Abstract:Selenocysteine (Sec) is a primary kind of reactive selenium species in cells whose antioxidant roles in a series of liver diseases have been featured. However, it is difficult to determine Sec in living cells and in vivo due to its high reactivity and instability. This work reports a ratiometric near‐infrared fluorescent probe (Cy‐SS) for qualitative and quantitative determination of Sec in living cells and in vivo. The probe is composed of heptamethine cyanine fluorophore, the response unit bis(2‐hydroxyethyl) disulfide, and the liver‐targeting moiety d‐galactose. Based on a detection mechanism of selenium–sulfur exchange reaction, the concentrations of Sec in HepG2, HL‐7702 cells, and primary mouse hepatocytes is determined as 3.08 ± 0.11 × 10−6m, 4.03 ± 0.16 × 10−6m and 4.34 ± 0.30 × 10−6m, respectively. The probe can selectively accumulate in liver. The ratio fluorescence signal of the probe can be employed to quantitatively analyze the fluctuation of Sec concentrations in cells and mice models of acute hepatitis. The experimental results demonstrate that Sec plays important antioxidant and anti‐inflammatory roles during inflammatory process. And the levels of intracellular Sec have a close relationship with the degree of liver inflammation. The above imaging detections make this new probe a potential candidate for the accurate diagnosis of inflammation.

Abstract:As a common gaseous signaling molecule, hydrogen sulfide (H2S) plays a vital role in physiology and pathology. The development of fluorescent probes for detecting H2S has attracted widespread attention. However, most of the reported fluorescent probes with nitrobenzoxadiazole (NBD) as the recognition group have been widely used to simultaneously detect biothiols and H2S, instead of specifically detecting H2S. Herein, a novel NBD-based near-infrared (NIR) fluorescent probe named CX-N for the detection of H2S is synthesized. The selectivity of CX-N for H2S is significantly higher than that for biothiols and other potential interferences. After reacting with H2S, CX-N shows a significant increase in NIR fluorescence (75-fold), large Stokes shift (155 nm) and fast response (4 min). And the possible response mechanism of CX-N to H2S is given and confirmed by HPLC and HRMS. Based on the low cytotoxicity of CX-N, it has been used for H2S imaging in live cells and zebrafish. More importantly, CX-N has also been successfully applied for the real-time imaging of H2S in inflammatory and tumor mice based on its NIR emission, which provides a reliable platform for the specific recognition of H2S in complex biological systems.

Abstract:Thiols, such as cysteine (Cys), homocysteine (Hcy), glutathione (GSH), hydrogen sulfide (H2S) and thiophenol are metabolically correlated with each other via redox reactions. Due to the similarity of chemical properties between Cys, Hcy, GSH, H2S and thiophenol, it's very challenging to develop an effective methodology to differentiate them. In this work, a triple-emission fluorescent probe, NCQ, was reported for the simultaneous detection Cys/Hcy, GSH/H2S and thiophenol with high sensitivity and selectivity. The solution of NCQ displayed distinct fluorescent signals toward Cys/Hcy, GSH/H2S and thiophenol: blue and green for Cys/Hcy, blue for GSH/H2S, blue and red for thiophenol. Through the blue-green-red emission color combination, Cys/Hcy, GSH/H2S and thiophenol could be discriminatively detected in solution and in living cells.

Abstract:Biothiols such as cysteine (Cys), homocysteine (Hcy), glutathione (GSH) and hydrogen sulfide (H2S) are widely found in mammalian cells. They are closely related to the production and metabolic pathways and play very important roles in physiological and pathological activities. Therefore, the quantitative detection of these biothiols is of great significance. Although many fluorescent probes have been successfully used to track biothiols in biological samples, the fluorescence method for simultaneously detecting these biothiols using separated fluorescence emission channels under single wavelength excitation is still immature. In this work, we prepared the conjugate of seminaphthorhodafluor (SNARF) dye and 7-nitro-1,2,3-benzoxadiazole (NBD) using as a simple long-wavelength fluorescent probe SNARF-NBD for specific detection of biothiols. Cys/Hcy and GSH/H2S were identified by two separated fluorescence emission channels under single wavelength excitation, which showed good selectivity and sensitivity. In addition, SNARF-NBD has low cytotoxicity and shows good imaging ability in living cells and zebrafish.

