Abstract:Photodynamic therapy (PDT) is a promising singlet oxygen ((1)O2) mediated clinical treatment for many tumors. As the source of (1)O2, oxygen plays an important role in the curative effect of PDT. However, the facts of photochemical depletion of oxygen and the intrinsic hypoxic microenvironment of tumors remain the major challenges. In this work, a novel photosensitizer carrier with oxygen self-compensating ability was designed for PDT. It was synthesized via chemical conjugation of hemoglobin (Hb) to polymeric micelles formed by triblock copolymers of poly(ethylene glycol)-block-poly(acrylic acid)-block-polystyrene (PEG-b-PAA-b-PS). The PEG-b-PAA-b-PS and resultant micelles in aqueous solution were comprehensively characterized by means of FTIR, (1)H NMR, GPC, DLS, TEM, and fluorescence spectroscopy. The oxygen-binding capacity and antioxidative activity of the Hb conjugated micelles were evaluated via UV-vis spectroscopy. In addition, compared with the control micelles without Hb, the Hb conjugated photosensitizer carrier was able to generate more (1)O2 and exert greater photocytotoxicity on Hela cells in vitro.

Abstract:Photodynamic therapy (PDT) is recognized to be a promising strategy for anticancer treatment. Considering the progressive application of PDT in clinical trials, highly efficient and photostable photosensitizers (PSs) are in strong demand. Aggregation-induced emission (AIE) based PSs are promising phototheranostic materials for tumor imaging and PDT due to their high fluorescence efficiency and photostability. Herein, a mitochondria-targeted PS, TPA-2TCP with AIE characteristics is developed by adopting an acceptor-π-donor-π-acceptor (A-π-D-π-A) structure. The untypical sequence of the electron donors and electron acceptors endows the derived AIE PS with evident redshift of the absorption and emission, and efficient generation of reactive oxygen species. With the positively charged pyridinium groups, nanoparticulated AIE PS (TPA-2TCP NPs) exhibits high cell binding efficiency towards 4T1 breast cancer cells, leading to the massive cell death via the apoptotic pathway under white light irradiation, demonstrating its potential application in cancer imaging and PDT.
Mitochondria-targeting and ROS-sensitive smart nanoscale supramolecular organic framework for combinational amplified photodynamic therapy and chemotherapy

Abstract:Owing to their reversibly dynamic features, and the regularity of their architectures, supramolecular organic frameworks (SOFs) have attracted attention as new porous materials. Herein, we propose a smart SOF platform for enhanced photodynamic therapy, where the SOF with a superior mitochondria-targeting capability could be cleaved by reactive oxygen species (ROS) produced by itself for highly enhancing PDT. Moreover, it can further work as a platform for carrying chemo-therapeutic drug doxorubicin for synergistic chemo-photodynamic therapy. The SOF is constructed by combining a tetra-β-cyclodextrin-conjugated porphyrin photosensitizer and a ROS-sensitive thioketal linked adamantane dimer utilizing a host-guest supramolecular strategy. The unique supramolecular framework not only completely resolves the aggregation caused quenching of porphyrin photosensitizers but also endows them with significantly enhanced water-solubility. The in vitro and in vivo results demonstrate that the SOF could be targeted onto mitochondria by confocal imaging, and dissociated by ROS generated by itself, leading to autonomous release of porphyrin photosensitizers and DOX for high anti-cancer activity. It is believed that the strategy using a SOF has the potential of being used to construct versatile agents for combined therapies.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Statement of significance</h3>: Photosensitizers are the essential element in photodynamic therapy. However, typical photosensitizers commonly encounter poor water-solubility, non-specific tumor-targeting, aggregation-caused quenching (ACQ), which seriously reduce PDT efficacy. A mitochondria-targeting and ROS-sensitive supramolecular organic framework (SOF) is designed for photodynamic therapy in cancer treatment, which could completely overcome the bottleneck in the applications of photosensitizers (PSs). The SOF is constructed by combining a tetra-β-cyclodextrin-conjugated porphyrin photosensitizer and a ROS-sensitive thioketal linked adamantane dimer unit utilizing a host-guest supramolecular strategy. The unique supramolecular framework not only completely resolves the aggregation caused quenching of porphyrin photosensitizers but also endows them with significantly enhanced water-solubility. Moreover, the SOF can be readily functionalized to incorporate the anti-cancer agent Doxorubicin and mitochondria targeting molecules through respective physical encapsulation and host-guest interactions.

