Abstract:The catalytic activity of nanocrystal catalysts depends strongly on their structures. Herein, we report three distinct structures of Fe3O4 nanocrystals, cluster spheres, octahedra, and triangular plates, prepared by a similar hydrothermal procedure. Additionally, the three Fe3O4 nanostructures were used as peroxidase nanomimetics and the correlation between the catalytic activities and the structures was first explored by using 3,3',5,5'-tetramethyl-benzidine and H2O2 as peroxidase substrates. The results showed that the peroxidase-like activities of the Fe3O4 nanocrystals were structure dependent and followed the order cluster spheres > triangular plates > octahedra; this order was closely related to their preferential exposure of catalytically active iron atoms or crystal planes. Such investigation is of great significance for peroxidase nanomimetics with enhanced activity and utilization.

Abstract:Ferroptosis is a new form of iron-dependent programmed cell death discovered in recent years, which is caused by the accumulation of lipid peroxidation (LPO) and reactive oxygen species (ROS). Recent studies have shown that cellular ferroptosis is closely related to tumor progression, and the induction of ferroptosis is a new means to inhibit tumor growth. Biocompatible Fe3O4 nanoparticles (Fe3O4-NPs), rich in Fe2+ and Fe3+, act as a supplier of iron ions, which not only promote ROS production but also participate in iron metabolism, thus affecting cellular ferroptosis. In addition, Fe3O4-NPs combine with other techniques such as photodynamic therapy (PDT); heat stress and sonodynamic therapy (SDT) can further induce cellular ferroptosis effects, which then enhance the antitumor effects. In this paper, we present the research progress and the mechanism of Fe3O4-NPs to induce ferroptosis in tumor cells from the perspective of related genes and chemotherapeutic drugs, as well as PDT, heat stress, and SDT techniques.

Abstract:Ferroptosis is characterized by iron accumulation and lipid peroxidation. However, a clinical dose of Fe3O4 nanoparticles could not cause effective ferroptosis in tumors, and the mechanism is yet to be completely understood. In this study, using RNA-seq data, we found that tumor cells could feedback-activate the antioxidant system by upregulating Nrf-2 expression, thus avoiding ferroptosis caused by Fe3O4 nanoparticles. We also found that DHJS (a probe for ROS generation) can antagonize Nrf-2 expression when it synergizes with Fe3O4 nanoparticles, thus inducing ferroptosis in tumor cells. Considering these findings, we created a biomimetic hybrid cell membrane camouflaged by PLGA-loaded Fe3O4 and DHJS to treat osteosarcoma. The hybrid cell membrane endowed the core nanoparticle with the extension of blood circulation life and enhanced homologous targeting ability. In addition, DHJS and Fe3O4 in nanoparticles prompted synergistically lethal ferroptosis in cancer cells and induced macrophage M1 polarization as well as the infiltration of CD8(+) T cells and dendritic cells in tumors. In summary, this study provides novel mechanistic insights and practical strategies for ferroptosis induction of Fe3O4 nanoparticles. Meanwhile, the synthesized biomimetic nanoparticles exhibited synergistic ferroptosis/immunotherapy against osteosarcoma.
Three Birds with One Stone: A Ferric Pyrophosphate Based Nanoagent for Synergetic NIR-triggered Photo/chemodynamic Therapy with Glutathione Depletion

