Abstract:In recent years, near-infrared (NIR) spectroscopy has gained wide acceptance in different fields by its advantages over other analytical techniques. NIR techniques have the ability to record spectrum for solid and liquid samples with rapid speed and non-destruction. This article discusses the features and development of NIR spectroscopy in the last few years; the analysis procedure and some mathematical tools to acquire specific information; some NIR techniques and new equipment that adopted in agriculture fields; the application of NIR in quality inspection of agricultural products, such as cereal, fruits, vegetables, and animal products; the application prospect of NIR spectroscopy in modern agriculture. As a rapid-response analytical tool, NIR spectroscopy may plays an important role in quality inspection of agricultural products.

Abstract:There is a lack of methods to analyze large numbers of traditional Chinese medicine (TCM) samples quickly and efficiently. Here we present a near infrared (NIR) spectroscopy-based method for the rapid analysis of Honeysuckle extracts. NIR spectra of 124 Honeysuckle extract samples produced within a four-year period were collected in the diffuse reflection mode and pretreated with multiplicative scatter correction (MSC), smoothing and derivative. Using partial least squares regression (PLSR), including PLS1 and PLS2, we established and validated the calibration models of six organic acids. Our results showed that both accuracy and precision of the NIR spectroscopy-based method meet the requirements of a traditional Chinese medicine (TCM) factory. This new method has the potential to save time, efforts and cost associated with the analysis of large quantities of unknown samples and will be particularly applicable to TCM factories for cost reduction.
A Study on the Use of Near-Infrared Spectroscopy for the Rapid Quantification of Major Compounds in Tanreqing Injection

Abstract:In this paper we describe the strategy used in the development and validation of a near infrared spectroscopy method for the rapid determination of baicalin, chlorogenic acid, ursodeoxycholic acid (UDCA), chenodeoxycholic acid (CDCA), and the total solid contents (TSCs) in the Tanreqing injection. To increase the representativeness of calibration sample set, a concentrating-diluting method was adopted to artificially prepare samples. Partial least square regression (PLSR) was used to establish calibration models, with which the five quality indicators can be determined with satisfied accuracy and repeatability. In addition, the slope/bias (S/B) method was used for the models transfer between two different types of NIR instruments from the same manufacturer, which is contributing to enlarge the application range of the established models. With the presented method, a great deal of time, effort and money can be saved when large amounts of Tanreqing injection samples need to be analyzed in a relatively short period of time, which is of great significance to the traditional Chinese medicine (TCM) industries.

Abstract:A method for rapid quantitative analysis of four kinds of Tanreqing injection intermediates was developed based on Fourier transform near infrared (FT-NIR) spectroscopy and partial least squares (PLS) algorithm. The NIR spectra of 120 samples were collected in transflective mode. The concentrations of chlorogenic acid, caffeic acid, luteoloside, baicalin, ursodesoxycholic acid (UDCA), and chenodeoxycholic acid (CDCA) were determined with the HPLC–DAD/ELSD as reference method. In the PLS calibration, the NIR spectra were pretreated with different methods and the number of PLS factors used in the model calibration was optimized by leave-one-out cross-validation. The performance of the final PLS models was evaluated according to the root mean square error of calibration (RMSEC), root mean square error of cross-validation (RMSECV), root mean square error of prediction (RMSEP), BIAS, standard error of prediction (SEP), and correlation coefficients (R). The R values in the prediction sets were all higher than 0.93, and the SEPs for the 6 compounds are 1.18, 6.02, 2.71, 155, 126, 30.0mg/l, respectively. The established models were used for the liquid preparation process analysis of Tanreqing injection in three batches, and a model updating method was proposed for the long-term usage of the established models. This work demonstrated that NIR spectroscopy is more rapid and convenient than the conventional methods to analyze the intermediates of Tanreqing injection, and the presented method is helpful to the implementation of process analytical technology (PAT) in pharmaceutical industry of Chinese Medicines Injections.
Rapid monitoring approaches for concentration process of lanqin oral solution by near-infrared spectroscopy and chemometric models

