Abstract:BACKGROUND: Cereal fermentations have shown significant potential in improvement and design of the nutritional quality and health effects of foods and ingredients. In the present study, the effect of supplementary Lactobacillus plantarum dy-1 fermented barley (LFB) on obesity in high-fat diet (HFD)-induced obese rats was investigated.RESULTS: LFB treatment showed a lower rate of increase in body weight and percentage of body fat and a reversal of HFD-induced glucose intolerance, with ameliorated hyperinsulinemia, decreased levels of triglycerides and total cholesterol, and inhibited concentration of interleukin (IL)-1 beta, IL-6 and tumor necrosis factor-alpha. Moreover, LFB treatment also showed the strongest inhibition of nuclear factor (NF)-kB activation and exhibited the greatest effects in blocking the degradation of the inhibitor of NF-kB and inhibiting p38 and JNK1 phosphorylation compared with HFD and raw barley treatment.CONCLUSIONS: It was clear that Lactobacillus plantarum dy-1 fermentation significantly improves the anti-obesity properties of barley. The results establish the foundation for ameliorating diet-induced obesity of product with LFB as nutritional supplements. (C) 2016 Society of Chemical Industry

Abstract:We have previously reported an aqueous extract of fermented barley with Lactobacillus plantarum dy-1 (LFBE) has more efficient anti-obesity effect compared with that of Saccharomyces cerevisiae. To further explore associated effects of LFBE on body weight and body fat distribution, and lipid profiles related metabolic outcomes, serum metabolites were analysed using LC-MS-MS and partial least-squares-discriminant analysis (PLS-DA). Obese and lean groups were clearly discriminated from each other on PLS-DA score plot and major metabolites contributing to the discrimination were assigned as lipid metabolites (fatty acids), lipid metabolism intermediates (choline, betaine, carnitine and butyryl-carnitine), amino acids and citric acid. A high-fat diet increased lipid metabolites and decreased lipid metabolism intermediates, indicating that abnormal lipid metabolism induced by a high-fat diet resulted in fat accumulation via decreased β-oxidation. But LFBE can inhibit fat accumulation by reducing lipid metabolites and increasing lipid metabolism intermediates. Furthermore, the level changes of these metabolites can be used to assess the risk of obesity and the therapeutic effect of obesity management.

Results demonstrated that LFBE and its main bioactivate compounds inhibited fat accumulation partly in a daf-2-dependent mechanism. Antiobesity Effect of Lactiplantibacillus Plantarum Fermented Barley Extracts Via the Interactions with Gut Microbiota of the Obese Adult Humans

Abstract:Extracts of fermented barley with Lactiplantibacillus plantarumdy-1 (LFBE) have been reported to exert antiobesity activity in vivo and in vitro. However, whether it worked in clinical trials remained uncovered. In the current study, we conducted a single-blind experiment by enrolling on some obese adult humans with hyperlipemia to test the effects of products containing LFBE (No. ChiCTR1800019614 by the Chinese Clinical Trial Registry). Results indicated that LFBE intervention could ameliorate the symptoms of the obese, characterized by the decrease of body fat percentage, visceral fat area, and serum lipid levels. More interestingly, products containing the beta-glucan ingredients had similar antiobesity effects. 16S rRNA sequencing revealed that LFBE modulated the community structure of the gut microbiota of the obese adults as shown by the diversity analysis. For gut microbiota composition, LFBE significantly increased the Bacteroides genus while decreasing Streptococcus and Haemophilus genus. At the species level, beneficial microorganisms such as Bifidobacterium_longum_subsp_longum, Alistipes_sp_AL-1, Bacteroides_plebeius, and Bacteroides_vulgatus were enriched by LFBE, which was different from the effects of beta-glucan. Furthermore, Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis indicated that LFBE particularly regulated the steroid biosynthesis and hypertrophic cardiomyopathy pathways. Furthermore, correlation analysis suggested that body fat percentage and visceral fat area were significantly positively correlated with Desulfovibrio and Sutterella, the basal metabolic rate was negatively correlated with Haemophilus, and fasting blood glucose was positively correlated with Dialister. In all, this clinical study demonstrated the antiobesity function of LFBE in humans, probably via its interactions with the gut microbiota, and LFBE could be developed as a prebiotic ingredient with antiobesity effects for a healthy diet. Transcriptomics Reveals the Anti-Obesity Mechanism of Lactobacillus Plantarum Fermented Barley Extract