Abstract:Strategies utilizing reactive oxygen species (ROS) to cause cell death are widely practiced for cancer therapy, including photodynamic therapy (PDT) and chemodynamic therapy (CDT). However, the efficiency of PDT alone is greatly hindered by tumor hypoxia, laser penetration depth and the relatively low oxidation performance of O-1(2) and its subsequent products H2O2. Meanwhile, CDT is limited by the deficiency of endogenous H2O2 in cancer cells. If the ROS, produced in PDT process, served as the substrate in CDT, the total ROS production would be greatly enhanced. However, the increased ROS could be scavenged by the overexpressed glutathione (GSH) in cancer cells, impairing the effect of PDT and CDT. Thus, in this study, a self-reinforcing ferric pyrophosphate based nanoagent (FeP-ZnPc) for synergetic NIR-triggered photo/chemodynamic therapy with glutathione depletion ability was constructed. The constructed FeP-ZnPc exhibited "triple use": PDT, CDT and GSH depletion function. Upon internalized into cancer cells, the FeP-ZnPc could release Fe3+ which underwent a redox reaction with GSH, resulting in GSH depletion and the production of Fe2+. Upon laser, Zinc phthalocyanine (ZnPc) in the FeP-ZnPc could generate ROS for PDT directly. Moreover, the elevated ROS particularly H2O2 could serve as the substrate of CDT, accelerating the Fenton reaction process, producing the strongest oxidants, hydroxyl radical (center dot OH) and leading to ROS burst to expedite cell death. This combination strategy of enhancing ROS production and inhibiting ROS elimination could eventually break down the redox homeostasis and enhance the ROS-mediated cancer therapy.

Abstract:Cancer treatments based on reactive oxygen species (ROS) have emerged as effective clinical methods. However, the generation of ROS is significantly suppressed owing to the hypoxic tumor microenvironment. ROS generated in an oxygen-independent manner provides a novel strategy for cancer therapy. Here, we use an initiator as therapeutic drug to realize light controlled generation of free radicals. The initiator was loaded onto Fe3O4 nanoparticles. Upon the irradiation of near infrared light, the temperature of the nanoparticles increased, inducing the decomposition of the initiator to produce alkyl radicals. This therapeutic system showed remarkable damage to cells in hypoxic environment.

Abstract:Ferrite nanoparticles have been widely used in the biomedical field (such as magnetic targeting, magnetic resonance imaging, magnetic hyperthermia, etc.) due to their appealing magnetic properties. In tumor acidic microenvironment, ferrite nanoparticles show intrinsic peroxidase-like activities, which can catalyze the Fenton reaction of hydrogen peroxide ( H 2 O 2 ) to produce highly toxic hydroxyl free radicals (• OH ), causing the death of tumor cell. Recent progresses in this field have shown that the enzymatic activity of ferrite can be improved via converting external field energy such as alternating magnetic field and near-infrared laser into nanoscale heat to produce more • OH , enhancing the killing effect on tumor cells. On the other hand, combined with other nanomaterials or drugs for cascade reactions, the production of reactive oxygen species (ROS) can also be increased to obtain more efficient cancer therapy. In this review, we will discuss the current status and progress of the application of ferrite nanoparticles in ROS-mediated cancer therapy and try to provide new ideas for this area.

Synergistic ROS Generation via Core-Shell Nanostructures with Increased Lattice Microstrain Combined with Single-Atom Catalysis for Enhanced Tumor Suppression

Abstract:This study emphasizes the innovative application of FePt and Cu core-shell nanostructures with increased lattice microstrain, coupled with Au single-atom catalysis, in significantly enhancing •OH generation for catalytic tumor therapy. The combination of core-shell with increased lattice microstrain and single-atom structures introduces an unexpected boost in hydroxyl radical (•OH) production, representing a pivotal advancement in strategies for enhancing reactive oxygen species. The creation of a core-shell structure, FePt@Cu, showcases a synergistic effect in •OH generation that surpasses the combined effects of FePt and Cu individually. Incorporating atomic Au with FePt@Cu/Au further enhances •OH production. Both FePt@Cu and FePt@Cu/Au structures boost the O2 → H2O2 → •OH reaction pathway and catalyze Fenton-like reactions. This enhancement is underpinned by DFT theoretical calculations revealing a reduced O2 adsorption energy and energy barrier, facilitated by lattice mismatch and the unique catalytic activity of single-atom Au. Notably, the FePt@Cu/Au structure demonstrates remarkable efficacy in tumor suppression and exhibits biodegradable properties, allowing for rapid excretion from the body. This dual attribute underscores its potential as a highly effective and safe cancer therapeutic agent.

