Abstract:Osteoporosis is a debilitating disease that increases with age and is characterized by increased bone fragility. However, dietary factors can influence the development of osteoporosis. For example, high fat diets (HFD) have been shown to have detrimental effects on bone density and quantity. However, the effects of HFD on bone microarchitecture have not been extensively studied. The architecture of the bone is important because it provides information regarding the quality of the bone and not just quantity. Therefore, the purpose of this study was to investigate the effects of HFD on bone microarchitecture in mice when compared to low fat diet fed mice. Five-week-old mice were randomized into two different treatment groups, control and HFD (n = 6 per group). The control group were fed AIN-93 G growing rodent diet (20% protein, 65% carbohydrate (CHO), 15% fat) while the HFD group was fed a standard HFD ordered from Research Diets Inc. Cat #D12451M (20% protein, 35% CHO, 45% fat) for 20 weeks. After 20 weeks the animals were sacrificed and left tibial bone specimen were prepared for analysis via micro-computed tomography. Data were analyzed using one-way ANOVA to detect differences between groups. Mean values ± SEM for bone measurements (control, HFD) are as follows. Total Volume (TV) (6.04 mm3 ± 0.077, 6.16 mm3 ± 0.12 mm), bone volume (BV) (2.33 mm3 ± 0.054, 1.89 mm3 ± 0.06), BV/TV (0.386% ± 0.011, 0.31% ± 0.01), structural model index (SMI) (–0.687 ± 0.097, –0.35 ± 0.09), connective density (Conn.D) (775.83 ± 51.2, 693.8 ± 91.7), trabecular number (Tb.N) (7.52 mm ± 0.135, 6.23 mm ± 0.32), trabecular thickness (Tb.Th) (0.059 mm ± 0.0009, 0.056 mm ± 0.00069), and trabecular separation (Tb.Sp) (0.139 mm ± 0.003, 0.18 mm ± 0.0063). BV, BV/TV, Tb.N and Tb.Th values were all significantly higher (P < 0.05) in the control group versus the HFD group while SMI and Tb.Sp were significantly lower (P < 0.05). These results agree with previous studies showing that HFD has detrimental effects on bone. This study demonstrated that HFD negatively impacts the microarchitecture of bone, specifically trabecular number, thickness, and separation which are important for overall structure, strength, and flexibility of bone. Future studies should focus on mechanisms underlying the role of HFD on the microarchitecture of bone. None

Abstract:Objective:To evaluate the effect of fatty liver induced by high fat diet in C57 mice on bone microstructures and the possible mechanism. Methods:A total of 20 C57 male mice(4 weeks old)were enrolled and divided into two groups, fed for 12 weeks either with standard chow(control;n=9)or with high-fat diet to induce fatty liver(HFD;n=11). Blood, femur, tibia and liver samples were collected after sacrifice. Plasma alanine aminotransferase(ALT)and aspartate aminotransferase(AST)concentration were examined. The liver was stained with H&E and Oil red-O to determine if the mice were suffering from fatty liver and to measure the amount of triglyceride(TG)and total cholesterol(TC). The bone samples were scanned using a micro-CT system in a high-resolution scanning mode. Total volume(TV), bone volume(BV), total bone volume fraction(BV/TV), connectivity density(Conn.D), structural model index(SMI), trabecular number(Tb.N), trabecular thickness(Tb.Th), trabecular spacing(Tb. Sp), apparent density(Ap.Dens), material density(Mat.Dens)and degree of anisotropy(DA)were compared between control and HFD groups. The number of marrow fat drops was measured by some of the femur with HE staining. Results:In the HFD group, all mice were proved with fatty liver by liver H&E and oil red O staining. Compared to the control group, the HFD group had lower levels of TV[(2.128 ± 0.591)mm3 vs.(3.570 ±0.330)mm3, P<0.001)], Tb.N[(1.769 ± 0.218)/mm vs.(2.284 ± 0.726)/mm, P=0.030)], SMI(2.950 ± 0.242 vs. 3.820 ± 0.729, P=0.004)and higher levels of Tb.Sp[(0.595 ± 0.083)mm vs.(0.472 ± 0.116)mm, P=0.013]in femur. The HFD group also had lower levels of TV[(1.127 ± 0.338)mm3 vs(. 1.741±0.683)mm3, P=0.017]and SMI(2.431 ± 0.501 vs. 3.188 ± 0.465, P=0.003)in tibia. Bone marrow histomorphological analysis showed that the number of adipose drop(AD)per mm2 bone marrow was significantly higher in the HFD group compared with control mice(5.5 ± 2.2 vs. 2.6 ± 0.5, P=0.042). AD was positively with TV(Femur:r=-0.756, P=0.030; Tibia:r=-0.771, P=0.025). Conclusion:Our results suggest an adverse effect of fatty liver on bone microstructure in C57 mice. Bone marrow adiposity may be a factor underlying this physiopathologic process.
Effects of High-Fructose Corn Syrup on Bone Health and Gastrointestinal Microbiota in Growing Male Mice

