Abstract:Cell therapies have progressed to cures for hematopoietic disorders, neurodegenerative diseases, and cancer. However, only some patients can benefit from cell therapies even with prior screening. Due to the limited clinical methods to monitor the in vivo therapeutic functions of these transferred cells over time, the uncertain prognosis is hard to attenuate. Positron emission tomography (PET) cell tracking can provide comprehensive dynamic and spatial information on the proliferation status and whole-body distribution of the therapeutic cell. In this work, we designed and synthesized the first SNAP-tagged PET radiotracer. SNAP tag is an O 6-alkylguanine-DNA alkyltransferase that can form an irreversible bond with 18F-BG-surface for in vivo cell tracking based on a reporter gene system. 18F-BG-surface was obtained by the F-Al radiolabeling method in 32 ± 7% radiochemical yield and showed a high in vitro stability in mouse serum. SNAP-tagged cells could be selectively targeted by 18F-BG-surface both in vitro (4.81 ± 0.08%AD/106 cell vs 2.26 ± 0.10%AD/106 cell) and in vivo (1.90 ± 0.05 vs 0.55 ± 0.02% ID/g, p < 0.01).

Abstract:In this study, a calibration model based on Near-infrared spectroscopy (NIR) technique and chemometrics method was developed for rapid and non-destructive detecting the polysaccharide contents of lentinula edodes samples collected from different regions. The polysaccharide contents of these samples were firstly determined by standard phenol-sulphruic acid method. Then, NIR spectra of these samples were collected by using Fourier transform infrared spectrometry. Based on these experimental data, a random forest method was further used to distinguish the regions of these samples, with a classification accuracy of 96.6%. After that, a rapid, accurate, and quantitative model was established for predicting the polysaccharide contents of these samples. In the model establishing process, some signal pre-treatment methods were optimized, and the validation results with highest determination coefficient (R2) and low root mean square errors of prediction (RMSEP) were, 0.925 and 0.720, respectively. These results showed that combined NIR technique with chemometrics was an effective and green method for lentinula edodes quality control.

Abstract:The 18 kDa translocator protein (TSPO) is a viable target for imaging of inflammation in brain. In the recent past we have explored a pharmacophore skeleton acetamidobenzoxazolone for positron emission tomography (PET) imaging of TSPO expression in brain. Here we evaluated a new radioligand for visualization of TSPO, namely [C-11]N-(2-methoxyoxyphenyl)-N-methyl-2-(5-nitro-2-oxobenzo[d]oxazol-3(2H)-yl)acetamide ([C-11]N '-MPB). This PET ligand exhibited high binding affinity towards TSPO (K-i = 4.9 nM) and a suitable lipophilicity (log D) of 2.08 for brain imaging. A biodistribution study on mice showed high accumulation of radioactivity in TSPO-rich organs, such as the lungs, heart, kidneys, and adrenal glands. Metabolite analysis of rat brain homogenate showed 98% intact [C-11]N '-MPB at 30 min after injection. To determine the specific binding of the radioligand with TSPO on neuroinflammation of the brain, in vitro autoradiography and PET studies were performed in an ischemic rat model. In vitro autoradiography indicated significantly increased binding on the ipsilateral side compared with that on the contralateral side of ischemic rat brains. This result was supported firmly by the contrast of radioactivity in PET images. Displacement experiments with PK11195 minimized the difference in radioactivity uptake between the two sides. In summary, [C-11]N '-MPB is a potential PET imaging radioligand for TSPO and, consequently, for gaining more insight for up-regulation of microglia during neuroinflammation.
Study on the pharmacokinetics of mulberry fruit polysaccharides through fluorescence labeling

Abstract:A sensitive and efficient fluorescence labeling method was developed and validated for the microanalysis and detection of polysaccharides. Fluorescein isothiocyanate (FITC) was successfully labeled on mulberry fruit polysaccharides (MFP) through a reductive amination reaction with the assistant of tyramine. The fluorescent labeled polysaccharides (FMFP) was identified by fluorescence, UV-visible, flourier transform infrared (FT-IR) and 1H NMR spectrum. Results demonstrated that the labeling efficiency of FMFP was 0.32%, and the FMFP was stable in simulated digestion fluid without cytotoxicity. The pharmacokinetics and biodistribution after administration were analyzed in rats, which indicated that the FMFP obtained could be absorbed in a short time (tmax 0.50 h) but eliminated slowly (t1/2 8.77 ± 1.38 h). At 24 h after administration, the polysaccharide could be tested mainly in intestine, stomach, liver and kidney. The FITC labeling method lays a foundation for investigating the absorption and metabolism of MFP, and provides references for the microanalysis research of bioactive polysaccharides.
Preparation and Preliminary Molecular Imaging Study of 124I In-Situ Labeled Organic Melanin Nanoparticles

