Abstract:Objective: To develop an appropriate nanocarrier for intratumoral drug delivery,paclitaxel loaded cationic nanoemulsomes(PTX-CTAB-NES) was prepared and investigated in vitro.Methods: PTX-CTAB-NES was prepared by solvent evaporation method.The formulation was optimized by central composite design.The morphology of PTX-CTAB-NES was observed by transmission electron microscopy(TEM).The mean particle size and zeta potential were measured by laser particle size analyzer.The state of drug in nanoemulsomes was confirmed by X-ray diffraction(XRD).Drug loading and encapsulation efficiency were determined by HPLC.Dialysis method at 37 ℃ was employed to investigate the in vitro release of PTX-CTAB-NES.In vitro cytotoxicity of PTX-CTAB-NES was evaluated in MCF-7 cells.Results: The obtained PTX-CTAB-NES was approximately spherical in shape with average particle size of 94.0 nm and zeta potential of +48.3 mV.The drug loading and encapsulation efficiency were 98.0% and 3.25%,respectively.The release of PTX-CTAB-NES was slowed down and the IC50 to MCF-7 human breast carcinoma cells was reduced compared with taxol injection.Conclusion: PTX-CTAB-NES with high positive charge and encapsulation efficiency,slow release as well as advanced cytotoxicity would be a promising nanocarrier for intratumoral drug delivery.

Abstract:To develop an appropriate carrier for intratumoral drug delivery, cetyltrimethylammonium bromide (CTAB) modified nanoemulsome (CTAB-NES) was designed and prepared by solvent evaporation method. Coumarin-6 was chosen as the fluorescent probe and the conventional nanoemulsome (NES) without CTAB modification served as a control. The results demonstrated that CTAB-NES had a smaller particle size of 71.9 +/- 4.32 nm, considerate positive zeta potential of +48.7 +/- 0.2 mV, preferably entrapment efficiency of 97.483 +/- 0.693% and the release of coumarin-6 in 24 h was little. The in vitro cytotoxicity of CTAB-NES to the CHO cells and MCF-7 cells increased consistently with concentrations and was higher than that of NES, especially to the cancer cells. Both the fluorescence microscopy images and HPLC assay verified that the cellular uptake of CTAB-NES in MCF-7 cells was much higher than that of NES, and the uptake was time-, concentration- and temperature- dependent. The uptake mechanism results demonstrated that the internalization of CTAB-NES and NES involved clathrin- and caveolae-mediated endocytosis while macropinocytosis only influenced the uptake of CTAB-NES in MCF-7 cells for CTAB could mediate adsorptive pinocytosis. Thus, CTAB-NES with high positive charge and good intracellular uptake ability could be a promising drug carrier for intratumoral drug delivery.

Abstract:Polymeric micelles hold great promise in nanocarrier delivery systems for anticancer drugs. A mixed micelle composed of deoxycholic acid-modified chitooligosaccharide (COS-DOCA) and methoxy poly(ethylene glycol)-polylactide copolymer (mPEG-PDLLA) was designed and synthesized for encapsulation of paclitaxel. The PTX-loaded mixed micelles have high drug-loading efficiency and encapsulation efficiency as well as small size (∼40nm) and narrow size distribution. The PTX-loaded COS-DOCA/mPEG-PDLLA mixed micelles showed the enhanced antitumor efficacy in A549 lung xenograft model.
An Optimized Two-Vial Formulation Lipid Nanoemulsion of Paclitaxel for Targeted Delivery to Tumor.

