Influence of Surface Chemistry on the Biological Feature of Giant Cell Tumor of Bone Stromal Cells in Vitro

Abstract:In this study, self-assembled monolayers of alkanethiols on gold (SAMs) with different terminal chemical groups, including methyl (-CH3), amino (-NH2), hydroxyl (-OH) and carboxyl (-COOH) groups, were tested for their effects on behavioral of giant cell tumor of bone stromal cells (GCTSCs) in vitro. Atomic force microscopy confirmed the similar density of different functional surfaces. Fluorescence microscopy, flow cytometry, and scan electron microscopy (SEM) were used to analyse the morphological and biological alterations of GCTSCs. The results showed surface chemistry strongly influenced cell morphology, attachment, proliferation and apoptosis. The GCTSCs cultured on different chemical groups exhibited morphological changes at cellular level, the cells cultured on -CH3 surface exhibited the smallest contact area and rounded morphology, while those on -NH2, -OH and -COOH surfaces had much larger contact area and flatter morphology. After 5 days of culture, the proliferation of GCTSCs on those surfaces decreased in the following sequence: -COOH > -NH2 > -OH >> -CH3. Additionally, -CH3 group exhibited significantly higher toxicity than -NH2, -OH and -COOH groups, and it also showed the ability to inhibit GCTSCs adhesion, growth and promote cell death, providing a new way for design of bone implants to prevent the recurrence of GCT after surgery.