Abstract:Intracellular reactive oxygen species (ROS) have been extensively shown to play an important role in the regulation of cell proliferation and cell cycle progression. The effects of endogenous ROS on the proliferation and differentiation of cancer stem cells (CSCs) have received increasing attention because of the unique properties of these cells that allow them to drive tumor growth and evade conventional cancer therapies. In this study, poly(L-Lysine) (PLL)-modified Fe(3)O(4) nanoparticles were synthesized to label CSCs derived from U251 glioblastoma multiform. A featured peroxidase-like activity within PLL-modified Fe(3)O(4) nanoparticles that could greatly reduce intracellular H(2)O(2) activity was identified. We also found that PLL-modified Fe(3)O(4) nanoparticles could accelerate the progression of CSC cell cycle, probably due to the impaired activity of endogenous ROS in CSCs. These results show that growth and proliferation of CSCs could be promoted by Fe(3)O(4) nanocarriers in an ROS-dependent manner, and Fe(3)O(4) nanocarriers may be suitable for certain tumor therapies as a drug delivery system.
Copper-induced Synthesis of Versatile FeOx Nanozymes for Catalytic Cancer Therapy

Abstract:The recent advancement of nanocatalysis offers great promise for cancer therapy, and the development of efficient nanozymes for catalytic biological reactions is highly desirable, but still challenging. Herein, we report the synthesis of ultra-small FeOx nanozymes which is enabled by the introduction of Cu2+. The obtained nanozymes exhibit versatile catalytic activities integratively for effective cancer treatment. In particular, the as-synthesized nanozymes display pH-dependent peroxidase (POD)-like catalytic activity and approach the optimal performance at around pH = 4, endowing it with the possibility for the precise treatment of tumors with acidic microenvironment. Furthermore, the FeOx nanozyme possesses a promising glutathione (GSH) depleting capability and allows to reverse the abnormal GSH level in the tumor issues. Additionally, the fabricated nanozymes are able to catalyze the decomposition of hydrogen peroxide (H2O2) to O-2, allowing to alleviate the hypoxic environment of the tumors and improving the function of immune system for the treatment of tumors. More importantly, the FeOx nanozymes showed remarkable stability and maintained the enzymatic activity after six months in storage. Collectively, the FeOx nanozyme was demonstrated as a promisingly effective nanomedicine for tumor treatment by 4T1 cells, providing insight into the rational design of efficient nanozymes for cancer treatment.

Abstract:While Fe3O4 nanoparticles are frequently exploited for the generation of reactive oxygen species (ROS) in the cancer treatment, its therapeutic performance within a yolk–shell nanostructure for Fenton reaction remains unexplored. To investigate the therapeutic potential, a nanoreactor (YSN@ZnPc@HA) is fabricated by loading zinc phthalocyanine (ZnPc) into a yolk–shell nanocomposite (YSN) consisting of Fe3O4 nanoparticles encapsulated by MgSiO3, followed by modifying the silica surface with hyaluronic acid (HA). ZnPc is a typical photosensitizer for photodynamic therapy; Fe3O4 nanoparticles are responsible for Fenton reaction‐based generation of toxic ·OH radicals and also enable magnetic resonance imaging; and HA serves as tumor‐targeting ligand. The obtained YSN@ZnPc@HA is fully characterized by several techniques. Its therapeutic performance is studied both in vitro and in vivo. The experimental results indicate that Fe3O4 nanoparticles in YSN@ZnPc@HA can successfully enhance the therapeutic efficiency of ZnPc by selectively producing ROS in cancer cells. In addition, YSN@ZnPc@HA shows potent T2‐weighted magnetic resonance imaging capacity. These results collectively demonstrate that the YSN@ZnPc@HA nanoparticles can be used as a robust delivery platform for cancer theranostics.
Nanozyme for tumor therapy: Surface modification matters

