Mechanism study on behavior of albumin microbubbles in myocardium undergoing ischemia-reperfusion

Abstract:Objective To investigate the behavior of albumin microbubbles in myocardium undergoing ischemia reperfusion(I R) and the mechanism responsible for this phenomenon. Methods The myocardial transit rate of albumin microbubbles was measured in open chest dogs undergoing I R with intermittent harmonic myocardial contrast echocardiography(MCE). In vitro studies were performed in which activated or nonactivated leukocytes were incubated with albumin microbubbles and observed under microscopy. The suspensions of leukocytes and microbubbles which contained or absent of integrins were analyzed with flow cytometry. Results Albumin microbubbles persisted within the myocardium undergoing I R. Five min later microbubbles were incubated with leukocytes, the number of cells interacting with microbubbles was greater for activated cells than for nonactivated cells( 20.3 ± 2.7 vs 4.5 ± 1.4 ,P< 0.01 ). Albumin microbubbles attached to the surface of leukocytes which activated by PMA, were phagocytosed at 15 min, and remained intact for up to 30 min. Microbubbles′ attachment was inhibited notably by blocking the leukocyte β 2 integrin Mac 1 (P< 0.01 ) and inhibited by VLA 4mAb slightly (P< 0.05 ).Conclusions The possible mechanism of albumin microbubbles that persists within the myocardium undergoing I R is due to β 2 integrin and VLA 4 induced microbubbles attaching to and phagocyted by activated leukocyted after I R, phagocytosed microbubbles can remain at the region of inflammation, also are responsible to ultrasound.