Abstract:Developing biocompatible, multifunctional and in-situ labeling nanoplatform is high challenging for molecular imaging. Organic derivates melanin nanoparticles (MNPs) holds great potential to be multimodal contrast agents, and have been used for photoacoustic imaging, magnetic resonance imaging, and Cu-64 PET imaging with simple modifications. In order to extend MNPs application in molecular imaging, here a novel radio-nuclide was applied to in-situ labeling of MNPs. Large numbers of active dihydroxyindole/indolequinone groups and natural binding ability of MNPs enabled them to have the ability to label different types of radionuclides which have unique half-life and functions, especially long-life elemental nuclide. This project explored the in-situ labeling methods of organic melanin nanoparticles with a promising diagnostic radionuclides named Iodine-124 (I-124), and using this novel multifunctional organic nanoparticles for preliminary molecular imaging studies. Generally, ultrafine particle size melanin nanoparticles (5.5 nm in diameter) were prepared by ultrasonication method using naturally occurring melanin, then PEG(3500) which had amino group at both ends was used as a stabilizer agent to obtain PEG-MNP nanocarriers (7.5 nm in diameter) with better water solubility and stability. The nanoparticles were full characterized by dynamic light scattering (DLS), transmission electron microscope (TEM) and H-1 NMR, respectively. Then, one kind of elemental nuclide was labeled. Classic iodine labeled method with N-Bromo Succinimide (NBS) was used as oxidant to oxidize active dihydroxyindole/indolequinone ring of PEG-MNP for electrophilic substitution reaction labeling I-124 (100.8 h). This reaction rate is extremely fast (60 s reaction time) and high labelling yield (>99%). The I-124 was labeled successfully and in-situ labeled PEG-MNP nanocarriers were obtained. After that, I-124 and I-124-PEG-MNP were used to further preclinical evaluation by micro-PET imaging. Micro-PET images were collected at 2 h, 24 h and 48 h after intravenous injection 7.4 MBq I-124 and I-124-PEG-MNP in normal Kunming mice (n=3). The ROI target area of heart, liver and thyroid were delineated for semi-quantitative analysis. Then, in order to verify the imaging ability of I-124-PEG-MNP in solid tumor. We built human pancreatic cancer BxPC3 xenograft model (n=3), and Micro-PET scans were performed at different time points. Results showed that the labeling rate of I-124 on PEG-MNP was 99.9%. And the radiochemical purity in vitro stability of I-124-PEG-MNP in 96 h was more than 90%. Micro-PET images showed that I-124-PEG-MNP had no obvious thyroid uptake which indicated no de-marking in mice. The radio-distribution of I-124 and I-124-PEG-MNP was substantially different in liver and thyroid (P<0.001). In vivo semi-quantitative analysis showed that the radio uptakes of organs were consistent with the distribution of nanoparticles. And the PET imaging of xenograft mice showed that I-124-PEG-MNP can utilize the enhanced permeability and retention effect (EPR) to be significantly enriched at the tumor and retained in the tumor site for more than 48 h. PEG-MNP has the ability to label long half-life nuclide I-124. This research provides an experimental basis for further construction of long-circulation multimodal imaging probes.

Abstract:<p>Lentinan (LNT), a typical triple helix β-glucan, has been widely used as drug and biomaterial. However, its pharmacokinetics <em>in vivo</em> is rarely reported, which severely limits its further development and application. The aim of this study is to establish a sensitive method for detecting LNT in biosamples and to evaluate the plasma level, tissue distribution and metabolic degradation of LNT in rats. 5-([4,6-Dichlorotriazin-2-yl] amino) fluorescein (DTAF) was labelled to LNT. After purification and identification, FLNT was intravenously administered to rats at dose of 32 mg/kg. LNT was predominantly incorporated into the liver and liver microsomes were used to study the degradation mechanism of LNT in the liver. The results showed that two cytochrome P450 (CYP450) enzymes subtypes (CYP2D6 and CYP2C9), as well as epoxide hydrolase, were involved in the metabolic degradation of LNT. These findings provide a pharmacokinetic reference for further study and application of LNT and other β-glucans.</p>