Abstract:As the next generation of artificial enzymes, nanozymes have shown unique properties compared to its natural counterparts, such as stability in harsh environment, low cost, and ease of production and modification, paving the way for its biomedical applications. Among them, tumor catalytic therapy mediated by the generation of reactive oxygen species (ROS) has made great progress mainly from the peroxidase-like activity of nanozymes. Fe3O4 nanozymes, the earliest type of nanomaterial discovered to possess peroxidase-like activity, has consequently received wide attention for tumor therapy due to its ROS generation ability and tumor cell killing ability. However, inconsistent results of cytotoxicity were observed between different reports, and some even showed the scavenging of ROS in some cases. By collectively studying these inconsistent outcomes, we raise the question whether surface modification of Fe3O4 nanozymes, either through affecting peroxidase activity or by affecting the biodistribution and intracellular fate, play an important role in its therapeutic effects. This review will go over the fundamental catalytic mechanisms of Fe3O4 nanozymes and recent advances in tumor catalytic therapy, and discuss the importance of surface modification. Employing Fe3O4 nanozymes as an example, we hope to provide an outlook on the improvement of nanozyme-based antitumor activity.
Fe3S4 nanozyme inhibits tumor growth by synergistic effects of ferroptosis and apoptosis

Abstract:Although apoptosis therapy offers huge potential for cancer treatment, its efficacy is still limited by KRAS mutations and insufficient H2O2 supply in tumor cells and considered a critical issue to overcome. Herein, we developed a novel peroxidase nanozyme (Fe3S4) with excellent H2O2 affinity and can effectively catalyze ·OH generation from H2O2, which not only induces apoptosis but also activates ferroptosis of tumor cells. Interestingly, Fe3S4 nanozyme administration disturbs iron metabolism, and causes abundant Fe2+ accumulation in tumor cells, which amplifies Fenton reaction and further promotes tumor cell apoptosis and ferroptosis. Additionally, Fe3S4 nanozyme possesses ultrahigh glutathione peroxidase-like activity and can convert GSH into GSSG. The depletion of GSH sensitizes tumor cells to Fe3S4-mediated ferroptosis. With the assistance of ferroptosis, apoptosis activating by Fe3S4 can more effectively to inhibit tumor growth, and even apoptosis-resistant tumors are no different. Thus, this strategy provides a superior platform for strengthening the therapeutic effect of nanozymes.

Abstract:The hypoxic tumor environment prevents the generation of reactive oxygen species (ROS), reducing the therapeutic efficiency. We construct oleylamine (OA) coated CaO2/Fe3O4 nanocomposites to realize oxygenindependent generation of ROS and high efficient treatment of cancer. In the tumor site, CaO2 reacts with water to generate H2O2, which can be catalized by Fe2+ that is produced by Fe3O4, to form highly toxic hydroxyl radicals (center dot OH). To inhibit the premature reaction, CaO2/Fe3O4 nanoparticles were coated with pH sensitive OA. The nanocomposites exhibited remarkable tumor growth inhibition ability and favorable biocompatibility, holding a great potential for hypoxic tumor therapy.
Fe/Ni Layered Double Hydroxide Biocatalysts Inhibit Tumor Growth Through ROS and Ferroptosis Signaling Pathway

Abstract:Nanocatalysts provide a platform for developing anti-tumor strategies by activating intratumoral chemical catalysis to produce highly toxic reactive oxygen species (ROS). However, the biological effect of nanocatalysts on tumor cells, which is pivotal for effective nanotherapy treatment, remains poorly understood. Herein, we present a general and facile method of synthesizing Fe/Ni-layered double hydroxides (LDHs) for enhanced apoptosis- and ferroptosis-induced cancer nano-therapy, as well as a potential mechanism underlying the antitumor effect based on transcriptome sequencing. An analysis of the chemical structure suggests that the Fe atoms interact strongly with the Ni atoms in Fe/Ni-LDH. Compared with Fe-OH and Ni-OH, Fe/Ni-LDH exhibited significantly better ROS-catalytic activity, with a V-max of 5.015 x 10(-2) mu M.s(-1), and generated massive amounts of center dot OH and O-2(center dot-) radicals, enabling outstanding chemo-dynamic therapy. Both in vitro and in vivo experiments indicated a robust apoptosis-induced cell death pathway, increased intracellular oxidative stress, and accumulation of lipid peroxide (LPO), promoting ferroptosis-involved cancer-cell death, which was further confirmed by transcriptome sequencing. The Fe/Ni-LDH biocatalyst not only provides a robust nanoplatform for tumor suppression but also encourages further exploration and applications of nanobiocatalysts.
Peroxidase-like Activity of Fe3O4@carbon Nanoparticles Enhances Ascorbic Acid-Induced Oxidative Stress and Selective Damage to PC-3 Prostate Cancer Cells.

