Abstract:Professor Zhi-Fei Dai obtained his Ph.D. degree at Chinese Academy of Sciences in Beijing. He is now a professor at Biomedical Engineering Department, Peking University. His research is focused on contrast agent, nanomedicine and molecular imaging. He is a chief scientist for National Key Research and Development Program of China. He works as a member of editorial board for several international and national journals such as Bioconjugate Chemistry, Theranostics, Journal of Interdisciplinary Nanomedicine, IET Nanobiotechnology, Chinese Journal of Nuclear Medicine and Molecular Imaging and so on. He is now serving as a vice chairman in Chinese Society of Biomedical Engineering in Photonics, a standing committee member in Ultrasound Molecular Imaging, Chinese Association of Ultrasound in Medicine and Engineering, an executive member of the council in Chinese Society for Functional Materials, and a committee member in the Biomedical Engineering in Ultrasonics, the Acoustic Society of China. Abstract:Precise nanomedicine has been extensively explored for efficient cancer imaging and targeted cancer therapy, as evidenced by a few breakthroughs in their preclinical and clinical explorations. Here, we demonstrate the recent advances of intelligent cancer nanomedicine, and discuss the comprehensive understanding of their structure-function relationship for smart and efficient cancer nanomedicine including various imaging and therapeutic applications, as well as nanotoxicity. In particular, a few emerging strategies that have advanced cancer nanomedicine are also highlighted as the emerging focus such as tumor imprisonment, supramolecular chemotherapy, and DNA nanorobot. The challenge and outlook of some scientific and engineering issues are also discussed in future development. We wish to highlight these new progress of precise nanomedicine with the ultimate goal to inspire more successful explorations of intelligent nanoparticles for future clinical translations.

Abstract:Nanotechnology is a new technology for observing and manipulating atoms and molecules in the range of 0.1-100 nm. The development of nanotechnology provides a new way for biomedicine. Based on the unique physical and chemical properties of nanomaterials, the novel biosensor based on bio-nanotechnology can be used for early diagnosis and therapy of tumors. Nowadays, the application of nanomedicine is mainly focused on the following points: tissue engineering and regenerative medicine, the accurate diagnosis and real-time monitoring of the disease, novel imaging and analysis techniques, and multifunctional drug delivery system and targeting therapy. Nanotechnology will provide effective means for accurate diagnosis and treatment of cancer. This article reviews the new progress of nanotechnology in cancer diagnosis and treatment.

Abstract:Cancer is a serious health problem which increasingly causes morbidity and mortality worldwide. It causes abnormal and uncontrolled cell division. Traditional cancer treatments include surgery, chemotherapy, radiotherapy and so on. These traditional therapies suffer from high toxicity and arouse safety concern in normal area and have difficulty in accurately targeting tumour. Recently, a variety of nanomaterials could be used for cancer diagnosis and therapy. Nanomaterials have several advantages, e.g. , high concentration in tumour via targeting design, reduced toxicity in normal area and controlled drug release after various rational designs. They can combine with many types of biomaterials in order to improve biocompatibility. In this review, we outlined the latest research on the use of bioresponsive nanomaterials for various cancer imaging modalities (magnetic resonance imaging, positron emission tomography and phototacoustic imaging) and imaging-guided therapy means (chemotherapy, radiotherapy, photothermal therapy and photodynamic therapy), followed by discussing the challenges and future perspectives of this bioresponsive nanomaterials in biomedicine.

Abstract:Cancer is a major public health problem in the world. There is a great need to apply novel technologies and drugs to revolutionize multiple aspects of cancer diagnosis and therapy. Advances in nanotechnology and nanomaterials have the potential to achieve the objective of early diagnosis and early therapy of cancer in the future. During the past few years, fullerene and its derivatives have been considered as some of the most promising nanomaterials because of their unique properties that enable a variety of medicinal applications. They can deliver drugs or small therapeutic molecules to the cancer cells. In this review, we will discuss how fullerene derivatives have been introduced into the field of cancer diagnosis and therapy. It will be highlighted that fullerene derivatives are used as anti-tumor drugs. Furthermore, preparation, characterization, pharmacokinetics and bio-distribution of fullerene and its derivatives reported in recent years will be summarized.

Abstract:Anticancer nanomedicines have been studied over 30 years, but fewer than 10 formulations have been approved for clinical therapy today. Despite abundant options of anticancer drugs, it remains challenging to have agents specifically target cancer cells while reducing collateral toxicity to healthy tissue. Nanocompartments that can be selective toward points deeply within malignant tissues are a promising concept, but the heterogeneity of tumor tissue, inefficiency of cargo loading and releasing, and low uniformity of manufacture required from preclinical to commercialization are major obstacles. Technological advances have been made in this field, creating engineered nanomaterials with improved uniformity, flexibility of cargo loading, diversity of surface modification, and less inducible immune responses. This review highlights the developmental process of approved nanomedicines and the opportunities for novel materials that combine insights of tumors and nanotechnology to develop a more effective nanomedicine for cancer patients.