Abstract:The discovery of new intravenous drug delivery carrier for water-insoluble drug is a challenging task. In this paper, novel two-vial formulation of paclitaxel (PTX)-loaded lipid nanoemulsions (TPLEs) with particle sizes of 110nm (TPLE-1), 220nm (TPLE-2) and 380nm (TPLE-3), which were formed by mixing a PEG400 solution of PTX and 10% (w/w) blank lipid emulsions (BLEs) with different particle size prior to use, were developed and comparatively evaluated for their pharmaceutics, pharmacokinetics, biodistribution, in vitro and in vivo anticancer efficiency. Among them, TPLE-1 displayed higher PTX-loading, slower PTX-release and larger PTX-distribution in oil-phase, significantly reduced extraction by RES organs, increased tumor-uptake, showed stronger cytotoxicity against MCF-7 cells and more potent anticancer efficacy on MCF-7 tumor-bearing nude mice, and had greater plasma AUC0-∞ value, smaller plasma clearance (CL), longer mean residence time (MRT) and elimination half-life (T1/2) in SD rats. It also exhibited the same in vivo efficacy as Taxol® and even produced less hemolysis and intravenous irritation. Moreover, its LD50 was 4.3-fold higher than that of Taxol®. All results demonstrate that TPLE-1 is a promising candidate drug due to its high tumor-accumulation and effectiveness, low toxicity, good safety and druggability in clinical application for the cancer therapy.

Abstract:Aim:To prepare paclitaxel-loaded cationic chitosan micelles(PTX-CCM) and to study the biodistribution of PTX-CCM and paclitaxel injection(PTX-INJ) in mice.Methods:PTX-CCM was prepared by dialysis.PTX-CCM and PTX-INJ were administered to mice through caudal veins at a dose of 20 mg/kg.The RP-HPLC method was established to determine the PTX levels in the plasma and other tissues of mice.The tissues distribution and targeting effenciency were evaluated by pharmacokinetic parameters(AUC,MRT) and targeting parameters(Re,Ce and Te).Results:The drug loading and drug encapsulation of PTX-CCM were 26.40% and 76.23%,respectively,and the diameters and Zeta potential were 164.2 nm and +23.7 mV,respectively.In comparison to PTX-INJ group,the AUCs of PTX in the liver,spleen and lung had risen 5.65,91.20 and 2.53 times respectively in PTX-CCM group.Meanwhile,the MRT were 1.83,8.40 and 1.16 times of those of control group respectively.The relative exposure(Re) and ratio of maximum concentration(Ce) for PTX-CCM in these tissues were far more than 1 and targeting efficiecy(Te) was much higher than that of control group,while PTX in the heart and kidney decreased significantly and AUC were 61.00% and 89.07% of those of control group,meanwhile,c_(max) were 52.62% and 52.94% of those of control group respectively.Conclusion:PTX-CCM shows excellent drug loading capabilities with a amount of cationic charges on their surface.It showsd a higher targeting efficiency in the liver,spleen and lung in vivo and accumulates in the tissues for relatively longer time while the levels of PTX-CCM in the heart and kidney tissues are significantly reduced which might improve the treatment efficacy of PTX and decrease the side effects.

Abstract:Paclitaxel (PTX)-loaded poly(N-vinylpyrrolidone)-b-poly(ε-caprolactone) (PVP-b-PCL) nanoparticles with high drug payload were successfully prepared by a modified nano-precipitation method and characterized by transmission electron microscopy (TEM), atomic force microscopy (AFM), dynamic light scattering (DLS) and zeta potential. The satisfactory drug loading content (>25%) and high encapsulation efficiency (>85%) were achieved. The in vivo real-time biodistribution of PTX-loaded nanoparticles was investigated using near-infrared fluorescence (NIRF) imaging. The antitumor effect of PTX-loaded nanoparticles was evaluated, both, in vitro on three different cancer cell lines and in vivo on hepatic H22 tumor bearing mice model via intravenous administration (i.v.). It is found that PTX-loaded nanoparticles exhibit significant superior in vivo antitumor effect than the commercially available Taxol® formulation by combining the tumor volumes and survival rates measurement, intravital positron emission tomography and computed tomography (PET/CT) imaging.
In vivo evaluation of novel chitosan graft polymeric micelles for delivery of paclitaxel.

