Abstract:A rigorous preselection of identified compounds by in vitro cellular screening is necessary prior to using the drug candidates for the further time consuming and parallel monitoring stage. In this paper we introduced an integrated cell based chip which can detect changes of electrophysiological parameters by using microelectronic chips such as light-addressable potentiometric sensors(LAPS), micro-electrodes array sensors(MEAS) and interdigitated array sensor(IDA) etc. to record the extracellular metabolism chemical and biological substances, action potential and living characteristics of cells representing the different response. In this research we presented novel ideas including an integrating multifunctional cell sensors chip, obtaining the living cells as the source of cell sensors with cardiac myocytes. Further developments will be done by culturing living olfactory and taste cells on surface of the devices. Now, we have completed the fabrication of the sensor and obtained some promising experiments results in different sensor units. This research will be interesting and useful for parallel extracellular monitoring.
A Novel Glucose Sensor System with Au Nanoparticles Based on Microdialysis and Coenzymes for Continuous Glucose Monitoring

Abstract:A novel glucose sensor based on microdialysis technique has been developed for continuous glucose monitoring. It includes: (1) The microdialysis sampling and interval perfusion system, it can prevent electrode from being fouled and it also provides enough time for glucose molecule traversing the microdialysis membrane to establish a dynamic balance, and the sensor can be recovered well, too. (2) A “sandwich” structure glucose sensor based on an aqueous glucose oxidase (GOD) and catalase solution, it is easy to replace the inactive enzyme solution. Au interdigital array (IDA) microelectrodes and microgrooves were fabricated on the silicon wafer for electrochemistry determinations. (3) Continuous glucose monitoring system. The results of in vitro experiments show this glucose sensor has short response time, high sensitivity and good linearity. To improve the stability of liquid enzyme, aqueous colloidal gold nanoparticles was mixed with coenzymes solution. The result of experiments shows the repeatability of sensor was improved, and the sensitivity of sensor was enhanced. To the best of our knowledge, this is the first demonstration that aqueous colloidal gold nanoparticles enhance the activity of aqueous enzymes.

Abstract:Cell-based biosensors incorporate cells as sensing elements that convert a change in immediate environment to signals conducive for processing. It characterizes with high sensitivity, excellent selectivity and fast response. Cell-based biosensors have been implemented for a number of applications ranging from pharmaceutical screening to environmental pollutant detection. This work demonstrate a novel cell-based biosensor following the principle of light-addressable potentiometric sensor and field effect transistor for detecting the action potential of single living cell under drugs. The experimental results agree with our expectation.

Abstract:Biofuel cells allow for constructing sensors that leverage the specificity of enzymes without the need for an external power source. In this work, we design a self-powered glucose sensor based on a biofuel cell. The redox enzymes glucose dehydrogenase (NAD-GDH), glucose oxidase (GOx), and horseradish peroxidase (HRP) were immobilized as biocatalysts on the electrodes, which were previously engineered using carbon nanostructures, including multi-wall carbon nanotubes (MWCNTs) and reduced graphene oxide (rGO). Additional polymers were also introduced to improve biocatalyst immobilization. The reported design offers three main advantages: (i) by using glucose as the substrate for the both anode and cathode, a more compact and robust design is enabled, (ii) the system operates under air-saturating conditions, with no need for gas purge, and (iii) the combination of carbon nanostructures and a multi-enzyme cascade maximizes the sensitivity of the biosensor. Our design allows the reliable detection of glucose in the range of 0.1–7.0 mM, which is perfectly suited for common biofluids and industrial food samples.

Abstract:Nondestructive, high-efficiency, and on-demand intracellular drug/biomacromolecule delivery for therapeutic purposes remains a great challenge. Herein, a biomechanical-energy-powered triboelectric nanogenerator (TENG)-driven electroporation system is developed for intracellular drug delivery with high efficiency and minimal cell damage in vitro and in vivo. In the integrated system, a self-powered TENG as a stable voltage pulse source triggers the increase of plasma membrane potential and membrane permeability. Cooperatively, the silicon nanoneedle-array electrode minimizes cellular damage during electroporation via enhancing the localized electrical field at the nanoneedle-cell interface and also decreases plasma membrane fluidity for the enhancement of molecular influx. The integrated system achieves efficient delivery of exogenous materials (small molecules, macromolecules, and siRNA) into different types of cells, including hard-to-transfect primary cells, with delivery efficiency up to 90% and cell viability over 94%. Through simple finger friction or hand slapping of the wearable TENGs, it successfully realizes a transdermal biomolecule delivery with an over threefold depth enhancement in mice. This integrated and self-powered system for active electroporation drug delivery shows great prospect for self-tuning drug delivery and wearable medicine.

