This work was aim to assess the effects of a cardiovascular serum marker, oxidized low-density lipoprotein (ox-LDL) in PCa. METHODS Concentrations of ox-LDL in peripheral blood samples from 75 patients with localized or metastatic PCa or noncancerous controls was investigated by enzyme-linked immunosorbent assay analysis (ELISA). In vitro studies investigated the effects of ox-LDL in different PCa cell lines (LNCaP and PC-3), and phosphorylation protein chips were used to compare altered cellular signal pathways stimulated by treatment of ox-LDL. Expression patterns of OLR1, the ox-LDL receptor in cancer cells, were analyzed by tissue microarray in paired biopsy samples of PCa mentioned above, and compared to the clinical pathological characters in the cohort. The external validation was conducted using The Cancer Genome Atlas (TCGA) database. RESULTS Peripheral ox-LDL levels were significantly higher in metastatic PCa. In vitro, Ox-LDL increased the proliferation, apoptosis as well as the migration and invasion capabilities of LNCaP and PC-3, in a dose-dependent manner. Phosphorylation protein chips indicated exogenous ox-LDL stimulated the activation of cellular nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kB), P27, c-Myc, ¦Â-catenin, STAT1/STAT3, and caspase-3. Dose-response experiments showed that ox-LDL was able to increase OLR1 protein expression in a dose-dependent manner, and OLR1 expressions in PCa tissues were correlated to peripheral ox-LDL levels. OLR1 was associated with high Gleason Score (>7) and metastatic disease both in the present and the TCGA cohort. CONCLUSIONS Ox-LDL may promote metastasis in PCa, by acting on the proliferation, apoptosis as well as the migration and invasion capabilities of cancer cells.
