Abstract:Concentrated γ-Oryzanol nanodispersions were fabricated using milli-Q water (no emulsifier) or 0.1% (w/w) polysorbate 80 (T80), modified lecithin (ML) or sodium caseinate (SC) as emulsifiers. The freshly prepared nanodispersions had comparable particle diameter (118 to 157 nm), γ-Oryzanol concentration (1.75 to 1.92 mg mL-1) and free-radical scavenging activity (59 to 62%) and had negative ζ-potentials (-22 to -59 mV), indicating that both γ-Oryzanol and emulsifier coexisted on the particles' interface. The nanoparticles had superior physicochemical stability up to 30 days of storage at 5 °C and were successfully autoclaved without excessive growth or aggregation. Nevertheless, they showed distinct physical stability upon storage at specific environmental conditions, which affected their In-vitro gastrointestinal digestion. Comprehensively, emulsifier-free nanodispersions were sensitive to acidic pH, NaCl and CaCl2 addition. ML and SC coated nanoparticles were sensitive to Ca2+ ions, while T80 stabilized nanodispersions resisted to all environmental stresses, resulting in optimal simulated intestinal absorption.

Abstract:Gamma-oryzanol (GO) is a bioactive compound that, due to its biological characteristics, can be added to a food matrix. However, the bioactive compound is difficult to incorporate due to its low solubility and stability. A nanoemulsion allows substances to be packaged in nanometric sizes, improving their bioavailability. In this work, a GO nanoemulsion was developed using high-energy techniques. The methodological process began with the formulation of the coarse emulsion, where the emulsifiers (sodium caseinate and citrus pectin), diluent (rice bran oil), and pH were varied to find the most stable formulation. The coarse emulsion was subjected to four high-energy techniques (conventional homogenization, high-pressure homogenization, ultra-high-pressure homogenization, and ultrasonication) to reduce the droplet size. A physical-stability test, rheological-behavior test, image analysis, and particle-size-and-distribution test were conducted to determine which was the best technique. The formulation with the highest stability (pH 5.3) was composed of 87% water, 6.1% sodium caseinate, 0.6% citrus pectin, 6.1% rice bran oil, and 0.2% GO. The ultrasonic treatment obtains the smallest particle size (30.1 ± 1 nm), and the high-pressure treatment obtains the greatest stability (TSI < 0.3), both at 0 and 7 days of storage. High-energy treatments significantly reduce the droplet size of the emulsion, with important differences between each technique.

Abstract:In this work, the soy protein/chitosan-based microsphere (SCM), as a stable biocompatible vehicle of oleanolic acid (OA), was constructed for the first time under acidic conditions, and OA was firstly used for quality maintenance of minimally processed products. The effect of soybean protein isolate/chitosan ratio, particle concentration and oil fraction on emulsion stability was studied. Additionally, the interfacial characteristics of SCM with and without loading OA were determined by cryo-SEM. Results demonstrated that the encapsulation efficiency of SCM embedded with 0.15% OA (SCM-OA0.15%) rated up to 87.53% +/- 2.23%, and SCM-OA showed smaller particle size, greater stability, and better liquid-state behavior, as well as the enhanced antibacterial and antioxidant properties in comparison to the control and SCM group. The minimally processed apples were used to evaluate the potential application of SCM-OA delivery system and it was evident that SCM-OA contributed to reducing weight loss, maintaining total phenol content and inhibiting color browning of minimally process apples. These findings shed new light on the emerging biocompatible vehicles of plant-derived bioactive substance.
Self-stabilizing performance of γ-oryzanol in oil-in-water emulsions and solid dispersions

Abstract:The surface activity of γ-oryzanol was evaluated by the pendant drop method (PDM), and its self-stabilizing properties were investigated by high-pressure homogenization (HPH) and solvent displacement method (SDM). Emulsions prepared by HPH were highly unstable due to the poor surface-active character of γ-oryzanol as identified by the PDM. In contrast, solid dispersions fabricated by SDM had comparable particle size to those prepared using Tween 80 (T80) as surfactant, and were stable up to 30 days of storage at 4 °C. The self-stabilizing properties of γ-oryzanol were attributed to the mechanism of spontaneous particle formation in SDM and to the ability of γ-oryzanol molecules to prevent particles aggregation by electrostatic repulsion. The outcome of this study indicates the potential of encapsulating selected bioactive compounds, such as γ-oryzanol, in stable colloidal systems by SDM without adding emulsifier(s), regardless of their surface-active character.