Abstract:In this work we developed a fully integrated biofuel cell on a microchip, which consisted of glucose dehydrogenase supported (carbon nanotubes/thionine/gold nanoparticles)(8) multilayer as the anode, and the (carbon nanotubes/polylysine/laccase)(15) multilayer as the cathode. The as-obtained biofuel cell produced open circuit potential 620 mV and power density 302 microW cm(-2), showing great potential as a small power resource of portable electronics. Most importantly, for the first time we demonstrated the feasibility of developing a self-powered biosensor based on the inhibitive effect on microchip enzyme biofuel cell. With cyanide employed as the model analyte, this method showed a linear range of 3.0 x 10(-7) to 5.0 x 10(-4) M and a detection limit with 1.0 x 10(-7) M under the optimal conditions. The detection limit was lower than the acceptable cyanide concentration in drinking water (1.9 x 10(-6) M) according to the World Health Organization (WHO). This self-powered sensor was successfully used to detect the cyanide concentration in a real sample, cassava, which is the main carbohydrate resource in South America and Africa. This presented biosensor combined with a resistor and a multimeter demonstrated the general applicability as a fast and simple detection method in the determination of endogenous biological cyanide.

Abstract:Tumors are the leading killers of human health, however, diagnosis on tumors are neither timely nor accurate due to the high detection cost and high technical requirements. For cancer diagnosis and treatment, self-powered biosensors based on enzymatic biofuel cells have the advantages of simple structure, easy miniaturization, instantaneous response to analytes, and high economic applicability. It is of great significance for the development of portable and personalized biosensors. Over the past twenty years, a number of prominent self-powered biosensors have been created to diagnose tumor via analyzing the corresponding biomarkers with low cost. What's more, some intelligent self-powered biosensors with integrated diagnostic and therapeutic functions have also been reported. In this review, the focus of this work is on the development of self-powered biosensors in tumor diagnosis and intelligent regulation from the aspects of design and working principles. Furthermore, the challenge for the future work is also analyzed.

Abstract:Miniaturized bioelectronics have great potential in smart devices and on-body biomedical applications, but their development is hindered by the limited availability of a sustainable energy supply. Enzymatic biofuel cells show promise as a power source, but integrating them into practical platforms presents challenges. To address this, we explore a range of materials and strategies for embedding energy-harvesting devices into flexible platforms. For instance, we present a self-powered bioelectronics mask that includes a biofuel cell, a supercapacitor, and a biosensor for detecting glucose levels, all powered by carbon nanotube-based printed biofuel cells. We have also engineered functional materials and customized inks for flexible, single-enzyme-based energy harvesting and self-powered biosensing devices. Additionally, we introduce a battery-free biosensing system that monitors metabolites through in vivo analysis. A self-powered glucose biofuel cell/biosensor integrated into a small circuit performs energy harvesting, biosensing, and wireless human-body communication, allowing for direct biochemical analysis of metabolic profiles. Furthermore, we developed a lab-in-a-mouth sensing device that analyzes thiocyanate levels directly and noninvasively. Carbon electrodes were screen-printed and integrated with mouthguards, and the resulting sensor demonstrates high sensitivity and selectivity for detecting salivary thiocyanate ions. We also discuss the challenges and opportunities in the fields of biosensors and on-demand sensors to further advance these technologies. It is envisioned that developments in biosensors and bioelectronics have significant potential in modern healthcare, including presymptomatic, preventive, and administrational medicine.
A Self-Powered Optogenetic System for Implantable Blood Glucose Control

