC separation was conducted using an Agilent InfinityLab Poroshell 120 EC-C18 column (2.1 mm × 30 mm, 2.7 μm) with a mobile phase consisting of 0.1% formic acid and acetonitrile (90:10, v/v) at a flow rate of 0.6 mL/min. Isocratic elution was employed, and the analytes were detected by MS. Through careful optimization and selection of LC-MS conditions, the analysis time was reduced to 1 min, demonstrating the method’s efficiency. The developed method was validated and exhibited excellent specificity, linearity, limit, precision, accuracy, and stability in quantifying aesculin and aesculetin in the Cortex Fraxini samples. The analysis result revealed the presence of aesculin (ranging from 3.55 to 18.8 mg/g) and aesculetin (ranging from 1.01 to 16.2 mg/g) in all ten batches of Cortex Fraxini samples. Compared to the reported LC methods, this approach substantially reduces the total analysis time and requires a minuscule volume of organic solvents. An “Analytical Eco-Scale” assessment was used to evaluate the different assay methods of Cortex Fraxini. The current LC-MS method scored an impressive 90; it was better than the other four reports’ LC methods. Thus, the developed LC-MS method is rapid and green, which is helpful for the quality evaluation of Cortex Fraxini.

Abstract:A novel pre-treatment was proposed for the simultaneous determination of aflatoxins, ochratoxin A and zearalenone in foodstuffs using high-performance liquid chromatography with fluorescence detection. The analytical procedure was based on a first step using a quick, easy, cheap, effective, rugged, and safe based extraction procedure, followed by salting out and purification with a C18 solid-phase extraction column as interference removal clean-up. Subsequently, collected supernatant was subjected to dispersive liquid-liquid microextraction. Response surface methodology based on central composite design was employed to optimize conditions in the microextraction procedure. Under the optimum conditions, satisfactory analytical performance with recoveries ranging from 63.22 to 107.6% were achieved in different types of cereals and beans, as well as desirable precisions (0.81-8.13%). Limits of detections and quantifications for these six mycotoxins ranging from 0.03 to 13 mu g/ kg and 0.22 to 44 mu g/ kg, respectively, were obtained. Finally, the established method was successfully validated by four certified reference materials (P = 0.897 > 0.05) and applied to 79 samples from local markets.
Preparation of chitosan microextraction probe with hollow fiber as shape template

Abstract:Preparation of a novel coating is one of the keys to solid phase microextraction. A new method was proposed to prepare a chitosan microprobe with a film thickness of several dozen micrometers with the phase inverse technique by using a microporous polyacrylonitrile (PAN) hollow fiber with a diameter of 1.5 mm as template. The dilute acetic acid solution of chitosan and glutaraldehyde was added into the hollow fiber, a quartz fiber was inserted into the hollow fiber as supporter and then the hollow fiber was placed in the NaOH solution, because of the porosity of hollow fiber, NaOH diffused and contacted with acetic acid, then the chitosan thin film was formed. For intensifying the adsorption of acidic compounds, the probe was then modified with polyethyleneiminate (PEI). The SEM photos showed that the probe obtained with this process was 17 μm thick. The PEI-modified chitosan probes showed stronger adsorption of furomeside. Using NaOH as desorber, a concentration factor over 10 was observed and the solute concentration in the desorber was linear with that in the original samples. The operation is convenient and easy to realize automation.
Optimization of Ultrasonic-assisted Extraction of Soybean Isoflavones by Response Surface Methodology and Simultaneous Preparation of Protein Isolates and Oligosaccharides

Abstract:An effective and green strategy was developed to obtain isoflavones from soybean.Soy protein isolates and oligosaccharides were also obtained from soybean dregs simultaneously.Firstly,the optimal extraction conditions were investigated by single factor experiments and RSM optimization.A high extraction amount(2 104.25 μg·g-1)and yield(85.34%)of isoflavones were achieved under the optimal extraction conditions:ultrasonic power of 250 W,70% ethanol,70℃,120 min.Secondly,the purified isoflavones of 1 838.00 μg·g-1 was obtained with purity of 62.09% and recovery of 74.54% via three simple purification steps.Finally,the amount of soy protein isolates and oligosaccharides obtained from soybean dregs achieved 216.25 mg·g-1 and 73.75 mg·g-1,respectively.There is no waste liquid and solid generated from the whole process.The soybean could be utilized comprehensively through this strategy.

Abstract:A confirmatory and reliable procedure has been developed for extraction and determination of Cnidii fructus by accelerated solvent extraction (ASE) and high-performance liquid chromatography coupled with photodiode array, electrospray ionisation ion trap tandem mass spectrometry and time of flight mass spectrometry (HPLC-PDA-ESI-ITMSn/TOF-MS). The determination method enabled the characterisation of sixteen bioactive components in C. fructus and quantification of three major coumarins, namely osthole, imperatorin and isopimpinellin. Response surface methodology (RSM) was employed to optimise the extraction parameters yielding the optimum conditions of ASE (extraction temperature 122 degrees C, extraction time 5 min and two static cycles). And the total contents of three major coumarins extracted by ASE under the optimum conditions was significantly higher than those by reflux and ultrasonic extraction (P< 0.05) with better reproducibility. At last, the proposed method coupled with pattern recognition was applied to analysis of C fructus from eight different regions in China. (C) 2013 Elsevier Ltd. All rights reserved.