Preparation and Bioevaluation of 18F-Labeled Small-Molecular Radiotracers via Sulfur(VI) Fluoride Exchange Chemistry for Imaging of Programmed Cell Death Protein Ligand 1 Expression in Tumors

Abstract:Nowadays, one of the most effective methods of tumor immunotherapy is blocking programmed cell death protein 1/programmed cell death protein ligand 1 (PD-1/PD-L1) immune checkpoints. However, there is still a significant challenge in selecting patients to benefit from immune checkpoint therapies. Positron emission tomography (PET), a noninvasive molecular imaging technique, offers a new approach to accurately detect PD-L1 expression and allows for a better prediction of response to PD-1/PD-L1 target immunotherapy. Here, we designed and synthesized a novel group of aryl fluorosulfate-containing small-molecule compounds (LGSu-1, LGSu-2, LGSu-3, and LGSu-4) based on the phenoxymethyl-biphenyl scaffold. After screening by the time-resolved fluorescence resonance energy transfer (TR-FRET) assay, the most potent compound LGSu-1 (half maximal inhibitory concentration (IC50): 15.53 nM) and the low-affinity compound LGSu-2 (IC50: 189.70 nM) as a control were selected for 18F-radiolabeling by sulfur(VI) fluoride exchange chemistry (SuFEx) to use for PET imaging. [18F]LGSu-1 and [18F]LGSu-2 were prepared by a one-step radiofluorination reaction in over 85% radioconversion and nearly 30% radiochemical yield. In B16-F10 melanoma cell assays, [18F]LGSu-1 (5.00 ± 0.06%AD) showed higher cellular uptake than [18F]LGSu-2 (2.55 ± 0.04%AD), in which cell uptake could be significantly blocked by the nonradioactivity LGSu-1. In vivo experiments, micro-PET imaging of B16-F10 tumor-bearing mice and radiographic autoradiography of tumor sections showed that [18F]LGSu-1 was more effectively accumulated in the tumor due to the higher binding affinity with PD-L1. The above experimental results confirmed the potential of the small-molecule probe LGSu-1 as a targeting PD-L1 imaging tracer in tumor tissues.

Abstract:The purpose of this study was to develop an efficient synthetic method for labeling d -Glucosamine with indium-111 ( 111 In), and to investigate the imaging properties of the resulting radiotracer in MDA-MB-468 xenograft models using single-photon emission computed tomography (SPECT). The precursor compound, 2-(4-Isothiocyanato benzyl)-1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid-deoxyglucosone (DOTA-DG), was synthesized from 2- d -Glucosamine. DOTA-DG was labeled with 111 In within 20 min. The labeling efficiency and radiochemical purity of 111 In-DOTA-DG were >95 and >96 % as determined by radio-HPLC. SPECT imaging studies were performed using nude mice bearing MDA-MB-468 mammary tumors after intravenous injection of at a dose of 1.11 MBq (0.1 mL) 111 In-DOTA-DG. Tumors were clearly delineated by SPECT imaging at 120 min after injection.

Abstract:Rare-earth-based nanoparticles have attracted increasing attention for their unique optical and magnetic properties. However, their application in bioimaging has been limited to photoluminescence bioimaging and magnetic resonance imaging. To facilitate their use in other bioimaging techniques, we developed a simple, rapid, efficient and general synthesis strategy for (18)F-labeled rare-earth nanoparticles through a facile inorganic reaction between rare-earth cations and fluoride ions. The (18)F-labeling process based on rare-earth elements was achieved efficiently in water at room temperature with an (18)F-labeling yield of >90% and completed within 5 min, with only simple purification by aqueous washing and centrifugation, and without the use of organic agents. The effectiveness of (18)F-labeled rare-earth nanoparticles was further evaluated by positron emission tomography (PET) imaging of their in vivo distribution and application in lymph monitoring. In addition, this strategy is proposed for the creation of a dual-model bioimaging technique, combining upconversion luminescence bioimaging and PET imaging.

