Abstract:The development and utilization of environment-friendly energy is imminent with global energy shortage and climate change becoming increasingly serious. The advancement and utilization of environment-friendly energy, including photocatalytic hydrogen production, electrocatalytic pollutant removal, and the development of electrochemical energy storage devices, have emerged as prominent research topics. A variety of modern analysis techniques provide strong support in these related studies, such as X-ray absorption spectroscopy (XAS) for investigating the active site geometry and electronic structure of photocatalysts, synchrotron radiation X-ray photoelectron spectroscopy (SR-XPS) for real-time monitoring of interatomic electron excitation and transfer in photocatalysts, in situ electron paramagnetic resonance spectroscopy (EPR) for the detection of free radicals in photocatalysis reactions, Raman spectroscopy (RS) for analyzing the interfacial structure of electrochemical reactions, and in situ transmission electron microscopy (TEM) for studying catalyst structure and reactivity in photocatalytic reactions. While these techniques provide abundant information for the mechanistic aspects of the associated chemical processes, it remains challenging to conduct further identification and structural analysis of the reaction intermediates at a molecular level. Due to the ability in rapid identification of the components in complex system, mass spectrometry (MS) has been widely used in the new energy fields such as photocatalytic reaction, electrocatalytic reaction, and the development of electrochemical energy storage devices. This review introduced the application of mass spectrometry techniques, including liquid chromatography-mass spectrometry (LC-MS), gas chromatography-mass spectrometry (GC-MS), matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS), differential electrochemical mass spectrometry (DEMS), and electrospray ionization mass spectrometry (ESI-MS), in the detection of transient intermediates and products as well as the exploration of reaction mechanisms. Modern mass spectrometry offers critical support for the mechanism analysis of photocatalytic reactions aimed at the mineralization of organic pollutants, hydrogen generation, CO2 reduction, N2 fixation, and organic synthesis. It also provides distinct advantages for real-time product monitoring, isotopic labeling analysis, and the capture and identification of short-lived intermediates in electrocatalytic processes, such as hydrogen evolution, oxygen evolution, CO2 reduction, N2 reduction, and organic synthesis. By analyzing reaction products and intermediates in electrochemical energy storage devices through mass spectrometry, it is possible to elucidate the nature of energy storage and conduct rational design of high-performance materials. Modern mass spectrometry facilitates both offline detection of products and online identification of reaction intermediates, enhancing the understanding of reaction kinetics and elucidating mechanisms when combined with theoretical calculations. Thus, modern mass spectrometry has become a powerful tool for the study of reaction mechanisms. The challenges and prospects of mass spectrometry in the fields of photocatalytic degradation, photocatalytic synthesis, electrocatalytic reduction, electrocatalytic synthesis, lithium-ion batteries, and lithium-sulfur batteries were also discussed in this review. While current mass spectrometry techniques can detect transient intermediates, challenges persist in identifying the reaction intermediates with lower concentrations and reduced stability. Therefore, optimizing the sampling systems and mass analyzers is crucial to improve the sampling speed, transfer efficiency, and analysis throughput. Understanding complex reaction mechanisms requires correlating reaction intermediates with various conditions, such as reaction time, radiation wavelength, and potential. This necessitates a spatial integration between the reactor and the mass spectrometer to preserve the true form of intermediates and ensure accurate signal transmission that reflects the pertinent reaction parameters. Integrating mass spectrometry to analyze the morphology and structure of reaction intermediates and products with theoretical calculations may enable the construction of a comprehensive, multi-dimensional reaction theory system. Ultimately, employing this theoretical framework to advance experimental research could represent a novel idea in the research filed.