Abstract:Abstract Previous studies have shown that the cysteine hydropersulfide (Cys-SSH) as the sulfur donor is crucial to sulfur-containing cofactors synthesis. Recently, a selective and sensitive near-infrared ratiometric fluorescent chemosensor Cy-DiSe has been designed and synthesized to detect Cys-SSH spontaneously. Herein, by means of the density functional theory (DFT) and time-dependent density functional theory (TD-DFT) approaches, the sensing mechanism has been thoroughly explored. According to our calculations, the experimental data have been reproduced. The results indicate the intramolecular charge transfer (ICT) is the reason for changes in fluorescence wavelengths. Compared with the chemosensor Cy-DiSe, the larger energy gap of Cy induced by ICT mechanism leads to the blue-shift of the absorption and emission spectra, which guarantees that Cy-DiSe can become a ratiometric fluorescent chemosensor to detect Cys-SSH.

Abstract:Cysteine is an essential amino acid for humans and can be generated from the conversion of methionine or the interconversions with cystine. The detection of cysteine in the living system is of great importance for the diagnosis of related diseases. Based on the classical cysteine response mechanism, the sulfhydryl group of cysteine undergoes Michael addition with the acrylate to rapidly respond to form a seven-membered ring that specifically recognizes cysteine. In the present work, we designed a novel ratiometric fluorescent probe N-1, which contained the benzo[cd]indol fluorophore and the response group acrylate. As the concentration of cysteine increased, the fluorescence of I485 was significantly enhanced while the fluorescence intensity at 635 nm was weakened. Moreover, probe N-1 showed excellent sensitivity and selectivity to Cys with the calculated LOD of 4.0 × 10-9 M. Probe N-1 was successfully used for imaging cysteine in the living Hela cells with low cytotoxicity, indicating that this ratiometric fluorescent probe would be a novel and useful tool to monitor cysteine in the physiological or pathological processes.

Abstract:Cysteine (Cys) is a crucial biothiol that acts a significant function in food samples and biological systems, including plant roots and living cells. Hence, we developed a novel colorimetric and near-infrared ratiometric fluorescent probe (CT), composed of coumarin and tetrahydroacridine-conjugated indole salt, for the detection of Cys. Upon reaction with Cys, the probe undergoes a specific N-substitution reaction, resulting in a notable colorimetric change and a significant ratiometric fluorescent response in both visible and near-infrared emission channels. These dual-channel ratiometric fluorescence changes are completely independent, enabling the probe to obtain great selectivity, sensitivity, and exceptional detection accuracy. Leveraging these attributes, the probe was employed to provide accurate quantitative analysis of Cys in food samples. Furthermore, confocal imaging demonstrated that the probe could monitor both exogenous and endogenous Cys levels in living cells and track Cys changes in plant roots under heavy metal stress. This work presents a dependable and accurate imaging solution for tracking and identifying Cys of real food, plants, and living cells.

Abstract:A long-wavelength fluorescent probe NR-CY was developed for simultaneous identification of cysteine/glutathione and sulphide by combining the derivative of Nile red with 7-nitrobenzofurazan. The response of NR-CY to thiols is regulated by intramolecular charge transfer and photoinduced electron transfer mechanisms. For sulphide at 560 nm, cysteine at 475 nm and glutathione at 425 nm, different absorbance increases can be observed. NR-CY can detect cysteine at fluorescence emission 543 nm and distinguish sulphide from other analytes by kinetic experiments at 636 nm. The probe showed a rapid response to these thiols (cysteine was 90 s and sulphide was 30 s). In addition, NR-CY has been successfully applied to live MCF-7 cell imaging.

Abstract:Ratiometric near-infrared (NIR) fluorescent probes are attractive tools for bioimaging applications. Although many fluorescent probes have been developed for detection of the biologically important cysteine (Cys), ratiometric NIR fluorescent probes for Cys are very rare, particularly those with significant fluorescence enhancement in the NIR region. In this work, a new ratiometric NIR fluorescent probe with significant NIR fluorescent turn-on changes was reported for rapid, colorimetric and ratiometric fluorescent detection of Cys. This probe is able to detect Cys in aqueous solution under mild conditions, with high selectivity and sensitivity for Cys over various analytes including the similar structured homocysteine (Hcy) and glutathione (GSH). The detection limit of this probe for Cys was found to be as low as 0.2 μM, which is far below the normal content of Cys (30–200 μM) in cells. Besides the excellent sensing properties, this probe shows low cytotoxicity and can be conveniently applied to image intracellular Cys in living cells, indicating it is a promising tool for biological applications.

