Transfection Of Vascular Smooth Muscle Cells With Novel Biodegradable Arginine Based Poly(Ester-Amide)S

Abstract:A novel family of synthetic biodegradable poly(ester amide)s (Arg-PEAs) were synthesized using arginine (Arg), diols and diacids as building blocks. The biosafety and functional properties of these polymers were tested in rat vascular smooth muscle cells, a major cell type participating in vascular diseases. Arg-PEAs showed high binding capacity for plasmid DNA with a trend that was reversibly related to the number of methylene groups in the diol segment. All Arg-PEAs transfected smooth muscle cells with an efficiency that was comparable to the commercial transfection reagent Superfect® according to the results from luciferase and GFP plasmid transfection. But unlike Superfect®, Arg-PEAs, over a large range of doses, had minimum effects on cell morphology, viability or apoptosis in MTT and cell death detection assay. Using rhodamine labeled plasmid DNA, we demonstrated that Arg-PEAs were able to deliver DNA, through an active endocytosis mechanism, into close to hundred percent of cells under optimal polymer-to-DNA weight ratios. However, a large portion of delivered DNA was trapped outside of the nucleus, in acidic endocytotic compartments, and subsequently was not expressed. These results suggest that Arg-PEAs, with further modifications to enhance endosome escape, are good candidates for non-viral gene carrier systems owning to their high cellular uptake nature and reliable biocompatibilities.
