Oxidative Damage and Apoptosis in Human Umbilical Vein Endothelial Cells Induced by Arachidonic Acid

Abstract:To decide whether arachidonic acid (AA) induce apoptosis in human umbilical vein endothelial cells (HUVECs) and the potential mechanism involved. Cells were treated with different concentrations of AA for 8, 16, 20 and 24h respectively. The cell viability and morphology were monitored by MTT assay and hoechst 33258 staining. Agarose gel electrophoresis detected DNA fragmentation. Flow cytometer showed the apoptotic ratio. Malondialdehyde (MDA) content and cellular reduced glutathione (GSH) level in HUVECs were measured to evaluate the oxidative damage degree. AA induced decrease in cell viability in concentration and time-dependent manners (P0.01). Cell incubated with AA for 24h showed typical morphological changes of apoptosis. A “DNA ladder” pattern was also observed. After treated HUVECs with AA for 20h in the range of 50~150μmol/L, flow cytometer showed “apoptotic peak” and the apoptosis ratio were 20.7%,38.6% and 52.5% respectively(P0.05). With increasing concentration of AA, the MDA contents improved accordingly but the GSH contents decreased (P0.05). These data demonstrated that arachidonic acid (50~150μmol/L) could induce apoptosis in HUVECs. The mechanism of its action might be related to lipid peroxidation.