Abstract:Sulfite from cysteine metabolism in living cells plays a crucial role in improving the water solubility of metabolic xenobiotics for their easier excretion in urine or bile. However, an imbalance of sulfite in vivo would lead to oxidative stress or age-related diseases, and an effective strategy for real-time imaging of cysteine metabolism in living cells is still lacking due to its low metabolite concentration and rapid reaction kinetics. Herein, a cyanine moiety based ratiometric fluorescence probe was developed for highly selective and sensitive detection of sulfite in aqueous solution and living cells. The free probe exhibited an orange emission color, and the fluorescence color would gradually change to blue once sulfite anions selectively reacted with the unsaturated carbon double bonds in the probe molecule. This ratiometric fluorescence manner endowed the probe excellent sensitivity with a detection limit of 0.78 nM, which was then explored to image the kinetic process of sulfite release in hepatic BRL cells after incubating with an excess amount of cysteine. This strategy opens new opportunities for revealing thiol-containing species metabolism and even quantitatively tracking their distributions in live cells or organelles.

Abstract:Hydrogen sulfide (H2S) has been confirmed as a significant endogenous gaseous signaling molecule involved in various physiological processes. In order to monitor H2S in living cells, a Forster resonance energy transfer (FRET) ratiometric probe based on quantum dot-cresyl violet was developed. In this work, the quantum dot nanospheres via a facile ultrasonication emulsion strategy, and the mixture chloroform solution containing hydrophobic quantum dots and COOH-functionalized amphiphilic polymer were successfully transferred into the oil-in-water micelle. The negatively charged quantum dot nanospheres with quantum dots embedded in the polymer matrixes were successfully fabricated after the evaporation of chloroform. And then, these quantum dot nanospheres were condensed with positively charged cresyl violet-azide (CV-N3) via electrostatic interaction to obtain the complexes (QDS-N3). The as-prepared QDS-N3 complexes were monodispersed nanospheres with an average diameter of about 120 nm. These complexes were taken up by the cell through endocytosis, and they were still stable even in wide pH range. In addition, the QDS-N3 complexes exhibited no cellular toxicity which was verified by MTT assay. In this ratiometric probe, CV-N3 as a FRET acceptor was conjugated to quantum dot nanospheres. The quantum dots emitted at 591 nm and served as the FRET donor; once the aryl azide on the CV-N3 was reduced by H2S to aniline, the probe emitted at 620 nm. The ratiometric probe allowed the elimination of interference of excitation intensity, intracellular environment and other factors. Furthermore, this method also offered a general protocol for preparing nanosensors for monitoring various small molecular in living cells.

Abstract:Cysteine is associated with various diseases in the organism. Monitoring Cys levels is important for the study of related diseases. Here, a ratiometric fluorescent probe based on a novel fluorophore was designed for detecting Cys. The fluorophore exhibits a large Stokes shift and good photostability. The probe QL‐A shows high specificity and has been used to visualize Cys in living cells. It is a potential tool for exploring the role of Cys in biological processes and diseases. As a biothiol, cysteine (Cys) is essential to both physiological and pathological processes and has been associated with many diseases, including neurological disorders, rheumatoid arthritis, and renal dysfunction. Therefore, the development of a high‐performance probe for detecting Cys levels can help prevent and diagnose disease. In this study, a ratiometric fluorescent probe based on a novel fluorophore was developed for detecting Cys, and it showed high specificity and a rapid response time toward Cys. This probe demonstrates excellent biocompatibility and has been utilized effectively for the imaging of Cys in living cells.

Abstract:A benzothiazole-based near-infrared (NIR) ratiometric fluorescent probe (HBT-Cys) was developed for discriminating cysteine (Cys) from homocysteine (Hcy) and glutathione (GSH). The probe was designed by masking phenol group in the conjugated benzothiazole derivative with methacrylate group that could be selectively removed by Cys, and therefore an intramolecular charge transfer (ICT) fluorescence was switched on in the NIR region. In the absence of Cys, the probe exhibited a strong blue fluorescence emission at 431 nm, whereas a NIR fluorescence emission at 710 nm was significantly enhanced accompanied by a decrease of emission at 431 nm in the presence of Cys, allowing a ratiometric fluorescence detection of Cys. The fluorescence intensity ratio (I710nm/I431nm) showed a good linear relationship with Cys concentration of 1-40 μM with the detection limit of 0.5 μM. The sensing mechanism was explored based on MS experimental analysis and DFT theoretical calculation. Moreover, the fluorescent probe was successfully used for fluorescence bioimaging of Cys in living cells.