Abstract:Currently, photosensitizers (PSs) that are microenvironment responsive and hypoxia active are scarcely available and urgently desired for antitumor photodynamic therapy (PDT). Presented herein is the design of a redox stimuli activatable metal-free photosensitizer (aPS), also functioning as a pre-photosensitizer as it is converted to a PS by the mutual presence of glutathione (GSH) and hydrogen peroxide (H2O2) with high specificity on a basis of domino reactions on the benzothiadiazole ring. Superior to traditional PSs, the activated aPS contributed to efficient generation of reactive oxygen species including singlet oxygen and superoxide ion through both type 1 and type 2 pathways, alleviating the aerobic requirement for PDT. Equipped with a triphenylphosphine ligand for mitochondria targeting, (mito)aPS showed excellent phototoxicity to tumor cells with low light fluence under both normoxic and hypoxic conditions, after activation by intracellular GSH and H2O2. The (mito)aPS was also compatible to near infrared PDT with two photon excitation (800 nm) for extensive bioapplications.

Abstract:Increasing interest has been noted in photosensitizer design. Currently, most small molecular photosensitizers have several drawbacks, including limited penetration depth and unsatisfying photostability. For addressing these limitations, Frequency upconversion luminescence (FUCL) small molecule was adopted as a photosensitizer design strategy for its deeper penetration, sensitive imaging, and better photostability. We developed a series of FUCL (NIR-NH2, NIR-NHEt, NIR-NEt2) molecules with rigid structures. Equipped with the most powerful electron donor, NIR-NEt2 exhibits the highest singlet oxygen generation, the strongest upconversion luminescence intensity under 808 nm irradiation and mitochondria-targeted ability. FUCL can be a superior molecular platform for image-guided photodynamic therapy of cancer cells.

Abstract:In this paper, a series of novel BODIPY-based photosensitizers have been designed and synthesized for photodynamic therapy. BODIPY3 was screened out as the most potential photosensitizer due to its excellent optical properties, high singlet oxygen efficiency and good photostability. However, as an organic photosensitizer, BODIPY3 still suffered from the drawbacks of insolubility and instability in aqueous system. In view of these problems, DSPE-PEG2000 was used to trap BODIPY3 into the hydrophobic core of micelles to obtain well-dispersing nano complexes BODIPY3-PEG3 in aqueous system. More importantly, BODIPY3-PEG3 not only has better solubility and stability in aqueous media but can generate significant singlet oxygen (1O2, one of the reactive oxygen species, the real cytotoxic agent in photodynamic therapy) in living cells and exhibit high light cytotoxicity to three cancer cell lines. The mechanism studies indicated the mitochondrial localization of BODIPY3-PEG3 was able to generate ROS in mitochondria, which further result in mitochondrial dysfunction and photoinduced apoptosis via caspase-8 and caspase-3 pathway.
Molecular Engineering Design of Enhanced Donor-Acceptor Therapeutic Reagent for Efficient Image-Guided Photodynamic Therapy

Abstract:The greatest barrier to the further development and clinical application of tumor image-guided photodynamic therapy (PDT), is the inconsistency between the fluorescence intensity and singlet oxygen generation yield of the photosensitizer under light excitation. Herein, a novel donor-acceptor (D-A) system is designed from the point of molecular selection by wrapping a classical porphyrin molecule (5,10,15,20-tetraphenylphorphyrin, H2 TPP) as an acceptor into conjugated polymer (Poly[N,N'-bis(4-butylpheny)-N,N'-bis(phenyl)benzidine], ADS254BE) as a donor through fluorescence resonance energy transfer (FRET) mechanism, which exhibits bright red emission centered at 650 nm (quantum yield, 0.12), relatively large Stoke shift of 276 nm, enhanced singlet oxygen generation rate of 0.73, and excellent photostability. The investigations on distribution and killing effect of nanomaterials in cancer cells reveal that ADS254BE/H2 TPP NPs can accumulate in the cytoplasm for imaging while simultaneously producing a large amount of singlet oxygen to remarkably kill cancer cells, which can be used for real-time image-guided PDT. In the xenograft tumor model, real-time imaging and long-term tracing in tumor tissue with ADS254BE/H2 TPP NPs disclose that the growth of lung cancer in mice can be effectively inhibited during in situ imaging. From the standpoint of molecular engineering design, this work provides a feasible strategy for novel D-A systems to improve the development of image-guided PDT.

