Abstract:Surface-enhanced Raman scattering (SERS) signals strongly rely on the interactions and distance between analyte molecules and metallic nanostructures. In this work, the use of a gold nanoparticle (AuNP)-embedded metal-organic framework was introduced for the highly sensitive SERS detection. The AuNPs were in situ grown and encapsulated within the host matrix of MIL-101 by a solution impregnation strategy. The as-synthesized AuNPs/MIL-101 nanocomposites combined the localized surface plasmon resonance properties of the gold nanoparticles and the high adsorption capability of metal-organic framework, making them highly sensitive SERS substrates by effectively preconcentrating analytes in close proximity to the electromagnetic fields at the SERS-active metal surface. We discussed the fabrication, physical characterization, and SERS activity of our novel substrates by measuring the Raman signals of a variety of model analytes. The SERS substrate was found to be highly sensitive, robust, and amiable to several different target analytes. A SERS detection limit of 41.75 and 0.54 fmol for Rhodamine 6G and benzadine, respectively, was demonstrated. The substrate also showed high stability and reproducibility, as well as molecular sieving effect thanks to the protective shell of the metal-organic framework. Subsequently, the potential practical application of the novel SERS substrate was evaluated by quantitative analysis of organic pollutant p-phenylenediamine in environmental water and tumor marker alpha-fetoprotein in human serum. The method showed good linearity between 1.0 and 100.0 ng/mL for p-phenylenediamine and 1.0-130.0 ng/mL for alpha-fetoprotein with the correlation coefficients of 0.9950 and -0.9938, respectively. The recoveries ranged from 80.5% to 114.7% for p-phenylenediamine in environmental water and 79.3% to 107.3% for alpha-fetoprotein in human serum. These results foresee promising application of the novel metal-organic framework based composites as sensitive SERS-active substrates in both environmental and clinical samples.

Abstract:Aggregation of metal nanoparticles plays an important role in surface enhanced Raman scattering（SERS）. Here, a strategy of dynamically aggregating/releasing gold nanoparticles is demonstrated using a gold-nanofilm-coated nanofiber, with the assistance of enhanced optical force and plasmonic photothermal effect. Strong SERS signals of rhodamine 6 G are achieved at the hotspots formed in the inter-particle and film-particle nanogaps. The proposed SERS substrate was demonstrated to have a sensitivity of 10-12 M, reliable reproducibility, and good stability.

Abstract:Single-molecule detection via surface-enhanced Raman scattering (SERS) has raised great interest over the past decade. The usual approach toward this goal is to harness the strong surface plasmon resonance of light with complex metallic nanostructures, such as particle aggregates, two-particle gaps, sharp tips, or particles with sharp apexes. Here we propose another route toward the goal by introducing gain medium into single metal nanoparticles with simple geometry. Our calculations show that cubic gold nanobox particles that contain a gain material within the core can create an extremely high enhancement factor of local field intensity larger than 10(8) and a SERS enhancement factor on the order of 10(16)-10(17).

Abstract:Metal-organic frameworks (MOFs) have attracted widespread interest due to their unique and unprecedented advantages in microstructures and properties. Besides, surface-enhanced Raman scattering (SERS) technology has also rapidly developed into a powerful fingerprint spectroscopic technique that can provide rapid, non-invasive, non-destructive, and ultra-sensitive detection, even down to single molecular level. Consequently, a considerable amount of researchers combined MOFs with the SERS technique to further improve the sensing performance and broaden the applications of SERS substrates. Herein, representative synthesis strategies of MOFs to fabricate SERS-active substrates are summarized and their applications in ultra-sensitive biomedical trace detection are also reviewed. Besides, relative barriers, advantages, disadvantages, future trends, and prospects are particularly discussed to give guidance to relevant researchers.

Abstract:Since the discovery of surface enhanced Raman scattering (SERS), the choice of SERS-active materials has been limited mainly to metals, especially gold and silver in the visible spectrum. Although non-metals can also be SERS-active by forming nanostructures or composite structures with SERS-active materials, the mechanism behind it is still unclear and there is no perfect technique to study it. In this work, by constructing a SERS structure on a flexible polydimethylsiloxane film, we provide a way to study the effect of non-metallic nanostructures on Raman enhancement by attaching the above film onto flat and nanostructured surfaces. it was found that a nanoporous silicon surface contributes to an additional, up to five times, Raman enhancement. The pore depth and pore size also influence the observed Raman enhancement. These findings will help us not only to understand the mechanism of SERS involving non-metallic nanostructures, but also to design more efficient SERS structures for various applications.
Mesoporous Nanostructures Encapsulated with Metallic Nanodots for Smart SERS Sensing