Abstract:Ascorbic acid (AA) is capable of inhibiting cancer cell growth by perturbing the normal redox state of cells and causing toxic effects through the generation of abundant reactive-oxygen species (ROS). However, the clinical utility of AA at a tolerable dosage is plagued by a relatively low in vivo efficacy. This study describes the development of a peroxidase-like composite nanoparticle for use in an AA-mediated therapeutic strategy. On the basis of a high-throughput, one-pot solvothermal approach, Fe3O4@C nanoparticles (NPs) were synthesized and then modified with folic acid (FA) on the surface. Particular focus is concentrated on the assessment of peroxidase-like catalytic activity by a chromogenic reaction in the presence of H2O2. The carbon shell of Fe3O4@C NPs contains partially graphitized carbon and thus facilitates electron transfer in the catalytic decomposition of H2O2, leading to the production of highly reactive hydroxyl radicals. Along with magnetic responsiveness and receptor-binding specificity, the intrinsic peroxidase-like catalytic activity of Fe3O4@C-FA NPs pronouncedly promotes AA-induced oxidative stress in cancer cells and optimizes the ROS-mediated antineoplastic efficacy of exogenous AA. In vitro experiments using human prostate cancer PC-3 cells demonstrate that Fe3O4@C-FA NPs serve as a peroxidase mimic to create hydroxyl radicals from endogenous H2O2 that is yielded in response to exogenous AA via an oxidative stress process. The usage of a dual agent leads to the enhanced cytotoxicity of PC-3 cells, and, because of the synergistic effect of NPs, the administrated dosage of AA is reduced markedly. However, because normal cells (HEK 293T cells) appear to have a higher capacity to cope with additionally generated ROS than cancer cells, the NP-AA combination shows little damage in this case, proving that selective killing of cancer cells could be achieved owing to preferential accumulation of ROS in cancer cells. A possible ROS-mediated mechanism is discussed to elucidate the pharmaceutical profile of the NP-AA agent. In general, this foundational study reveals that the peroxidase-like nanomaterials are applicable for modulating oxidative stress for the selective treatment of cancer cells by generating a high level of endogenous ROS.

Abstract:The ultrasmall nanoparticles easily lead to a more seriously response than larger nanoparticles because of their physicochemical features. It is essential to understand their cytotoxicity effects for their further application. Here, we used ultrasmall 9 nm Fe3O4 NPs to explore its cytotoxicity mechanism on breast cancer cells. We demonstrated 9 nm Fe3O4 NPs was effectively internalized into cells and located in nucleus, subsequently, it inhibited DNA synthesis through inducing S-phase arrest. Moreover, 9 nm Fe3O4 NPs induced ROS production and oxidative damage by disturbing the expression of antioxidant-related genes (HMOX-1, GCLC and GCLM), which resulted in the enhancement of cells apoptosis and inhibition of cell proliferation, suggesting its potential to be used as therapeutic drug.

Abstract:Photodynamic therapy (PDT) that utilizes apoptosis induced by reactive oxygen species (ROS) has received extensive attention in practical cancer therapy. However, the hypoxic microenvironment of solid tumors significantly limits the efficacy of therapy. Approaches that overcome the barriers to PDT in hypoxic conditions by simultaneously producing ROS exogenously and improving the oxygenation of tumors have never been studied. Herein, an activatable ROS platform was designed that uses the high reactivity of peroxidase-like Fe3O4 toward endogenous hydrogen peroxide (H2O2) to concurrently generate OH as a therapeutic agent and provide O-2 for oxygen-dependent PDT. Multifunctional chitosan-encapsulated Fe3O4 nanoparticles modified with CuS and porphyrin (FCCP NPs) were fabricated to achieve multimodal imaging and synergetic therapy. The FCCP NPs possess enhanced intrinsic peroxidase mimetic activity to produce ROS and O-2 from endogenous H2O2. Multimodal imaging in vivo, including photoacoustic imaging (PAI), magnetic resonance imaging (MRI), photoluminescence imaging (PLI), and photothermal imaging (PTI), exploits the tumor-targeting property of FCCP NPs upon intravenous injection. It can induce cancer cell death with remarkable efficiency both in vitro and in vivo via synergetic treatment with PDT and photothermal therapy (PTT). This study demonstrates the promise of the activatable generation of ROS and O-2 for PDT with nanotechnology to overcome a current deficiency in cancer therapies.
Enhanced highly toxic reactive oxygen species levels from iron oxide core–shell mesoporous silica nanocarrier-mediated Fenton reactions for cancer therapy