Abstract:Qualitative and quantitative detection methods based on near-infrared spectroscopy (NIRs) have been proposed in the process analysis of traditional Chinese medicine in recent years. In this study, rapid monitoring methods were developed for quality control of concentration process of lanqin oral solution (LOS). Partial least squares regression (PLSR) method was adopted to construct quantitative models for epigoitrin, geniposide, baicalin, berberine hydrochloride and density. Simultaneously, the genetic algorithm joint extreme learning machine (GA-ELM) was first applied in qualitative analysis of NIRs to distinguish end point of concentration process. Results of PLSR models were satisfactory with the relative standard error of calibration valued at 3.80%, 3.75%, 3.79%, 11.5% and 1.22% for epigoitrin, geniposide, baicalin, berberine hydrochloride and density respectively, and the residual predictive deviation values were higher than 3. For qualitative analysis, the GA-ELM model obtained 100% prediction accuracy. The PLSR quantitative models and the end point discrimination model constructed by GA-ELM correspond with the requirements of practical applications. The results indicate that NIRs in combination with chemometrics has great potential in improving the efficiency in production. (c) 2020 Elsevier B.V. All rights reserved.
A Feasibility Study on the Non-Invasive Analysis of Bottled Compound E Jiao Oral Liquid Using Near Infrared Spectroscopy

Abstract:The aim of this study was to explore the feasibility of near infrared (NIR) spectroscopy for the quantitative analysis of Compound E Jiao oral liquid, a popular oral TCM preparation, through intact glass vials. Transmission spectra were acquired with a sample holder tailored to the instrument's sample compartment. Total flavonoids, total saponins and soluble solid contents were considered as quality indicators of this oral preparation and calibration models were built using partial least-squares regression (PLSR) algorithm. The sample set was comprised of production samples as well as laboratory samples prepared by a concentrating-diluting method so as to extend the concentration range beyond the production variation. The developed models gave root mean square error of prediction (RMSEP) of 0.084 mg mL(-1) for total flavonoids, 0.014 mg mL(-1) for total saponins and 0.21% for soluble solid contents. To better evaluate the prediction performance, an independent set of ten production samples was introduced for the external validation. The models are compared with those built with the transflectance spectra collected in the destructive way. The results of paired t-test and F-test at a significance level of 0.05 revealed that the two sets of models exhibited comparable model performance. The overall results demonstrate that the proposed non-destructive method can be used as an advantageous alternative in the quality inspection of Compound E Jiao oral liquid production. Abstract:Near infrared spectroscopy combined with chemometrics was investigated for the fast determination of total organic carbon (TOC) and soluble solids contents (SSC) of Tanreqing injection intermediates. The NIR spectra were collected in transflective mode, and the TOC and SSC reference values were determined with Multi N/C UV HS analyzer and loss on drying method. The samples were divided into calibration sets and validation sets using the Kennard–Stone (KS) algorithm. The Dixon test, leverage and studentized residual test were studied for the sample outlier analysis. The selection of wavebands, spectra pretreated method and the number of latent variables were optimized to obtain better results. The quantitative calibration models were established with 3 different PLS regression algorithms, named linear PLS, non-linear PLS and concentration weighted PLS, and the net result was defined as the average of the predicted values of the different calibration models. The overall results indicated that the presented method is more powerful than single multivariable regression method, characterized by higher mean recovery rate (MRR) of the validation set, and can be used for the rapid determination of TOC and SSC values of Tanreqing injection intermediates, which are two important quality indicators for the process monitoring.

Abstract:Due to the advantages of fast and noninvasive, requiring almost no sample preparation, supplying chemical and physical information, near infrared (NIR) spectroscopy is widely used in the pharmaceutical industry. In the life cycle of an established analytical method, rigorous validation should be performed before routine use. However,there is no detailed experimental standards or decision rules for the NIR methods validation currently, and various approaches were applied to validate the established NIR quantification models in literatures.In this paper, progress on the validation of NIR methods developed for the pharmaceutical application was reviewed, concentrating on the standard documents and main ideas of validation.It will provide references for the standardized application of NIR spectroscopy in the pharmaceutical industries.

Abstract:The industry of traditional Chinese medicine (TCM) encounters problems like quality fluctuation of raw materials and unstandardized production process.Near infrared (NIR) spectroscopy technology is widely used in quality control of TCM because of its abundant information, fast and nondestructive characters.The main applications include quantitative analysis of Chinese medicinal materials, intermediates and Chinese patent medicines;the authenticity of TCM, species, origins and manufacturers;monitoring and control of the extraction, alcohol precipitation, column chromatography and blending process.This article reviews the progress on the application of NIR spectroscopy technology in TCM field.In view of the problems existing in the application, the article proposes that the standardization of NIR analysis method should be developed according to specific characteristics of TCM, which will promote the application of NIR technology in the TCM industry.