Abstract:In virtue of uniform mesopores and core–shell nanoarchitectures, metallic nanodot-encapsulated hollow mesoporous nanostructures have shown promising potential in various applications. However, their fabrication with versatile tunability of the encapsulated metallic content has been a challenge. Herein, we have prepared metallic nanodot-encapsulated hollow mesoporous silica nanoparticles (M-HMSNPs) with adjustable inner metallic components. The sacrificial template of polystyrene (PS) nanoparticles precoated with metals (Au/Ag/Pt) is fully wrapped with mesoporous silica (mSiO2). The metallic nanodots are formed during the template removal process by calcination. The type and content of the encapsulated nanodots can be readily and precisely controlled by the initially deposited metallic layers. We demonstrate the application of the gold (Au) nanodot-loaded HMSNPs (denoted Au-HMSNPs) as smart surface-enhanced Raman spectroscopy (SERS) probes, which can screen between big molecules and small analytes. With the aid of a Raman reporter, the SERS probe can successfully quantify H2O2, which is used to distinguish cancer cells in vitro. Further integrated with enzymes, the SERS chips of specificity are prepared and used to detect corresponding substrates of glucose and uric acid, responsively. Abstract:Surface-enhanced Raman spectroscopy (SERS) is a powerful novel analytical tool which integrates high levels of sensitivity for trace analysis of chemical and biomolecular species due to the massive enhancement of Raman signals by using nanometre-sized metal particles. However, SERS can be envisaged as an analytical tool only if substrates with strong, predictable and reproducible SERS enhancement can be produced. Here we have developed one simple Ar+ ions sputtering technology to prepare gold nano-cones array on silicon substrates as surface-enhanced Raman scattering (SERS)-active substrates. The tip of the gold cone-structures exhibited an extremely sharp curvature with an apex diameter of 20 nm and the interior apex angle of the nanocones was around 20 degrees. These samples were evaluated as potential SERS substrates using Rhodamine 6G molecules as molecule probe and exhibited SERS enhancement factor of greater than 10, originated from the localized electron field enhancement around the apex of cones and the surface plasmon coupling of periodic structures.

Abstract:In this work, we designed a process to assemble gold nanoparticles onto a three-dimensional (3D) polymer surface, which can then be monitored using surface-enhanced Raman scattering (SERS). This work is the first demonstration of the assembly of gold nanoparticles on a filter film and in situ measurement with Raman spectroscopy. Herein, a polyhexamethylene adipamide (Nylon66) film embedded in the organic filter film was used as a template to fabricate a tunable SERS-active substrate. A "hotspot"-rich gold-nanoparticle-decorated polymer substrate for SERS was prepared; this substrate exhibited high sensitivity in trace detection of targets. The study was conducted using 4-mercaptobenzoic acid as a probe molecule with the aim of comparing the scattering efficiency and the homogeneity of the Raman signal on selected substrates. In addition, we used the gold-decorated polymer film to detect a biotin-avidin complex. The most powerful advantage of the proposed microanalytical device is the in situ SERS application. The 3D nanoporous structures described in this work hold strong potential for use in various applications such as environmental monitoring and biomolecule detection.

Abstract:Large-area gold and silver nanoparticles inside periodical silicon microhole arrays were prepared via simple electrodeposition. Such metal nanoparticles within Si microholes showed a strong surface-enhanced Raman scattering (SERS) effect, which enabled the detection of rhodamine at 10(-8) M and 10(-9) M on substrates of AuNPs and AgNPs, respectively. The SERS activity of the substrates depended on the metal particle morphology. Microfabrication combined with electrodeposition appears to be a viable approach for preparing reproducible SERS Substrates for sensitive Raman detection.

Abstract:The performance of surface-enhanced Raman scattering (SERS) active substrates highly depends on its structure and components. Herein, for the first time, a novel zeolitic imidazolate framework-8 (ZIF-8)/Ag nanoparticles composite with flower-like architecture is demonstrated. By using tannic acid as structure-directing agent, well-controlled flower-like ZIF-8 could be obtained. Through in-situ reduction, Ag nanoparticles with uniform grain distribution were directly grown on the surface of ZIF-8 nanosheets. The synergy effects of preconcentration and plasmonic hot spot allow the picomolar detections and high reproducibility of the SERS method. As a result, the minimum detection limit of rhodamine 6G can be as 10(-14) mol L-1 and the stability and repeatability are satisfactory. This work provides a facile and general method to develop plasmonic ZIF-8 hybrid with controlled morphology for potential applications in bioanalysis, chemical sensing and catalysis.