Abstract:The use of the appropriate emulsifier is essential for forming a stable nanoemulsion delivery system that can maintain the sustained release of its contents. Health concerns have prompted the search for natural biopolymers to replace traditional synthetic substances as emulsifiers. In this study, an oregano essential oil (OEO) nanoemulsion-embedding system was created using soybean protein isolate (SPI), tea saponin (TS), and soy lecithin (SL) as natural emulsifiers and then compared to a system created using a synthetic emulsifier (Tween 80). The results showed that 4% Tween 80, 1% SPI, 2% TS, and 4% SL were the optimal conditions. Subsequently, the influence of emulsifier type on nanoemulsion stability was evaluated. The results revealed that among all the nanoemulsions, the TS nanoemulsion exhibited excellent centrifugal stability, storage stability, and oxidative stability and maintained high stability and encapsulation efficiency, even under relatively extreme environmental conditions. The good stability of the TS nanoemulsion may be due to the strong electrostatic repulsion generated by TS molecules, which contain hydroxyl groups, sapogenins, and saccharides in their structures. Overall, the natural emulsifiers used in our study can form homogeneous nanoemulsions, but their effectiveness and stability differ considerably.

Abstract:In this study, the stability mechanism of grape seed oil/rice hydrolyzed protein emulsion was studied. The grape seed oil (10% v/v) and rice hydrolyzed protein (2% w/v) were homogenized under high pressure to prepare the emulsion. It was observed by CLSM and Multiple light scatterometer that the emulsion had long-term storage stability, and the average particle size of droplets was 0.984–1.363 μm. ζ-potential ranged from −37.733 mV to −25.633 mV. It is found that the emulsion has strong resistance to temperature, ions and other environmental factors from the macroscopic and microscopic structure, and no emulsion stratification phenomenon occurs. The composite emulsion can be used in the field of food industry and fine chemical industry, which can provide nutrition and functionality of products, its research has certain value and has a wide space for development.

Abstract:Unsaturated guluronate oligosaccharide (GOS) is generated via alginate-derived polyguluronate (PG) degradation by alginate lyase, followed by formation of a double bond between C-4 and C-5 at the nonreducing end. In this study, GOS was first used as a stabilizer to fabricate O/W nanoemulsions loaded with resveratrol (GOS-RES). Our results revealed that both the GOS-RES and normal O/W resveratrol nanoemulsions (water-RES) showed small droplet sizes and narrow size distributions under certain experimental conditions. However, the particle size and stability of the GOS-RES were slightly greater than those of the water-RES in acidic and neutral environments and at high temperatures. Furthermore, the GOS-RES exhibited a better sustained release effect for resveratrol than the water-RES. Moreover, the GOS-RES showed a significant superoxide radical scavenging effect. All these results demonstrated that GOS has good prospects for preparing nanoemulsions to encapsulate hydrophobic nutrients, which could be applied as food-grade components in beverages and other foods.

Abstract:This study was to improve the physical and oxidative stability of sodium caseinate (NaCas)-stabilized oil-in-water (O/W) emulsions with arabinoxylan hydrolysates (AXHs). AXHs with different molecular sizes were prepared using xylanase treatment for 0 (AXH0), 30 (AXH30), 60 (AXH60), and 120 (AXH120) min, respectively. Compared with the emulsion without AXHs, all AXHs emulsions showed increased coalescence stability, evidenced by no change occurred in the droplet size with the pH variation from 7.0 to 5.0. Moreover, at pH 7.0 and 5.0, AXH60 and AXH120 emulsions showed no flocculation , coalescence, or creaming before and after 21 d storage. All the AXH samples showed excellent antioxidant capacities, demonstrated by the slow accumulation of lipid hydroperoxides and thiobarbituric acid reactive substances during storage. In sum, rice bran arabinoxylans hydrolyzed ≥60 min possess a potential as effective antioxidants to form physically and oxidatively stable O/W emulsions at pH above the emulsifier pI, and substances with high antioxidant activity below the emulsifier pI still need to be explored.