Abstract:A water-insoluble anti-tumor agent, Paclitaxel (PTX) was successfully incorporated into polymeric micelles formed from new graft copolymers, N-octyl-N-(2-carboxyl-cyclohexamethenyl) chitosan derivatives(OCCC). The objective of this study was to optimize and characterize the novel PTX micelle (PTX-M). Furthermore, the pharmacokinetics, NIR-imaging, biodistribution, and anti-tumor effects of PTX-M were evaluated. The results showed that the PTX-M had high drug loading (43.25%). The V(d) and t(1/2 beta) of PTX-M were increased by 11.4- and 2.83-fold, respectively, but the plasma AUC of PTX-M was 3.8-fold lower than that of Taxol. Biodistribution study indicated that PTX-M was widely distributed into most tissues, most of them found in liver, spleen, lung, and kidney. This result was similar with NIR(near-infrared)-imaging. Tumor growth was significantly inhibited after intravenous injection of PTX-M. PTX-M showed the enhanced anti-tumor effect and non-toxic effects compared with Taxol, the commercial product of PTX. Therefore, the chitosan-derived micelle system offered a stable and effective platform for cancer chemotherapy with PTX.
Preparation and characterization of paclitaxel palmitate albumin nanoparticles with high loading efficacy: an in vitro and in vivo anti-tumor study in mouse models

Abstract:Background: Combination of the prodrug technique with an albumin nano drug-loaded system is a novel promising approach for cancer treatment. However, the long-lasting and far-reaching challenge for the treatment of cancers lies in how to construct the albumin nanometer drug delivery system with lead compounds and their derivatives. Methods: In this study, we reported the preparation of injectable albumin nanoparticles (NPs) with a high and quantitative drug loading system based on the NabTM technology of paclitaxel palmitate (PTX-PA). Results: Our experimental study on drug tissue distribution in vivo demonstrated that the paclitaxel palmitate albumin nanoparticles (Nab-PTX-PA) remained in the tumor for a longer time post-injection. Compared with saline and paclitaxel albumin nanoparticles (Abraxane®), intravenous injection of Nab-PTX-PA not only reduced the toxicity of the drug in normal organs, and increased the body weight of the animals but maintained sustained release of paclitaxel (PTX) in the tumor, thereby displaying an excellent antitumor activity. Blood routine analysis showed that Nab-PTX-PA had fewer adverse effects or less toxicity to the normal organs, and it inhibited tumor cell proliferation more effectively as compared with commercial paclitaxel albumin nanoparticles. Conclusions: This carrier strategy for small molecule drugs is based on naturally evolved interactions between long-chain fatty acids (LCFAs) and Human Serum Albumin (HSA), demonstrated here for PTX. Nab-PTX-PA shows higher antitumor efficacy in vivo in breast cancer models. On the whole, this novel injectable Nab-PTX-PA has great potential as an effective drug delivery system in the treatment of breast cancer.
A Self-Assembling Nanoparticle for Paclitaxel Delivery in Ovarian Cancer

Abstract:Paclitaxel (PTX) is one of the most effective chemotherapeutic drugs for the treatment of a variety of cancers. However, it is associated with serious side effects caused by PTX itself and the Cremophor EL emulsifier. In the present study, we report the development of a well-defined amphiphilic linear-dendritic copolymer (named as telodendrimer) composed of polyethylene glycol (PEG), cholic acid (CA, a facial amphiphilic molecule) and lysine, which can form drug-loaded core/shell micelles when mixed with hydrophobic drug, such as PTX, under aqueous condition. We have used PEG(5k)-CA(8), a representive telodendrimer, to prepare paclitaxel-loaded nanoparticles (PTX-PEG(5k)-CA(8) NPs) with high loading capacity (7.3 mg PTX/mL) and a size of 20-60 nm. This novel nanoformulation of PTX was found to exhibit similar in vitro cytotoxic activity against ovarian cancer cells as the free drug (Taxol) or paclitaxel/human serum albumin nanoaggregate (Abraxane). The maximum tolerated doses (MTDs) of PTX-PEG(5k)-CA(8) NPs after single dose and five consecutive daily doses in mice were approximately 75 and 45 mg PTX/kg, respectively, which were 2.5-fold higher than those of Taxol. In both subcutaneous and orthotopic intraperitoneal murine models of ovarian cancer, PTX-PEG(5k)-CA(8) NPs achieved superior toxicity profiles and anti-tumor effects compared to Taxol and Abraxane at equivalent PTX doses, which were attributed to their preferential tumor accumulation, and deep penetration into tumor tissue, as confirmed by near infrared fluorescence (NIRF) imaging.

