Abstract:Regioselective functionalization of C-H bonds has attracted the attention of chemists for recent years. However, transition metal is indispensable in most of C-H activation and often expensive. Here, we describe a metal-free intramolecular heteroarene C(sp2)−H amination from unprotected arylamine. Exposure of 2-(Pyridin-3-yl)aniline to posstium tert-butoxide was discovered to access N-heterocycles without additives, which unlocks new breakthrough. This process is applicable for the synthesis of a wide variety of N-heterocycles including natural products and bioactive compounds. Mechanistic studies have established that these reactions proceed through a SNArH process. Such a unique transformation opens a door toward weak reactivity arylamine regioselective intreamolecular C-H amination with heterocyclic compounds.