Abstract:The application of near-infrared (NIR) spectroscopy for on-line monitoring of the extraction process of red paeony root was investigated. For NIR measurements, two fiber optic probes designed to transmit NIR radiation through a 2mm pathlength flow cell were employed to collect spectra in real-time. Partial least squares regression (PLSR) calibration models were developed for the parameters of interest: total solid, paeoniflorin, and benzoic acid. The influences of flow rate and air bubble on the NIR spectra and calibration models were also investigated. The established models were used for on-line and real-time monitoring of extraction process, and a model updating method was proposed for the long-term usage of the developed models. Furthermore, both the moving block of standard deviation (MBSD) and relative concentration changing rate (RCCR) methods were used to identify the end point of extraction process. The results of this particular application of implementing NIR spectroscopy to monitor extraction process are very encouraging. Successful models have been built and applied on-line, which proffers real-time data and instant feedback about the extraction course, and in turn, provides improved control.

Abstract:In this study, the application of near-infrared (NIR) spectroscopy for online monitoring of the Moluodan extraction process was investigated. Paeoniflorin, the main active component of Moluodan, was chosen as the quality index. Samples were partitioned into calibration and validation sets by random select and set partitioning based on joint X-Y distances algorithm (SPXY), respectively. Wavelengths for modeling were selected by manual method and competitive adaptive reweighted sampling (Cars) algorithm. Particle swarm optimizationbased least square support vector machines (PSO-LS-SVM) and partial least squares models were both established for quantitative analysis to determine the content of paeoniflorin online. At last, 8 models were obtained according to the combination of these algorithms and the SPXY-Cars-PSO-LS-SVM model had the best quantitative analysis performance compared with the other 7 models. Specifically, in the developed SPXY-Cars-PSO-LS-SVM model, the determination coefficients of the calibration R2c and validation R2v sets were 0.99 and 0.95, respectively, and the root mean square errors of the calibration and validation sets were 0.012 and 0.024 mg/ mL, respectively, and the relative standard errors of the calibration and validation sets were 2.84% and 6.34%, respectively. These results suggested that the appropriate sample partition, wavelength selection, and regression analysis methods in this study, namely, the SPXY-Cars-PSO-LS-SVM algorithm combined with NIR spectroscopy, could be an effective and real-time approach for online monitoring of the extraction process of Moluodan.

Abstract:In recent years, the near infrared (NIR) spectroscopy has gained wide acceptance within the quantitative analysis of traditional Chinese medicine (TCM). However, the lack of technical standards is the bottleneck problem in this process. To address this issue, standardization study of the NIR spectroscopy based method for the quantitative analysis of TCM is needed, in which the specific characteristics of TCM should be given full considerations. The main research contents include:the scope definition for the application of NIR spectroscopy in the TCM quantitative analysis field, the selection criteria for the sample pretreatment and spectral acquisition conditions, the rules for the model optimization and evaluation, and the regulations for the model update and transfer. In this paper, some foreign studies in the agricultural areas are reviewed for reference. Different chemometrics methods reported in the literature are investigated and compared systematically. This research is important actual significance to the theoretical development of NIR spectroscopy analytical techniques, and will effectively promote the application of the technology in the TCM industry. Furthermore, it is beneficial to improve the technical level of TCM quality control, and can also be used as references to achieve similar purposes for other natural products.
On-line Monitoring of Traditional Chinese Medicinal Powder Blending Process by Using Near-Infrared Spectroscopy

Abstract:OBJECTIVE To establish a rapid method based on near-infrared diffuse reflectance spectroscopy(NIR) to monitor the blending process of traditional Chinese medicinal powder on-line.METHODS NIR spectra were collected during the blending processes of mixtures consisting of Red Peony Root powder and defatted Spine Date Seed powder.Multivariate calibration models based on PLS regression were developed to correlate the collected spectra with the content of Red Peony Root powder.The optimal calibration model was applied to monitor the blending process in real time with noninterference.RESULTS After spectral pretreatment using multiplicative signal correction(MSC),first derivative and Savitzky-Golay filter,Red Peony Root powder contents were quantified using PLS algorithm in the spectral ranges of 5 900-5 950 cm-1,6 700-6 750 cm-1 and 6 950-7 100 cm-1.The correlation coefficient of the calibration model was 0.999 0.The RMSEC,RMSEP and RMSECV were 1.14,1.16 and 1.26,respectively.CONCLUSION It was demonstrated that a miniature near-infrared analyzer could be used to monitor the contents of the constituents and the blending homogeneity of traditional Chinese medicinal powder in real time,and to rapidly determine the end-point of blending process.In addition,it can be used to judge whether the physical state of powder mixture conforms to the standard.Therefore,NIR spectroscopy is considered to be an appropriate PAT tool for end-point control of blending process of traditional Chinese medicinal powder.
Application of Near Infrared Spectroscopy Combined with Particle Swarm Optimization Based Least Square Support Vactor Machine to Rapid Quantitative Analysis of Corni Fructus