Abstract:Recently, nanomedicine without drug carriers has attracted many pharmacists' attention. A novel paclitaxel-s-s-paclitaxel (PTX-s-s-PTX) conjugate with high drug loading (∼78%, w/w) was synthesized by conjugating paclitaxel to paclitaxel by using disulfide linkage. The conjugate could self-assemble into uniform nanoparticles (NPs) with 1,1-dioctadecyl-3,3,3,3-tetramethylindotricarbocyanine iodide (DiR) encapsulated within the core of PTX-s-s-PTX NPs for photothermal therapy (PTT). The DiR-loaded self-assembled nanoparticles (DSNs) had a mean diameter of about 150 nm and high stability in biological condition. A disulfide bond is utilized as a redox-responsive linkage to facilitate a rapid release of paclitaxel in tumor cells. DSNs indicated significant cytotoxicity as a result of the synergetic chemo-thermal therapy. DSNs were featured with excellent advantages, including high drug loading, redox-responsive releasing behavior of paclitaxel, capability of loading with photothermal agents, and combinational therapy with PTT. In such a potent nanosystem, prodrug and photothermal strategy are integrated into one system to facilitate the therapy efficiency.
Paclitaxel Prodrug Nanomedicine for Potential CT-imaging Guided Breast Cancer Therapy

Abstract:Nanotheranostics, which combine the therapeutic and diagnostic functions in one integrated system, have received extensive attentions in cancer treatments because they enable non-invasive diagnosis, tumor-targeted drug delivery, and real-time monitoring of therapeutic response. However, due to the high systemic toxicity of commonly used chemotherapeutics, current treatment still has limitations. Herein, to simultaneously achieve safe cancer therapy and therapeutic response monitoring, an iodinated prodrug strategy was proposed. 2,3,5-Triiodobenzoic acid (TIBA) was used to modify both paclitaxel (PTX) and the polymeric vehicle, so that the encapsulation efficiency of PTX could be increased and the systemic toxicity could be reduced. As-prepared prodrug nanoparticles could accumulate passively in the tumor site and promptly release loaded drugs in response to the overexpressed GSH in cancer cells, which then caused efficient cell cycle arrest and apoptosis like that of the parent PTX. With this rational design, safe and efficient antitumor therapy and real-time computer tomography (CT) imaging could be simultaneously realized, facilitating potential CT imaging-guided therapy of metastatic breast cancer.
The Preparation, Characterization, and Pharmacokinetic Studies of Chitosan Nanoparticles Loaded with Paclitaxel/Dimethyl-Beta-Cyclodextrin Inclusion Complexes

