Abstract:A new chromone derivative, aspergione A (1), along with seven known metabolites, was isolated from a mangrove endophytic fungus, Aspergillus sp. GXNU-B1, which was collected from mangrove Acanthus ilicifolius L. Their structures and the absolute configuration of 1 were elucidated based on the analysis of HR-ESI-MS, NMR, and ECD calculation. Compounds 1-8 were evaluated for their anti-inflammatory effects on the production of nitricoxide (NO). Compounds 1 and 8 have potent inhibitory effects against NO production in activated macrophages with IC50 values of 38.26 and 44.30 μM, respectively.

Abstract:From the marine sponge Xestospongia exigua, fungal isolates of Aspergillus versicolor (Vuill)Triab were obtained. Isolation and purification of ethyl acetate extracts from culture filtrates of the fungus led to yield six new chromone derivatives namely aspergione A, aspergione B, aspergione C, aspergione D. aspergione E, aspergione F. The structures of all the new compounds were established on the basis of extensive spectroscopy (UV, MS, H-1 and C-13 NMR, COSY, HMQC and HMBC) analysis.

Abstract:A new sesquiterpenoid derivative, named aspergiketone (1), along with seven known compounds (2-8) were isolated from the coastal saline soil fungus Aspergillus fumigatus. Their structures were elucidated by spectroscopic analysis, and by comparison of experimental and reported data. The absolute configuration of compound 1 was defined by X-ray diffraction analysis. Compound 1 was cytotoxic towards HL-60 and A549 cell lines with IC50 values of 12.4 and 22.1 mu M, respectively.

Abstract:Asperginine (1), an alkaloid possessing a rare skeleton, was isolated from the cultural broth of the marine fungus Aspergillus sp. (Z-4) isolated from the gut of the marine isopod Ligia oceanica. The planar structure and relative configuration of 1 was determined by analysis of NMR and mass spectral data. Its absolute configuration was elucidated by Marfey's method, together with NOESY correlations of key hydrogen atoms. The cytotoxicity against prostate cancer PC3 and human HCT116 was assayed by the MTT method. Unfortunately, asperginine did not show any activity.

Abstract:A novel polycyclic aspochalasin, aspochalazine A (1), was isolated along with three related known compounds 2-4 from the culture broth of Aspergillus sp., which was found in the gut of a marine isopod Ligia oceanica. The structures were determined on the basis of ESI-MS, NMR spectrum data and single-crystal X-ray diffraction. This is the first report about aspochalasin derivative with an azabicyclo. Cytotoxicity against the prostate cancer PC3 cell line was assayed by MTT method. Aspochalazine A showed weak activity. A plausible biosynthetic pathway for aspochalazine A was proposed. Abstract:Aspergiolide A (1), an anthraquinone derivative with naphtho[1,2,3-de]chromene-2,7-dione skeleton, has been isolated from cultures of a marine-derived fungus Aspergillus glaucus. The structure and stereochemistry of 1 was determined by NMR and X-ray diffraction analyses. Compound 1 showed selective cytotoxicities against A-549, HL-60, BEL-7402, and P388 cell lines.

Abstract:From the marine sponge Xestospongia exigua collected in Indonesia the fungus Aspergillus versicolor was isolated. Following cultivation in a seawater-based medium seven new angular tricyclic chromone derivatives (1-7) were obtained from the mycelia and culture filtrate. Compounds 2-7 contain an additional dihydropyran ring system which is replaced by a pyridine ring in 1. The structures of the new natural products were established on the basis of extensive one- and two-dimensional NMR spectroscopic studies (H-1, C-13, COSY, HMQC, HMBC, NOE difference spectra) as well as on mass spectral analysis.

Abstract:Chemical investigation of the fermentation extract of the mangrove endophytic fungus Aspergillus sp. GXNU-A1, isolated from Acanthus ilicifolius L., discovered an undescribed pair of enantiomers (asperphenyltones A and B (±1)), together with four previously described metabolites: nodulisporol (2), isosclerone (3), 2,3,4-trihydroxy-6-(hydroxymethyl)-5-methylbenzyl alcohol (4), and 4,6-dihydroxy-5-methoxy-7-methyl-1,3-dihydroisobenzofuran (5). Analyses of the 1D and 2D NMR spectroscopic data of the compounds supported their structural assignments. The presence of the asperphenyltones A and B, which are a pair of enantiomers, was established by HR-ESI-MS, 1D and 2D NMR data and confirmed by single-crystal X-ray diffraction analysis. Metabolites 1-5 were evaluated for their anti-inflammatory effects on the production of nitric oxide (NO), and 1, 3, and 4 showed significant potential inhibitory activities against NO production in activated macrophages with IC50 values of 26-40 μM, respectively.

