Abstract:Peroxynitrite (ONOO-) has been proven to participate in various physiological and pathological processes, and may also be a contributing factor in many diseases. In view of this, there is a need to develop detection tools for unambiguously tracking a small amount of endogenous ONOO- to reveal its exact mechanisms. In this paper, a colorimetric and red-emitting fluorescent probe Red-PN, based on a rhodamine-type fluorophore and hydrazide reactive site is described. The probe Red-PN possesses the advantages of rapid response (within 5 s), visual color change (from colorless to pink), preeminent sensitivity (detection limit = 4.3 nM) and selectivity. Because of these outstanding performances, it was possible to accurately detect endogenous ONOO-. It was encouraging that the probe Red-PN could be used effectively for tracking the relatively low levels of endogenous and exogenous ONOO-in living cells and zebrafish. Thus, it is envisioned that the probe Red-PN would have promising prospects in applications for imaging ONOO- in a variety of biological settings.

Abstract:As a reactive nitrogen species (RNS), peroxynitrite (ONOO-) is known for its crucial functions in biological systems. Excessive formation of ONOO- is very closely related to the pathogenesis of a series of diseases, and thus, effective methods are needed for highly sensitive detection of ONOO-. Herein, a simple fluorescent probe RHHP-PN was proposed to detect ONOO-, and manifested obvious color changes from colorless to red in less than 10?s. Besides, it is not only suitable for quantitative detection of ONOO-, but also satisfactory in distinguishing ONOO- from other related species. What is particularly noteworthy is the fluorescence bioimaging for tracing endogenous and exogenous ONOO- in living cells and zebrafish. Therefore, probe RHHP-PN could be used as an imaging tool to visualize intracellular ONOO- in biological systems.

Abstract:Peroxynitrite (ONOO(-)), the product of a radical combination reaction of nitric oxide and superoxide, is a potent biological oxidant involved in a broad spectrum of physiological and pathological processes. Herein we report the development, characterization, and biological applications of a new fluorescent probe, HKGreen-4, for peroxynitrite detection and imaging. HKGreen-4 utilizes a peroxynitrite-triggered oxidative N-dearylation reaction to achieve an exceptionally sensitive and selective fluorescence turn-on response toward peroxynitrite in chemical systems and biological samples. We have thoroughly evaluated the utility of HKGreen-4 for intracellular peroxynitrite imaging and, more importantly, demonstrated that HKGreen-4 can be efficiently employed to visualize endogenous peroxynitrite generated in Escherichia coli-challenged macrophages and in live tissues from a mouse model of atherosclerosis. This probe should serve as a powerful molecular imaging tool to explore peroxynitrite biology under a variety of physiological and pathological contexts.

Abstract:Peroxynitrite (ONOO−), as one of the important reactive oxygen/nitrogen species, is associated with many physiological and pathological activities. Therefore, effective detection methods with high sensitivity and rapid response time are essential to reveal its functions. A novel Rhodol-based fluorescent probe RN is constructed and synthesized by introducing of 1,1-dimethylhydrazone as a recognition group. Due to the oxidative breakage of the CN bond by ONOO−, the probe has significant fluorescence enhancement change with excellent detection properties such as rapid response time, high sensitivity and selectivity. Moreover, probe RN show low cytotoxicity and is successfully capable of imaging endogenous and endogenous ONOO− in living cells. These properties indicate that RN has great application potential for imaging of ONOO− in biological systems.

Abstract:A fluorescent probe, HKGreen-2, has been developed based on a specific reaction between ketone and peroxynitrite (ONOO(-)). This probe is highly sensitive and selective for the detection of peroxynitrite not only in abiotic but also in biological systems. With this probe, we successfully detected peroxynitrite generated in murine macrophage cells activated by phorbol 12-myristate 13-acetate (PMA), interferon-gamma (IFN-gamma), and lipopolysaccharide (LPS). This new probe will be a useful tool for studying the roles of peroxynitrite in biological processes.

Abstract:Peroxynitrite is widely present in organisms and closely related to many pathophysiological functions. Therefore, it is of great physiological significance to develop capable probes for detecting ONOO − . In this work, a novel fluorescent probe B-Ch was designed based on the intramolecular charge transfer (ICT) effect. By means of molecular engineering, the replacement from diethylamine group to hydroxyl group has improved the detection sensitivity of the probe. After the addition of ONOO − , the solution color and fluorescence showed noticeable changes, which were visible to the naked eye. The probe showed excellent advantages: visualization, good selectivity, low sensitivity (22.4 nM), good stability and biocompatibility, exogenous and endogenous imaging of ONOO − in HeLa cells.