Abstract:A novel biocompatible and biodegradable drug-delivery nanoparticle (NP) has been developed to minimize the severe side effects of the poorly water-soluble anticancer drug paclitaxel (PTX) for clinical use. PTX was loaded into the hydrophobic cavity of a hydrophilic cyclodextrin derivative, heptakis (2,6-di-O-methyl)-β-cyclodextrin (DM-β-CD), using an aqueous solution-stirring method followed by lyophilization. The resulting PTX/DM-β-CD inclusion complex dramatically enhanced the solubility of PTX in water and was directly incorporated into chitosan (CS) to form NPs (with a size of 323.9-407.8 nm in diameter) using an ionic gelation method. The formed NPs had a zeta potential of +15.9-23.3 mV and showed high colloidal stability. With the same weight ratio of PTX to CS of 0.7, the loading efficiency of the PTX/DM-β-CD inclusion complex-loaded CS NPs was 30.3-fold higher than that of the PTX-loaded CS NPs. Moreover, it is notable that PTX was released from the DM-β-CD/CS NPs in a sustained-release manner. The pharmacokinetic studies revealed that, compared with reference formulation (Taxol(®)), the PTX/DM-β-CD inclusion complex-loaded CS NPs exhibited a significant increase in AUC(0→24h) (the area under the plasma drug concentration-time curve over the period of 24 hours) and mean residence time by 2.7-fold and 1.4-fold, respectively. Therefore, the novel drug/DM-β-CD inclusion complex-loaded CS NPs have promising applications for the significantly improved delivery and controlled release of the poorly water-soluble drug PTX or its derivatives, thus possibly leading to enhanced therapeutic efficacy and less severe side effects.

Abstract:Polymeric nanoparticles (NPs) have great potential application in achieving targeted delivery of anticancer drugs. Paclitaxel (PTX) loaded NPs were developed using biodegradable methoxy poly (ethylene glycol)-poly (ε-caprolactone) (MPEG-PCL) diblock copolymer by solid dispersion technique without toxic organic solvent. The lyophilized powder has been stored at room temperature for more than six months and still unchanged. PTX-loaded MPEG-PCL nanoparticles (PTX-NPs) displayed that the highest drug loading of PTX was about 25.6% and entrapment efficiency was over 98%, and the optimized average diameter and polydispersity index (PDI) were about 27.6 ± 0.1 nm and 0.05, respectively. Moreover, experimental results shown PTX-NPs had sustained-release effects and its curve fitting followed the Higuchi model. The maximum tolerated dose (MTD) of PTX-NPs after single dose in Balb/c mice was above 80 mg PTX/kg body weight (b.w), which was 2.6-fold higher than that of Taxol(®) (30 mg PTX/kg b.w). The levels of PTX administrated PTX-NPs had obvious distinction to Taxol(®) in plasma, liver, spleen, kidneys, lungs, heart and tumor. Especially, the concentration of PTX in tumor administrated PTX-NPs was higher than administration of Taxol(®). All results suggested that we had contrived a simple, biodegradable, effective and controllable drug delivery system for paclitaxel.

Current Cancer Drug Targets

Abstract:BACKGROUND:Improving poorly soluble drugs into druggability was a major problem faced by pharmaceutists. Nanosuspension can improve the druggability of insoluble drugs by improving the solubility, chemical stability and reducing the use of additives, which provided a new approach for the development and application of the insoluble drugs formulation. Paclitaxel (PTX) is a well-known BCS class IV drug with poor solubility and permeability. Also, many studies have proved that paclitaxel is a substrate of the membrane-bound drug efflux pump P-glycoprotein (P-gp), therefore it often shows limited efficacy against the resistant tumors and oral absorption or uptake. OBJECTIVE:To manufacture an enhanced-penetration PTX nanosuspension (PTX-Nanos), and evaluate the physicochemical property, pharmacokinetics and tissue distribution in vivo and cytotoxic effect in vitro. METHODS:PTX-Nanos were prepared by microprecipitation-high pressure homogenization, with a good biocompatibility amphiphilic block copolymer poly(L-phenylalanine)-b-poly(L-aspartic acid) (PPA-PAA) as stabilizer. RESULTS:The PTX-Nanos had a sustained-dissolution manner and could effectively reduce plasma peak concentration and extend plasma circulating time as compared to PTX injection, markedly passively targeting the MPS-related organs, such as liver and spleen. This unique property might enhance treatment of cancer in these tissues and reduce the side effects in other normal tissues. Moreover, the hybrid stabilizers could enhance penetration of PTX in PTX-Nanos to multidrug resistance cells. CONCLUSION:To sum up, our results showed that the optimal formula could improve the solubility of PTX and the stability of the product. The PTX-Nanos developed in this research would be a promising delivery platform in cancer treatment.