Abstract:Fermentation by lactic acid bacteria can improve the nutritional value and biological function of cereal. Our previous studies have confirmed that Lactobacillus plantarum fermented barley extract (LFBE) can alleviate obesity caused by high-fat diet (HFD) in rats, while the precise mechanism remains unclear. Herein, we explored the effect of LFBE on the adipose tissue in obese rats and its mechanism via transcriptomics technology. Results showed that administration of LFBE in obese rats for 8 weeks significantly alleviated weight gain, reduced fasting blood glucose, and inhibited lipid accumulation. Transmission electron microscope (TEM) observation of adipose tissue found that LFBE held the ability to maintain mitochondria integrity and functionality. Transcriptomics analysis revealed that LFBE increased the expressions of mitochondrial beta-oxidized-related genes, while inhibiting the expressions of fatty acid synthesis-related genes. Furthermore, KEGG (Kyoto Encyclopedia of Genes and Genomes) enrichment analysis and western blotting studies confirmed that LFBE mainly enhanced the energy consumption of adipocytes through the phosphorylation of AMP-Activated Protein Kinase (AMPK) and the mitochondrial proliferation pathway regulated by peroxisome proliferative activated receptor, gamma, coac-tivator 1 alpha (PGC1 alpha). Taken together, these findings indicated that LFBE could ameliorate HFD-induced obesity by activating AMPK/PGC1 alpha axis regulated signaling pathways.
Lactobacillus Plantarum Dy-1 Fermented Barley Extraction Activates White Adipocyte Browning in High-Fat Diet-Induced Obese Rats.

Abstract:High-fat diet (HFD)-induced obesity is caused by the imbalance between energy intake and expenditure. Here, we studied the inhibitory effects of aqueous extracts of fermented barley with Lactobacillus plantarum dy-1 (LFBE) and beta-glucan (BGL) on the obesity induced by HFD. Both LFBE and BGL significantly decreased body weight, suppressed visceral lipid accumulation, improved blood lipid profile, and glucose tolerance in HFD rats. BGL showed no thermogenic capacity, while LFBE enhanced the expression of Uncoupling Protein 1 (UCP1), and brown-specific mRNA (PRDM16, PGC1-α, and CIDEA) levels in brown adipose tissue (BAT) and white adipose tissues (WAT) of HFD rats. In addition, LFBE increased the expression of key genes involved in mitochondria biosynthesis and the mitochondrial respiration function. Further, we demonstrated that proteins extracted from LFBE (LFBE-P) were responsible for triggering brown markers to some extent. In conclusion, LFBE alleviates HFD-induced obesity by activating thermogenic fat bioenergetics and mitochondria biosynthesis. PRACTICAL APPLICATIONS: Barley is one of the most productive crops with pretty low utilization. Our group committed to exploring the application and nutritional value of barley. This work aimed to explore improvements in nutritional function of barley after fermentation by Lactobacillus plantarum dy-1. Our study found that oral administration of LFBE help turning white adipose tissue into a thermogenesis state and activate heat generation function of brown adipose tissue. Its characteristics mentioned above significantly inhibited the body weight and blood lipid of high-fat diet rats. Further, we evidenced that LFBE-P were responsible for triggering brown markers in 3T3-L1 cells. We believe our research plays a great part to relieving high-fat diet-induced obesity and type 2 diabetes with functional diet supplementation.
Metabolomics Reveal the Regulatory Effect of Polysaccharides from Fermented Barley Bran Extract on Lipid Accumulation in HepG2 Cells