Abstract:Here, we explored the correlation between gut microbiota and bone health and the effects of high-fructose corn syrup (HFCS) on both. Sixteen 3-week-old male C57BL/6J mice were randomly divided into two groups and given purified water (control group) or 30% HFCS in water (HFCS group) for 16 weeks. The effects of HFCS were assessed via enzyme-linked immunosorbent assays, histopathological assays of colon and bone, and 16S rDNA sequence analysis of gut microbiota. The serum of HFCS group mice had lower levels of bone alkaline phosphatase (BALP), bone Gla protein (BGP), insulin-like growth factor 1 (IGF-1), and testosterone, and higher levels of type I collagen carboxyl-terminal telopeptide (ICTP) and tartrate-resistant acid phosphatase (TRAP) than that of the control group. HFCS caused trabecular bone damage by decreasing trabecular number and thickness and increasing trabecular separation. The HFCS group colons were shorter than the control group colons. The HFCS-fed mice showed mild, localized shedding of epithelial cells in the mucosal layer, focal lymphocytic infiltration of the lamina propria, mild submucosal edema, and loosely arranged connective tissue. The HFCS group displayed lower abundance and altered composition of gut microbiota. The abundance of Defluviitaleaceae UCG-011, Erysipelatoclostridium, Ruminococcaceae UCG-009, Lactobacillus, Blautia, and Parasutterella increased, positively correlating with BALP, BGP, IGF-1, and testosterone levels, and negatively correlating with ICTP and TRAP levels. Our study revealed a potential diet-gut microbiota-bone health axis.
Effects of long-term high-fat diet on bone mineral density and intestinal flora

Abstract:Objective:To investigate the effects of long-term high-fat diet on bone mineral density and intestinal flora in mice.Methods:Sixteen male C57BL/6 mice were randomly divided into control group (NC group) and high-fat group (HF group). After 24 weeks of high-fat feeding, biochemical indicators such as blood glucose and blood lipids were detected, bilateral femurs were taken and bone microstructure was analyzed with micro-computered tomography (micro-CT), and changes of intestinal microbial composition and proportion were revealed using 16S rDNA sequencing technology.Results:Compared with the control group, the serum total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) in HF group were significantly increased. Micro-CT uncovered that the bone mineral density (Tb.BMD), bone volume fraction (BV/TV) and the number of trabecular bone (Tb.N) decreased, yet structural model index (SMI) and the trabecular fraction (Tb.Sp) increased in the HF group mice. The gut microbiota 16S rDNA sequence analysis showed that the proportion of Proteobacter was significantly increased and the proportions of pachycete, warty microbacterius, and actinomycete were reduced in HF group at the phyla level. The proportion of Bacteroidetes S24-7_norank in the NC group was significantly higher than that in the HF group, and the multilevel discriminant analysis of species differences (LEfSe) identified that the difference was significant, yet the proportion of Bacteroides, Pseudo-Prevotella, Desulfovibrio, Altobacter, and Helicobacter in the HF group were higher than those in the NC group, which were significant differences in Altobacter and Helicobacter at genus level.Conclusion:Long-term high-fat feeding can cause the destruction of femoral trabecular structure, decrease in the number of trabeculus bones, and bone mineral density in C57BL/6 mice. It also leads to significant changes in the composition and proportion of the intestinal flora.

