Abstract:Grape seeds is a popular health food with various biological activities around the world. Grape seeds polysaccharides (GP) have antioxidant, anti-inflammatory, immunomodulatory and hypolipidemic effects, but little is known about the effects of GP on the gut microbiota. Therefore, we aim to investigate the fermentation properties of polysaccharides isolated from grape seeds and its effect on the mice's gut microbiota. We obtained polysaccharides from grape seeds by water extraction and alcoholic precipitation. We simulated the digestion and fermentation of GP in human body, and used bioinformation approach to study its effects on the mice's gut microbiota. In the simulated fermentation experiment of GP, the administration of GP significantly decreased the relative abundance of Fusobacterium and Allisonella , and significantly increased the relative abundance of Bacteroides at genus level. In addition, GP altered the structure and proportion of intestinal microbial community in Kunming mice, especially increased the relative abundance of Lachnospiracea _NK4A136_group (p = 0.008239). In conclusion, the effect of polysaccharides from grape seeds on gut microbiota was investigated in this study, and the results suggested that GP has the potential to become a functional food prebiotic. It provided the basis and insight for further exploration of grape seed polysaccharides, and provided a new idea for the development of functional food prebiotics.

Abstract:Proanthocyanidins is major polyphenols and widely existed in plants,and will produce anthocyanin under heating in acidic conditions.Proanthocyanidins has a very high resistance to oxidation and strong radical scavenging ability,and it also has anti-inflammatory,anti-cancer,anti-aging functions.Therefore,it widely used in medicine,food,health care and cosmetics.There are abundant proanthocyanidins in grape seeds,extracting proanthocyanidins from grape seeds by ethanol can fully use of the wine making by-products.The extraction rate of proanthocyanidins and its DPPH free radical scavenging capacity are two indexes in determining the optimum extraction process.The experiment provides support for the further development and utilization of proanthocyanidins.The optimum extraction process conditions determined by single-factor and orthogonal test are as follows:solid-liquid ratio of 1 ∶ 10,extraction time 90min,extraction temperature 20℃,ethanol concentration 70％,pH=4.Under the above conditions,the extraction amount of proanthocyanidins was 1.584rmg/g,and DPPH free radical scavenging capacity was 12.41 μmol/g.

Abstract:An extraction process of polyphenol from wine grape seeds was studied in this paper. Water was used as solvent and the material studied consisted of wine grape seeds, which was obtained from the pomace from wine vinification. Before extraction grape seed oil was squeezed out. Different factors were considered, such as grinding degree, material-water ratio, extract temperature, time, and frequency. Based on single factor tests, orthogonal test was done. Results showed that temperature was the most important factor, then frequency, time, and material-water ratio in turn, while grinding degree's effect on the extraction was not statistically evident. The optimum extraction conditions were as follows: 100 ℃ of water temperature, 1:9 of material-water ratio, 1.5h of extract time and 3 times. The concentration of the solution was about 1mg/mL. GSPE powder was produced by freeze drying, and the rate of production was about 3.3%.

Abstract:Few investigations have been conducted to evaluate pressurized liquid extraction (PLE) technology as a sustainable method for the recovery of phenolic compounds of grape seed by-products. In this study, PLE combined with an experimental design was evaluated for optimizing the sustainable extraction of phenolic compounds from grape seed by-products. The solvent ethanol content (X1, 0–100%), temperature (X2, 20–100 °C) and time (X3, 1–11 min) were studied as independent experimental factors. Yield, TPC, antioxidant activity and phenolic composition were analyzed as optimized dependent variables. Two optimal extraction conditions at different temperatures (20 °C and 100 °C) were found, but thermal degradations at 100 °C allowed for selecting the optimal condition as 75% ethanol, 11 min and 20 °C. The optimal extracts showed high phenolic content (TPC = 350.80 ± 3.97 mg GAE/g extract) and antioxidant activity (ABTS, 9.31 ± 0.33 mmol Trolox/g extract), mainly composed of polymeric and mono-oligomeric flavan-3-ols. The digestion process reduced the TPC and antioxidant activity due to the low bioaccessibility of the flavan-3-ols, mainly as catechin, epicatechin and polymeric proanthocyanidin losses during the digestion process. However, increases in the antioxidant activity of the basolateral side (DDPH, 0.061 ± 0.000 mmol Trolox/g extract) were determined after in vitro transepithelial transport, which is a consequence of bioavailable catechin and epicatechin and reduced amounts of dimer B2, dimer B1, epicatechin gallate and gallic acid. Consequently, PLE combined with hydroalcoholic solvents at a low temperature resulted in a valuable methodology to obtain sustainable extracts from grape seed by-products (contributing to the circular economy), containing bioavailable phenolic compounds, which are able to increase the antioxidant status.