Abstract:The determination of nanoparticle (NP) stability and degradation in vivo is essential for the accurate evaluation of NP biodistribution in medical applications and for understanding their toxicological effects. Such determination is particularly challenging because NPs are extremely difficult to detect and quantify once distributed in a biological system. Radiolabelling with positron or gamma emitters and subsequent imaging studies using positron emission tomography (PET) or single-photon emission computerised tomography (SPECT) are some of the few valid alternatives. However, NPs that degrade or radionuclides that detach or are released from the NPs can cause artefact. Here, submicron-sized poly(lactide-co-glycolide) nanoparticles (PLGA-NPs) stabilised with bovine serum albumin (BSA) were dual radiolabelled using gamma emitters with different energy spectra incorporated into the core and coating. To label the core, 111In-doped iron oxide NPs were encapsulated inside PLGA-NPs during NP preparation, and the BSA coating was labelled by electrophilic substitution using 125I. After intravenous administration into rats, energy-discriminant SPECT resolved each radioisotope independently. Imaging revealed different fates for the core and coating, with a fraction of the two radionuclides co-localising in the liver and lungs for long periods of time after administration, suggesting that NPs are stable in these organs. Organ harvesting followed by gamma counting corroborated the SPECT results. The general methodology reported here represents an excellent alternative for visualising the degradation process of multi-labelled NPs in vivo and can be extended to a wide range of engineered NPs.

Abstract:Lentinan (LNT) is a highly functional biomacromolecule, and its sulfated derivatives (sLNT) exhibit improved pharmacological properties compared to LNT. In the present study, LNT extracted from the fermentation broth of Lentinus edodes was sulfated using two methods, conventional heating and novel microwave radiation, and their biological activities were evaluated using an antioxidant system and an antiproliferation cell model. Our results indicate that the two sulfated derivates have similar physicochemical properties and spectroscopic profiles. Nevertheless, microwave radiation produced a higher yield of sLNT, with a shorter processing time and lower degradation. In contrast to LNT, the sLNT derivatives exhibited a strong antioxidant activity in vitro. In addition, the derivatives exhibited a significant antiproliferative effect on leukemia cells, whereas LNT exhibited the opposite outcome.

Abstract:Quantitative nuclear imaging techniques are in high demand for various disease diagnostics and cancer theranostics. The non-invasive imaging modality requires radiotracing through the radioactive decay emission of the radionuclide. Current preclinical and clinical radiotracers, so-called nuclear imaging probes, are radioisotope-labeled small molecules. Liposomal radiotracers have been rapidly developing as novel nuclear imaging probes. The physicochemical properties and structural characteristics of liposomes have been elucidated to address their long circulation and stability as radiopharmaceuticals. Various radiolabeling methods for synthesizing radionuclides onto liposomes and synthesis strategies have been summarized to render them biocompatible and enable specific targeting. Through a variety of radionuclide labeling methods, radiolabeled liposomes for use as nuclear imaging probes can be obtained for in vivo biodistribution and specific targeting studies. The advantages of radiolabeled liposomes including their use as potential clinical nuclear imaging probes have been highlighted. This review is a comprehensive overview of all recently published liposomal SPECT and PET imaging probes.

Studies on two new lymph imaging agents and their clinical application

Abstract:Liposome and PG kit were labeled with the99mTcO4 to form99mTc-liposome and99mTc-PG which are two non-invasive, safe diagnostic drugs. The labeling efficiency of the agents are more than 95%. The99mTc imaging agents in the PLN as compared with the other different organs showed that the ratio value of PLN to other organ is more than 149 times after 30 min i.d. injection of99mTc-liposome and the ratio value is more than 30.19 times after 2 h i.d. injection of99mTc-PG. The studies of the pharmacokinetics and mechanism of imaging have been carried out. The99mTc-liposome was administrated to more than 140 patients and99mTc-PG was administrated to more than 5000 patients. The clinical tests were satisfactory and no side-effect was shown.

Abstract:With the development of PD-1/PD-L1 immune checkpoint inhibitor therapy, the ability to monitor PD-L1 expression in the tumor microenvironment is important for guiding therapy. This study was performed to develop a novel radiotracer with optimal pharmacokinetic properties to reflect PD-L1 expression in vivo via single-photon emission computed tomography (SPECT) imaging. [99mTc]Tc-HYNIC-WL12-tricine/M (M = TPPTS, PDA, ISONIC, 4-PSA) complexes with high radiochemical purity (>97%) and suitable molar activity (from 100.5 GBq/μmol to 300 GBq/μmol) were prepared through a kit preparation process. All 99mTc-labeled HYNIC-WL12 radiotracers displayed good in vitro stability for 4 h. The affinity and specificity of the four radiotracers for PD-L1 were demonstrated both in vitro and in vivo. The results of biodistribution studies displayed that the pharmacokinetics of the 99mTc-HYNIC-conjugated radiotracers were significantly influenced by the coligands of the radiotracers. Among them, [99mTc]Tc-HYNIC-WL12-tricine/ISONIC exhibited the optimal pharmacokinetic properties (t1/2α = 8.55 min, t1/2β = 54.05 min), including the fastest clearance in nontarget tissues, highest tumor-to-background contrast (e.g., tumor-to-muscle ratio, tumor-to-blood ratio: 40.42 ± 1.59, 14.72 ± 2.77 at 4 h p.i., respectively), and the lowest estimated radiation absorbed dose, highlighting its potential as a clinical SPECT imaging probe for tumor PD-L1 detection.