Abstract:In this study, the cyclodextrin polypeptide (R8-CMβCD) was successfully synthesized by the conjugation of a cell-penetrating peptide (R8) with carboxymethyl-β-cyclodextrin (CMβCD) via the carbon diamine reaction. Then, paclitaxel-loaded nanoparticles (PTX@R8-CMβCD NPs) was prepared. Results of transmission electron microscopy (TEM) showed that PTX@R8-CMβCD NPs were spherical with smooth surfaces and an average diameter about 144 nm. The amount of PTX released from NPs was less than 20% at pH7.4, but it increased significantly to 80% in the weakly acidic cytoplasm of tumors (pH5.0). Furthermore, PTX@R8-CMβCD NPs promoted the cellular uptake of PTX. Further studies on the mechanism showed that cellular uptake of PTX@R8-CMβCD NPs could rely on multiple pathways. In addition, the NPs had the ability to inhibit P-gp efflux pumps. Cytotoxicity tests showed that the NPs had no side effects. Taken together, PTX@R8-CMβCD NPs is an effective anticancer drug delivery system, and the material (R8-CMβCD) may be a promising anti-cancer drug carrier.
Construction of glutathione-responsive paclitaxel prodrug nanoparticles for image-guided targeted delivery and breast cancer therapy

Abstract:Paclitaxel (PTX) remains an essential drug in the treatment of breast cancer. To improve metabolic stability and real-time monitoring of drug location, we develop a visualized nano-prodrug. Novel hyaluronic acid (HA)-coated glutathione (GSH)-sensitive chitosan (CS)-based nano-prodrug (HA/TPE-CS-SS-PTX NPs) with aggregation-induced emission effects (AIE) were accomplished. The prodrug NPs (drug loading 29.32%, particle size 105 nm, regular sphericity) exhibit excellent fluorescence stability. The prodrug NPs could target tumor cells with high expression of CD44 and decompose in the presence of high concentrations of glutathione. In vitro evaluations revealed that the prodrug NPs have significant cytotoxicity on 4T1 cells, and due to their excellent AIE characteristics, their position in cells can be tracked. Moreover, the prodrug NPs also shown superior anti-tumor effects in vivo experimental. Overall, the HA/TPE-CS-SS-PTX NPs we constructed have excellent bio-imaging capabilities and can be served as a potential nanomedicine for PTX delivery against breast cancer.

Superior Intratumoral Penetration of Paclitaxel Nanodots Strengthens Tumor Restriction and Metastasis Prevention

Abstract:Recently discovered intratumoral diffusion resistance, together with poor solubility and nontargeted distribution of chemotherapeutic drugs, has significantly impaired the performance of cancer treatments. By developing a well-designed droplet-confined/cryodesiccation-driven crystallization approach, we herein report the successful preparation of nanocrystallites of insoluble chemotherapeutic drug paclitaxel (PTX) in forms of nanodots (NDs, ≈10 nm) and nanoparticles (NPs, ≈70 nm) with considerably high drug loading capacity. Superficially coated Pluronic F127 is demonstrated to endow the both PTX nanocrystallites with excellent water solubility and prevent undesired phagocyte uptake. Further decoration with tumor-penetrating peptide iRGD, as expected, indiscriminatively facilitates tumor cell uptake in traditional monolayer cell culture model. On the contrary, distinctly enhanced performances in inward penetration and ensuing elimination of 3D multicellular tumor spheroids are achieved by iRGD-NDs rather than iRGD-NPs, revealing the significant influence of particle size variation in nanoscale. In vivo experiments verify that, although efficient tumor enrichment is achieved by all nanocrystallites, only the iRGD-grafted nanocrystallites of ultranano size realize thorough intratumoral delivery and reach cancer stem cells, which are concealed inside the tumor core. Consequently, much strengthened restriction on progress and metastasis of orthotopic 4T1 mammary adenocarcinoma is achieved in murine model, in sharp contrast to commercial PTX formulation Taxol.