Abstract:A novel method was developed for the rapid determination of multi-indicators in corni fructus by means of near infrared (NIR) spectroscopy. Particle swarm optimization (PSO) based least squares support vector machine was investigated to increase the levels of quality control. The calibration models of moisture, extractum, morroniside and loganin were established using the PSO-LS-SVM algorithm. The performance of PSO-LS-SVM models was compared with partial least squares regression (PLSR) and back propagation artificial neural network (BP-ANN). The calibration and validation results of PSO-LS-SVM were superior to both PLS and BP-ANN. For PSO-LS-SVM models, the correlation coefficients (r) of calibrations were all above 0.942. The optimal prediction results were also achieved by PSO-LS-SVM models with the RMSEP (root mean square error of prediction) and RSEP (relative standard errors of prediction) less than 1.176 and 15.5% respectively. The results suggest that PSO-LS-SVM algorithm has a good model performance and high prediction accuracy. NIR has a potential value for rapid determination of multi-indicators in Corni Fructus.
[Study on fast quality control in extracting process of Paeonia lactiflora using near infrared spectroscopy].

Abstract:OBJECTIVE:To set up a novel method for fast analysis of active components in water extracting process of Paeonia lactiflora with near infrared spectroscopy (NIRS). METHOD:HPLC was used as the reference method to determine the content of Paeoniflorin and a multivariate calibration model based on PLS algorithm was developed to analyze the correlation between the spectra and the corresponding values determined by the reference method. RESULT:The correlation coefficient of the calibration model was 0.996 2, and the predicted coefficient was 0.989 5. The RMSEC and RMSEP were 0.109 g x L(-1) and 0.138 g x L(-1), respectively, and the RSEP was 5.6%. CONCLUSION:The method mentioned above is proved to be convenient, rapid and nondestructive, accurate and reliable, and is applicable for fast analysis and monitoring of active components in extraction process of traditional Chinese medicine.

Abstract:The analysis of infrared spectroscopy of substances is a non-invasive measurement technique that can be used in analytics. Although the main objective of this study is to provide a review of machine learning (ML) algorithms that have been reported for analyzing near-infrared (NIR) spectroscopy from traditional machine learning methods to deep network architectures, we also provide different NIR measurement modes, instruments, signal preprocessing methods, etc. Firstly, four different measurement modes available in NIR are reviewed, different types of NIR instruments are compared, and a summary of NIR data analysis methods is provided. Secondly, the public NIR spectroscopy datasets are briefly discussed, with links provided. Thirdly, the widely used data preprocessing and feature selection algorithms that have been reported for NIR spectroscopy are presented. Then, the majority of the traditional machine learning methods and deep network architectures that are commonly employed are covered. Finally, we conclude that developing the integration of a variety of machine learning algorithms in an efficient and lightweight manner is a significant future research direction.

Abstract:Near infrared (NIR) spectroscopy is a non-destructive detection technology involving NIR spectral data acquisition and chemometric treatment. The use of an NIR spectrometer is evidently crucial in this regard; however, traditional benchtop NIR spectrometers considerably limit usage scenarios. Accordingly, the miniaturization of spectrometers with high level performance has become a research trend. Various commercial products have been developed, and new techniques have been applied to produce more portable NIR spectrometers. This paper reviews the main types of commercial portable NIR spectrometers and summarizes as well as compares their specifications. Moreover, new techniques for promoting miniaturization and the prospects for future development are introduced.

Abstract:Near-infrared spectroscopy is a widely-used nondestructive analytical tool, but the disadvantages of weak band intensities and greatly overlapped bands have limited its application. In this review, several methods to overcome the drawbacks of NIR spectroscopy, such as second-derivative spectrum, two-dimensional correlation spectroscopy and chemometrics, are introduced. The applications of NIR spectroscopy in the studies of the structures and contents of both proteins and polymers with amide groups are discussed. All these show that the methods help a lot of in the qualitative and quantitative analysis of near-infrared spectroscopy and broaden the potential application of NIR spectroscopy.