Abstract:Algae oil, enriched with omega-3 long-chain polyunsaturated fatty acids (omega-3 LC-PUFA), is known for its health benefits. However, protection against lipid oxidation as well as masking of unpleasant fishy malodors in algae oil enriched foods is a big challenge to achieve. In this study, we firstly achieved a one-pot ultrasound emulsification strategy (alternative heating-homogenization) to prepare phytosterol structured thermosensitive algae oil-in-water nanoemulsion stabilized by quillaja saponin. After spray drying, the resulting algae oil powders from the structured nanoemulsion templates exhibit an excellent reconstructed behavior, even after 30 d of storage. Furthermore, an enhanced oxidative stability was obtained by reducing both the primary and secondary oxidation products through formulation with beta-sitosterol and gamma-oryzanol, which are natural antioxidants. Following the results of headspace volatiles using dynamic headspace-gas chromatography-mass spectrometry (DHS-GC-MS), it was clear that the structured algae oil-loaded nanoemulsion and powder had lower levels of fishy off-flavour (e.g., (Z)-heptenal, decanal, ethanone, and hexadecenoic acid), whereas the control emulsion and oil powder without structure performed worse. This study demonstrated that the structure from phytosterols is an effective strategy to minimize the fishy off-flavour and maximize oxidative stability of both algae oil nanoemulsions and spray-dried powders, and opens up the possibility of formulation design in polyunsaturated oil encapsulates as novel delivery systems to apply in functional foods and beverages.

Abstract:The thermodynamic instability of food nanoemulsions may occur during production and storage due to environmental stress or reaction with other constituents, appearing in the forms of sedimentation, creaming or oil oxidation. Such instabilities reduce the tendency to consumption and threaten consumers' health. The present study aimed to review the effects of emulsifiers on the physicochemical stability of oil-in-water nanoemulsions. Emulsifiers were selected considering several factors, including the preparation and processing methods, nature, concentration, hydrophilic-lipophilic balance, molecular weight, particle surface charge, ambient temperature, pro-oxidants and antioxidants, and type of the food matrix. Emulsifiers often include proteins, esters of polyoxyethylene (e.g., Tween), fatty acids, polysaccharides, and phospholipids, which play a key role in the formation and stability of food emulsions through adsorption at the oil–water interface. Determining the impact of emulsifiers on the control of instability at the interface contributes to the production of the healthiest, safest, and most stable nanoemulsion-based structures. Since emulsifiers have individual structures and different functional properties, they should be selected based on the desired product properties. Therefore further research is required to identify novel emulsifiers or a combination of emulsifiers with different properties for food applications.

Abstract:ABSTRAC T The practical usage of Osmanthus essential oil (OEO) in foods is limited due to its poor solubility in water, higher volatility, and instability. This research aimed to prepare OEO nano-emulsion (OEO-NE) by high-pressure homogenization technique and characterize its nano-form (particle size, zeta potential, structure, FTIR, and thermodynamic stability). The antibacterial activity of OEO was researched using network pharmacology and molecular docking. Furthermore, the antibacterial and antioxidant activities of OEO-NE and OEO were researched. The OEO-NE exhibited long-term storage stability compared to the OEO, with particle size changing to 40.90 nm - 68.09 nm and zeta potential changing to -38.37 mV - -35 mV over 31 storage days at 4 degrees C and 25 degrees C. Dihydro- beta -ionone and linalool oxide showed better binding affinity for S. aureus TyrRS than E. coli 24 kDa. In addition, nano-emulsification effectively stabilizes the antibacterial and antioxidant activity of OEO. Therefore, OEO-NE is an effective strategy to enhance stability and bioavailability.
Oleanolic acid nanoparticles-stabilized W/O Pickering emulsions: Fabrication, characterization, and delivery application

Abstract:Water-in-oil (W/O) Pickering emulsions have wide applications in the food industries. However, the existing W/O Pickering particles have disadvantages such as lack of bioactivity and poor stability. In this study, naturally occurring bioactive oleanolic acid (OA) was used as a novel emulsifier for W/O emulsions. Results revealed that rod-like OA could formulate into spherical nanoparticles by self-assembly, and then be anchored onto the oil-water interface to stabilize the emulsions. Besides, both OA concentration and internal water fraction (φ) had significant effect on the properties of emulsions. Furthermore, the resulted emulsions exhibited potential application as carriers for epigallocatechin-3-gallate (EGCG), which significantly improved its UV and thermal stability. Meanwhile, it could effectively protect EGCG from gastric digestion, and controlled release in the intestine. This work demonstrated the successful application of OA as a stabilizer for W/O emulsions, and provided valuable insight into its potential as delivery system for hydrophilic instable compounds.