Abstract:Peroxynitrite is one of the important reactive oxygen species in the human body and is closely related to the physiological and pathological processes of many diseases. Therefore, the development of probes to detect peroxynitrite is important for diagnostic and pathologic studies of many diseases. In this work, a ratiometric probe was designed using benzopyran as the recognition site, and the sensitivity and selectivity of the probe were tuned by modification of substituents on benzopyran. Upon reaction with peroxynitrite, the color of the solution changes to the naked eye (from blue to yellow), and the fluorescence changes from red to blue. The probe SJ has the advantages of large Stokes shift (237 nm), fast response (≤10 s), wide linear range, good selectivity, low detection line (21.3 nm), and low cytotoxicity. Probe SJ has been successfully used for bioimaging of endogenous and exogenous peroxynitrite.

Abstract:Peroxynitrite (ONOO−), as an important reactive oxygen species (ROS), holds great potential to react with a variety of biologically active substances, leading to the occurrence of various diseases such as cancer and neurodegenerative diseases. In this work, we developed a novel mitochondria-localized fluorescent probe, HDBT-ONOO−, which was designed as a mitochondria-targeting two-photon fluorescence probe based on 1,8-naphthylimide fluorophore and the reactive group of 4-(bromomethyl)-benzene boronic acid pinacol ester. More importantly, the probe exhibited good biocompatibility, sensitivity, and selectivity, enabling its successful application in imaging the generation of intracellular and extracellular ONOO−. Furthermore, exogenous and endogenous ONOO− products in live zebrafish were visualized. It is greatly expected that the designed probe can serve as a useful imaging tool for clarifying the distribution and pathophysiological functions of ONOO− in cells and zebrafish.

Abstract:Peroxynitrite is a highly reactive molecule involved in cell signaling and pathological processes. We hereby report a novel genetically encoded probe, pnGFP, which can selectively sense peroxynitrite. A boronic acid moiety was site-specifically introduced into circularly permuted fluorescent proteins. By examining different protein templates followed with site-targeted random mutagenesis, we identified a selective peroxynitrite sensor, which is essentially unresponsive to other common cellular redox signaling molecules. The new probe has been genetically introduced into mammalian cells to image peroxynitrite at physiologically relevant concentrations.

Abstract:Abnormal peroxynitrite (ONOO−) expression is closely associated with the development of various pathological processes. In this study, we devised a series of fluorescent probes, namely DHU-DCM-1, DHU-DCM-2, and DHU-DCM-3, tailored specifically for monitoring ONOO− through rationally molecular design. By modulating the electron-donating site within the fluorophore structure, the optimal probe DHU-DCM-3 exhibited excellent ratiometric detecting performance. DHU-DCM-3 displayed high selectivity and sensitivity towards ONOO−, effectively distinguishing it from other reactive oxygen species. Notably, DHU-DCM-3 was capable of labeling mitochondrial ONOO−, producing an enhanced fluorescence ratio (F656/F550), thereby allowing for the visualization of ONOO− fluctuations in both exogenous and endogenous contexts within living cells. The utility of DHU-DCM-3 was further demonstrated in various applications. For instance, through detecting elevated levels of ONOO−, senile C. elegans was successfully identified by utilizing DHU-DCM-3. Moreover, relying on its ratiometric fluorescence, DHU-DCM-3 was proved to be valuable in accurately diagnosing liver injury in mice with varying degrees of severity. Overall, we have constructed a ratiometric fluorescent probe for detecting and imaging ONOO− in both subcellular organelle and in vivo, highlighting its potential significance in various biomedical applications.

Abstract:A novel fluorescent probe for peroxynitrite, PN600, was rationally designed on the basis of a unique fluorophore assembly approach. PN600 is a green-emitting coumarin derivative. Upon oxidation by peroxynitrite, PN600 is transformed into a highly fluorescent red-emitting resorufin derivative via an orange-emitting intermediate. This three-channel signaling capability enables PN600 to differentiate peroxynitrite from other reactive oxygen and nitrogen species, including hypochlorite and hydroxyl radical. Moreover, PN600 is membrane permeable and compatible with common TRITC filter sets and displays low cytotoxicity. Therefore, PN600 is a promising candidate for in vitro peroxynitrite imaging.