Abstract:Objective: The purpose of this work was to prepare a stable paclitaxel nanosuspension and test it for potential use as a targeted chemotherapeutic. Different particle coatings were employed to assess their impact on cellular uptake in vitro. In vivo work was then performed to demonstrate efficacy in tumor-bearing mouse models. Materials and method: Paclitaxel nanosuspensions were prepared using a homogenization process and coated with excipients. Surface charge was measured by zeta potential, potency by high-performance liquid chromatography, and solubility using an in-line UV probe. Cellular uptake studies were performed via flow cytometry. In vivo experiments were performed to determine residence time, maximum tolerated dose, and the efficacy of paclitaxel nanosuspensions (Paclitaxel-NS). Results: A stable paclitaxel nanosuspension was prepared and coated with various excipients. Studies in mice showed that the nanosuspension was well-tolerated and at least as effective as the IV Taxol control in prolonging mouse survival in a head and neck cancer model as well as an ovarian cancer model with a lower overall drug dose than the traditional IV administration route. Conclusions: The paclitaxel nanosuspension is suitable for cellular uptake. The nanosuspension was effective in prolonging life in two separate xenograft orthotopic murine cancer models through two separate routes of administration.

Abstract:Paclitaxel (PTX), one of the most potent anticancer agents, has showed a remarkable activity against varieties of tumors. However, the bioavailability of PTX is quite low due to its poor aqueous solubility. Moreover, the emerging multidrug resistance (MDR) in cancer to PTX remains a major obstacle for successful chemotherapy. In order to address these problems, we developed self-assembly of biotinylated poly(ethylene glycol)-poly(curcumin) (Biotin-PEG-PCDA) for PTX delivery (termed as PTX-BPC NPs) with the application of mPEG2K-P(CL-co-LLA) as an emulsifier. The loading content and encapsulation efficiency of PTX were 13.2% and 92.0%, respectively. In vitro drug release study showed that PTX-BPC NPs could degrade rapidly and then release the PTX payload in a 10 mM glutathione (GSH) environment. Compared with free PTX, PTX-BPC NPs exhibited enhanced anticancer efficacy (IC50(MCF-7/ADR cells), 17.28 µg/mL vs. 1.15 µg/mL). In addition, these biotin-modified nanoparticles could also significantly reverse PTX resistance by suppressing the over-expression of P-gp, thus resulting in increased intracellular drug accumulation and reduced drug efflux in MCF-7/ADR cells, which showed a great anticancer effect.

Abstract:To ensure the delivery of antitumor drugs to tumor site and quick release in tumor cells,we designed and prepared pH-sensitive polymeric micelles by combining cationic ring-opening polymerization of 2-ethyl-2-oxazoline (EOz) with vitamin E succinate (VES),and then encapsulating paclitaxel (PTX) into the micelles self-assembled by poly(2-ethyl-2-oxazoline)-vitamin E succinate (PEOz-VES).The structure of the synthesized PEOz-VES was confirmed by 1H NMR spectrum,and the molecular weight measured by GPC was 1212 g/mol.The pKa of PEOz-VES with a low critical micelle concentration of (5.84±0.02) mg/Lwas determined to be 6.01.The PTX-loaded PEOz-VES polymeric micelles prepared by film hydration method were characterized to have a nanoscaled size of about 30 nm in diameter,a positive Zeta potential of 4.86 mV and uniform spherical morphology by TEM observation.The drug loading content and encapsulation efficiency were (2.63±0.16)％ and (84.1+3.38)％,respectively.The in vitro release behavior of PTX from PEOz-VES rnicelles in PBS displayed pH-dependent pattern and was gradually accelerated with decrease of pH value,implying that the micelles could distinguish endo/lysosomal pH and tumor extracellular pH from physiological pH by accelerating drug release.Therefore,the designed PEOz-VES micelles might have significant promise for anti-cancer drug delivery.