Abstract:Eighty percent of grape production is destined for the wine industry, which generates various types of waste, of which grape pomace is the main one, accounting for 50–60% of waste created during processing. This waste could be a promising source of bioactive compounds (e.g., flavonoids and tannin), which are known for their antioxidant properties. Although these byproducts pose disposal challenges, they can be utilized as a substrate for solid-state fermentation bioprocess using co-cultures, where different microorganisms can interact and complement each other, improving the efficiency of metabolite production or substrate degradation. This study investigates the extraction of phenolic compounds and the antioxidant activity of the compounds from grape pomace in the solid-state fermentation bioprocess, comparing fungal and yeast monocultures, and then exploring the use of two co-cultures (P. stipites/A. niger GH1 and S. cerevisiae/A. niger) on the flavonoid extractive process. Fermentation kinetics were evaluated over 120 h, with sampling done every 12 h. Initially, yeasts were used to reduce the content of simple sugars in the medium, and fungus was added at 24 h into the process due to its ability to produce a broad spectrum of extracellular enzymes, allowing a higher efficiency in substrate degradation. Competition or antagonism during co-culture leads to significantly higher production of compounds, which are recovered using different solvents. The evaluation included phenolic compounds (total polyphenols, condensed tannins, and total flavonoids), antioxidant activity (DPPH●/FRAP), molecular characterization (HPLC-MS), and structural microscopy during the bioprocess. The highest titers obtained were 62.46 g/L for total flavonoids and 32.04 g/L for condensed tannins, using acetone as the solvent in co-culture with P. stipitis after 120 h of fermentation. Characterization identified 38 compounds, highlighting families of flavonols, hydroxybenzoic acids, and hydroxycinnamic acids. The co-culture of P. stipitis and A. niger GH1 significantly improved the extraction yield of bioactive compounds through solid-state fermentation.

Abstract:Grape pomace seeds contain abundant phenolic compounds, which are also present in both soluble and insoluble forms, similar to many other plant matrices. To further increase the extractable soluble phenolics and their antioxidant activities, grape pomace seeds were fermented with different fungi. Results showed that solid-state fermentation (SSF) with Aspergillus niger, Monascus anka, and Eurotium cristatum at 28 °C and 65% humidity had a significantly positive impact on the release of soluble phenolics in grape pomace seeds. Specifically, SSF with M. anka increased the soluble phenolic contents by 6.42 times (calculated as total phenolic content) and 6.68 times (calculated as total flavonoid content), leading to an overall improvement of antioxidant activities, including DPPH (increased by 2.14 times) and ABTS (increased by 3.64 times) radical scavenging activity. Furthermore, substantial changes were observed in the composition and content of individual phenolic compounds in the soluble fraction, with significantly heightened levels of specific phenolics such as chlorogenic acid, syringic acid, ferulic acid, epicatechin gallate, and resveratrol. Notably, during M. anka SSF, positive correlations were identified between the soluble phenolic content and hydrolase activities. In particular, there is a strong positive correlation between glycosidase and soluble phenols (r = 0.900). The findings present an effective strategy for improving the soluble phenolic profiles and bioactivities of grape pomace seeds through fungal SSF, thereby facilitating the valorization of winemaking by-products.