Abstract:Marine fungi are a promising source of novel bioactive natural products with diverse structure. In our search for new bioactive natural products from marine fungi, three new phenone derivatives, asperphenone A-C (1-3), have been isolated from the ethyl acetate extract of the fermentation broth of the mangrove-derived fungus, Aspergillus sp. YHZ-1. The chemical structures of these natural products were elucidated on the basis of mass spectrometry, one- and two-dimensional NMR spectroscopic analysis and asperphenone A and B were confirmed by single-crystal X-ray crystallography. Compounds 1 and 2 exhibited weak antibacterial activity against four Gram-positive bacteria, Staphylococcus aureus CMCC(B) 26003, Streptococcus pyogenes ATCC19615, Bacillus subtilis CICC 10283 and Micrococcus luteus, with the MIC values higher than 32.0 mu M.

Abstract:One new anthraquinone derivative, named 8-O-methylnidurufin ( 1 ), together with five known analogues ( 2 – 6 ), have been isolated from a gorgonian-derived fungus, Aspergillus sp. The structures were elucidated by combined spectroscopic methods including 1D and 2D NMR spectral data. All isolated metabolites were evaluated for their cytotoxic and antibacterial activities in vitro. Compound 2 showed significant cytotoxic activity against K562 and HL-60 cell lines with IC 50 values of 0.87 and 1.46 μM, respectively.

Abstract:Four new isochromane derivatives (1-4) along with the known peniciphenol (5) and (R)-2-(hydroxymethyl)-3-(2-hydroxypropyl)phenol (6) were isolated from the EtOAc extract of the fermentation broth of the mangrove fungus, Aspergillus ustus 094102. The structures of the new compounds including the absolute configuration were elucidated on the basis of spectroscopic analysis, CD and ECD calculation. Compounds 1 and 2 exhibited alpha-glucosidase inhibition and antioxidation against DPPH radical with IC50 values of 1.4 mM and 25.7 mu M, respectively.

Abstract:Three new asperentin-type compounds, 6-O-alpha-D-ribosylasperentin (1) and 6-O-alpha-D-ribosyl-8-O-methylasperentin (2) and 5-hydroxyl-6-O-methylasperentin (3), along with asperentin (4) and its known analogues (5-9), were isolated from a halotolerant Aspergillus sp. strain F00785, an endotrophic fungus from marine alga. Their structures were determined using extensive NMR and HRESIMS spectroscopic analysis, including the X-ray crystallographic data for the assignment of the absolute configurations of compound 9. Compound 4 exhibited highly potent inhibitory activity against crop pathogens, Colletotrichum gleosporioides Penz. and Colletotrichum gleosporioides (Penz.) Sacc.

Abstract:A new xanthenone derivative, 3-hydroxy pinselin (1), together with five known analogues (2-6) were isolated from the marine-derived fungus Aspergillus versicolor MF160003. Their structures were identified by extensive 1D- and 2D-NMR, and high-resolution mass spectrometry data. Compounds 5 and 6 showed moderate bioactivities against BCG with MIC values of 40 and 20 mu g/mL, respectively.

These structures were elucidated by spectroscopic analysis and the absolute configuration of 1 was unambiguously determined by ECD calculation. Abstract:Two new polyketides, aspergchromones A (1) and B (2), together with five known compounds, secalonic acid D (3), noreugenin (4), (3S)-5-hydroxymellein (5), (4S)-6-hydroxyisosclerone (6), and (-)-regiolone (7), were isolated from the ethyl acetate extract of marine sponge-derived fungus Aspergillus sp. SCSIO XWS03F03. Their structures were elucidated by means of spectroscopic techniques (1D and 2D NMR, MS, UV, and IR). The absolute configurations of the new compounds were established by ECD calculations. Compound 3 showed moderate antimicrobial activity.

Abstract:Two new meroterpenoids, aspergienynes O and P (1 and 2), one new natural compound, aspergienyne Q (3), and a new α-pyrone derivative named 3-(4-methoxy-2-oxo-2H-pyran-6-yl)butanoic acid (4) were isolated from the mangrove endophytic fungal strain Aspergillus sp. GXNU-Y85, along with five known compounds (5–9). The absolute configurations of those new isolates were confirmed through extensive analysis using spectroscopic data (HRESIMS, NMR, and ECD). The pharmacological study of the anti-proliferation activity indicated that isolates 5 and 9 displayed moderate inhibitory effects against HeLa and A549 cells, with the IC50 values ranging from 16.6 to 45.4 μM.

Abstract:Four new chromones, phomochromenones D–G (1–4), along with four known analogues, diaporchromone A (5), diaporchromanone C (6), diaporchromanone D (7), and phomochromenone C (8), were isolated from the culture of Phomopsis asparagi DHS-48 from Chinese mangrove Rhizophora mangle. Their structures were elucidated on the basis of comprehensive spectroscopic analysis. The absolute configurations of 1 and 4 were assigned on the basis of experimental and calculated electronic circular dichroism (ECD) data, and those of enantiomers 2 and 3 were determined by a modified Mosher’s method and basic hydrolysis. To the best of our knowledge, phomochromenones D–F (1–4) possessing a 3-substituted-chroman-4-one skeleton are rarely found in natural sources. Diaporchromone A (5) showed moderate to weak immunosuppressive activity against T and/or B lymphocyte cells with IC50 of 34 μM and 117 μM.