Abstract:Objective To study the optimal prescription screening and preparation technology of oryzanol solid self-microemulsion.Methods The formulation ratio of oryzanol self-microemulsion was screened by using the pseudo-ternary phase diagram and homogeneous design with the dissolution as index.Also the solid adsorbent was investigated and the prescription form of solid self-microemulsion was finally determined.Results The prescription ratio of the drug delivery system for oryzanol self-microemulsion was ethyloleate∶ OP-10∶ EL-35∶ Transcutol∶ oryzanol(19.4∶ 33.26∶ 33.26∶ 11.08∶ 3).The solid adsorbent was mannitol∶ micronized silica gel(1∶ 2).Conclusion A good prescription for self-emulsifying of oryzanol was established,oral capsules formulation was prepared and the development of oryzanol solid self-microemulsion was accomplished.

Abstract:A current challenge in the area of food emulsion is the design of microstructure that provides controlled release of volatile compounds during storage and consumption. Here, a new strategy addressed this problem at the fundamental level by describing the design of organogel-based emulsion from the self-assembly of β-sitosterol and γ-oryzanol that are capable of tuning volatile release. The results showed that the release rate (v0), maximum headspace concentrations (Cmax) and partition coefficients (ka/e) above structured emulsions were significantly lower than unstructured emulsions and controlled release doing undergo tunable though the self-assembled interface and core fine microstructure from internal phase under dynamic and static condition. This result provides an understanding of how emulsions can behave as delivery system to better design novel food products with enhanced sensorial and nutritional attributes.

Abstract:In this work, high internal phase emulsions (HIPEs) stabilized by naturally derived cellulose nanocrystals (CNC) and gelatinized soluble starch (GSS) were fabricated to stabilize oregano essential oil (OEO) in the absence of surfactant. The physical properties, microstructures, rheological properties, and storage stability of HIPEs were investigated by adjusting CNC contents (0.2, 0.3, 0.4 and 0.5 wt%) and starch concentration (4.5 wt%). The results revealed that CNC-GSS stabilized HIPEs exhibited good storage stability within one month and the smallest droplets size at a CNC concentration of 0.4 wt%. The emulsion volume fractions of 0.2, 0.3, 0.4 and 0.5 wt% CNC-GSS stabilized HIPEs after centrifugation reached 77.58, 82.05, 94.22, and 91.41 %, respectively. The effect of native CNC and GSS were analyzed to understand the stability mechanisms of HIPEs. The results revealed that CNC could be used as an effective stabilizer and emulsifier to fabricate the stable and gel-like HIPEs with tunable microstructure and rheological properties.

Abstract:To extend the application of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) in liquid food, and to improve the physicochemical stability, we prepared stable DHA and EPA nanoemulsions, which can be used for large scale production. The Emulsion Phase Inversion (EPI) method was used to prepare nanoemulsions as safe and convenient with low-energy emulsification method. The type of oil, the type of surfactant and the concentration of surfactant have the most significant effect on particle size. Within 20 days, the nanoemulsions have good physical stability under different storage conditions, and the retention rate of DHA/EPA can be stabilized at >60%. Furthermore, different types of natural antioxidants were added and the effect was detected by analyzing the peroxide value. Among them, tea polyphenols showed the most significant effect. The data obtained showed enhanced nanoemulsions stability, increased application in food and actual large-scale production.