Abstract:Traditional cancer therapeutics have been criticized due to various adverse effects and insufficient damage to targeted tumors. The breakthrough of nanoparticles provides a novel approach for upgrading traditional treatments and diagnosis. Actually, nanoparticles can not only solve the shortcomings of traditional cancer diagnosis and treatment, but also create brand-new perspectives and cutting-edge devices for tumor diagnosis and treatment. However, most of the research about nanoparticles stays in vivo and in vitro stage, and only few clinical researches about nanoparticles have been reported. In this review, we first summarize the current applications of nanoparticles in cancer diagnosis and treatment. After that, we propose the challenges that hinder the clinical applications of NPs and provide feasible solutions in combination with the updated literature in the last two years. At the end, we will provide our opinions on the future developments of NPs in tumor diagnosis and treatment.

Abstract:Cancer remains a significant risk to human health. Nanomedicine is a new multidisciplinary field that is garnering a lot of interest and investigation. Nanomedicine shows great potential for cancer diagnosis and treatment. Specifically engineered nanoparticles can be employed as contrast agents in cancer diagnostics to enable high sensitivity and high-resolution tumor detection by imaging examinations. Novel approaches for tumor labeling and detection are also made possible by the use of nanoprobes and nanobiosensors. The achievement of targeted medication delivery in cancer therapy can be accomplished through the rational design and manufacture of nanodrug carriers. Nanoparticles have the capability to effectively transport medications or gene fragments to tumor tissues via passive or active targeting processes, thus enhancing treatment outcomes while minimizing harm to healthy tissues. Simultaneously, nanoparticles can be employed in the context of radiation sensitization and photothermal therapy to enhance the therapeutic efficacy of malignant tumors. This review presents a literature overview and summary of how nanotechnology is used in the diagnosis and treatment of malignant tumors. According to oncological diseases originating from different systems of the body and combining the pathophysiological features of cancers at different sites, we review the most recent developments in nanotechnology applications. Finally, we briefly discuss the prospects and challenges of nanotechnology in cancer.
Nanoscale Radiotheranostics for Cancer Treatment: From Bench to Bedside

Abstract:In recent years, the application of radionuclides-containing nanomaterials in cancer treatment has garnered widespread attention. The diversity of nanomaterials allows researchers to selectively combine them with appropriate radionuclides for biomedical purposes, addressing challenges faced by peptides, small molecules, or antibodies used for radionuclide labeling. However, with advantages come challenges, and nanoradionuclides still encounter significant issues during clinical translation. This review summarized the recent progress of nanosized radionuclides for cancer treatment or diagnosis. The discussion began with representative radionuclides and the methods of incorporating them into nanomaterial structures. Subsequently, new combinations of nanomaterials and radionuclides, along with their applications, were introduced to demonstrate their future trends. The benefits of nanoradionuclides included optimized pharmacokinetic properties, enhanced disease-targeting efficacy, and synergistic application with other treatment techniques. Besides, the basic rule of this section was to summarize how these nanoradionuclides can truly impact the diagnosis and therapy of various cancer types. In the last part, the focus was devoted to the nanoradionuclides currently applicable in clinics and how to address the existing issues and problems based on our knowledge.

Abstract:The field of nanomedicines and nanomaterials has been rapidly developing with several considerable breakthroughs in the treatment of cancer. However, some challenges may hinder their clinical translation, such as controversial enhanced permeability and retention (EPR) effect, potential nanosafety issues, limited large-scale production of nanomedicines with complex prescription. Herein, an overview of currently emerged cancer nanomedicines is summarized, that mainly focus on the recent progress in chemotherapy, phototherapy (photothermal therapy (PTT) and photodynamic therapy (PDT)), radiation therapy, chemodynamic therapy (CDT), immunotherapy (including vaccine delivery) and combined therapy. Furthermore, the exist challenges of cancer nanomedicines are discussed, and several perspectives based on our group’s research experience are put forward, including nanomedicines with non-intravenous administration route, enrichment of nanomedicines knowledge repository and advanced strategies (predictive toxicology, high throughput screening, multiomics and cellular pharmacokinetics, data mining, etc.) to achieve the goal of “safe-by-design”, and simple component, multimodal theranostic nanosystems to facilitate large-scale production.