Abstract:Diabetes treatment and rehabilitation are usually a lifetime process. Optogenetic engineered designer cell-therapy holds great promise in regulating blood glucose homeostasis. However, portable, sustainable, and long-term energy supplementation has previously presented a challenge for the use of optogenetic stimulation in vivo. Herein, we purpose a self-powered optogenetic system (SOS) for implantable blood glucose control. The SOS consists of a biocompatible far-red light (FRL) source, FRL-triggered transgene-expressing cells, a power management unit, and a flexible implantable piezoelectric nanogenerator (i-PENG) to supply long-term energy by converting biomechanical energy into electricity. Our results show that this system can harvest energy from body movement and power the FRL source, which then significantly enhanced production of a short variant of human glucagon-like peptide 1 (shGLP-1) in vitro and in vivo. Indeed, diabetic mice equipped with the SOS showed rapid restoration of blood glucose homeostasis, improved glucose, and insulin tolerance. Our results suggest that the SOS is sufficiently effective in self-powering the modulation of therapeutic outputs to control glucose homeostasis and, furthermore, present a new strategy for providing energy in optogenetic-based cell therapy.

Abstract:As an important carbon and energy source, glucose is a biomarker of many diseases, such as diabetes, peritoneal carcinomatosis and peritonitis. Nevertheless, real-time glucose detection strategy with single portable device to monitor different diseases was seldom established. In this study, a Pt-Ni nanocomposite and silica nanoporous membrane modified electrode with a miniaturized electrochemical circuit board was developed for glucose detection to monitor different diseases. Through construction of Pt-Ni nanocomposite, the sensitivity of glucose detection was improved obviously compared with Pt electrode and Ni electrode. Silica nanoporous membrane eliminated the negatively charged interferences, such as uric acid and ascorbic acid, simultaneously resisting the interference of albumin. Modified with these nanomaterials, the glucose electrode realized 1-40 mM detection in artificial blood and 0.2-15 mM detection in artificial ascites respectively, displaying good selectivity and stability. Miniaturized electrochemical circuit board was established to execute electrochemical test and realize data interaction with a Bluetooth-enabled smartphone. Furthermore, the accuracy of this portable system was validated with biochemical analyzer in clinical samples, including diabetic whole blood and pathological ascites. In this way, the system could offer real-time and rapid opportunities towards dynamic evaluation of diabetes, peritoneal carcinomatosis and peritonitis.
Towards a Self-Powered Amperometric Glucose Biosensor Based on a Single-Enzyme Biofuel Cell

Abstract:This paper describes the study of an amperometric glucose biosensor based on an enzymatic biofuel cell consisting of a bioanode and a biocathode modified with the same enzyme—glucose oxidase (GOx). A graphite rod electrode (GRE) was electrochemically modified with a layer of Prussian blue (PB) nanoparticles embedded in a poly(pyrrole-2-carboxylic acid) (PPCA) shell, and an additional layer of PPCA and was used as the cathode. A GRE modified with a nanocomposite composed of poly(1,10-phenanthroline-5,6-dione) (PPD) and gold nanoparticles (AuNPs) entrapped in a PPCA shell was used as an anode. Both electrodes were modified with GOx by covalently bonding the enzyme to the carboxyl groups of PPCA. The developed biosensor exhibited a wide linear range of 0.15–124.00 mM with an R2 of 0.9998 and a sensitivity of 0.16 μA/mM. The limit of detection (LOD) and quantification (LOQ) were found to be 0.07 and 0.23 mM, respectively. The biosensor demonstrated exceptional selectivity to glucose and operational stability throughout 35 days, as well as good reproducibility, repeatability, and anti-interference ability towards common interfering substances. The studies on human serum demonstrate the ability of the newly designed biosensor to determine glucose in complex real samples at clinically relevant concentrations.

Abstract:Information related to the diverse and dynamic metabolite composition of the small intestine is crucial for the diagnosis and treatment of various diseases. However, our current understanding of the physiochemical dynamics of metabolic processes within the small intestine is limited due to the lack of in situ access to the intestinal environment. Here, we report a demonstration of a battery-free ingestible biosensing system for monitoring metabolites in the small intestine. As a proof of concept, we monitor the intestinal glucose dynamics on a porcine model. Battery-free operation is achieved through a self-powered glucose biofuel cell/biosensor integrated into a circuit that performs energy harvesting, biosensing, and wireless telemetry via a power-to-frequency conversion scheme using magnetic human body communication. Such long-term biochemical analysis could potentially provide critical information regarding the complex and dynamic small intestine metabolic profiles.