Abstract:Abstract Precise diagnosis of lymph node metastasis is important for therapeutic regimen planning, prognosis analysis and probably better outcomes for cancer patients. In this work, 68Ga-labeled amphiphilic alternating copolymers nanoparticles with different rigid ligands were synthesized as positron emission tomography (PET) probes for lymph node metastasis imaging. The labeling efficiency and stability of nanoparticles was improved with increased rigidity of coordination unit. PU(68Ga-L-MDI-PEG) nanoparticles (PU(68Ga-L-MDI-PEG) NPs) with the strongest rigidity of coordination unit exhibited the lowest critical micelle concentration, the best 68Ga labeling efficiency and stability. During in vivo lymph node metastasis imaging, PU(68Ga-L-MDI-PEG) NPs led to different accumulations in normal lymph nodes (N-LN) and tumor metastasized sentinel lymph nodes (T-SLN), which resulted in different PET signal presentation, making it feasible to differentiate N-LN from T-SLN. In comparison, small molecule probe 68GaL had poor lymph node accumulation, not only making it difficult to find lymph nodes on PET/computed tomography scan, but also tough to distinguish N-LN from metastatic ones. Overall, this work provides a reference for design of 68Ga labeled polymeric nanoparticles with high chelation efficiency and stability, as sensitive PET probes for lymph node imaging.

Radioisotope Post-Labeling Upconversion Nanophosphors for in Vivo Quantitative Tracking.

Abstract:Lanthanide based upconversion nanophosphors (UCNPs) attracted increasing attention for potential applications in bioimaging, while its in vivo behaviors are not clear until now due to no available quantification imaging tools. Herein, we developed a unique rare-earth cation-exchange-based postlabelling method to introduce 153Sm into the lattice of UCNPs, providing this 153Sm-postlabeling UCNP having bifunction of radioactive property and upconversion luminescence under excitation at 980 nm laser. This 153Sm-postlabelling method shows rapid treatment time of <1 min, high labeling yield of >99%, and without usage of organic solvents. More importantly, this 153Sm-postlabelling method is also suitable for most of rare earth nanoparticles to track their in vivo behaviors. The dynamic quantification studies of the in vivo fate of the rare-earth nanoparticles were further investigated by radioactive detection method such as single-photon emission computed tomography (SPECT) and gamma counter. The imaging results revealed that UCNPs were mainly captured by the mononuclear phagocyte system (liver and spleen). The amount of nanoparticles in liver arrived at its peak quicker and was about 15 fold of that in spleen. And the nanoparticles will be slowly excreted with the bile. Therefore, the concept of postlabeling 153Sm onto lanthanide-based UCNPs may serve as a facile strategy of fabricating multifunctional nanoprobes for upconversion luminescence (UCL) and SPECT dual-modality imaging.

Abstract:The interest in developing new drug carriers for delivery to the liver using natural polysaccharides with a high galactose content has necessitated the study of the pharmacokinetics and tissue distribution of these polysaccharides. In this paper, a new method was established for the microanalysis of Angelica sinensis polysaccharide (ASP) in biosamples. Fluorescein-labelled ASP (FA) was rapidly eliminated from the bloodstream and distributed to the liver with high specificity following intravenous injection. The analysis of the hepatocellular localization demonstrated that FA was predominantly endocytosed by the parenchymal cells, an observation consistent with the results obtained from microscopy studies. Additionally, FA showed a high affinity for asialoglycoprotein receptor-rich cells, while minimal binding of FA to asialoglycoprotein receptor-poor cells was observed. Moreover, the absorption of FA was markedly inhibited by the co-administration of neogalactosylalbumin (NGA) both in vivo and in vitro. To allow for the visualization of the systemic circulation of ASP, 99mTc-DTPA-ASP was synthesized and in vivo imaging was performed with single photon emission computed tomography (SPECT). It also showed a high aggregation of 99mTc-DTPA-ASP in liver. These results suggest that the distribution of ASP to the liver occurs via asialoglycoprotein receptor (ASGPR) mediated endocytosis and ASP could potentially be applied as a new carrier for delivering drugs to the liver.
Preclinical characterization of [ 18 F]D 2 -LW223: an improved metabolically stable PET tracer for imaging the translocator protein 18 kDa (TSPO) in neuroinflammatory rodent models and non-human primates