Abstract:A high-fat diet (HFD) is defined as a diet that contains lipids that account for more than 30% of the total energy intake, and current research has documented cases with intakes as high as 45% and 60%. There is a view that patients who have a tendency to consume a HFD are more susceptible to various kinds of diseases, including osteoporosis, metabolic syndrome, coronary heart disease, and cancer. Thus, hypotheses have been proposed that a HFD may serve as a significant risk factor for bone loss and osteoporosis. A plethora of studies has suggested a relationship between a HFD and bone health. Moreover, high fat has a vital effect on the bone structure and bone health, and intestinal flora imbalances and intestine barrier deterioration, inflammation, oxidative stress, adipokine changes, and bone marrow fat tissue (BMFT) accumulation are thought to be potential mechanisms. Most research has demonstrated that a HFD diminishes bone mineral density and bone microstructure. Some studies, however, showed that a HFD contributes to achieving peak bone mass, which is associated with weight gain. As diet is modifiable, lifestyle changes and medication can help bone improvement, as well as alleviating bone loss associated with a HFD. This review aims to give a comprehensive understanding of the relationship between a HFD and bone health, which might provide strategies to improve bone health by varying daily dietary components and building a healthy lifestyle. We also hope that further treatments for diet-related bone loss can be put forward.
High-fat diet causes undesirable bone regeneration by altering the bone marrow environment in rats

Abstract:Objective: Diet structure has changed greatly over the last few decades, and high-calorie diets have become an integral part of people's daily diet, as well as the important cause of obesity in society. Several organ systems, including the skeletal system, are seriously affected by high-fat-diets (HFD) in the world. There is, however, still a lack of knowledge about the effects of HFD on bone regeneration and the possible mechanisms involved. In this study, the difference in bone regeneration between rats under HFD and low-fat-diets (LFD) was evaluated by monitoring the process of bone regeneration in distraction osteogenesis (DO) model animals, as well as the possible mechanisms. Methods: A total of 40 Sprague Dawley (SD) rats (5 weeks old) were randomly divided into HFD group (n=20) and LFD group (n=20). Except for feeding methods, there were no differences between the two groups in terms of treatment conditions. All animals received the DO surgery eight weeks after starting to feed. After a delay of 5 days (latency phase), the active lengthening phase was performed for 10 days (0.25 mm/12 h), and the consolidation phase followed for 42 days. An observational study of bone included radioscopy (once a week), micro-computed tomography (CT), general morphology, biomechanics, histomorphometry, and immunohistochemistry. Result: The results showed that HFD group had a higher body weight than LFD group after 8, 14, and 16 weeks of feeding. Furthermore, at the final observation, there were statistically significant differences between LFD group and HFD group in terms of total cholesterol (TC), triglycerides (TG), low-density lipoprotein (LDL), and high-density lipoprotein (HDL) levels. Additionally, observations on bone regeneration showed a slower regeneration and a lower biomechanical strength in HFD group than LFD group, based on radiography, micro-CT, general morphology, biomechanics, histomorphometry, and immunohistochemistry. Conclusion: In this study, HFD resulted in elevated blood lipids, increased adipose differentiation at the bone marrow level, and delayed bone regeneration. The pieces of evidence are beneficial to better understand the association between diet and bone regeneration and to adjust the diet optimally for fracture patients.

Abstract:Background Accumulating evidence indicates that high-fat diet (HFD) is a controllable risk factor for osteoporosis, but the underlying mechanism remains to be elucidated. As a primary biological barrier for nutrient entry into the human body, the composition and function of gut microbiota (GM) can be altered rapidly by HFD, which may trigger abnormal bone metabolism. In the current study, we analyzed the signatures of GM and serum metabolomics in HFD-induced bone loss and explored the potential correlations of GM and serum metabolites on HFD-related bone loss. Methods We conducted a mouse model with HFD-induced bone loss through a 12-week diet intervention. Micro-CT, Osmium-μCT, and histological analyses were used to observe bone microstructure and bone marrow adipose tissue. Quantitative Real-Time PCR was applied to analyze gene expression related to osteogenesis, adipogenesis, and osteoclastogenesis. Enzyme-linked immunosorbent assay was used to measure the biochemical markers of bone turnover. 16s rDNA sequencing was employed to analyze the abundance of GM, and UHPLC-MS/MS was used to identify serum metabolites. Correlation analysis was performed to explore the relationships among bone phenotypes, GM, and the metabolome. Results HFD induced bone loss accompanied by bone marrow adipose tissue expansion and bone formation inhibition. In the HFD group, the relative abundance of Firmicutes was increased significantly, while Bacteroidetes , Actinobacteria , Epsilonbacteraeota , and Patescibacteria were decreased compared with the ND group. Association analysis showed that thirty-two bacterial genera were significantly related to bone volume per tissue volume (BV/TV). One hundred and forty-five serum metabolites were identified as differential metabolites associated with HFD intervention, which were significantly enriched in five pathways, such as purine metabolism, regulation of lipolysis in adipocyte and cGMP-PKG signaling pathway. Sixty-four diffiential metabolites were matched to the MS2 spectra; and ten of them were positively correlated with BV/TV and five were negatively correlated with BV/TV. Conclusions These findings indicated that the alternations of GM and serum metabolites were related to HFD-induced bone loss, which might provide new insights into explain the occurrence and development of HFD-related osteoporosis. The regulatory effects of GM and metabolites associated with HFD on bone homeostasis required further exploration.

