Abstract:Recent studies are increasingly uncovering the profound (and unexpected) influence of the biological corona on the in vivo behaviour of nanomaterials. Accurate analysis of corona activity is crucial for future nanomedicine design, but the major techniques for protein analysis need adjustment to make them fit the seminatural features of corona proteins and the size of nanomaterials. Efforts are being put into optimising electrophoresis, Western blotting, and mass spectroscopy for corona analysis. Innovative use of nonprotein analytical techniques (e.g., flow cytometry) and cutting-edge tools (e.g., immune electron microscopy) are providing new possibilities for corona analysis. Ongoing studies are focussing on tailor-made modifications to existing techniques, aimed at translating them into competent and collaborative tools for dissecting corona activities. The recent discovery of the biological corona is revolutionising our understanding of the in vivo behaviour of nanomaterials. Accurate analysis of corona bioactivity is essential for predicting the fate of nanomaterials and thereby improving nanomedicine design. Nevertheless, current biotechniques for protein analysis are not readily adaptable for analysing corona proteins, given that their conformation, activity, and interaction may largely differ from those of the native proteins. Here, we introduce and propose tailor-made modifications to five types of mainstream bioanalytical methodologies. We specifically illustrate how these modifications can translate existing techniques for protein analysis into competent tools for dissecting the composition, bioactivity, and interaction (with both nanomaterials and the tissue) of corona formed on specific nanomaterial surfaces. It is designed to selectively precipitate or pull down a target protein (e.g., Protein X), together with another protein (e.g., Protein Y) believed to bind with Protein X, from a mixture of proteins (e.g., cell lysate). Normally, the specific antibody of Protein X (anti-X) or that of Protein Y (anti-Y) is incubated with the protein mixture. Solid microbeads precoated with Protein A/G (an engineered protein that can bind IgG) are then added, which can bind the anti-X or anti-Y to precipitate the Protein X–Protein Y complex. Co-IP is widely used to examine the binding affinities and functions of the target protein. See Figure 3A in the main text for a scheme of the principle of co-IP. a layer of complex biological molecules (mainly proteins) adsorbed onto the surface of nanoparticles that have entered the body. Increasing evidence suggests that the corona can form rapidly and dynamically and on diverse types of nanomaterial surfaces. The corona proteins fully cover the surface of the nanomaterials and endow them with new biological functions in the body. a classical technique used to separate nucleic acids or proteins, based on their different sizes and charges. a plate-based, quantitative assay to determine the concentration of proteins; also based on the antigen–antibody interaction. The antibody is linked with enzymes, which can generate a measurable product through incubation with its substrate. a commonly used technology for detecting cells expressing, or labelled with, fluorescence. The two- (or multi-) dimension analysis of the physical and chemical characteristics, such as side scattering and fluorescence intensity, can reflect the difference between samples. a commonly used assay in molecular biology to detect the presence of specific proteins. Usually following protein electrophoresis, WB is based on immunoblotting the antigen–antibody interaction and subsequent luminesence application to visualise the bands of target proteins.