Abstract:Grape pomace (GP) is a major byproduct worldwide, and it is well known for its bioactive compounds, such as fibers and phenolic compounds, that are popular for their impact upon human health, including gastrointestinal health. The objective of this work was to evaluate the chemical composition and biological activities of an enzymatic GP extract, as well as to investigate how gastrointestinal digestion (GID) modulates these properties. GP extract was previously produced using an enzymatic cocktail with xylanase activity and was then exposed to simulated conditions of GID, characterized for its chemical composition, and screened for antimicrobial, prebiotic, and antioxidant activities. The safety of this ingredient after GID was also assessed. GP extract presented high contents of dietary fiber and other carbohydrates, including xylooligosaccharides, in addition to minerals and phenolic compounds. In vitro simulated GID revealed that xylobiose was resistant to gastric conditions, unlike phenolic compounds. The use of 2% (w/v) of this ingredient proved to be a potential carbon source that could be fermented by Lactobacillus and Bifidobacterium spp, even after digestion. The extract also exhibited strong antioxidant and antimicrobial activities against Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa; however, after GID, the antioxidant capacity decreased, and the antimicrobial capacity was strongly reduced or lost. Furthermore, the extract safety was also guaranteed on Caco-2 intestinal cells. This novel and green GP extract proved to be composed of relevant bioactive molecules, including xylooligosaccharides, polyphenols, organic acids, and minerals, which provided different biological properties; it has potential applications in the food industry such that it can be used as an ingredient in the development of new functional foods.

Abstract:With the grape seeds as the raw material to study the effect of affecting factors and extraction process of polysaccharide.Polysaccaride was extracted from grape seeds by hot water extraction,the content of total polysaccharides was measured by phenol-sulfuric acid method.The effects of temperature,liquid-solid ratio and time on the extraction were analyzed,the best condition and production process of polysaccharide extracted from grape seeds were deter mined.The optimum technical condition of polysaccharides from grape seeds was as followed:temperature 95 ℃,solid-liqid ratio 40 g/L,time 3 h.The yeild of polysaccharides was 1.36 % under the conditions.The method of extraction and deter mination of polysaccharides from grape seeds is simple,reliable and stable.

Abstract:In this paper,the process of extracting polyphenol with ethanol from grape residue was studied. The result showed that the appropriate processing condition was extracting proportion 1∶6,extracting temperature 70 ℃,extracting time 12 h,extracting 3 times and alcohol concentration 70%,the rate of extracting polyphenol was about 80%;the total quantity of polyphenol in grape residue was 8.41%. At the same time,the effect of temperature was studied on the extraction rate of Res in grape residue.

Abstract:Fecal samples from 20 healthy adults were collected for in vitro fermentation experiments to investigate the effects of combined probiotics on the utilization of grape seed extract in humans. After fermenting for 24 h, short-chain fatty acids, metabolites, and gut microbiota composition were analyzed. Short-chain fatty acids in the grape seed extract probiotics group were significantly higher than those in the grape seed extract group. Probiotics significantly enhanced the conversion and utilization of catechins and epicatechins in grape seed extract group and increased the production of 3-hydroxyphenylacetic acid. The 16S rRNA sequencing results revealed that compound probiotics significantly increased the relative abundance of Lacticaseibacillus, HT002, Bifidobacterium, and Lactobacillus and reduced that of Escherichia-Shigella. Our findings showed considerable individual variability in the metabolic utilization of grape seed extract in humans. The consumption of probiotics appears to significantly enhance the utilization.
Polyphenols from Grape Skin:Optimization of Ultrasonic-assisted Extraction Process by Response Surface Methodology and Antioxidant Activity Evaluation

