Abstract:This paper presents an approach to the preparation of a new glycerylsilane with glycerol and tetraethyl orthosilicate, using Amberlyst 15 (dry), an acidic ion-exchange resin, as catalyst. Trypsin was selected as a model enzyme to study the biocompatible properties of the resultant glycerylsilane by comparing the properties of trypsin in solution, trypsin entrapped in tetraethyl orthosilicate and the resultant glycerylsilane derived sol-gel matrix. The enzyme entrapped into the glycerylsilane derived sol-gel glass exhibits improved properties, which is not round in free enzyme and conventional sol-gel glass and the result indicates the glycerylsilane has good biocompatible properties.

Abstract:Three-dimensional branched copolymer, with N,N′-methylene bisacrylamide as the crosslinker and 3-allyloxy-2-hydroxy-1-propane sulfonic acid sodium salt as the monomer, was grafted from silica particles by thiol-ene click reaction. The obtained hydrophilic material with sulfonic acid groups was successfully applied for chromatography separation and glycopeptide enrichment. The separation mechanism was proven as the mixed mode of hydrophilic interaction and cation-exchange by investigating the effect of various chromatographic factors on the retention of polar analytes. By such mixed-mode chromatography, nucleosides, nucleobases, and acidic compounds were successfully separated. The column efficiency was up to 136,000 theoretical plates m−1 for cytidine, which was much higher than those of previous reports. Furthermore, benefitting from the large amount of hydrophilic groups provided by the branched copolymer, the material was used for the selective enrichment of glycopeptides. Results demonstrated the great potential of such material for chromatography separation and glycoproteome analysis.Graphical abstractThe branched copolymer modified HILIC/cation-exchange particles Sil@Poly(AHPS-co-MBAAm) were prepared via thiol-ene click copolymerization reaction. Such Sil@Poly(AHPS-co-MBAAm) particles showed great performance in the separation of polar compounds and the enrichment of glycopeptides.

Abstract:A glutathione (GSH)-functionalized silica material was prepared using divinyl sulfone activation chemistry (named SiO2-DVS-GSH). The successful synthesis of the SiO2-DVS-GSH material was confirmed by FT-IR, elemental analysis, and zeta potential analysis. The effects of water content, pH value, and salt concentration in the mobile phase on the model compound (uracil, uridine, cytosine, cytidine, guanosine, xanthosine, orotic acid) retention was studied, and a hydrophilic interaction liquid chromatography (HILIC) retention feature together with electrostatic interaction of the SiO2-DVS-GSH material was observed. The prepared stationary phase was further applied for the separation of oligosaccharide. In addition, the SiO2-DVS-GSH material displayed remarkable selectivity and specificity for the sialylated N-glycopeptides’ enrichment from bovine fetuin tryptic digests, even at a mass ratio of 1:1000 (w/w) to bovine serum albumin (BSA, non-glycosylated protein), showing superior performance compared to commercial ZIC-HILIC material. Moreover, the SiO2-DVS-GSH material behaved well in the N-glycopeptides’ enrichment from human serum, demonstrating its promising potential for glycoproteomics of complex biological samples.Graphical AbstractA glutathione (GSH)-functionalized silica material was prepared using divinyl sulfone activation chemistry, and it shows remarkable selectivity and specificity for the sialylated N-glycopeptides’ enrichment.

Abstract:In this study, core-shell mesoporous silica-carbon composite microspheres (Sil@MC) were prepared by one-step coating of the phenol formaldehyde polymer (PF) on SiO2 surface and by carbonizing the PF polymer under nitrogen atmosphere. The morphology observation of the Sil@MC stationary phase showed that it had good monodispersity. Surface area (302 m2/g), mean pore diameter (9.5 nm), and pore volume (0.63 cm3/g) of Sil@MC materials were also measured by pore structure analysis. The results showed that the Sil@MC was successfully immobilized on the silica particles via copolymerization and carbonization. As a stationary phase of HPLC, the Sil@MC column was filled by a slurry method. The Sil@MC materials formed after calcination of SiO2 coated with phenolic resin could be used for the separation of four polar sugar compounds (D-(+)-glucosamine hydrochloride, glucose, D-(+)-trehalose dihydrat and raffinose) with the mobile phase of acetonitrile-water (containing 0.1% (v/v) formic acid). However, the material formed by calcinating SiO2 without coating phenolic resin could not separate these polar sugar compounds by HPLC-MS. Finally, the representative oligosaccharide isomers of raffinose, melezitose and stachyose, nystose, and human milk oligosaccharide isomers, such as 3'-sialyllactose, 6'-sialyllactose and lacto-N-newtetraose, lacto-N-tetraose, were successfully separated by the Sil@MC column with good peak shapes. The results demonstrates that silica-carbon composites derived from phenolic resin have potential application in polar compounds chromatographic separation.