Abstract:With people aging, osteoporosis is expected to increase notably.Nutritional status is a relatively easily-modified risk factor, associated with many chronic diseases, and is involved in obesity, diabetes, and coronary heart disease (CHD), along with osteoporosis.Nutrients, such as fats, sugars, and proteins, play a primary function in bone metabolism and maintaining bone health.In Western nations, diets are generally high in saturated fats, however, currently, the nutritional patterns dominating in China continue to be high in carbohydrates from starch, cereals, and sugars.Moreover, high fat or high sugar (fructose, glucose, or sucrose) impart a significant impact on bone structural integrity.Due to diet being modifiable, demonstrating the effects of nutrition on bone health can provide an approach for osteoporosis prevention.Most researchers have reported that a high-fat diet consumption is associated with bone mineral density (BMD) and, as bone strength diminishes, adverse microstructure changes occur in the cancellous bone compartment, which is involved with lipid metabolism modulation disorder and the alteration of the bone marrow environment, along with an increased inflammatory environment.Some studies, however, demonstrated that a high-fat diet contributes to achieving peak bone mass, along with microstructure, at a younger age.Contrary to these results, others have shown that a high-fructose diet consumption leads to stronger bones with a superior microarchitecture than those with the intake of a high-glucose diet and, at the same time, research indicated that a high-fat diet usually deteriorates cancellous bone parameters, and that the incorporation of fructose into a high-fat diet did not aggravate bone mass loss.High-fat/high-sucrose diets have shown both beneficial and detrimental influences on bone metabolism.Combined, these studies showed that nutrition exerts different effects on bone health.Thus, a better understanding of the regulation between dietary nutrition and bone health might provide a basis for the development of strategies to improve bone health by modifying nutritional components.
Fructose Consumption Does Not Worsen Bone Deficits Resulting from High-Fat Feeding in Young Male Rats

Abstract:Dietary-induced obesity (DIO) resulting from high-fat (HF) or high-sugar diets produces a host of deleterious metabolic consequences including adverse bone development. We compared the effects of feeding standard rodent chow (Control), a 30% moderately HF (starch-based/sugar-free) diet, or a combined 30%/40% HF/high-fructose (HF/F) diet for 12weeks on cancellous/cortical bone development in male Sprague–Dawley rats aged 8weeks. Both HF feeding regimens reduced the lean/fat mass ratio, elevated circulating leptin, and reduced serum total antioxidant capacity (tAOC) when compared with Controls. Distal femur cancellous bone mineral density (BMD) was 23–34% lower in both HF groups (p<0.001) and was characterized by lower cancellous bone volume (BV/TV, p<0.01), lower trabecular number (Tb.N, p<0.001), and increased trabecular separation versus Controls (p<0.001). Cancellous BMD, BV/TV, and Tb.N were negatively associated with leptin and positively associated with tAOC at the distal femur. Similar cancellous bone deficits were observed at the proximal tibia, along with increased bone marrow adipocyte density (p<0.05), which was negatively associated with BV/TV and Tb.N. HF/F animals also exhibited lower osteoblast surface and reduced circulating osteocalcin (p<0.05). Cortical thickness (p<0.01) and tissue mineral density (p<0.05) were higher in both HF-fed groups versus Controls, while whole bone biomechanical characteristics were not different among groups. These results demonstrate that “westernized” HF diets worsen cancellous, but not cortical, bone parameters in skeletally-immature male rats and that fructose incorporation into HF diets does not exacerbate bone loss. In addition, they suggest that leptin and/or oxidative stress may influence DIO-induced alterations in adolescent bone development.