Abstract:As one of the most essential active nitrogen species, the abnormal metabolism of ONOO - in biological systems is considered to be associated with multiple diseases. Therefore, real-time in situ monitoring of organisms' ONOO - levels is highly significant for the in-depth investigation of their molecular mechanisms and early diagnosis of related diseases. Here, we reported a fluorescent probe, IPPN3 , based on a novel trifluoroacetamide-based strategy, enabling sensitive and rapid detection of ONOO - in aqueous solutions. Leveraging the specific oxidative hydrolysis reaction of ONOO - towards trifluoroacetamide moiety, the probe exhibits a substantial ratiometric fluorescence response to ONOO - with notable advantages including high selectivity, low detection limit (31.6 nM) and instantaneous response (less than 10 seconds). Furthermore, additional live-cell imaging results demonstrate the capability of IPPN3 for real-time exogenous and endogenous ONOO - fluorescence imaging in both live HeLa cells and zebrafish. The successful development of the IPPN3 may offer a new tool for tracking ONOO - levels in biological organisms in real time and exploring their biological functions.

Abstract:One kind of novel dicyanisophorone-based colorimetric fluorescent probe 3 has been prepared for recognizing peroxynitrite in aqueous condition. Large bathochromic shift of the absorption band is observed upon titration with ONOO–, inducing a clear solution color change from yellow to pale pink which enabling "naked-eye" detection possible. Moreover, probe 3 can react instantly with ONOO–accompanied by significant fluorescence enhancement at 621 nm while the detection limit is as low as 37 nM. Most importantly, probe 3 exhibits high selectivity and sensitivity towards ONOO– over other competitive ions in the aqueous solutions. Probe 3 has also been successfully applied in living MCF-7 cells and the result suggests that probe 3, could be applied as a potential candidate for the detection of ONOO–.

Abstract:Peroxynitrite is a short-lived endogenous reactive species and plays important roles in many physiological and pathological processes. In this work, we synthesized a near-infrared probe based on the structure of benzothiazole derived cyanine for determination of peroxynitrite (ONOO-). The designed probe specifically reacted with ONOO- through oxidative cleavage of conjugated C˭C double bonds and generating the non-fluorescent product. Meanwhile, the characteristic absorption of the probe at 630 nm greatly decreased after reaction with ONOO-, accompanied by drastic color change from bright blue to green yellow, which exhibited a distinct visual feature. It was demonstrated that the probe could be used to measure ONOO- in a dose-response manner and had a detection limit lower as 26 nM. Furthermore, the probe Cy-SN was applied for the imaging of endogenous ONOO- in living cells by confocal microscopy, which showed good cell permeability and low cytotoxicity. Successful application of probe for exogenous colorimetric detection and endogenous fluorescence imaging of ONOO- is suggesting its great potential applications in biological analysis.

Abstract:Peroxynitrite (ONOO(-)) and hypochlorous acid (HOCl) are two highly reactive oxygen species generated in biological systems. The overproduction of peroxynitrite or hypochlorous acid is implicated in a broad array of human pathologies including vascular, immunological, and neurodegenerative diseases. However, unambiguous detection of these reactive oxygen species has been relatively difficult due to their short biological half-lives and multiple reaction pathways. Based on their specific chemical reactions, we have developed fluorescent probes HKGreen-1 and HKOCl-1 for highly sensitive detection of peroxynitrite and hypochlorous acid, respectively. Both probes have been demonstrated to be able to discriminate corresponding reactive species from other reactive oxygen and nitrogen species (ROS and RNS) in not only chemical systems but also biological systems. The endogenous production of peroxynitrite in neuronal cells under oxygen-glucose deprivation (OGD) conditions has been visualized for the first time by utilizing HKGreen-1 probe, whilst the endogenous production of hypochlorous acid in macrophage cells upon stimulation with LPS, IFN-gamma, and PMA has been imaged by utilizing HKOCl-1 probe.

Abstract:Developing a reliable tool for monitoring peroxynitrite (ONOO-) in the complex organisms is critical to uncover its detailed cellular functions in numerous physiological and pathological processes. Thus, a novel ratiometric fluorescent probe was reasonably designed for ONOO-, in which the rhodol-HBT conjugate dye was chosen as a fluorophore, and hydrazine moiety was adopted as recognition receptor of ONOO-. The probe exhibited outstanding sensitivity, specificity, and a fast response towards ONOO-. More importantly, the probe with a large emission shift (Delta lambda(em)= 127 nm) has been proven to be capable of tracking ONOO- with high resolution in live cells and zebrafish. This work provides a simple and reliable tool for detecting and imaging ONOO- in the living systems, and thus we anticipate that the designed probe could be used as a powerful imaging tool in unraveling biological functions of ONOO-.

Abstract:A novel hepatocyte-targeting near-infrared fluorescent probe named Gal-NIR is developed. Gal-NIR shows ratiometric response to ONOO- with high sensitivity and selectivity. Moreover, the probe can accurately target the hepatocyte, and thus is used for assessing drug-induced hepatotoxicity and its remediation by using hepatoprotective medicines in living cells and mice.
