Abstract:Efficient copper-catalyzed 1,2-difunctionalization of alkenes with commercially available BrCH2Cl as a chloromethylating source was carried out, in which mild conditions, high reactivity, excellent functional-group tolerance, and late-stage modification of a bioactive molecule are demonstrated. This strategy offers a solution for the diverse syntheses of nitrogen-containing terminal alkyl chlorides, a common synthetic handle that is promising for multiple derivatizations. Mechanistic studies indicate that a chloromethyl radical is involved in the catalytic cycle.

Abstract:A novel and efficient free-radical-promoted copper-catalyzed decarboxylative alkylation of α,β-unsaturated carboxylic acids with ICH2CF3 and its analogues has been developed. This methodology provides a convenient access to the synthesis of allylic trifluoromethyl and β-CF3 ketone containing compounds as well as other biologically useful fluorinated molecules and materials.

Abstract:Functionalized ketene dithioacetals are important intermediates in organic synthesis.α-Functionalization reactions based on them provide potential precursors for the construction of usefully carbo-and heterocyclic compounds.In this paper,a versatileα-bromination of functionalized ketene dithioacetals was developed.In the presence of H2 O,the reactions of functionalized ketene dithioacetals(1) with CuBr 2 in THF at room temperature gave variousα-bromo ketene dithioacetals(2) in excellent yields with high regioselectivities.The mechanism of thisα-bromination is proposed to undergo an addition of bromine formed in situ to compound 1 and sequontial elimination of theα-proton.The simplicity of execution,mild conditions,inexpensive,easily usable brominating agent and high yields,make this synthetic method attractive for practical applications.

Abstract:A copper-catalyzed dichloromethylazidation of alkenes using commercially available BrCCl2H as a stoichiometric dichloromethylating reagent has been reported. Mechanistic studies indicated that a dichloromethyl radical was involved in the catalytic cycle. This transformation has demonstrated mild conditions, high reactivity, excellent functional-group tolerance, and a broad substrate scope, and thus it has offered a new solution for difunctionalization of alkenes to make diverse transformable dichloromethylated molecules.

Abstract:α-substituted ketones are important chemical targets as synthetic intermediates as well as functionalities in in natural products and pharmaceuticals. We report the α-acetylation of C(sp3)-H substrates R-H with arylmethyl ketones ArC(O)Me to provide α-alkylated ketones ArC(O)CH2R at RT with tBuOOtBu as oxidant via copper(I) β-diketiminato catalysts. Proceeding via alkyl radicals R•, this method enables α-substitution with bulky substituents without competing elimination that occurs in more traditional alkylation reactions between enolates and alkyl electrophiles. DFT studies suggest the intermediacy of copper(II) enolates [CuII](CH2C(O)Ar) that capture alkyl radicals R• to give R-CH2C(O)Ar under competing dimerization of the copper(II) enolate to give the 1,4-diketone ArC(O)CH2CH2C(O)Ar.

Abstract:A copper-catalyzed highly regio- and stereoselective silaboration of alkynes was developed. In this work, direct cis-difunctionalization of alkynes was realized with silaboronate reagent and copper catalyst in aprotic solvents. The regiodivergent silaborations were controlled by tuning the copper catalysts and phosphine ligands used in reactions. This protocol provides an efficient and practical method to synthesize the multisubstituted functionalized alkenes with specific stereoselectivity.

Abstract:Alkyl constitutes one of the most widely used groups in organic synthesis. Here, the development of a series of thianthrenium salts, which act as reliable alkylation reagents, are ready to engage in copper-catalyzed Sonogashira reaction to build C(sp 3 )-C(sp) bonds under mild photoinduced conditions. Diverse alkyl thianthrenium salts, methyl and disubstituted thianthrenium salts are employed with great functional breadth, since sensitive Cl, Br and I are compatible and challenging to achieve by conventional approaches. The generality of the developed alkyl reagents has also been demonstrated in copper-catalyzed Kumada reactions.

The method is based on a copper(I)-catalyzed three-component reaction of a terminal alkyne, diazoesters and aldehydes. Abstract:Allenenitriles bearing different synthetically versatile functional groups have been prepared smoothly from 5-alkynyl fluorosulfonamides in decent yields with an excellent chemo- and regio-selectivity under redox neutral conditions. The resulting allenenitriles can be readily converted to useful functionalized heterocycles. Based on mechanistic study, it is confirmed that this is the first example of radical-based non-activated propargylic C-H functionalization for allene syntheses.

Abstract:The formal C–C bond insertion into aldehydes is an attractive methodology for the assembly of homologated carbonyl compounds. However, the homologation of aldehydes has been limited to diazo approach and the enantioselective reaction was rarely developed. Herein, we report an asymmetric formal C–C bond insertion into aldehydes through diyne cyclization strategy. In the presence of Cu(I)/SaBOX catalyst, this method leads to the efficient construction of versatile axially chiral naphthylpyrroles in moderate to excellent yields with good to excellent enantioselectivities. This protocol represents a rare example of asymmetric formal C–C bond insertion into aldehydes using non-diazo approach. The combined experimental and computational mechanistic studies reveal the reaction mechanism, origin of regioselectivity and stereoselectivity. Notably, the chiral phosphine ligand derived from synthesized axially chiral skeleton was proven to be applicable to asymmetric catalysis.