Abstract:Objective: The aim of the current study was to investigate the effects of obesity, induced via a high-fat diet, on bone metabolism in rats.Methods: Two hundred healthy Wistar male rats aged 4 weeks were fed a standard diet and a high-fat diet. At specific time points (week 0, 4, 6, 8, and 10), plasma was collected to determine the levels of glucose and lipid metabolism. Additionally, enzyme-linked immunoassays were performed to determine the plasma levels of adipocyte and bone metabolism factors. Micro-CT imaging was used to determine the parameters of bone metabolism. At 10th week, immunohistochemistry evaluation of femoral bone samples was performed to determine the expression of adipocyte factors.Result: Receptor activator of nuclear factor kB ligand (RANKL) was positively correlated with levels of triglyceride (TG), free fatty acids (FFA), and tumor necrosis factor alpha (TNF-alpha) (P < 0.05), while receptor activator of the NF-kappa B (RANK) showed a positive correlation with TG, FFA, TNF-alpha and leptin (LPT) (P < 0.05). CT imaging demonstrated that bone mineral density and trabecular thickness were elevated compared to controls before 6 weeks, but these values were found to be lower in rats fed a high fat diet in the following weeks (P < 0.05). Immunohistochemistry showed that the expression of TNF-alpha, Interleukin- 6 (IL-6) and peroxisome proliferator activated receptor-gamma (PPAR-gamma) were increased and the expression of adiponectin (APN) were diminished in rats fed a high-fat diet compared to controls at 10 weeks (p < 0.05).Conclusion: With obesity intensifies, the release of FFA cause inflammation factor increase, resulting in bone parameters decreased. High-fat diet and caffeine interact to modulate bone microstructure and biomechanics in mice

Abstract:Aims: Although excessive fat and caffeine intake are independent risk factors for bone microstructural and functional disturbances, their association remains overlooked. Thus, we investigated the impact of high-fat diet (HFD) and caffeine alone and combined on serum lipid profile, bone microstructure, micromineral distribution and biomechanical properties. Methods: Forty female C57BL/6 mice were randomized into 4 groups daily treated for seventeen weeks with standard diet (SD) or HFD (cafeteria diet) alone or combined with 50 mg/kg caffeine. Key findings: The association between HFD and caffeine reduced the weight gain compared to animals receiving HFD alone. Caffeine alone or combined with HFD increases total and HDL cholesterol circulating levels. HFD also reduced calcium, phosphorus and magnesium bone levels compared to the groups receiving SD, and this reduction was aggravated by caffeine coadministration. From biomechanical assays, HFD combined with caffeine increased bending strength and stiffness of tibia, a finding aligned with the marked microstructural remodeling of the cortical and cancellous bone in animals receiving this combination. Significance: Our findings indicated that HFD and caffeine interact to induce metabolic changes and bone microstructural remodeling, which are potentially related to bone biomechanical adaptations in response to HFD and caffeine coadministration.

Abstract:Obesity-induced inflammation can lead to an imbalance in bone formation and resorption. Our previous studies have demonstrated that apple polyphenols (APs) can reduce body weight and inflammation. But their effect on bone is still unclear. In this study, we investigated the protective effects of APs on bone loss in mice with high-fat-diet (HFD)-induced obesity. Forty male C57BL/6J mice were divided into control group (10% fat diet), HFD group (60% fat diet), resveratrol group (60% fat diet), and AP group (60% fat diet). Micro-computed tomography revealed a significant increase in bone volume fraction and bone mineral density, and more trabecular bone and less trabecular bone separation in the AP group compared with the HFD group. In addition, serum tumor necrosis factor (TNF)-α, interleukin-6 (IL-6), and tartrate-resistant acid phosphatase (TRACP) levels were decreased; runt-related transcription factor 2 (Runx2) levels were increased; the collagen area was enlarged; and femur biomechanical property was enhanced in the AP group compared with the HFD group. APs significantly increased the ratio of osteoprotegerin to the receptor activator for the nuclear factor-κB ligand (OPG/RANKL) compared with the HFD group. Resveratrol could also improve the glucolipid regulation, but poorer osteogenic promotion was found compared to APs. The present study demonstrated that APs prevent loss of bone mass induced by obesity, which has potential implications for the prevention and treatment of obesity-related osteoporosis.