Abstract:Barley bran has potential bioactivities due to its high content of polyphenols and dietary fiber, etc. Fermentation has been considered as an effective way to promote the functional activity of food raw materials. In this study, polysaccharides from barley bran extract fermented by Lactiplantibacillus plantarum dy-1 (FBBE-PS) were analyzed, and its effects on lipid accumulation and oxidative stress in high-fat HepG2 cells induced by sodium oleate were evaluated. The results showed that the molecular weight decreased and monosaccharide composition of polysaccharides changed significantly after fermentation. In addition, 50 μg/mL FBBE-PS could reduce the triglyceride (TG) content and reaction oxygen species (ROS) level in high-fat HepG2 cells by 21.62% and 30.01%, respectively, while increasing the activities of superoxide dismutase (SOD) and catalase (CAT) represented by 64.87% and 22.93%, respectively. RT-qPCR analysis revealed that FBBE-PS could up-regulate the lipid metabolism-related genes such as ppar-α, acox-1 and cpt-1α, and oxidation-related genes such as nrf2, ho-1, nqo-1, sod1, cat, etc. The metabolomics analysis indicated that FBBE-PS could alleviate lipid deposition by inhibiting the biosynthesis of unsaturated fatty acids, which is consistent with the downregulation of scd-1 expression. It is demonstrated that fermentation can alter the properties and physiological activities of polysaccharides in barley bran, and FBBE-PS exhibited an alleviating effect on lipid deposition and oxidative stress in high-fat cells.

Anti-obesity Action of Fermented Barley Extracts with Lactobacillus Plantarum Dy-1 and Associated MicroRNA Expression in High-fat Diet-induced Obese Rats

Abstract:Objective To further explore associated effects of Lactobacillus plantarum dy-1（LFBE） on obesity and lipid metabolism at the gene expression level, the expression of micro RNAs（mi RNAs） was investigated in the liver of high-fat diet（HFD） induced obese rats.Methods Three groups of animal models were established. Changes in mi RNA expression in the liver of each group were analyzed by microarray and RT-q PCR, complemented by bioinformatics. Palmitateinduced hepatocellular carcinoma（Hep G2） cells were used as a model to validate the test.Results LFBE treatment groups and HFD groups were observed to be distinctly different with respect to rates of increase in body weight and body fat percentage and triglyceride（TG） and total cholesterol（TC） levels in serum and liver. In addition, the LFBE group showed upregulation of ten mi RNAs and downregulation of five mi RNAs in the liver. Downregulation of mi R-34 a and mi R-212 was observed in the livers of the LFBE group. Gene ontology and kyoto encyelopedia of geues and genomes（KEGG）pathway analysis showed that possible target genes of the deregulated mi RNAs were significantly enriched in the adrenergic and HIF-1 signaling pathways.Conclusion These results demonstrate that LFBE might regulate the expression of mi RNAs in order to inhibit obesity and fatty liver.
Fermented Barley Β‐glucan Regulates Fat Deposition in Caenorhabditis Elegans

Abstract:BACKGROUND: Barley contains a relatively high concentration of the mixed-linkage (1 -> 3) (1 -> 4) beta-glucan, which has been reported to be a functional food with prebiotic potential. In the current study we compared the properties of two neutral barley beta-glucans, obtained from raw barley: raw barley beta-glucan (RBG) and Lactobacillus plantarum dy-1-fermented barley (FBG). RESULTS: Molecular characteristics revealed that the molecular weight of barley beta-glucan decreased from 1.13 x 10(5) D to 6.35 x 10(4) D after fermentation. Fermentation also improved the water / oil holding capacity, solubility, and swelling capacity of barley beta-glucan. Both RBG and FBG significantly improved the locomotive behavior of nematodes, thereby increasing their energy consumption and reducing fat deposition - the effect was more significant with FBG. These effects could potentially depend on nhr-49, TGF-daf-7 mediated pathways and so on, in which nhr-49 factor is particularly required. CONCLUSION: These results suggested that fermentation may enhance in vitro physiological activities of barley beta-glucan, thereby altering the effects on the lipid metabolism in vivo. Abstract:Highland barley is a well-known cereal in Qinghai-Tibet Plateau area with high nutritional value, which has been reported to be a health-promoting grain for the obesity and the diabetes. Fermentation by certain microbiota can improve the flavor property and nutritional characteristics. In the present study, Lactiplantibacillus plantarum and Saccharomyces cerevisiae were singly or jointly applied to ferment highland barley, and the profile of volatile substances and lipid-lowering effects of the respective extracts were analyzed. Results indicated that either L. plantarum or S. cerevisiae or co-fermentation could consume the polysaccharides of highland barley to provide energy, and dramatically increase the contents of total protein and polyphenol. Gas chromatography-mass spectrometry (GC-MS) analysis revealed that the presence of S. cerevisiae was critical for production of the pleasant flavors, especially for the ethyl ester substances including hexadecanoic acid ethyl, hexanoic acid ethyl ester and so on. Meanwhile, we found that fermented highland barley extracts by L. plantarum exhibited stronger lipid-lowering effects in Caenorhabditis elegans than that by S. cerevisiae, while the co-fermentation not only emitted pleasant odors but also exerted high hypolipidemic function. In all, co-fermentation by L. plantarum and S. cerevisiae was proposed to be a promising processing to improve the flavor and functional properties of highland barley.