Abstract:Surface-enhanced Raman scattering (SERS) has proven to be promising for the detection of trace analytes; however, the precise nanofabrication of a specific and sensitive plasmonic SERS-active substrate is still a major challenge that limits the scope of its applications. In this work, gold nanoparticles are self-assembled into densely packed two-dimensional arrays at a liquid/liquid interface between dimethyl carbonate and water in the absence of template controller molecules. Both the simulation and experiment results show that the particles within these film-like arrays exhibit strong electromagnetic coupling and enable large amplification of Raman signals. In order to realize the level of sensing specificity, the surface chemistry of gold nanoparticles (Au NPs) is rationally tailored by incorporating an appropriate chemical moiety that specifically captures molecules of interest. The ease of fabrication and good uniformity make this platform ideal for in situ SERS sensing of trace targets in complex samples.

Abstract:Metal–organic framework (MOF) compounds are centered on metal ions or metal ion clusters, forming lattices with a highly ordered periodic porous network structure by connecting organic ligands. As MOFs have the advantages of high porosity, large specific surface area, controllable pore size, etc., they are widely used in gas storage, catalysis, adsorption, separation and other fields. SERS substrate based on MOFs can not only improve the sensitivity of SERS analysis but also solve the problem of easy aggregation of substrate nanoparticles. By combining MOFs with SERS, SERS performance is further improved, and tremendous research progress has been made in recent years. In this review, three methods of preparing MOF-based SERS substrates are introduced, and the latest applications of MOF-based SERS substrates in biosensors, the environment, gases and medical treatments are discussed. Finally, the current status and prospects of MOF-based SERS analysis are summarized.

Composite Structure Based on Gold-Nanoparticle Layer and HMM for Surface-Enhanced Raman Spectroscopy Analysis

Abstract:Hyperbolic metamaterials (HMMs), supporting surface plasmon polaritons (SPPs), and highly confined bulk plasmon polaritons (BPPs) possess promising potential for application as surface-enhanced Raman scattering (SERS) substrates. In the present study, a composite SERS substrate based on a multilayer HMM and gold-nanoparticle (Au-NP) layer was fabricated. A strong electromagnetic field was generated at the nanogaps of the Au NPs under the coupling between localized surface plasmon resonance (LSPR) and a BPP. Additionally, a simulation of the composite structure was assessed using COMSOL; the results complied with those achieved through experiments: the SERS performance was enhanced, while the enhancing rate was downregulated, with the extension of the HMM periods. Furthermore, this structure exhibited high detection performance. During the experiments, rhodamine 6G (R6G) and malachite green (MG) acted as the probe molecules, and the limits of detection of the SERS substrate reached 10−10 and 10−8 M for R6G and MG, respectively. Moreover, the composite structure demonstrated prominent reproducibility and stability. The mentioned promising results reveal that the composite structure could have extensive applications, such as in biosensors and food safety inspection.

Abstract:Surface-enhanced Raman scattering (SERS) has attracted great interest due to its remarkable enhancement, excellent sensitivity, and the "fingerprinting" ability to produce distinct spectra for detecting various molecules. Noble metal nanomaterials have usually been employed as SERS-active substrates because of their strong SERS enhancement originated from their unique surface plasmon resonance (SPR) properties. Because the SPR property depends on metal material's size, shape, morphology, arrangement, and dielectric environment around metal nanostructures, the key to wider applications of SERS technique is to develop plasmon-resonant structures with novel geometries to enhance Raman signals and to control the periodic ordering of these structures over a large area to obtain reproducible Raman enhancement. This review presents a general view on the theory background of SERS effect and several basic concepts and focuses on recent progress in engineering metallic nanostructures with various morphologies using versatile methods for improving SERS properties. Their potential applications in the field of chemical detection and biological sensing are overviewed.