Abstract:A hybrid ultramicroelectrode containing one micro-carbon electrode and one empty micro-channel was employed to be a micro-electrochemical cell and a mass spectrometric nanospray emitter. This setup can combine MS with an electrode directly and provide in situ information about an electrochemical reaction. The mechanisms proposed by Bard et al. for a Ru(bpy)3 2+ (bpy 1⁄4 2,20-bipyridine) electrochemiluminescence (ECL) system were confirmed by the MS detection of key intermediates. The short-lived diimine intermediate of electrochemical oxidation of uric acid was also detected, which affirms that the novel technique is able to catch fleeting intermediates. These experimental results demonstrate that this new method is simple, easy to implement and can be coupled with many commercial mass spectrometric instruments to provide very useful information about electrochemical reactions.

Abstract:Hyphenation of electrochemistry (EC) and mass spectrometry has become a powerful tool to study redox processes. Approaches that can achieve this hyphenation include integrating chromatography/electrophoresis between electroinduced redox reactions and detection of products, coupling an EC flow cell to a mass spectrometer, and performing electrochemical reactions inside the ion source of a mass spectrometer. The first two approaches have been well reviewed elsewhere. This Minireview highlights the inherent electrochemical properties of many mass spectrometry ion sources and their roles in the coupling of electrochemistry and mass spectrometric analysis. Development of modified ion sources that allow the compatibility of electrochemistry with ionization processes is also surveyed. Applications of different in-source electrochemical devices are provided including intermediate capturing, bioanalytical studies, nanoparticle formation, electrosynthesis, and electrode imaging.

Abstract:Electrochemical reaction is a continuous dynamic process, accompanied by generation of short-lived intermedi-ates and complex structural substances. Therefore, precisely and effectively capturing the products of the reaction process is helpful to accurately deduce its reaction mechanism, optimize the reaction parameters and improve the reaction efficiency. At present, the mainstream electrochemical on-line monitoring techniques include spectroscopy, cyclic voltammetry and linear polarization curves. These methods are capable to detect the structure and composition changes of most substances in the reaction process. However, in order to more systematically and accurately grasp the information of all products, the real-time and in situ reaction monitoring technologies needs to be further expanded. Mass spectrometry (MS) has the advantages of high sensitivity, good selectivity, rapid response time and structural analysis, making itself an ideal research method for electrochemical reactions. In recent years, more and more reports on the study of electrochemical reaction by MS have been published. In particular, ambient ionization sources such as electrospray ionization (ESI) and its derived ionization techniques developed for electrochemistry have become a research hotspot. This review introduced the recently published electrochem-istry-mass spectrometry (EC-MS) techniques, and described the electrochemical ion sources that designed and developed for different types of electrochemical reactions.

Abstract:A new mode of the scanning electrochemical microscope (SECM) operation was developed that combines reagent delivery from the nanopipette with electron transfer at the conductive substrate and ion transfer across the liquid/liquid interface supported at the nanopipette tip. This approach offers potential advantages for measurements of heterogeneous electron transfer kinetics and reaction rate imaging by enabling straightforward separation of the contributions of surface topography and reactivity features to the tip current. It addresses some other long-standing technical issues, including sensitive probing of low signal sources (e. g., immobilized enzymes or catalyst particles) without diffusional broadening and the elimination of the elevated background signal in generation/collection-type experiments. The high spatial resolution attainable in electron transfer/ion transfer mode experiments and the absence of redox mediator species in the bulk solution are advantageous for studies of biological cells.

Abstract:Comprehensive SummaryElectrochemical mass spectrometry (EC‐MS) is a powerful tool to capture and analyze the intermediates and products during electrochemical reactions. This hyphenated technique combines electrochemistry with mass spectrometry using specific apparatuses, which helps researchers study mechanisms of redox reactions by in situ detecting chemical composition changes. Recently, various EC‐MS methods have been applied in a series of electrochemical reactions to reveal the mechanisms, mainly in the areas of electrochemical sensors, organic electrochemistry, and electrocatalysis. In this review, we intend to summarize the recent advances in real‐time analysis of different types of electrochemical reactions by EC‐MS and offer an outlook on the perspectives in these areas.