Abstract:OBJECTIVE:We aimed to explore how fermented barley extracts with Lactobacillus plantarum dy-1 (LFBE) affected the browning in adipocytes and obese rats.METHODS:In vitro, 3T3-L1 cells were induced by LFBE, raw barley extraction (RBE) and polyphenol compounds (PC) from LFBE to evaluate the adipocyte differentiation. In vivo, obese SD rats induced by high fat diet (HFD) were randomly divided into three groups treated with oral gavage: (a) normal control diet with distilled water, (b) HFD with distilled water, (c) HFD with 800 mg LFBE/kg body weight (bw).RESULTS:In vitro, LFBE and the PC in the extraction significantly inhibited adipogenesis and potentiated browning of 3T3-L1 preadipocytes, rather than RBE. In vivo, we observed remarkable decreases in the body weight, serum lipid levels, white adipose tissue (WAT) weights and cell sizes of brown adipose tissues (BAT) in the LFBE group after 10 weeks. LFBE group could gain more mass of interscapular BAT (IBAT) and promote the dehydrogenase activity in the mitochondria. And LFBE may potentiate process of the IBAT thermogenesis and epididymis adipose tissue (EAT) browning via activating the uncoupling protein 1 (UCP1)-dependent mechanism to suppress the obesity.CONCLUSION:These results demonstrated that LFBE decreased obesity partly by increasing the BAT mass and the energy expenditure by activating BAT thermogenesis and WAT browning in a UCP1-dependent mechanism.

Abstract:Fermented barley (FB) flour byLactobacillus plantarumis richer in dietary fiber, polyphenols, gamma-aminobutyric acid, and other biologically active ingredients. This study aimed to determine the impacts of fermented barley - wheat flour compound noodle (FBWN) on glucose and fat metabolism in subjects with metabolic syndrome. This was a single-blinded and parallel 10-week clinical trial study. Subjects were randomly assigned into the trial group (FBWN) and whole wheat noodles group (WWN), and were measured on the beginning of week 3 and the ending of week 10. The glucose level at 30 and 60 min was significantly decreased after FBWN intervention. Levels of fasting blood glucose, HbA1c, and TG were all declined after FBWN intervention compared to before in the trial group. Moreover, the fat mass, fat rate, and visceral fat were decreased by 6.48, 7.19, 6.3 kg after FBWN intervention, respectively, while muscle mass and basal metabolic rate rose 7.44 kg and 252.60 kcal. Practical applications Many studies have illustrated that the extraction of fermented barley held the activities of anti-obesity, antitumor, and so on. Moreover, this present study evaluated the effects of fermented barley byLactobacillus plantarumon patients with metabolic syndrome. Results indicated that FB benefits the subjects on improving plasma glycolipids and insulin sensitivity, decreasing visceral fat level, and increasing satiety. The findings showed that the products of FB may be beneficial to dietary manipulations, thus, reducing the burden of patients.