Abstract:In this study, iron oxide core-shell mesoporous silica nanoparticles (Fe3O4@MSN) were prepared via the hydrolysis of tetraethyl orthosilicate on the surfaces of Fe3O4 nanoparticles, and these were further conjugated with folate (PEG-FA) and mitochondrial targeting triphenylphosphonium (TPP) to form Fe3O4@MSN-TPP/PEG-FA. A reactive oxygen species (ROS) promoting synergistic combined chemotherapy platform was designed through Fe3O4@MSN-TPP/PEG-FA encapsulating doxorubicin (DOX) and 3-amino-1,2,4-triazole (AT) for cancer therapy. DOX could stimulate the activation of nicotinamide adenine dinucleotide phosphate oxidases (NOXs), which change oxygen into superoxide radicals, which could be further triggered to produce hydrogen peroxide (H2O2) using the superoxide dismutase (SOD) enzyme. AT, as a catalase inhibitor, was employed to inhibit catalase activity to protect the production of H2O2. Thereafter, H2O2 was catalyzed with the help of Fe2+/Fe3+ to form highly toxic free hydroxyl radicals through Fenton reactions, which could induce cell death via synergistic DOX therapy. From in vitro assays, the prepared DOX/AT-loaded Fe3O4@MSN-TPP/PEG-FA showed remarkable inhibition efficiency (3.23% cell viability and 88.1% cell apoptosis) towards MGC-803 cells. This work has created a novel approach to gradually promote the production of ROS and combine this with chemotherapy to enhance anticancer efficacy.

Abstract:In this study, we developed a novel Fe3O4 nanoparticles-doxorubicin (DOX)-Hyaluronic acid (HA) nanoparticles on the basis of firstly discovered "formed porous structure" in spontaneously assembled Fe3O4 nanoparticles. The Mechanism of Action (MOA) behind this porous DOX-loading cargo was tested and confirmed. A multi-functional Fe3O4-DOX + HA nanoparticle was further constructed by incorporating HA into our system. In vitro and in vivo studies exhibited that Fe3O4-DOX + HA owned enhanced antitumor efficacy with significantly prolonged survival time due to the combination of M-1 polarization ability of Fe3O4 nanoparticles, tumor killing effect of DOX and tumor and TAM-targeting effect of HA. All in all, our studies offer a novel strategy to develop a multifunctional antitumor system with a simple preparation method and an enhanced therapeutic outcome.
Porous Yolk-Shell Fe/Fe3O4 Nanoparticles with Controlled Exposure of Highly Active Fe(0) for Cancer Therapy.

Abstract:The iron-based Fenton-type reaction has drawn tremendous attention in cancer therapy. Compared with oxidized iron, Fe(0) possesses high catalytic activity but unstable for biomedical application. Here, we report a new strategy to stabilize Fe(0) via a porous yolk shell nanostructure of Fe/Fe3O4 (PYSNPs) in normal physiological condition, and to control the release of Fe(0) in tumor microenvironment for enhanced cancer therapy. These PYSNPs display superior tumor inhibition with the IC50 down to 20 μg/mL (over 1 mg/mL for iron oxide nanoparticles as control) for HepG2 cell. A single intravenous injection of as low as 1 mg/kg dosage is effective to suppress tumor growth in vivo. Moreover, the disintegration of PYSNPs in the acidic tumor microenvironment could cause significant change in MRI signal for contrast-enhanced diagnosis. Of note, the resulting Fe3O4 fragments are renal clearable with minimized side effect. In all, this work represented a nanoplatform to stabilize and selectively deliver Fe(0) for highly effective cancer therapy.