Abstract:Eight-weeks old C57BL/6J male mice were treated with normal or high fat diet for4 weeks,and then bone mass,number and activity of osteoclasts,differentiation of osteoblasts from bone marrow mesenchymal cells and the change in lipid genes expression were studied.Compared with the normal diet treated group,bone mass of femur from high fat diet treated animal was decreased,but there were increased number and activity of osteoclasts and raised lipid genes expression levels.Besides,no obvious change was found in differentiation of osteoblasts from bone marrow mesenchymal cells.The loss of bone mass induced with high fat diet in young mice was mainly due to increased activity of osteoclasts and bone resorption.

Abstract:The transdifferentiation of bone and fat is a new insight in studying the increasingly bone marrow fat in the process of osteoporosis of elderly or menopause crowd which is increasing in prevalence. The loss of bone mass in osteoporosis is multifactorially determined and includes genetic, hormonal and environmental determinants. Although it has long been considered whether the transdifferentiation process does exist in vivo and whether it could be find in the same individual, interaction between skeleton and adipose tissue has been proved pre-clinically and clinically by increasing evidence. Here we focus on the current understanding of the transdifferentiation between bone and fat, the molecular interactions and future clinical implications of recent studies linking the transdifferentiation to bone metabolism diseases. Furthermore, a set of recommendations of bone and fat transdifferentiation on bone metabolism are also presented to facilitate evaluation of this magic process.

Abstract:Objective To provide a novel insight in bone tissue sampling.MethodsTwenty femoral heads were extracted from sample bank of OP patients with femoral neck fracture, and they were matched with the feature of female, age gap less than 5 years and similar size of femoral head (diameter differ less than 2 mm) and were randomly assigned to two groups (A and B). Subchondral bone columns were then extracted from these femoral heads using two different methods (A for standard method while B for conventional method). The morphometric characterization of column specimens, the axis deflection angle and density parameters were compared between two groups.Results Trabeculae extracted in conventional way displayed a heterogeneous distribution as long as multiple lacerated ends. The axis deflection angle (ADA,α°) of conventional group was significantly greater than that of standard group (P=0.006). The density parameters in inferior layers were significantly higher than those in superior layers in both groups. The coefficient of uniformity of standard group was significantly better than that of conventional group in bone mineral density (BMD,P=0.000 8), bone / trabecular volumn (BT/TV,P=0.003 4), trabecular thickness (Tb.Th,P=0.000) and trabecular number (Tb.N,P=0.002 9).Conclusion The inconformity of the spatial position caused by conventional sampling method may affect the evaluation of trabecular microstructure changes, which should be considered as an important experimental bias that will influence the reliability of the results.
Rapid Transition from a High-Fat, High-Fructose to a Low-Fat, Low-Fructose Diet Reverses Gains in Bone Mass and Strength

Abstract:Purpose: Obesity is thought to negatively impact bone quality and strength despite improving bone mineral density. We hypothesized that 1) continuous consumption of a high-fat, high-sugar (HFS) diet would impair bone quality and strength, and 2) a change from an HFS diet to a low-fat, low-sugar (LFS) would reverse HFS-induced impairments to bone quality and strength. Methods: Six-week-old male C57Bl/6 mice ( n = 10/group) with access to a running wheel were randomized to an LFS diet or an HFS diet with simulated sugar-sweetened beverages (20% fructose in place of regular drinking water) for 13 wk. HFS mice were subsequently randomized to continuing HFS feeding (HFS/HFS) or transition to the LFS diet (HFS/LFS) for four additional weeks. Results: HFS/HFS mice exhibited superior femoral cancellous microarchitecture (i.e., greater BV/TV, Tb.N, Tb.Th, and decreased Tb.Sp) and cortical bone geometry (i.e., lower Ct.CSA and pMOI) compared with all other groups. At the femoral mid-diaphysis, structural, but not material, mechanical properties were greatest in HFS/HFS mice. However, HFS/HFS exhibited greater femoral neck strength only when compared with mice assigned to diet transition (HFS/LFS). Osteoclast surface and the percentage of osteocytes staining positive for interferon-gamma were greater in HFS/LFS mice, consistent with reduced cancellous microarchitecture postdiet transition. Conclusions: HFS feeding enhanced bone anabolism and structural, but not material, mechanical properties in exercising mice. A change from an HFS to LFS diet returned the bone structure to that of continuously LFS-fed mice while compromising strength. Our results indicate rapid weight loss from obese states should be performed with caution to prevent bone fragility. A deeper analysis into the altered bone phenotype in diet-induced obesity from a metabolic standpoint is needed.
High-fat diet exacerbates the loss of bone fracture toughness in aging for C57BL/6JN mice

