Abstract:OBJECTIVE: To comstruct a siRNA vector targeting human breast cancer metastasis suppressor 1(BRMS1) gene and identify it,soas to provide the foundation for further study on BRMS1.METHODS: The specific pre-siRNA single stranded DNA oligos for BRMS1 were designed and synthesized,then via annealing and ligating in order.The recombined interfering plasmids pcDNA6.2-GW/EmGFP-BRMS1-siRNA was constructed which included pre-siRNA of BRMS1.After identified by sequencing,pcDNA6.2-GW/EmGFP-BRMS1-siRNA was cotransfected into human embryonic kidney(HEK) 293 cells with pDsRed2-N1-BRMS1(including red fluorescin-BRMS1 fusion gene) by mediating liposome.The RNAi effects were assessed by an inverted fluorescence microscope and flow cytometry assay.RESULTS: Two siRNA sequences for BRMS1 were determined.Two recombined plasmids including pre-siRNA were constructed respectively.After cotransfection,the inrerted fluorecma microscope showed that the expression of BRMS1 in transfected cells was depressed significantly by both siRNA(s)(P0.05),and the FCM assay showed that the interfering efficiencies were 67.33% and 76.67% respectively.CONCLUSIONS: The effective siRNA expression vector targeting human BRMS1 gene has been successfully constructed,which lays the foundation of study on the function and action mechanisms of BRMS1 by RNAi.