Abstract:We have developed a ratiometric fluorescent probe based on ICT mechanism for imaging intracellular sulfite (SO32) in HeLa cells. The probe could selectively stain cells with scarce cytotoxicity and excellent photostability. The probe displayed a high selectivity for sulfite over other anions and active small biological molecules including cysteine (Cys), homocysteine (Hcy) and glutathione (GSH) at room temperature. The reaction could complete within 30 s and detection limit was as low as 76 nM, which is superior to some reported probes. The detecting mechanism was proved by MS, H-1 NMR and theoretical calculation. The probe can also detect endogenous sulfite in HepG2 cells. (C) 2016 Elsevier B.V. All rights reserved.

Abstract:Small-molecule biothiols, including cysteine (Cys),homocysteine (Hcy), and glutathione (GSH), participate invarious pathological and physiological processes. It is still a challenge to simultaneously distinguish Cys and Hcy because of their similar structures and reactivities, as well as the interference from the high intramolecular concentration of GSH. Herein, anovelfluorescent probe,CySI, based on cyanine and thioester was developed to differentiate Cys and Hcy through a single-wavelength excitation and two distinctly separated emissionchannels. The probe exhibited a turn-onfluorescence responseto Cys at both 625 nm (the red channel) and 740 nm (the near-infrared channel) but only showedfluorescence turn-on to Hcy at740 nm (the near-infrared channel) and nofluorescent response toGSH. With the aid of built-in self-calibration of single excitation and dual emissions, simultaneous discriminative determinations ofCys and Hcy were realized through red and near-infrared channels.CySIexhibited excellent selectivity toward Cys and Hcy with afast response. This probe was further exploited to visualize exogenous Cys and Hcy in cells through dual emission channels underone excitation. Moreover, it could efficiently target mitochondria and was applied to monitor the endogenous Cys fluctuations independently in mitochondria through the red emission channel.

Abstract:Hydrogen sulphide (H2S), the third endogenous gaseous signalling molecule, has attracted attention in biochemical research. The selective detection of H2S in living systems is essential for studying its functions. Fluorescence detection methods have become useful tools to explore the physiological roles of H2S because of their real-time and non-destructive characteristics. Herein we report a near-infrared fluorescent probe, NIR-HS, capable of tracking H2S in living organisms. With high sensitivity, good selectivity and low cytotoxicity, NIR-HS was able to recognize both the exogenous and endogenous H2S in living cells. More importantly, it realized the visualization of endogenous H2S generated in cells overexpressing cystathionine β-synthase (CBS), one of the enzymes responsible for producing endogenous H2S. The probe was also successfully applied to detect both the exogenous and endogenous H2S in living mice. The superior sensing properties of the probe render it a valuable research tool in the H2S-related medical research.

Abstract:Small molecular biothiols, such as cysteine (Cys), homocysteine (Hcy), reduced glutathione (GSH), and hydrogen sulfide (H^2S), play crucial parts in regulating the redox balance of life activities, regulating normal physiological activities and preventing various diseases. Quantitative analysis of these important small molecular substances is very important for revealing their diverse physiological and pathological effects. Although many fluorescent probes have been reported to detect biothiols in cells, it is still not sufficiently advanced to detect biothiols with separated fluorescence emission peak by same wavelength excitation. In our work, we designed a simple conjugate of Nile red and NBD (7-nitro-1,2,3- benzoxadiazole) as long-wavelength fluorescent probe NR-NBD for the simultaneous discrimination of these biothiols at single wavelength excitation. Probe NR-NBD could efficiently discriminate Cys/Hcy, GSH and H^2S by two separated fluorescence emission channels and absorption spectra. Importantly, probe NR-NBD has excellent specificity and sensitivity towards the monitoring of endogenous/exogenous Cys/Hcy and GSH/H2S in living cells and zebrafish.