Abstract:The elderly are at increased risk of bone fracture and more often consume poor quality diets, such as high-fat diet (HFD). We hypothesized that HFD would exacerbate the loss of bone fracture resistance in aging. To test this hypothesis, we fed 5-month and 22-month C57BL/6JN mice of both sexes HFD or low-fat diet for 8 weeks. Aging and HFD lowered bone fracture toughness, while only aging reduced bone strength. At the tissue-scale, aging increased bone mineral maturity, while HFD altered bone mineralization and maturity as well as collagen structure. Cortical bone metabolism was also dysregulated with aging and HFD. Dysregulated pathways in aging and HFD were related to cellular function and viability, or glucose regulation, respectively. Aging with HFD also impacted osteoclast and adipocyte abundance and osteocyte viability. Together, these data demonstrate that HFD may exacerbate the loss of bone matrix quality and fracture resistance in aging.

Abstract:Western diets are becoming increasingly common around the world. Western diets have high omega 6 (ω-6) and omega 3 (ω-3) fatty acids and are linked to bone loss in humans and animals. Dietary fats are not created equal; therefore, it is vital to understand the effects of specific dietary fats on bone. We aimed to determine how altering the endogenous ratios of ω-6:ω-3 fatty acids impacts bone accrual, strength, and fracture toughness. To accomplish this, we used the Fat-1 transgenic mice, which carry a gene responsible for encoding a ω-3 fatty acid desaturase that converts ω-6 to ω-3 fatty acids. Male and female Fat-1 positive mice (Fat-1) and Fat-1 negative littermates (WT) were given either a high-fat diet (HFD) or low-fat diet (LFD) at 4 wk of age for 16 wk. The Fat-1 transgene reduced fracture toughness in males. Additionally, male BMD, measured from DXA, decreased over the diet duration for HFD mice. In males, neither HFD feeding nor the presence of the Fat-1 transgene impacted cortical geometry, trabecular architecture, or whole-bone flexural properties, as detected by main group effects. In females, Fat-1-LFD mice experienced increases in BMD compared to WT-LFD mice; however, cortical area, distal femur trabecular thickness, and cortical stiffness were reduced in Fat-1 mice compared to pooled WT controls. However, reductions in stiffness were caused by a decrease in bone size and were not driven by changes in material properties. Together, these results demonstrate that the endogenous ω-6:ω-3 fatty acid ratio influences bone material properties in a sex-dependent manner. In addition, Fat-1 mediated fatty acid conversion was not able to mitigate the adverse effects of HFD on bone strength and accrual.

Abstract:A diabetogenic high fat diet (HFD) can be used to induce insulin resistance and obesity in animal models; however, its effects on bone are unknown. We investigated the effects of long term HFD on bone in ovariectomized (OVX) female rats. We used 12-week-old female rats divided randomly into four groups: sham operation (sham), sham operation with HFD (SHFD), OVX and OVX with HFD (OVX + HFD). Ovaries were removed in the OVX and OVX + HFD groups and the SHFD and OVX + HFD groups were fed a HFD for 28 weeks. Serum estrogen, testosterone, lipid, adiponectin, leptin, tartrate-resistant acid phosphatase (TRAP) and N-mid fragment of osteocalcin (N-MID-OT) levels were measured. Structure, apoptosis and specific transcription factors in bone were evaluated using pathologic, densitometric and immunohistochemical analysis. Body weight, serum leptin, TRAP and testosterone levels were increased, while serum N-MID-OT, estrogen and adiponectin levels were decreased in the SHFD, OVX and OVX + HFD groups. Expression of BCL2-associated X protein, caspase-3, matrix metalloproteinase-9 and calcitonin was increased, while bone mineral density (BMD) and content (BMC) in femurs and lumbar spine, and expression of B cell lymphoma 2, type 1 collagen and osteocalcin were decreased in the bones of the SHFD, OVX and OVX + HFD groups. All indices were greatest in the OVX + HFD group and HFD produced a detrimental effect on bone in both normal and OVX rats, which may be due to increased apoptosis in bone and increased leptin and decreased adiponectin levels in serum. The effects of HFD and OVX may be synergistic.