Abstract:Nanoemulgels are novel formulations of great interest for their use as dual-release systems and as fat substitutes in foods. Lemon essential oil, with a large number of benefits due to its antimicrobial, antifungal, and medicinal properties, and low methoxyl pectin, a natural polysaccharide capable of gelling by adding divalent ions such as calcium, are very appropriate ingredients to produce nanoemulgels with potential applications in industries such as cosmetics, agrochemistry, pharmaceuticals, or food. In this work, lemon-essential-oil-in-water nanoemulgels containing low methoxyl pectin derived from citrus peels were prepared following a three-step process that involves the preparation of a nanoemulsion, a pectin gel, and the mixture of both. In the first stage, the stirring time and the rotational rate employed during the mixing step were assessed. Once the preparation protocol was established, the pectin gel/nanoemulsion mass ratio was investigated. Different techniques were combined to evaluate the influence of the processing and the composition variables on the particle size distribution, mean diameters, flow curves, and physical stability of different emulgels obtained. It was found that the processing variables studied, stirring time, and rotational rate, do not influence the mean particle size of the emulgel, with values matching those of the starting nanoemulsion. However, 3 min and 200 rpm were selected for exhibiting the lowest TSI values. Regarding the composition, a higher content of pectin gel caused a higher viscosity, and therefore a higher physical stability, with the 75P/25E emulgel being the most stable. Aggregation of gel particles, because the pectin gel was really a sheared gel, was the main responsible contributor to the results obtained. This work highlights the importance of the preparation and formulation variables to develop stable, innovative formulations based on nanoemulgels.

Preparation and Characterization of Fish Oil Pickering Emulsions Stabilized by Resveratrol-Loaded Gliadin/Chitin Nanocrystal Composite Nanoparticles

Abstract:Unsaturated fatty acids present in fish oil offer various physiological benefits to the human body. However, their susceptibility to oxidation severely limits their potential applications. The purpose of this study was to develop Pickering emulsions stabilized from a composite of resveratrol-loaded gliadin nanoparticles and oxidized chitin nanocrystals (GR/OC) to protect fish oil from oxidation. The effects of the GR/OC composite on the characterizations of fish oil Pickering emulsions were investigated, including the microstructure, physicochemical properties (stability and rheological behavior), and digestion properties in vitro. The results revealed that an increased concentration of the GR/OC composite significantly reduced the droplet size and improved the ambient stability of the emulsions (in terms of pH, ionic strength, temperature, and storage time). Confocal laser scanning microscopy images depicted that the GR/OC nanoparticles were uniformly dispersed at the interface between water and fish oil (W-O interface). This distribution formed a protective envelope around the droplets. Remarkably, the addition of 2% GR/OC nanoparticles stabilized the Pickering emulsions and showed the most positive effect on the antioxidant capacity compared to that of the control group. These stabilized emulsions maintained lower peroxide values and acid values, which were 1.5 times less than those of the blank control during the 14 day accelerated oxidation experiment. Furthermore, the Pickering emulsions stabilized by GR/OC nanoparticles exhibited the ability to protect fish oil from contamination by gastric juices and facilitate the intestinal absorption of omega-3 polyunsaturated fatty acids. The findings suggest that these GR/OC-stabilized Pickering emulsions offer a promising alternative for delivering fish oils in various industries, including the food industry.

Abstract:In this study, flaxseed oil was used as embedding material and flaxseed gum as sole emulsifier to prepare flaxseed oil nanoemulsions. The storage temperature (4–37 °C, pH 5.8, 0 mmol/L), pH (3–11, 4 °C, 0 mmol/L), NaCl (100–500 mmol/L, pH 5.8, 4 °C) on the stability of flaxseed oil nanoemulsions were investigated in terms of particle size, ζ -potential, creaming index, microscopy, rheological characteristics, surface tension, and nanoemulsion color as characterization. The results showed that temperature significantly affected the stability of flaxseed oil nanoemulsions, and the stability decreased with increasing temperature. The characteristics of flaxseed oil nanoemulsion were relatively stable at pH 5 and pH 7, while the stability was poor under pH 3 and alkaline conditions. As the strength of the ionic increased, unstable aggregation occurred (NaCl >200 mmol/L). In conclusion, the stability of flaxseed oil nanoemulsion was better at 4 °C, pH 5–7, and NaCl <200 mmol/L. These results provide useful information to facilitate the design and preparation of stable package delivery systems for flaxseed oil in food and other industries.