The contents of total polysaccharide, polyphenol, and protein in the four LFBEs significantly changed as the fermentation time went by and exerted different inhibitory effects on the proliferation of HT-29 cells. Results indicated that LFBE (YangSi No.3) inhibited proliferation of HT-29 cells in a time- and dose-dependent manners. The scanning electron micrograph illustrated that LFBE caused representative apoptotic trait and flow cytometric analysis suggested that LFBE brought about apoptosis by ceasing cell cycle at S phase. Western-blotting results indicated that LFBE promoted apoptosis was relevant to the regulation of apoptosis-related proteins, such as B-cell lymphoma-2 (Bcl-2), Bcl-2-associated X protein (Bax), and the release of Cytochrome-C from mitochondria. Practical applications Abundant studies have reported that extracts of fermented barley held the activities of anti-obesity, antitumor, and so on. However, little information about the comparison in the chemical profile and antiproliferation property among different barley varieties (namely, YangSi barley No.1, YangSi barley No.3, DaZhong 88-91, XiYin No.2) was observed. Results indicated that LFBE (YangSi No.3 barley) exhibited the best inhibitory property by inducing the apoptosis of HT-29 cells. These findings may be beneficial to select a higher nutritional value barley and optimize the fermentation conditions to maximize the bioactive concentration expected in foods for the human.
Lactiplantibacillus Plantarum Fermented Barley Extracts Ameliorate High-Fat-diet-induced Muscle Dysfunction Via Mitophagy

Abstract:BACKGROUND A reduced level of fatty acid oxidation (FAO) by skeletal muscle leads to the accumulation of intermuscular fat (IMF), which is linked to impaired exercise capacity. Previously, we have reported that Lactiplantibacillus plantarum fermented barley extract (LFBE) has effective anti-obesity properties. In this study, the effects of LFBE on muscle were investigated. RESULTS LFBE improved running endurance and muscle strength, which was caused by the elevation of FAO in muscle. In addition, LFBE renovated muscle regeneration through the upregulation of paired box 7 and myogenic differentiation 1 expression avoiding the injury of skeletal muscle fibers. Furthermore, total polyphenol isolated from LFBE (FTP) reinforced mobility and showed a significant protective effect on maintaining muscle fiber morphogenesis in Caenorhabditis elegans. Transmission electron microscope observation suggested FTP induced mitophagy in C. elegans body wall muscle, which was strongly connected with enhanced FAO in mitochondria. CONCLUSIONS Our findings highlighted the beneficial bioactivities of FTP and its potential application for stimulating mitophagy and muscle function in obese individuals. Barley Protein LFBEP-C1 from Lactiplantibacillus plantarum dy-1 Fermented Barley Extracts by Inhibiting Lipid Accumulation in a Caenorhabditis elegans Model

Abstract:Objective: This study aimed to investigate the lipid-lowering activity of LFBEP-C1 in high glucose-fed Caenorhabditis elegans (C. elegans). Methods: In this study, the fermented barley protein LFBEP-C1 was prepared and tested for its potential anti-obesity effects on C. elegans. The worms were fed Escherichia coli OP50 ( E. coli OP50), glucose, and different concentrations of LFBEP-C1. Body size, lifespan, movement, triglyceride content, and gene expression were analyzed. The results were analyzed using ANOVA and Tukey's multiple comparison test. Results: Compared with the model group, the head-swing frequency of C. elegans in the group of LFBEP-C1 at 20 μg/mL increased by 33.88%, and the body-bending frequency increased by 27.09%. This indicated that LFBEP-C1 improved the locomotive ability of C. elegans. The average lifespan of C. elegans reached 13.55 days, and the body length and width of the C. elegans decreased after LFBEP-C1 intake. Additionally, LFBEP-C1 reduced the content of lipid accumulation and triglyceride levels. The expression levels of sbp-1, daf-2, and mdt-15 significantly decreased, while those of daf-16, tph-1, mod-1, and ser-4 significantly increased after LFBEP-C1 intake. Changes in these genes explain the signaling pathways that regulate lipid metabolism. Conclusion: LFBEP-C1 significantly reduced lipid deposition in C. elegans fed a high-glucose diet and alleviated the adverse effects of a high-glucose diet on the development, lifespan, and exercise behavior of C. elegans. In addition, LFBEP-C1 regulated lipid metabolism mainly by mediating the expression of genes in the sterol regulatory element-binding protein, insulin, and 5-hydroxytryptamine signaling pathways.
Anti-obesity effect of L-arabinose via ameliorating insulin resistance and modulating gut microbiota in obese mice