Abstract:Photodynamic therapy (PDT) often suffers from the exacerbated tumor hypoxia and the heterogeneous distribution of photosensitizers, leading to an inefficient ROS productivity and availability. In this work, a mitochondria targeted O2 economizer (designated as Mito‐OxE) is developed to improve PDT efficiency by alleviating tumor hypoxia and enhancing the subcellular localization of photosensitizers. Specifically, the photosensitizer of protoporphyrin IX (PpIX) is modified with the hydrophilic polyethylene glycol and the lipophilic cation of triphenylphosphine (TPP) to fabricate the biocompatible mitochondria targeted photosensitizers (designated as Mito‐PSs). And Mito‐OxE is prepared by using Mito‐PSs to load the mitochondrial oxidative phosphorylation inhibitors of atovaquone (ATO). Benefiting from the targeting capability of TPP, Mito‐OxE can selectively accumulate in mitochondria after cellular uptake. Subsequently, the mitochondrial respiration would be suppressed to with the participation of ATO, resulting in a local hypoxia mitigation for enhanced PDT. Compared with Mito‐PSs, Mito‐OxE maximizes the therapeutic effect against hypoxic tumors under light irradiation. This design of mitochondria targeted O2 economizer would advance the development of targeted drug delivery system for effective PDT regardless of hypoxic microenvironment.

Abstract:Photoimmunotherapy has been acknowledged to be an unprecedented strategy to obtain significantly improved cancer treatment efficacy. In this regard, the exploitation of high-performance multimodal phototheranostic agents is highly desired. Apart from tailoring electron donors, acceptor engineering is gradually rising as a deliberate approach in this field. Herein, we rationally designed a family of aggregation-induced emission (AIE)-active compounds with the same donors but different acceptors based on the acceptor engineering. Through finely adjusting the functional groups on electron acceptors, the electron affinity of electron acceptors and the conformation of the compounds were simultaneously modulated. It was found that one of the molecules (named DCTIC), bearing a moderately electrophilic electron acceptor and the best planarity, exhibited optimal phototheranostic properties in terms of light-harvesting ability, fluorescence emission, reactive oxygen species (ROS) production, and photothermal performance. For the purpose of amplified therapeutic outcomes, DCTIC was fabricated into tumor and mitochondria dual-targeted DCTIC nanoparticles (NPs), which afforded good performance in the fluorescence/photoacoustic/photothermal trimodal imaging-guided photodynamic/photothermal-synergized cancer immunotherapy with the combination of programmed cell death protein-1 (PD-1) antibody. Not only the primary tumors were totally eradicated, but efficient growth inhibition of distant tumors was also realized.

Abstract:We developed a mitochondrion-targeting MnII-terpyridine complex (MTP) for two-photon photodynamic therapy. MTP was subjected to two-photon excitation in the NIR region to generate 1O2 and hence produce a PDT effect. This use of MTP overcame the drawbacks of traditional PDT agents. The MnII center of MTP apparently catalyzed the H2O2-mediated production of ROS and oxygen, which subsequently promoted the 1O2 generation, furthering the PDT effect. Additionally, the mitochondrion targeting of MTP provided a good spatial condition for the generation of 1O2, which further promoted the PDT effect.

Abstract:Photodynamic therapy（PDT）is an attractive clinical technique for cancer treatment.However,the poor solubility and weak cellular internalization of the molecule-photosensitizers,as well as the exceedingly sho rt lifetime and limited diffusion distance of the generated reactive oxygen species（ROS）in cytoplasm directly restricted the therapeutic efficiency of conventional PDT.In this study,we proposed a facile strategy for improving PDT of cancer based on a mitochondria-targeted nanophoto sensitizer.The molecule-photo sensitizer chlorin e6 was covalently attached on the internal and exte rnal surfaces of the mesoporous silica nanoparticles.Then,the triphenylphosphonium was ancho red on the nanopa rticles fo r selectively targeting mitochondria.When irradiated with laser,the nanophotosensitizer can generate a large amount of ROS in mitochondria,thus causing the mitochondrial dysfunction and irreversible cell apoptosis.In vitro and in vivo studies demonstrated that the nanophotosensitizer could boost the treatment efficiency against cancer cells and xenograft tumor models.
Donor-Acceptor Modulating of Ionic AIE Photosensitizers for Enhanced ROS Generation and NIR-II Emission