Abstract:Electrochemical reaction is a continuous dynamic process, accompanied by electron transfer and generation of short-lived intermediates. The mechanism investigation of electrochemical interfaces is pivotal for the progression of energy storage, electro-organic synthesis, electrocatalysis, and electroanalysis. The valuable insights for optimizing performance, improving efficiency, and developing novel methodologies can be attained by elucidating the complicated pathways involved in the electrochemical processes. Thus, how to monitor transient intermediates formed at the solid-liquid interface becomes the critical point in unraveling these intricate processes. To this end, a series of in-situ analytical methods such as in-situ electron paramagnetic resonance spectroscopy (in-situ EPR), in-situ Raman spectroscopy, in-situ infrared spectroscopy (in-situ IR) and in-situ electrochemical mass spectrometry (in-situ EC-MS) have been exploited to probe the chemical transformations at the electrode/electrolyte interface. Among them, due to the superior specificity for interpreting molecular information, EC-MS has progressively gained extensive attention as a powerful and reliable tool for capturing intermediates and revealing the mechanisms in electrochemical reactions. In this review, the research progresses in the recent advancements in the detection of transient reaction intermediates through electrochemical mass spectrometry were presented. The discussion centered on elucidating the principle and configuration of the devices employed in EC-MS, with a specific focus on electrospray ionization mass spectrometry (ESI-MS), atmospheric pressure ionization mass spectrometry (AIMS) and time-of-flight secondary ion mass spectrometry (TOF-SIMS). In addition, the review delved into the wide-ranging applications of EC-MS in monitoring electrochemical processes in electro-organic synthesis, electrocatalysis, lithium-ion batteries, electrochemiluminescence, and bioelectrochemical reactions. Despite the remarkable progress has made, the challenges in the field of EC-MS are prospected. There is still an urgent need for further improvement of the experimental apparatus to enhance the sampling efficiency and time resolution to capture short-lived and low-concentration reaction intermediates. In the future, we anticipate that EC-MS will be combined with other in situ analytical techniques, such as in situ infrared or Raman spectroscopy, enabling multi-dimensional characterization and monitoring of electrochemical reaction intermediates.

Abstract:We report a new mass spectrometric method for detecting electrogenerated intermediates. This approach is based on simultaneous activation of electrospray ionization and redox reaction on a wireless bipolar ultramicroelectrode, which is fabricated in the tip of a quartz nanopipette. The hollow structure of the ultramicroelectrode permits rapid transferring the transient species from electrode–electrolyte interfaces into the gas phase for mass spectrometric identification on the time scale of microseconds. The long‐sought fleeting intermediates including TPrA.+, whose lifetime in solution is only 200 μs, and catecholamine o‐semiquinone radicals, the second‐order rate constant of which is typically 109 m−1 s−1, were successfully identified, helping clarify the previously hidden reaction pathways. Accordingly, our method may have wide applicability in exploring the dynamics of many electrochemical reactions, especially their ultrafast initial steps. A method for detecting electrogenerated intermediates is based on simultaneous activation of electrospray ionization and redox reaction on a wireless bipolar ultramicroelectrode. The hollow structure of the ultramicroelectrode permits rapid transferring the transient species from electrode–electrolyte interfaces into the gas phase for mass spectrometric identification on the time scale of microseconds.

Abstract:A droplet-scale, real-time electrochemical reaction screening platform based on droplet spray ionization mass spectrometry (DSI-MS) has been developed. The N,N-dimethylaniline (DMA) radical cation with a half-life of microseconds was readily detected by MS during the electrooxidation of DMA, and the subsequent reactions were followed in real time for minutes.