Abstract:Cancer continues to pose a severe threat to global health, making pursuing effective treatments more critical than ever. Traditional therapies, although pivotal in managing cancer, encounter considerable challenges, including drug resistance, poor drug solubility, and difficulties targeting tumors, specifically limiting their overall efficacy. Nanomedicine’s application in cancer therapy signals a new epoch, distinguished by the improvement of the specificity, efficacy, and tolerability of cancer treatments. This review explores the mechanisms and advantages of nanoparticle-mediated drug delivery, highlighting passive and active targeting strategies. Furthermore, it explores the transformative potential of nanomedicine in tumor therapeutics, delving into its applications across various treatment modalities, including surgery, chemotherapy, immunotherapy, radiotherapy, photodynamic and photothermal therapy, gene therapy, as well as tumor diagnosis and imaging. Meanwhile, the outlook of nanomedicine in tumor therapeutics is discussed, emphasizing the need for addressing toxicity concerns, improving drug delivery strategies, enhancing carrier stability and controlled release, simplifying nano-design, and exploring novel manufacturing technologies. Overall, integrating nanomedicine in cancer treatment holds immense potential for revolutionizing cancer therapeutics and improving patient outcomes.

Abstract:Key PointsAlthough first-generation cancer nanomedicines have been in clinical practice for more than 20 years, the total number of papers related to 'nanoparticle' on PubMed nearly doubled every 2 years between 2000 and 2014, and more than half of the papers on nanoparticles have been published in the past 5 years, indicating that our knowledge and arsenal of cancer nanomedicines is rapidly expanding.Cancer nanomedicines accumulate in solid tumours through the enhanced permeability and retention (EPR) effect, which is increasingly appreciated to be complex, dynamic and heterogeneous across tumours and even within the same tumour.Identifying biomarkers for the EPR effect may enable selection of cancer patients most likely to benefit from nanotherapeutics, prompting the development of personalized nanomedicine.Effective systemic delivery of nanotherapeutics to solid tumours requires a deeper understanding of the biological factors involved, such as nanoparticle–protein interaction, blood circulation, extravasation to and interaction with the perivascular tumour microenvironment (TME), tumour tissue penetration, tumour cell internalization and intracellular trafficking.The physicochemical properties of nanotherapeutics (for example, size, geometry, surface features, elasticity, stiffness, porosity, composition, targeting ligand and drug release kinetics) affect systemic delivery to tumours, thus determining the EPR effect and therapeutic outcomes.Targeting the TME and the premetastatic niche with nanotechnologies offers another promising strategy for cancer therapy.Addressing the challenges of controllable, reproducible and scalable nanoparticle synthesis will facilitate the clinical translation of cancer nanomedicines.

Abstract:In recent years, the incidence of cancer has been increasing year by year, and the burden of the disease and the economic burden caused by it has been worsening. Although chemotherapy, immunotherapy, targeted therapy and other therapeutic means continue to progress, they still inevitably have problems such as high toxicity and side effects, susceptibility to drug resistance, and high price. Photothermal therapy and photodynamic therapy have demonstrated considerable advantages in cancer imaging and treatment due to their minimally invasive and selective nature. However, their development has been constrained by challenges related to drug delivery. In recent times, drug delivery systems constructed based on supramolecular chemistry have been the subject of considerable interest, particularly in view of their compatibility with the high permeability and long retention effect of tumors. Furthermore, the advantage of dissociating the active ingredient under pH, light and other stimuli makes them unique in cancer therapy. This paper reviews the current status of supramolecular nanomedicines in cancer therapy, elucidating the challenges faced and providing a theoretical basis for the efficient and precise treatment of malignant tumors.

Abstract:As one of the most important cancer treatment strategies, conventional chemotherapy has substantial side effects and leads easily to cancer treatment failure. Therefore, exploring and developing more efficient methods to enhance cancer chemotherapy is an urgently important problem that must be solved. With the development of nanotechnology, nanomedicine has showed a good application prospect in improving cancer chemotherapy. In this review, we aim to present a discussion on the significant research progress in nanomedicine for enhanced cancer chemotherapy. First, increased enrichment of drugs in tumor tissues relying on different targeting ligands and promoting tissue penetration are summarized. Second, specific subcellular organelle-targeted chemotherapy is discussed. Next, different combinational strategies to reverse multidrug resistance (MDR) and improve the effective intracellular concentration of therapeutics are discussed. Furthermore, the advantages of combination therapy for cancer treatment are emphasized. Finally, we discuss the major problems facing therapeutic nanomedicine for cancer chemotherapy, and propose possible future directions in this field.
