Abstract:The incorporation of the privileged amino functionality is of paramount importance in organic synthesis. In contrast to the well-developed amination methods for alkenes, the dearomative amination of arenes is largely underexplored due to the inherently inert reactivity of arene π-bonds and selectivity challenges. Herein, we report an intermolecular dearomative aminofunctionalization via direct nucleophilic addition of simple amines to chromium-bound arenes. This multicomponent 1,2-amination/carbonylation reaction enables rapid access to complicated alicyclic compounds containing amino and amide functionalities from benzene derivatives under CO-gas-free conditions, which also represents the first application of nitrogen-based nucleophiles in η6 -coordination-induced arene dearomatizations.

Abstract:Catalytic asymmetric Diels−Alder reaction is one of the most powerful reactions in organic chemistry. It is still a challenge to achieve high and general exo-selectivity. A novel bispyrrolidine diboronate compound has been derived from the reaction of two molecules of oxazaborolidine with one molecule of water. Upon activation, it effectively catalyzes (0.4-5 mol % loading) the Diels–Alder reaction between a variety of mono- and di-carbonyl activated dienophiles and dienes with better than 20:1 regioselectivity, up to >99:1 enantioselectivity, and better than 20:1 exo/endo selectivity. Mechanistic studies show a remarkable nonlinearity in enantioselectivity, and a second-order kinetics with respect to the catalyst concentration. A model is proposed to rationalize the above observations. The reaction is expected to find wide applications in organic synthesis.
Inducing high exo selectivity in Diels-Alder reaction by dimethylborane substituent: a DFT study

Abstract:In this work, the role of Lewis acid-base (LAB) interaction on the stereoselectivity of the Diels-Alder (DA) reaction has been studied by DFT in gas and solution (dichloromethane) phases. The calculations were performed at the B3LYP/6-311G++ (d, p) level. Two different series of DA reactions were investigated: (1)-three mono-substituted cyclopentadienes + dimethyl(vinyl)borane; (2)-five α,β-unsaturated carbonyl compounds + cyclopenta-2,4-dien-1-yldimethylborane. The reacting diene and dienophile pairs were chosen to restrict LAB interaction to the exo reaction pathway. It was found that in some of the examined cases, the favorable LAB interaction is so strong that it can lead to a completely exo-selective DA reaction. Furthermore, a novel multistep synthetic method was hypothesized for preparing exo cycloadduct with near 100% stereoselectivity. Our results can open up new avenues toward the rational design of exo-selective DA reactions for synthesizing novel bioorganic compounds.

Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:AbstractA highly enantioselective catalytic system for exo‐Diels–Alder reactions was developed based on the newly discovered bispyrrolidine diboronates (BPDB). Activated by various Lewis or Brønsted acids, BPDB can catalyze highly stereoselective asymmetric exo‐Diels–Alder reactions of monocarbonyl‐based dienophiles. When 1,2‐dicarbonyl‐based dienophiles are used, the catalyst can sterically distinguish between the two binding sites, which leads to highly regioselective asymmetric Diels–Alder reactions. BPDB can be prepared as crystalline solids on a large scale and are stable under ambient condition. Single‐crystal X‐ray analysis of the structure for acid‐activated BPDB indicated that its activation involves cleavage of a labile B←N bond.

Abstract:Unusual anti-1,4-adducts of anthracene derivatives and anti-adducts of inert arenes with rigid scaffolds have been obtained via AlCl3-assisted Diels-Alder reaction in good to excellent yields under mild conditions. Further derivation of 1,4-adducts gave π-conjugated polymers which could act as sensors of explosive species. This highly efficient synthesis method provides versatile approaches to solid-state emissive π-conjugated polymers.

Abstract:To study the intramolecular Diels-Alder cycloadditon of 2H-chromen-2-one as a diene, a series of chiral N-allyl-N-benzylamides that contain a 2H-chromen-2-one moiety were designed for the synthesis of benzo[f]isoindol-1-ones via an intramolecular Diels-Alder and a subsequent retro-Diels-Alder cycloaddition with the expulsion of CO2. Both the yield (80%-89%) and absolute stereocontrol of the tandem reaction were high when an electron-withdrawing group was attached to the dienophile. The double bond in the styrene substructure remained in the products could be further derivatized by dihydroxylation. Abstract:The Diels-Alder reactions of ethyl alpha-bromoacrylate 1 with open-chain dienes 2 were conducted under thermal or Lewis acid-catalysis conditions. In most cases, the cyclic adducts of 1-bromocyclohex-3-enecarboxylates 3 were formed in high yields with good regio- and stereoselectivity. Subsequent E2-elimination by treatment with DBU provided the corresponding 1,3- or 1,4-cyclohexadienecarboxylates depending on the relative configuration of the products. Starting from myrcene (7-methyl-3-methyleneocta-1,6-diene) the reaction sequence afforded the ester precursor of Georgywood with good yields.

Abstract:Reactions of 1,2,3,4-tetrasubstituted 1,3-butadienes with maleic anhydride and other dienophiles were investigated with or without addition of Lewis acid. When the silylated 1,3-butadienes, such as 1,4-bis(trimethylsilyl)-1,3-butadienes or 1-trimethylsilyl-1,3-butadienes, were treated with maleic anhydride in the presence of 2equiv of AlCl3, multi-substituted 7-norbornenones of well-defined exo,exo-disubstituted patterns were produced by an unprecedented and synthetically useful tandem process. Although some tetrasubstituted 1,3-butadienes could react directly with maleic anhydride under relative harsh conditions to afford Diels–Alder cycloadducts, the reactions, in the presence of 1equiv of AlCl3, afforded the corresponding cycloadducts in higher yields under mild conditions. These results showed that the size and substitution pattern of substituents on the butadienyl skeleton played a very important role in the reactivity of butadienes as a partner for the Diels–Alder reaction and Lewis acid could promote and/or realize the process of a Diels–Alder reaction.