Abstract:An efficient and green ultrasonic-assisted micellar extraction coupled with ultra-high performance liquid chromatography with photodiode array (UHPLC-PDA) method was developed for the multi-ingredients quantitative analysis of Yangxinshi Tablet (YXST). The active ingredients were extracted from YXST using trehalose lipid biosurfactant solution as an environmentally friendly extraction solution. The response surface methodology (RSM) based on Box-Behnken design (BBD) was utilized to seek for the optimum extraction conditions of target analytes. When the concentration of trehalose lipid solution was 7 mg/mL, the liquid to solid was 125:1 (mL:g) and the extraction time was 40 min, the total extraction yield of eleven compounds (including puerarin, daidzin, ferulic acid, calycosin-7- O - β -D-glucoside, tetrahydropalmatine, coptisine, epiberberine, jatrorrhizine, berberine, palmatine chloride and icariin) obtained the maximum value. The relative standard deviations (RSD) of intra-day and inter-day precision were all less than 5.0%. The recoveries of all analytes were in the range of 95.1%-104% with the RSDs were all below 3.0%. Consequently, the ultrasonic-assisted micellar extraction coupled with UHPLC-PDA method could be successfully and efficiently applied to the extraction and analysis of target components in YXST.

Abstract:A new simple, fast and environmentally friendly deep eutectic solvent based dispersive liquid-liquid microextraction (DES-based DLLME) methodology assisted by vortex is presented for the separation and preconcentration of three elements (i.e., Fe, Cu and Pb) from edible oil samples (i.e., soybean, sunflower, rapeseed, sesame, and olive oil) prior to the determination by microwave-induced plasma optical emission spectrometry (MIP-OES). The deep eutectic solvent selected as extractant (i.e., choline chloride and ethylene glycol, 1:2) is synthesized and characterized by Fourier transform infrared spectroscopy (FT-IR), proton nuclear magnetic resonance spectroscopy ( 1 H NMR) and differential scanning calorimetry (DSC), and the extraction conditions are optimized by a two steps experimental design. Under the optimum extraction conditions (i.e., diluted sample weight: 8.6 g; DES volume: 100 μL; extraction time: 1 min; centrifugation time and speed: 3 min and 3000 rpm; and dispersion system: vortex) the analytical method presents excellent linearity (i.e., R 2 values higher than 0.99) in the range 10–500 μg kg −1 , repeatability (i.e., CV values lower than 9.2%), and limits of detection (LOD) values of 3, 2 and 0.7 μg kg −1 for Pb, Fe and Cu, respectively. None of the analytes displayed amounts over the upper limit permitted by law, and recovery values of all analytes evaluated in the different samples using external standard calibration were close to 100%, which excludes significant matrix effects. Finally, AGREEprep metric has been used to evaluate the method greenness (final score of 0.47) and it has been compared successfully with previous publications for the same type of analytes and matrices.

Abstract:The present study investigated the possibility of using near-infrared reflectance spectroscopy (NIRS) for quantitative determination of oligosaccharides, including sucrose, raffinose and stachyose in soybean products. One hundred and sixty samples (de-hulled soybean meal, extruded soybean meal, extruded full-fat soybean and fermented soybean meal) were collected and screened, and furthermore forty artificial soybean meal samples were prepared. Calibrations were performed by partial least squared algorithm with different derivatives, multiplicative scatter correction or normalization vector spectral pretreatment. The coefficients of determination for calibration (R-cv(2)), coefficients of determination for cross validation (R-cv(2)) and RPDcv in models were 0. 99, 0. 95, 0. 98; 0. 98, 0. 94, 0. 97; 7. 24, 4. 13, 5. 98; respectively. The results supposed that NIRS can be a quantitative determination method of oligosaccharides in soybean products except fermented soybean meal.
Analysis of forchlorfenuron and thidiazuron in fruits and vegetables by surface-enhanced Raman spectroscopy after selective solid-phase extraction with modified β-cyclodextrin