Abstract:Response surface methodology was employed to optimize the ultrasonic-assisted extraction of polyphenols from grape skin as a byproduct of wine making.On the basis of one-factor-at-a-time experiments,through which the optimal ultrasonic power and ethanol concentration were found to be 100 W and 40%,respectively,polyphenol yield was investigated by response surface methodology with respect to liquid-to-material ratio,extraction time and temperature at three levels.The optimal levels of liquid-to-material ratio,extraction time and temperature were determined to be 16:1(mL/g),57 min and 50 ℃.Under such conditions,the yield of grape skin polyphenols was(42.51 ± 1.21) mg/5 g.The antioxidant activity in vitro of the obtained extract was evaluated by DPPH free radical scavenging assay and potassium ferricyanide reduction assay using water-soluble VE as the control.Grape skin polyphenols were found to have powerful DPPH free radical scavenging capacity and reducing power in a linear dose-dependent manner.

Abstract:Vitis davidii Foex whole seed (VWS) is a by-product during the processing of grape products, which is rich in bioactive compounds that have great potential in the food industry. In this study, the bioactive compounds and antioxidant activity of VWS were determined, and their dynamic changes during in vitro colonic fermentation were also investigated after VWS subjected to in vitro simulated digestion. Results showed that VWS were rich in polyphenols (23.67 ± 0.52 mg GAE/g), flavonoids (13.13 ± 1.22 mg RE/g), and proanthocyanidins (8.36 ± 0.14 mg CE/g). It also had good DPPH and ABTS radical scavenging activity, which reached 82.10% and 76.10% at 1000 μg/mL. The alteration trend of the antioxidant activity during in vitro fermentation for 24 h was consistent with that of the content of bioactive substances, such as polyphenols, with the extension of fermentation time. The bioactive compounds and antioxidant activity showed a trend of increasing and then decreasing, reaching the highest value at 8 h. The high-throughput sequencing analysis of the regulatory effect of VWS on intestinal micro-organisms revealed that VWS influenced intestinal microbiota diversity. The relative abundance of beneficial microbiota, such as Blautia and Parabacteroides, increased by 4.1- and 1.65-fold after 24 h of fermentation compared with that of the control group. It also reduced Escherichia-Shigella by 11.23% and effectively reduced host inflammation, while increasing the contents of acetic acid, propionic acid, and other metabolites. Taken together, these results reveal the value of VWS utilization and provide new insights into the nutritional and microbiota modulation effects of VWS, which could therefore serve as a nutraceutical ingredient in health promotion.

Abstract:The extraction technology of proanthocyanidins from grape seeds was studied using response Surface Method(RSM),and acetone concentration,solid-liquid ratio,temperature and time were selected as experimental the factors.Based on the analysis of significance and interaction among different factors,the optimal condition of extraction technology was obtained as followed: acetone concentration was 75%,solid-liquid ratio was 1 ∶ 10,time was 44 min and temperature was 51 ℃,and the predicted value and measured value of proanthocyanidins yield was 2.90% and 2.88%,and the purity was 55.92%.

Abstract:Based on the single-factor experiments and the principle of Box-Behnken experiment design, extraction temperature, extraction time and the ratio between solvent and dreg were chosen to be the there factors. We used response surface design which was selected there factors and there levels to optimize the extraction condition of trans-resveratrol from grape pomace. The results showed that the there factors had noticeable correlation with extraction content and that the optimization extraction condition was that temperature is 69 ℃, time 61 min, the ratio 46 mL/g. On the condition of this, the theory content was 104.2859 μg/g.

In Vitro Fermentation Characteristics of Modified Dietary Fiber from Grape Pomace

Abstract:Human excrement extract was used as the inoculum to modify grape pomance dietary fiber in a mimic human intestinal environment by in vitro fermentation.The pH and the content of short-chain fatty acids(SCFAs) in modified dietary fiber obtained after different times of fermentation were determined.The results indicated that grape pomance dietary fiber was one of ideal carbon sources that could be exploited by intestinal microflora.SCFAs including acetic acid,propionic acid,butyrate acid and isobutyrate acid were produced during fermentation in vitro.Meanwhile,no obvious pH change in the fermentation broth of modified grape pomace dietary fiber was observed.However,the contents of total SCFAs and each SCFA were affected due to fermentation treatment.