Abstract:Modification of surface silanols is a topic of interest in the preparation of organo-functionalized silica particles. Herein, two novel contributions, mainly focused on separation science, were presented: i) the horizontal polymerization on silica surface by using the vinyl-triacetoxy silane and ii) a preparation of cysteine-based stationary phase via photo-click thiol-ene coupling. In the first derivatization step, the vinyl-triacetoxy silane was employed instead of conventional trichloro vinyl one. The one-step synthetic procedure needed imidazole as an activating agent in addition to the silica hydration. Modified silica particles offer a high loading of vinyl fragments and an extensive passivation of silanols such as to not require a subsequent end-capping procedure. The structural morphology of media was deeper characterized by combining infrared spectroscopy, solid-state nuclear magnetic resonance, and elemental analysis. A first application, the photo-click cysteine-based material was prepared by photo-click reaction and the stationary phase was employed in the separation of some conventional targets by hydrophilic interaction chromatography.

Abstract:There are great challenges in the field of natural product isolation and purification and in the pharmacological study of oligosaccharide monomers. And these isolation and purification processes are still universal problems in the study of natural products (NPs), traditional Chinese medicine (TCM), omics, etc. The same polymer-modified materials designed for the special separation of oligosaccharides, named Sil-epoxy-PEI and Sil-chloropropyl-PEI, were synthesized via two different methods and characterized by scanning electron microscopy combined with energy spectrum analysis, Fourier transform infrared spectroscopy, thermogravimetric analysis, zeta potential as well as surface area analysis, etc. Several nucleotide/nucleoside molecules with different polarities and selectivities were successfully isolated in our laboratory using stainless-steel columns filled with the synthesized material. In addition, the separation of saccharide probes and oligosaccharides mixtures in water extracts of Morinda officinalis were compared in HILIC mode. The results showed that the resolution of separations for the representative analytes of the Sil-epoxy-PEI column was higher than for the Sil-chloropropyl-PEI column, and the developed stationary phase exhibited improved performance compared to hydrothermal carbon, amide columns and other HILIC materials previously reported.

Abstract:Thiol-ene click chemistry was applied to immobilize polyacrylamide onto alkenyl-modified silica to prepare a novel stationary phase for hydrophilic interaction liquid chromatography. Polyacrylamide was synthesized by reversible addition-fragmentation chain transfer reaction. Alkenyl functionalities were introduced onto the silica by the reaction of silica with the reaction product of 3-isocyanate opropyl trimethoxy silane, allylamine and pyridine in toluene to obtain alkenyl-modified silica (EUS, E-thiol-ene, U-urea, S-silica). The alkenyl-modified silica was then reacted with polyacrylamide in methanol at 55 °C for 48 h in the presence of 2,2-azo diisobutyl amidine hydrochloride to prepare a novel stationary phase (TE-UPAM, TE-thiol-ene, U-urea, PAM-polyacrylamide), for hydrophilic interaction liquid chromatography. This new stationary phase “TE-UPAM” was characterized by element analysis and infrared spectra, and the results showed that, compared with EUS, the carbon content of TE-UPAM increased and the characteristic peaks of multiple amide existed at 1636 and 1570 cm−1. The effects of water content, salt concentration and pH of the mobile phase on the retention time of polar compounds were investigated. The results showed that the retention time of polar compounds decreased with the increase of water content. In contrast with acidic polar compounds, the retention time of neutral and basic polar compounds increased with the increase of salt concentration, and the retention time of basic polar compounds decreased with the decrease of pH value in the range of 3.3–4.8. The novel stationary phase behaved better than traditional silica stationary phase in the separation of uridine and bases. In addition, good separation of oligosaccharides by the novel stationary phase was also achieved, indicating great potential in the separation of polar compounds.

