The Role of IL-4 in the Induction of Tolerance by Pretransplant Infusion of Donor Liver Nonparenchymal Cell

Abstract:Objective To investigate the role of IL 4 in the induction of tolerance by pretransplant infusion of donor liver nonparenchymal cells (NPC). Methods C57BL/6(B6) mice received intravenous injection through tail vein of 2×10 7 liver NPC from C3H mice. After 48?h, cyclophosphamide in PBS was injected intraperitoneally into B6 mice in a dose of 200?mg/kg. Eighteen days after liver NPC intravenous injection, B6 mice undertook C3H mice skin graft transplant. The serum level of IL 4 in B6 mice was detected by ELISA methods on the different time points.Results The survival time of skin graft in 15 mice prolonged obviously with the mean survival time being (70± 17.2 ) days, the mean survival time of normal control was only (10± 0.4 ) days. The ELISA results showed that after NPC infusion, the level of IL 4 was also increased, especially in the longer tolerant mice group.Conclusion The high level of IL 4 plays a critical role in inducing and maintaining immune tolerance.