Abstract:Response surface methodology was applied to optimize the extraction of proanthocyanidins from seeds of the new grape cultivar Meili using an accelerated solvent extraction(ASE) system.The optimal extraction conditions were found to be 13 min,1161 psi,80% and 80 ℃ for extraction time,pressure,methanol concentration and temperature,respectively.The maximum predicted extraction yield of proanthocyanidins under the optimized conditions was 6.817% indicating a relative error of 7.79% as compared with the actual value of 6.286% obtained after the third repeated extraction.The purity of proanthocyanidins under the optimized conditions was 41.02%.

Abstract:Background and Objective: Vine tea (Ampelopsis grossedentata) may potentially perform multiple pharmacological roles, including antibacterial, anti-cancer, antioxidant, hepatoprotective and anti-hypertension functions. But effects of extraction polarity, time and temperature on the recovery of bioactive compounds along their pharmacological activity of vine tea extract have not been reported yet. The present study reports on the importance of extraction variables for obtaining vine tea extract aiding in purification and isolation of functional and polyphenolic compound and their pharmacological activity (polyphenols) from vine tea. Materials and Methods: The effects of extraction variables (time, temperature and polarity) on Total Phenolic Content (TPC), Total Flavonoid Content (TFC) and Condensed Tannin Content (CTC) were evaluated using single factor experiment. Antiradical capacity assay and radical-scavenging capacity 2,2-diphenyl-1-picrylhydrazyl (DPPH) were tested for evaluating the antioxidant activities of vine tea crude extract. To analyze data, one-way analysis of variance (ANOVA) was established followed by the Tukey's test and to test the correlation, Pearson correlation coefficient was tested. Results: Data explained that extraction polarity, extraction time and the temperature had a significant effect (p<0.05) on yield and their anti-radical activities. The optimized extraction parameters for TFA and CTC were 40% aqueous ethanol at 45 while 50 for TPC for 180 min. Whereas, maximum yield were 40.01 mu g Catechin Equivalent per gram (CE g(-1)) of Dry Weight (DW) for TFA, 15.12 mu g, Gallic Acid Equivalent per gram of DW (GAE g(-1)) for TPC and 12.70 mu g CE g(-1) DW for CTC. Ethanol concentration showed a significant effect (p<0.05) on extraction of phenolic compounds and their pharmacological activity especially antioxidant capacity and DPPH assay. Conclusion: This study showed that extraction variables were greatly influence on the pharmacological activities of vine tea crude extract and this study can be used as preliminary and key information to design central composite rotatable design for Response Surface Methodology (RSM). Moreover, this approach can be used to determine significant factors that influence functional compounds and their other pharmacological activities.
Effects of Grape Seed Oil Addition on Rumen Fermentation and Methanogenesis in Vitro

Abstract:A diet of 7 concentrate: 3 forage was used as substrate in vitro,treatments were consisted of control group(0) versus three levels of grape essential oils(100,150,250 μL/L),3 repeats in each group.In vitro gas production(GP) was recorded and methane production was determined 3,6,9,12 and 24 h after incubation.Ammonia nitrogen,volatile fatty acids(VFAs),protozoa and pH were measured after 24 h incubation.The results showed that the gas production,pH and acetate proportion were increased by supplementations of 100 and 150 μL/L grape seed oils(P0.05),where the methane production(P0.01),molar proportion of propionate,butyrate,valerate and BCVFA decreased.In addition,ammonia nitrogen and protozoa were decreased by supplementation of 150 μL/L grape seed oils,while,total volatile fatty acid increased by 100 μL/L grape seed oils.In conclusion,addition of grape seed oils at a proper level could change the fermentation pattern,increase the propionate acetate ratio and inhibit methanogenesis.