Abstract:In this study, we report a simple procedure for applying molecular imprinting functional groups to the inner surfaces of the template-synthesized sol-gel nanotubes for chemical separation of estrone. The silica nanotubes were synthesized within the pores of nanopore alumina template membranes using a sol-gel method by simultaneous hydrolysis of a silica monomer-imprinted molecule complex and tetraethoxysilane (TEOS). A covalent imprinting strategy was employed by generating a sacrificial spacer through the reaction of the isocyanate group of 3-(triethoxysilyl)propyl isocyanate and a phenol moiety of estrone to form a thermally cleavable urethane bond. This allowed us to remove the imprinted estrone by simple thermal reaction and to simultaneously introduce functional groups into the cavity formed by the silica nanotubes. Experiments indicated that estrone could be bound selectively by such an approach and have a binding affinity of 864 +/- 137 (n = 3).

Abstract:Boronic acids are important for effective separation of biological active cis-diols. For the purpose of constructing a new type of saccharide-sensitive material which can not only provide convenient separation but also improve the access of boronic acid to guest molecules, the fluorogenic boronic acid terminated, thermo-sensitive polymers (BA-polyNIPAm) were grafted to an alkyne modified silica gel through the exploitation of click chemistry. The BA-polyNIPAm grafted silica gel (BA-polyNIPAm-SG) was characterized by FT-IR, fluorescence spectra, fluorescence microscopy, elemental analysis (EA), thermal gravimetric analysis (TGA), scanning electron microscope (SEM) and so on. BA-polyNIPAm-SG displayed affinity binding ability for saccharides under physiological pH value and allowed saccharides to be conveniently separated from solution. The maximum binding capacities for fructose and glucose are 83.2 μmol/g and 70.4 μmol/g polymer, respectively. The intensity of fluorescence emission of BA-polyNIPAm-SG increased with the increasing of fructose concentration. The present study provides a new kind of composite material which contains moveable and flexible grippers for recognizing and binding guest molecules.

Abstract:The invention relates to a liquid chromatography stationary phase, with bonded phases of monosaccharide, disaccharide or oligosaccharide (polymerization degree of 3-10); in the structure of the invention, X is -OCH3 or -OCH2CH3 and Y is -OH or -NH2Ac; wherein n is equal to 1-10. The invention is characterized in that an azide group is firstly led in the surface of silica gel to prepare azide group silica gel intermediate; a 1, 3-ring addition reaction of the azid group and the alkyne group which is also the click chemistry is then carried out for 24-72 hours under the temperature of 20-80 DEG C; alkyne group sugar molecule is bonded to the surface of the azid group silica gel. The chromatography separation material provided by the invention has novel structure and high column efficiency and separation selectivity. The stationary phase is used as liquid phase chromatography separation material which has extremely good preservation and separation effects on the strong polar compounds under a hydrophilic chromatography mode, and is especially suitable for separating and analysing the strong polar compounds.

Abstract:A novel silica-based stationary phase with branched octadecyl groups was prepared by the sequential employment of the Michael addition reaction and photoinduced thiol-yne click chemistry with 3-aminopropyl-functionalized silica microspheres as the initial material. The resulting stationary phase denoted as SiO2 -N(C18)4 was characterized by elemental analysis, FTIR spectroscopy and Raman spectroscopy, demonstrating the existence of branched octadecyl groups in silica microspheres. The separations of benzene homologous compounds, acid compounds and amine analogues were conducted, demonstrating mixed-mode separation mechanism on SiO2 -N(C18)4 . Baseline separation of basic drugs mixture was acquired with the mobile phase of acetonitrile/H2 O (5%, v/v). SiO2 -N(C18)4 was further applied to separate Corydalis yanhusuo Wang water extracts, and more baseline separation peaks were obtained for SiO2 -N(C18)4 than those on Atlantis dC18 column. It can be expected that this new silica-based stationary phase will exhibit great potential in the analysis of basic compounds.

Abstract:Two kinds of modified amorphous silica, grafted vinyl trimethoxysilane (AH-171) and N-(n-butyl)-3-aminopropyl trimethoxysilane (DYNASYLAN 1189), were obtained from oil shale residue by the technical process of calcination and activation, alkaline dissolution, homogeneous sedimentation with ethylacetate and modification with silane coupling agents. The number of surface hydroxyl groups, modification degree, the structure and other properties of amorphous silica were investigated by chemical method, infrared analysis, nuclear magnetic resonance analysis, X-ray diffraction and scanning electron microscopy. The results show that amorphous silica treated with coupling agents AH-171 and DYNASYLAN 1189 is characterized by modification degrees of surface hydroxyl groups 93.7% and 71.2% and hydrophobic degrees 98% and 94%, respectively, as well as also by improved dispersion in organic solutions. This work gives a valid method to prepare surface modified amorphous silica from the oil shale residue.
