Functional Analysis of Arabidopsis Cryptochrome in ABA-regulated Seed Germination

Abstract:Cryptochromes( CRYs),a group of photolyase-like blue light photoreceptors widely found in animals,plants and microbes,mediate blue light signal transduction and regulate multiple processes of growth and development in plants. ABA plays an important role in seed germination and seedling development. It is reported that CRYs affect the seed germination and seedling development responding to stress in higher plants including wheat,rapeseed and sweet sorghum. However,the function and the molecular mechanism behind the effect of CRYs on ABA-regulated seed germination remain unclear. In this study,the seed germination of cry1 or cry1 cry2,the loss-of-function mutant of CRY1 or both of CRY1 and CRY2 of Arabidopsis,responding to ABA treatment,was explored.The transcripts of ABA signaling-related genes and the key enzyme genes of ABA metabolism in cry1 mutant were detected using real time fluorescence quantitative PCR. The results showed that the seed germination of cry1 mutant was more sensitive to ABA than that of the wild type. The germination rate between cry1 cry2 double mutant and the wild type displayed no significant difference. These results suggested that CRY1 functions as a negative regulator in the ABA-inhibited seed germination,and cry1 and cry2 play the opposite roles in this process.