Abstract:Smart drug delivery systems responding to glucose concentration of the surroundings are very critical for treating diabetes. Here, a closed-loop and glucose-responsive drug delivery system GOx-Au@ZIF-8 is constructed by encapsulating glucose oxidase (GOx), Au nanoparticles (AuNPs) and metformin into metal organic frameworks ZIF-8 through coprecipitation method. In this system, glucose is oxidized firstly by oxygen to generate gluconic acid and H2O2 under the catalysis of GOx, which decreases pH value of the surrounding. Thus, ZIF-8 collapses and releases the drug. Furthermore, the produced H2O2 is enzyme-free catalytic into H2O and O2 under the catalysis of AuNPs, which not only reduce the harm of reactive oxygen species but also achieved oxygen regeneration and recycling in the oxidation reaction of glucose. Finally, the excellent biocompatibility of GOxAu@ZIF-8 has been proven in vitro environment. The new closed-loop and glucose responsive system shows a new proof-of-concept for diabetes treatment, especially in hypoxic environment.
Dual-substrate, dual-mode and self-powered visual biosensor based on biofuel cell

Abstract:While lots of diseases and foods are related to two substrates, self-powered dual-substrate detection remains a great challenge. Here, we propose a portable self-powered visual biosensor based on a dual-substrate enzyme biofuel cell (EBFC) for glucose and ethanol detection. The integration of electrochemical and visual sensing can not only realize dual-substrate analysis but also implement dual-mode detection. The employed dual-substrate EBFC is composed of an alcohol dehydrogenase-based bioanode and a glucose oxidase/Prussian blue (PB) coupled biocathode. Since the proper redox potential and outstanding electrochromic property, PB serves as both the display screen for visual analysis and the biocathode for power generation. The constructed self-powered visual biosensor has wide detection range and low detection limit toward glucose and ethanol biosensing. Notably, the PB display screen could be recovered by the biosensor itself, making the biosensor reusable, integrating and miniaturization. For real sample analysis, glucose in serum and ethanol in wine can be successfully detected by visual inspection. In the meantime, the self-powered visual biosensor can distinguish different actual samples containing both glucose and ethanol. These further broaden the application range of self-powered biosensor and provide new insight for multiple substrates detection.

Abstract:Self-powered biosensors are innovative devices that can detect and analyze biological or chemical substances without the need for an external power source. These biosensors can convert energy from the surrounding environment or the analyte itself into electrical signals for sensing and data transmission. The self-powered nature of these biosensors offers several advantages, such as portability, autonomy, and reduced waste generation from disposable batteries. They find applications in various fields, including healthcare, environmental monitoring, food safety, and wearable devices. While self-powered biosensors are a promising technology, there are still challenges to address, such as improving energy efficiency, sensitivity, and stability to make them more practical and widely adopted. This review article focuses on exploring the evolving trends in self-powered biosensor design, outlining potential advantages and limitations. With a focal point on enzymatic biofuel cell power generation, this article describes various sensing mechanisms that employ the analyte as substrate or fuel for the biocatalyst's ability to generate current. Technical aspects of biofuel cells are also examined. Research and development in the field of self-powered biosensors is ongoing, and this review describes promising areas for further exploration within the field, identifying underexplored areas that could benefit from further investigation.

Light-driven ultrasensitive self-powered cytosensing of circulating tumor cells via integration of biofuel cells and a photoelectrochemical strategy

Abstract:Herein, a light-driven, membrane-less and mediator-less self-powered cytosensing platform via integration of biofuel cells (BFCs) and a photoelectrochemical strategy was developed for ultrasensitive detection of circulating tumor cells (CTCs). To construct cytosensors, an elaborately designed SH-Sgc8c aptamer/AuNP/g-C3N4 photoelectrode was used as an alternative anode for glucose oxidation, avoiding the introduction of anodic enzymes. Initially, glucose could favorably reach the photoanode surface and be easily oxidized by the photogenerated holes, while the photogenerated electrons would transfer to the biocathode and achieve biocatalytic reduction of O2, leading to a high EOCV. However, in the presence of CTCs, they could preferentially interact with the Sgc8c aptamer via specific recognition, and then complexes with large steric hindrance were immobilized on the photoanode surface, which could greatly affect the electron transfer between glucose and the photoanode surface. In this case, the EOCV decreased sharply. Encouragingly, this self-powered cytosensor exhibited an ultrasensitive response to the target CTCs in a wide concentration range from 20 to 2 × 105 cells mL-1 with a low detection limit of 10 cells mL-1 (S/N = 3), being superior to those of the reported methods. Moreover, this as-proposed self-powered cytosensor integrated with a photoelectrochemical strategy possessed unique advantages of not requiring an external power source, being anodic enzyme-free, having a simple construction process, facile miniaturization, and high selectivity and sensitivity, providing a promising and powerful tool for fundamental biochemical research and clinical diagnosis.