Abstract:The present work shows that the electrochemical properties of electrospray ionization (ESI) can be used to add functions to the process. As example, we show how the choice of the electrode material can be used to study interactions between metal ions and biomolecules in mass spectrometry (MS). In positive ionization MS, an electrospray device acts as anode, which implies oxidation reactions. Sacrificial electrodes (made of copper or zinc) are used to supply the electrospray current and to produce cations that are able to react on-line with compounds of interest. Thus, the interactions between copper ions and ligands or peptides were investigated by using a copper electrode. Another example is the in situ electrogeneration of a dinuclear zinc(II) complex for the mass tagging of phosphopeptides when working with a zinc electrode. In order to perform these reactions on the same microchip, a dual-channel microsprayer was used, where one channel was dedicated to the tag electrogeneration and the other to the infusion of a phosphopeptides solution. Finally, this dual-channel microsprayer was used to study complexation at liquid-liquid interfaces in biphasic ESI-MS, such as thioether crowns and lead ions or peptides and phospholipids complexes. These examples illustrate the use of electrochemistry and on-chip reactions in ESI-MS analysis.

Abstract:This manuscript presents a push-pull electrochemical scanner able to image reactivity of initially dry surfaces by scanning electrochemical microscopy (SECM) and to probe molecules present or generated at the surface by mass spectrometry (MS). The proof-of-concept is demonstrated by coupling SECM with matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) for imaging latent human fingerprints, which had been in contact with picric acid used here as a model explosive. The push pull electrochemical scanner has also been coupled with electrospray ionization mass spectrometry (ESI-MS) to assay the activity of surface spotted enzymes. These experimental studies are complemented by 3D finite element simulations solving Navier- Stokes and diffusion-convection differential equations to optimize the coupling between SECM imaging and mass spectrometry detection.

Abstract:The in situ molecular characterization of reaction intermediates and products at electrode electrolyte interfaces is central to mechanistic studies of complex electrochemical processes, yet a great challenge. The coupling of electrochemistry (EC) and mass spectrometry (MS) has seen rapid development and found broad applicability in tackling challenges in analytical and bioanalytical chemistry. However, few truly in situ and real-time EC-MS studies have been reported at electrode electrolyte interfaces. An innovative EC-MS coupling method named in situ liquid secondary ion mass spectrometry (SIMS) was recently developed by combining SIMS with a vacuum compatible microfluidic electrochemical device. Using this novel capability, we report the first in situ elucidation of the electro-oxidation mechanism of a biologically significant organic compound, ascorbic acid (AA), at the electrode electrolyte interface. The short-lived radical intermediate was successfully captured, which had not been detected directly before. Moreover, we demonstrated the power of this new technique in real-time monitoring of the formation and dynamic evolution of electrical double layers at the electrode electrolyte interface. This work suggests further promising applications of in situ liquid SIMS in studying more complex chemical and biological events at the electrode electrolyte interface.

Abstract:Monitoring of electrochemical (EC) reactions is crucial to obtain efficient, fast and inexpensive reactions for synthesis or analysis. Traditional characterizations by cyclic voltammetry or spectroscopic techniques are usually limited by the lack of detailed structural information on analytes. Without any sample pretreatment, ambient mass spectrometry (AMS) can directly provide structural information of multiple analytes in situ. The monitoring of EC reactions by AMS provides potentials for detecting short-lived intermediates to examine reaction mechanisms and discover new reactions. Therefore, these developments will give significant contributions to fields including biological chemistry, electrocatalysis, organic synthesis or battery fabrication. In this review, in light of the different configurations of electrodes (e.g., rotating waterwheel electrodes and ultramicroelectrodes) for different EC reactions, various designs of EC-coupled electrospray ionization-based AMS systems are summarized. In addition, the advantages, limitations and proposed suggestions for future developments are addressed.
In Situ Probing and Identification of Electrochemical Reaction Intermediates by Floating Electrolytic Electrospray Mass Spectrometry