Abstract:Surface-enhanced Raman scattering (SERS), due to its extreme sensitivity down to the single molecule level, has been a superior analytical technique for (bio)chemical sensing and monitoring. However, the reliability of SERS has been a major issue since its discovery and has not yet been fully addressed, which is largely due to the lack of properly designed SERS substrates and facile, cost-effective fabrication strategies of metallic nanostructures. Herein, we employ hierarchical metallic nanopatterns as active SERS substrates, which can be facilely fabricated by superplastic nanomolding of bulk metals with hierarchical anodic aluminum oxide templates. The SERS signals of such hierarchical metal substrates show both high sensitivity (SERS intensity of up to 10(6) with an enhancement factor of similar to 7.0 X 10(5)) and reproducibility (relative standard deviation as low as 7%) with an optimized configuration, which significantly outperforms that without hierarchical nanostructures. Our results reveal that the microcavities of the hierarchical molds can significantly reduce the nonuniform plastic deformation of the bulk metals caused by surface roughness, while the nanoholes in the microcavities yield densely packed nanopillars with exceptional SERS signals. These hierarchical metallic nanostructures are further applied for the detection of biomolecules and organic dyes with concentrations as low as 10(-10) M, whose SERS intensity still presents a prominent signal-to-noise ratio. Such facile and scalable fabrication of hierarchical metallic nanostructures not only sheds light on the key factors that affect the uniformity and sensitivity of SERS but also provides guidelines for rational design of superior SERS substrates with outstanding performances, which is of great implication for (bio)chemical sensing and pollutant monitoring.

Abstract:In this study, a novel composite Surface-Enhanced Raman Scattering (SERS) substrate is proposed for ultrasensitive detection. Consisting of gold sinusoidal nanograting and silver colloidal nanoparticles (AgNPs-AuSG), this type of SERS substrate is easy for fabrication by maskless laser interference lithography, and capable of providing large-scale ultra-high field enhancement, attributed to localized surface plasmons (LSPs) and surface plasmon polaritons (SPPs). The enhancement factor (EF) of this composite substrate is as high as up to 10 orders of magnitude in the simulation experiment. Experimental results show that this large-area, productive SERS substrate of AgNPs-AuSG has realized sensitive TNT and RDX detection with the limit of detection (LOD) of 10−10 M, which may be a potential candidate for trace explosives detection.

Abstract:In this work, we reported the fabrication of different sizes of polystyrene colloidal arrays which were used as a template to fabricate large areas of gold nanocaps (AuNCs) and gold nanobowls (AuNBs) through sputter deposition and solvent etching. The monolayer self-assembly process, formation mechanism of surface-enhanced Raman scattering (SERS)-active structures, and characterization of the SERS substrate were thoroughly studied and evaluated. Rhodamine 6G (R6G) was used as the target molecule to systematically compare the SERS signal changes with different substrate coatings. The AgNPs-AuNCs substrate that Ag nanoparticles (AgNPs) coated on the convex of AuNCs showed stronger and better-resolved spectra for the R6G molecule, a limit of detection (LOD) of R6G reached as low as 10-7 M and its SERS enhancement factor (EF) approached 4.05×108. The results demonstrated that the prepared substrate had promising potential as a cost-effective, robust, and rapid detection tool for SERS-based trace detection.

Abstract:Surface-enhanced Raman spectroscopy (SERS) has great potential for the analysis of molecules adsorbed on metals with rough surfaces or substrates with micro-/nanostructures. Plasmonic coupling between metal nanoparticles and the morphology of the rough metal surface can produce "hot spots" that enhance Raman scattering by adsorbed molecules, typically at micro- to nanomolar concentrations, although high enhancement factors can also facilitate single-molecule detection. This phenomenon is widely applicable for chemical analysis and sensing in various fields. In this review, the latest research progress on SERS micro-/nanosensors is evaluated, and the sensors are classified according to their individual functions. Furthermore, the design principles and working mechanisms of reported SERS-active micro-/nanostructured substrates are analyzed, and the design features adopted to overcome the difficulties associated with precision detection are explored. Finally, challenges and directions for future development in this field are discussed. This review serves as a design guide for novel SERS-active substrates.
Surface-enhanced Raman Scattering of Subwavelength Metallic Structures

Abstract:Surface-enhanced Raman scattering (SERS) makes the Raman signals, as fingerprints of different vibration modes of chemical bonds, significant in practical applications. Two main mechanisms, chemical and physical, are attributed to the SERS of molecules adsorbed on metals. The physical mechanism plays a major role in SERS, which is the focus of our paper. Recent SERS systems are mostly based on dimer structures, i. e. nanoparticle pairs, of noble metals. Large amplification of electrical field occurs in the gap of a dimer structure compared with a single nanoparticle. The above gap positions are called as "hot spots" of SERS. In addition, the reproducibility and reliability of SERS substrates are also important for practical applications. Here we use periodical subwavelength metallic structures to meet such needs, and develop other kinds of electrical field enhancement mechanisms. We present the electrical field enhancement of the band-edge mode of surface plasmon polariton, gap plasmon polariton mode, as well as their coupling mode. We choose one-dimensional subwavelength metallic structures to clarify the physical mechanism. Our purpose is to develop subwavelength metallic structures with even and intensive "hot spots", serving as ultrasensitive solid-state SERS substrates with excellent reproducibility and reliability.