Abstract:Diabetes mellitus and its associated secondary complications have become a pressing global healthcare issue. The current integrated theranostic plan involves a glucometer-tandem pump. However, external condition-responsive insulin delivery systems utilizing rigid glucose sensors pose challenges in on-demand, long-term insulin administration. To overcome these challenges, we present a novel model of antidiabetic management based on printable metallo-nucleotide hydrogels and optogenetic engineering. The conductive hydrogels were self-assembled by bioorthogonal chemistry using oligonucleotides, carbon nanotubes, and glucose oxidase, enabling continuous glucose monitoring in a broad range (0.5-40 mM). The optogenetically engineered cells were enabled glucose regulation in type I diabetic mice via a far-red light-induced transgenic expression of insulin with a month-long avidity. Combining with a microchip-integrated microneedle patch, a prototyped close-loop system was constructed. The glucose levels detected by the sensor were received and converted by a wireless controller to modulate far-infrared light, thereby achieving on-demand insulin expression for several weeks. This study sheds new light on developing next-generation diagnostic and therapy systems for personalized and digitalized precision medicine.
Self-Powered Diaper Sensor with Wireless Transmitter Powered by Paper-Based Biofuel Cell with Urine Glucose as Fuel

Abstract:A self-driven sensor that can detect urine and urine sugar and can be mounted on diapers is desirable to reduce the burden of long-term care. In this study, we created a paper-based glucose biofuel cell that can be mounted on diapers to detect urine sugar. Electrodes for biofuel cells were produced by printing MgO-templated porous carbon on which poly(glycidyl methacrylate) was modified using graft polymerization. A new bioanode was prepared through covalently modifying flavin-adenine-dinucleotide-dependent glucose dehydrogenase and azure A with pendant glycidyl groups of poly(glycidyl methacrylate). We prepared a cathode with covalently bonded bilirubin oxidase. Covalent bonding of enzymes and mediators to both the bioanode and biocathode suppressed elution and improved stability. The biofuel cell could achieve a maximum output density of 0.12 mW cm–2, and by combining it with a wireless transmission device, the concentration of glucose sensed from the transmission frequency was in the range of 0–10 mM. The sensitivity of the sensor was estimated at 0.0030 ± 0.0002 Hz mmol–1 dm3. This device is expected to be a new urine-sugar detection device, composed only of organic materials with a low environmental load and it can be useful for detecting postprandial hyperglycemia.This article has not yet been cited by other publications.

Glucose biosensor based on open-source wireless microfluidic potentiostat

Abstract:Wireless potentiostats capable of cyclic voltammetry and amperometry that connect to the Internet are emerging as key attributes of future point-of-care devices. This work presents an "integrated microfluidic electrochemical detector" (iMED) three-electrode multi-potentiostat designed around operational amplifiers connected to a powerful WiFi-based microcontroller as a promising alternative to more expensive and complex strategies reported in the literature. The iMED is integrated with a microfluidic system developed to be controlled by the same microcontroller. The iMED is programmed wirelessly over a standard WiFi network and all electrochemical data is uploaded to an open-source cloud-based server. A wired desktop computer is not necessary for operation or program uploading. This method of integrated microfluidic automation is simple, uses common and inexpensive materials, and is compatible with commercial sample injectors. An integrated biosensor platform contains four screen-printed carbon arrays inside 4 separate microfluidic detection chambers with Pt counter and pseudo Ag/AgCl reference electrodes in situ. The iMED is benchmarked with K3[Fe(CN)6] against a commercial potentiostat and then as a glucose biosensor using glucose-oxidising films of [Os(2,2'-bipyridine)2(polyvinylimidazole)10Cl] prepared on screen-printed electrodes with multi walled carbon nanotubes, poly(ethylene glycol) diglycidyl ether and flavin adenine dinucleotide-dependent glucose dehydrogenase. Potential application of this cost-effective wireless potentiostat approach to modern bioelectronics and point-of-care diagnosis is demonstrated by production of glucose oxidation currents, under pseudo-physiological conditions, using mediating films with lower redox potentials.