Abstract:The global prevalence of obesity, a chronically trophic metabolic disease, has garnered significant attention. L-arabinose, a unique functional sugar that improves insulin resistance and intestinal environment while promoting probiotic proliferation, was investigated in this study for its potential in preventing obesity induced by a high-fat and high-sugar (HFHS) diet in mice. L-arabinose group was intragastrically administered with 0.4 mL 60 mg/(kg bw) L-arabinose for 8 weeks. Metformin group was intragastrically administered at 0.4 mL 300 mg/(kg body weight (bw)), as a positive control group. Treatment with L-arabinose resulted in a reduction of various obesity symptoms, such as precented weight gain, increased liver/body ratio, decreased insulin, homeostasis model assessment for insulin resistance (HOMA-IR) index and lipopolysaccharide (LPS) levels, as well as improved insulin resistance, reduced fat volume, inhibited hepatic steatosis, and repaired the pancreas. The L-arabinose treatment also improved lipid metabolism and inflammatory response, as well as decreased the Firmicutes/Bacteroidetes ratio at the phylum level, and increased the relative abundance of Parabacteroides_gordonii and Akkermansia_muciniphila at the species level. Based on these results, L-arabinose could be a promising candidate for combating obesity and obesity-related diseases by regulating insulin resistance and gut microbiota.

Abstract:The phytochemical profiles of fermented barley with Lactiplantibacillus plantarum dy-1 (LFBE) and its antioxidant activity were investigated. Nine components including cinnamaldehyde, homovanillic acid, coumalic acid, quercetin, docosahexaenoic acid methyl ester, gallic acid, cafestol, 6-hydroxycaproic acid, and indole-3-lactic acid were identified by HPLC-MS/MS and evaluated as potential antioxidants in LFBE. DPPH and ABTS scavenging ability, superoxide dismutase (SOD) activity, reactive oxygen species (ROS) level in HepG2 cells, and the cellular antioxidant activity (CAA) were assessed. The results indicated that the nine components showed good radical scavenging capacities, most of them had the abilities of reducing ROS level and promoting SOD activity. In CAA tests, coumalic acid exhibited the highest value (11.51 mu mol of QE/100 g), while gallic acid showed the lowest value (3.47 mu mol of QE/100 g), which was not consistent with the trend of other evaluations. This study provided a basis for understanding the effects of fermentation on bioactivity of barley.

Abstract:Fermentation is an effective method for improving the nutritional quality and functional characteristics of grains. This study investigated changes in the structural, physicochemical, and functional properties of fermented barley dietary fiber (FBDF) exerted by Lactiplantibacillus plantarum dy-1 (Lp. plantarum dy-1) as well as its in vitro fecal fermentation characteristics. Lp. plantarum dy-1 fermentation remarkably changed the structure of FBDF, including the microstructure and monosaccharide components, correlating with improved water or oil retaining and cholesterol adsorption capacities. Additionally, Lp. plantarum dy-1 fermentation significantly (p < 0.05) promoted the release of bound phenolics from 6.24 mg g-1 to 6.93 mg g-1 during in vitro digestion, contributing to the higher antioxidant capacity and inhibitory activity of α-amylase and pancreatic lipase compared with those of raw barley dietary fiber (RBDF). A total of 14 phenolic compounds were detected in the supernatants of digestion and fermentation samples. During colonic fermentation, FBDF significantly increased the production of acetate, propionate, and butyrate (p < 0.05), inhibited the growth of Escherichia-Shigella, and promoted the abundance of SCFA-producing microbiota such as Faecalibacterium and Prevotella_9. In conclusion, Lp. plantarum dy-1 fermentation enhanced the physicochemical properties and in vitro fermentation characteristics of barley dietary fiber, representing a promising bioprocessing technology for modifying barley bran.