Abstract:Photosensitizers (PSs) with aggregation-induced emission (AIE) characteristics are competitive candidates for bioimaging and therapeutic applications. However, their short emission wavelength and nonspecific organelle targeting hinder their therapeutic effectiveness. Herein, a donor-acceptor modulation approach is reported to construct a series of ionic AIE photosensitizers with enhanced photodynamic therapy (PDT) outcomes and fluorescent emission in the second near-infrared (NIR-II) window. By employing dithieno[3,2-b:2',3'-d]pyrrole (DTP) and indolium (In) as the strong donor and acceptor, respectively, the compound DTP-In exhibits a substantial redshift in absorption and fluorescent emission reach to NIR-II region. The reduced energy gap between singlet and triplet states in DTP-In also increases the reactive oxygen species (ROS) generation rate. Further, DTP-In can self-assemble in aqueous solutions, forming positively charged nanoaggregates, which are superior to conventional encapsulated nanoparticles in cellular uptake and mitochondrial targeting. Consequently, DTP-In aggregates show efficient photodynamic ablation of 4T1 cancer cells and outstanding tumor theranostic in vivo under 660 nm laser irradiation. This work highlights the potential of molecular engineering of donor-acceptor AIE PSs with multiple functionalities, thereby facilitating the development of more effective strategies for cancer therapy.
Synthesis and evaluation of novel mitochondria-targeted, water-soluble phenoxazine-porphyrins for efficient photodynamic therapy

Abstract:In this study, we use cationic imidazole as the targeting group and phenoxazine as the electron donor, attaching phenoxazine to porphyrin via two different linking methods to synthesize two different phenoxazine-porphyrin photosensitizers (PSs), P1 and P2 , intended for photodynamic therapy (PDT) applications. Additionally, we synthesized a molecule, P3 , that does not contain the cationic imidazole group, to serve as a control PS. We comprehensively evaluate the performance of P1 and P2 through steady-state and transient spectroscopy analysis, theoretical calculations, photooxidation experiments, and in vitro cell experiments. The results showed that P1 and P2 exhibit excellent water solubility, longer triplet state lifetimes ( P1 at 200 μs, P2 at 170 μs), outstanding photostability, higher photooxidation rates and quantum yields of singlet oxygen ( 1 O 2 ). Notably, in the photooxidation experiment, P1 's photooxidation rate is 1.1 times that of P2 , and 1.5 times that of tetraphenyl zinc porphyrin (ZnTPP) without phenothiazine. This may be due to the larger Gibbs free energy difference between the singlet and triplet excited states in P1 (Δ G S-T of P1 is 0.87 eV, compared to 0.83 eV for P2 ), which results in a greater driving force for intersystem crossing (ISC) in P1 than in P2 . The introduction of the cationic imidazole group in P1 and P2 significantly enhances water solubility and tumor cell uptake compared to P3 , particularly showing higher intracellular accumulation and 1 O 2 generation capabilities in HepG2 tumor cells. Flow cytometry test results further confirmed the significant photocytotoxicity of P1 and P2 , with a marked decrease in survival rates after light exposure, where the survival rate of P1 nanoparticles (NPs) dropped to 41.7%, P2 NPs to 35.1%, and P3 NPs to 75.0%. The findings of this study provide crucial theoretical support for the design of new PSs.

Abstract:Developing efficient photosensitizers (PSs) that can generate type I reactive oxygen species (ROS) under illumination is considered an effective way to improve photodynamic therapy (PDT) outcomes due to the hypoxic nature of the tumor environment, but also is very challenging. Herein, a new PS of the multiarylpyrrole (MAP) derivative with a typical donor-acceptor structure was synthesized to efficiently generate type I ROS by using an acceptor-shielding strategy in their aggregated state. The enhanced generation mechanism of type I ROS originated from its ultralong triplet lifetime and the narrow singlet-triplet energy gap of the MAP. More importantly, type I ROS can transform protumoral M2 macrophages (M2) into antitumoral M1 macrophages (M1), which showed synergistic immunotherapy in in vivo experiments. Therefore, introducing shielding groups into acceptors provides general guidance for developing efficient PSs in the aggregation state for clinical PDT.
Deciphering Oxygen‐Independent Augmented Photodynamic Oncotherapy by Facilitating the Separation of Electron‐Hole Pairs