Abstract:Positron emission tomography (PET) targeting translocator protein 18 kDa (TSPO) can be used for the noninvasive detection of neuroinflammation. Improved in vivo stability of a TSPO tracer is beneficial for minimizing the potential confounding effects of radiometabolites. Deuteration represents an important strategy for improving the pharmacokinetics and stability of existing drug molecules in the plasma. This study developed a novel tracer via the deuteration of [ 18 F]LW223 and evaluated its in vivo stability and specific binding in neuroinflammatory rodent models and nonhuman primate (NHP) brains. Compared with LW223, D 2 -LW223 exhibited improved binding affinity to TSPO. Compared with [ 18 F]LW223, [ 18 F]D 2 -LW223 has superior physicochemical properties and favorable brain kinetics, with enhanced metabolic stability and reduced defluorination. Preclinical investigations in rodent models of LPS-induced neuroinflammation and cerebral ischemia revealed specific [ 18 F]D 2 -LW223 binding to TSPO in regions affected by neuroinflammation. Two-tissue compartment model analyses provided excellent model fits and allowed the quantitative mapping of TSPO across the NHP brain. These results indicate that [ 18 F]D 2 -LW223 holds significant promise for the precise quantification of TSPO expression in neuroinflammatory pathologies of the brain.

[Radiolabelling of a lung cancer-targeting small molecule polypeptide with (131)I and its radioactivity distribution in normal rabbits]

Abstract:Objective: To establish a labeling method for a specific lung cancer-targeting small molecule peptide cNGQGEQc with ¹³¹I and observe the radioactivity distribution of the labeled peptide in rabbits using single-photon emission computed tomography (SPECT). Methods: Chloramine-T method was used for ¹³¹I labeling of the tyrosine amino group on cNGQGEQc, and the labeling efficiency and radiochemical purity of ¹³¹I-cNGQGEQc were determined with paper chromatography. The stability of ¹³¹I-cNGQGEQc in saline and human serum was assessed after incubation in water bath at 37 degrees celsius; for 24 h. The octanol-water partition coefficient lg P (the radioactivity counting ratio of ¹³¹I-cNGQGEQc dissolved in 100 µl octanol or in 100 µl saline) was calculated. SPECT was performed in 3 male New Zealand white rabbits after intravenous injection of ¹³¹I-cNGQGEQc to observe the dynamic distribution of the peptide with the time-radioactivity curve (T-A curve) of the region of interest (ROI). Results: With a labeling efficiency of 90%, ¹³¹I-cNGQGEQc showed a radiochemical purity of was 95% after purification with HPLC. The radiochemical purity of ¹³¹I-cNGQGEQc was (93.12 ± 1.18)% and (88.34 ± 5.43)% after intubation in saline and human serum for 24 h, respectively. The octanol-water partition coefficient lg P of ¹³¹I-cNGQGEQc was -1.75, suggesting its hydrosolubility. In rabbits with intravenous injection of ¹³¹I-cNGQGEQc, SPECT visualized the kidneys at 1 min after the injection; the imaging of the heart and liver became attenuated at 5 min when the bladder was visualized with an increasing radioactivity. The radioactivity of the soft tissues began to fade at 30 min. No gallbladder visualization was detected, and the radioactivity of the abdomen remained low. No obvious radioactivity concentration was observed in the thyroid and stomach. The T-A curves of the ROI of all the tissues and organs descended over time. Conclusion: Radiolabeling of cNGQGEQc with ¹³¹I is simple and highly efficient. ¹³¹I-cNGQGEQc has good stability in vitro and good distribution characteristics for in vivo imaging, and is cleared mainly by renal excretion due to its hydrosolubility. These results provide experimental basis for further studies of diagnosis and therapy of lung cancer with targeting polypeptide.