Abstract:The in-depth study of electrochemical (EC) synthesis can require a powerful mass spectrometry (MS) analytical platform which can discover and identify fleeting intermediates in EC reactions. Here we report a floating electrolytic electrospray ionization (FE-ESI) strategy that can perform EC processes in a floating electrolytic cell and monitor intermediates by high-resolution MS. Compared with previous EC-MS methods, a significant advantage of FE-ESI-MS is that it allows one to modulate the electrolytic and electrospray process individually, ensuring its high sensitivity in discovering intermediates and universality to investigate redox reactions in different scenarios. This powerful platform has been successfully used to investigate the EC reductive coupling of p-tolylboronic acid and p-nitrotoluene. A series of nitrene intermediates were discovered and identified by FE-ESI-MS, indicating that a hidden mechanism involving nitrene formation might play a key role in EC reductive coupling process.

Abstract:Soft-ionization methods, namely electrospray ionization and laser desorption/ionization, are widely used to transfer large molecules as intact gas-phase ions either from a solution or from a solid substrate. During both processes, in-source electrochemical and photoelectrochemical reactions occur. These electrode reactions, which take place at interfaces, play important roles in influencing the ionization products, but they have received little attention. We show that having good control over both types of electrochemical reactions can lead to new analytical applications. Examples include online tagging by grafting of mass tags and in-source photooxidation of peptides.

Abstract:Molecular structure conversion concomitant with mass transfer processes at the electrode–electrolyte interfaces plays a central role in energy electrochemistry. Mass spectrometry, as one of the most intuitive, sensitive techniques, provides the capability to collect transient intermediates and products and uncover reaction mechanisms and kinetics. In situ time-of-flight secondary ion electrochemical mass spectrometry with inherent high mass and spatiotemporal resolution has emerged as a promising strategy for investigating electrochemical processes at the electrode surface. This review illustrates the recent advancements in coupling time-of-flight secondary ion mass spectrometry and electrochemistry to visualize and quantify local dynamic electrochemical processes, identify solvated species distribution, and disclose hidden reaction pathways at the molecular level. Moreover, the key challenges in this field are further discussed to promote new applications and discoveries in operando studying the dynamic electrochemical interfaces of advanced energy systems.

Abstract:Electrospray-related ion sources are promising for direct mass spectrometric analysis of complex samples, but current protocols suffer from complicated components and low analytical sensitivity. Here, we propose a surface charge-induced electrospray ionization (SCIESI) inspired by flashover on an insulator surface under high voltage. This protocol not only effectively avoids contact between the sample solution and metal electrode, but also allows completion of the entire analytical process in less than 40 seconds and limits of detection in the pictogram per milliliter range. SCIESI coupled to mass spectrometry can also be used to monitor electro-chemical processes, and a number of oxidation and reduction reactions have been studied, demonstrating that it is a powerful tool for understanding electrochemical reaction mechanisms.

Abstract:Desorption electrospray ionization mass spectrometry (DESIMS) previously has been used to capture and identify transient intermediates in electrochemical redox reactions on a platinum-covered rotating waterwheel. We present here two different setups that use a flat surface with porous carbon tape as the working electrode, where analyte-containing microdroplets from the DESI probe contacted with electrolyte supplied onto the surface. One setup had the conducting carbon tape in the form of a grooved inclined plane; the other one was in the form of a flat plane that had the conducting carbon tape as its front surface. Both these setups, which were relatively robust and easy to operate, overcame interference from the electrospray sheath gas that disturbs and dries the flowing electrolyte. By using the inclined-plane device, we observed radical cations and dimer species generated in the electrochemical oxidation of triphenylamine, diimine and imine alcohol in the electrochemical oxidation of uric acid, and the reductive cleavage of disulfide bonds in glutathione disulfide. By using the device with the flat carbon tape, we detected nitrenium ions generated in the electrochemical oxidation of N,N'-dimethyoxydiphenylamine and di-p-tolylamine. Our experience suggests that the flat porous carbon tape surface might be preferable over the inclined plane because of its ease of setup.