Abstract:Developing Type-I photosensitizers provides an attractive approach to solve the dilemma of inadequate efficacy of photodynamic therapy (PDT) caused by the inherent oxygen consumption of traditional Type-II PDT and anoxic tumor microenvironment. The challenge for the exploration of Type-I PSs is to facilitate the electron transfer ability of photosensitization molecules for transforming oxygen or H2O to reactive oxygen species (ROS). Herein, we propose an electronic acceptor-triggered photoinduced electron transfer (a-PET) strategy promoting the separation of electron-hole pairs by marriage of two organic semiconducting molecules of a non-fullerene scaffold-based photosensitizer and a perylene diimide that significantly boost the Type-I PDT pathway to produce plentiful ROS, especially, inducing 3.5-fold and 2.5-fold amplification of hydroxyl (OH•) and superoxide (O2−•) generation. Systematic mechanism exploration reveals that intermolecular electron transfer and intramolecular charge separation after photoirradiation generate a competent production of radical ion pairs that promote the Type-I PDT process by theoretical calculation and ultrafast femtosecond transient absorption (fs-TA) spectroscopy. By complementary tumor diagnosis with photoacoustic imaging and second near-infrared fluorescence imaging, this as-prepared nanoplatform exhibits fabulous photocytotoxicity in hypoxic tumor model of mice. We envision that this work will broaden the insight into high-efficiency Type-I PDT for cancer phototheranostics.

Abstract:The synthesis of water-soluble symmetric molecules with donor-acceptor-donor (D-A-D) structure is reported. The compound is connected by π bridge with 2-bromofluorene external polyethylene glycol 2000 as the shielding unit, and donor component and pyrrolopyrrole (DPP) as the acceptor unit. The D-A-D double donor fluorescent molecule P2-DPP is obtained by coupling reaction. The absorption peak and emission peak of the fluorescent molecule P2-DPP are 600 and 1020 nm, respectively. It has potential excellent imaging characteristics. It does not need to use nanoparticles formed by the DSPE-MPEG amphiphilic block to form micelles. The quantum yield reaches 0.6% and the penetration depth can reach 10 mm. The chemical is capable of achieving liver and renal metabolism. It has a good application prospect in the photothermal therapy of mouse tumors and realizes the integration of biological diagnosis and treatment.
H2O2-activated mitochondria-targeting photosensitizer for fluorescence imaging-guided combination photodynamic and radiotherapy

Abstract:Radiotherapy is a primary modality in cancer treatment but accompanied by severe side effects to healthy tissues and radiation resistance to some extent. To overcome these limitations, we developed a H2O2-responsive photosensitizer, CyBT, which could be activated by the upregulated H2O2 induced by radiotherapy, enabled near-infrared fluorescence imaging-guided combination photodynamic and radiotherapy. The synthesis of CyBT began with the covalent linkage of hemicyanine and a free radical TEMPO through click reaction, which demonstrated superior photodynamic properties. Shielding of fluorescence and photodynamic activity was achieved by incorporating phenylboronic acid pinacol ester. In X-ray irradiated tumor cells, the upregulation of H2O2 activated CyBT, thereby restoring its fluorescence and photodynamic activity. Additionally, the positive charge of CyBT facilitated its targeting to mitochondria within tumor cells for more efficiently trigger of cell apoptosis. CyBT was co-assembled with a polymer PEG-b-PDPA to form acid-responsive nanoparticles (NPs-CyBT). This formulation enhanced tumor targeting, improved water solubility of CyBT, and extended in vivo circulation time. Utilizing fluorescence imaging to guide photodynamic and radiotherapy, NPs-CyBT can accurately target solid tumors in mice, and lead to tumor elimination, suggesting that it is a potential strategy for the effective treatment of malignant tumors.

Abstract:Photodynamic therapy (PDT) has emerged as an attractive alternative in cancer therapy, but therapeutic effects suffer from low photosensitizing efficiency and poor retention of photosensitizes in cancer cells. This paper reports the photosensitizers which show absorption and emission in the long-wavelength region and high photosensitizing efficiency can be formed in situ in cells from 4,6-dibromothieno[3,4-b]thiophene derivative (TT-5-P) after white light irradiation. The self-oligomerization of TT-5-P is uptaken in cells upon light irradiation-induced cell apoptosis simultaneously and efficiently. In addition, the formation of oligomers (TT-5-Ps) enhances the retention time in cells remarkably, which is advantageous for boosting the photodynamic therapy efficiency. Moreover, the selectivity toward tumor cells of TT-5-P can be improved obviously via the formation of complex of TT-5-P with albumin. This in situ photoinduced self-oligomerization strategy can be utilized to design effective biomaterials for long-term imaging and improved therapy.