Abstract:β-Cyclodextrin and its derivatives can selectively bind to various organic molecules in its cavity and provide good applications in sample preparation. Surface-enhanced Raman spectroscopy is a sensitive technique and has received increasing attention in the last decade. Herein, 3,5-dimethyl phenyl carbamoylated β-cyclodextrin bonded silica gel was used as a ssorbent in solid-phase extraction to selectively enrich forchlorfenuron and thidiazuron followed by determination with surface-enhanced Raman spectroscopy. It showed excellent selectivity for forchlorfenuron and thidiazuron and the adsorption capacities were 40.0 and 30.0 μg/g, respectively. A rapid and sensitive method based on the modified β-cyclodextrin solid-phase extraction coupled with surface-enhanced Raman spectroscopy was developed. The linear ranges were 30.0-300.0 μg/L for forchlorfenuron and thidiazuron at 1005 and 640 cm(-1) , respectively. Both of the limits of detection were 15.0 μg/L, which were significantly lower than the maximum permitted by the National Standard. The recoveries of forchlorfenuron and thidiazuron were 78.9-87.9% for the spiked grape, kiwi, cucumber and tomato, with relative standard deviations of 8.1-13.2%. The results show that this method is sensitive, selective, and relatively time saving, and has great potential in the analysis of trace amounts of plant growth regulators in fruits and vegetables.
A fully green sample preparation method for synthetic antioxidants determination in edible oils based on natural feather fiber-supported liquid extraction

Abstract:The current study proposed a novel feather fiber-supported liquid extraction (FF-SLE) method for extracting analytes from oil samples. The natural feather fibers were used as the oil support material and directly loaded in the plastic tube of a disposable syringe to construct the low-cost extraction device (∼0.5 CNY). The edible oil without any pretreatment including dilution was added directly to the extraction device, followed by the addition of the green extraction solvent of ethanol. As an example, the proposed method was applied to extract nine synthetic antioxidants from edible oils. The optimized extraction conditions for processing 0.5 g of oil were obtained when the syringe dimension was 5 mL, the extraction solvent was 0.5 mL of ethanol, the amount of feather fibers was 200 mg of duck feather fibers and the static extraction time was 10 min. The applications to seven kinds of feathers and seven kinds of edible oils all indicated the excellent oil removal efficiencies (>98.0%). Combined with high-performance liquid chromatography-ultraviolet, a quantification method was validated with satisfied linearity (R 2 ≥0.994), accuracy (95.8-114.6%) and precision (≤8.3%) with the limits of detection ranging from 50 to 100 ng/g. The proposed FF-SLE method was simple, effective, convenient, low-cost, green and environmental-friendly for the extraction of analytes from oil samples prior to instrument analysis.

Abstract:In this study, an on-line supercritical fluid extraction (SFE) and supercritical fluid chromatography (SFC) method was developed for the rapid determination of oleanoic acid and ursolic acid in Chaenomelis Fructus. After optimization of the conditions, the two triterpenoid acids was obtained by SFE using 20% methanol as a modifier at 35°C in 8 min. They were resolved on a Shim-pack UC-X Diol column (4.6 × 150 mm, 3 μm) in 14 min (0 - 10 min, 5 - 10%; 10 - 14 min, 10% methanol in CO2) with a backpressure of 15 MPa at 40°C. The on-line SFE-SFC method could be completed within 40 min (10.79 mg/g dry plant, Rs = 2.36), while the ultrasound-assisted extraction and HPLC method required at least 90 min (3.55 mg/g dry plant, Rs = 1.92). This on-line SFE-SFC method is powerful to simplify the pre-processing and quantitative analysis of natural products.
Rapid-resolution HPLC with spectrometric detection for the determination and identification of isoflavones in soy preparations and plant extracts

A linear gradient elution (0 min 22% B, 1.0 min 80% B, 1.4 min 100% B, 1.8 min 22% B) using a mobile phase containing 0.2 % (v/v) acetic acid (solvent A) and methanol (solvent B) was applied on a Zorbax SB C18 column (1.8 microm particle size) at 80 degrees C. The method was verified using samples of bits of soy and methanolic extracts from Trifolium pratense, Iresine herbstii and Ononis spinosa plants. Pseudobaptigenin glucoside, irilone, prunetin, texasin, tlatlancuayin and other isoflavones, in addition to aglycones of isoflavones and their beta-glucosides and malonyl and acetyl derivatives, were identified by UV-VIS DAD and electrospray mass spectrometric (ESI-MS) detection in the extracts.

Abstract:The separation and extraction of chrysin from active ingredients of natural products are of great significance, but the existing separation and extraction methods have certain drawbacks. Here, chrysin molecularly imprinted nanofiber membranes (MINMs) were prepared by means of electrospinning using chrysin as a template and polyvinyl alcohol and natural renewable resource rosin ester as membrane materials, which were used for the separation of active components in the natural product. The MINM was examined using Fourier transform infrared (FT-IR) spectroscopy, scanning electron microscopy (SEM), and thermogravimetric analysis (TGA). The adsorption performance, adsorption kinetics, adsorption selectivity, and reusability of the MINM were investigated in static adsorption experiments. The analysis results show that the MINM was successfully prepared with good morphology and thermal stability. The MINM has a good adsorption capacity for chrysin, showing fast adsorption kinetics, and the maximum adsorption capacity was 127.5 mg·g−1, conforming to the Langmuir isotherm model and pseudo-second-order kinetic model. In addition, the MINM exhibited good selectivity and excellent reusability. Therefore, the MINM proposed in this paper is a promising material for the adsorption and separation of chrysin.
