Abstract:This study aimed to hydrolyze soy isolate protein (SPI) using five enzymes (alcalase, pepsin, trypsin, papain, and bromelain) in order to obtain five enzymatic hydrolysates and to elucidate the effect of enzymes on structural and biological activities of the resulting hydrolysates. The antioxidant and hypoglycemic activities of the soy protein isolate hydrolysates (SPIEHs) were evaluated through in silico analysis, revealing that the alcalase hydrolysate exhibited the highest potential, followed by the papain and bromelain hydrolysates. Subsequently, the degree of hydrolysis (DH), molecular weight distribution (MWD), amino acid composition, structure, antioxidant activities, and hypoglycemic activity in vitro of SPIEHs were analyzed. After enzymatic treatment, the particle size, polymer dispersity index (PDI), ζ-potentials, β-sheet content and α-helix content of SPIEHs was decreased, and the maximum emission wavelength of all SPIEHs exhibited red-shifted, which all suggesting the structure of SPIEHs was unfolded. More total amino acids (TAAs), aromatic amino acids (AAAs), and hydrophobic amino acids (HAAs) were found in alcalase hydrolysate. For 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, metal ion chelating activity, α-glucosidase inhibitory activity and α-amylase inhibitory activity, alcalase hydrolysate had the lowest IC50; alcalase hydrolysate and papain hydrolysate had the lowest IC50 for hydroxyl radical scavenging activity. Physiological activity of SPIEHs was evaluated thoroughly by 5-Axe cobweb charts, and the results revealed that alcalase hydrolysate exhibited the greatest biological activities.

Abstract:The effect of ultrafine comminution on physicochemical and adsorption properties of insoluble dietary fiber (IDF) from soybean hull was studied.The physicochemical properties included particle size,specific surface area,water-holding capacity,oil-holding capacity,viscosity curve,and cation-exchanging capacity.The in vitro adsorption capacity for fats,cholesterol,bile salts,glucose and heavy metals of IDF samples were determined.The results showed that the water-holding capacity and oil-holding capacity of IDF powder with a particle size of 16.24 μm were 2.31 and 5.27 g/g,respectively,which were 1.07 and 2.53 g/g higher than those of the sample with a particle size of 61.21 μm,respectively.The viscosity value and cation exchange capacity also showed significant changes.The adsorption capacity for fat,cholesterol,bile salt,glucose and heavy metals of ultrafine IDF were significantly increased especially for samples with a particle size of 28.27 and 40.17 μm.Conclusively,ultrafine comminution could significantly improve physicochemical and adsorption properties of insoluble dietary fiber from soybean hull,which would provide a theoretical reference for intensive processing of soybean hull.

Abstract:<section class="article-section__content"><p>We used ultrasound‐microwave comodification and enzyme modification (cellulase and hemicellulase) methods to extract soluble dietary fibers (SDFs) from black soybean hulls. Moreover, the structure, physical, and chemical properties, as well as the cholesterol‐binding capacity of SDFs before and after modification were analyzed. The average molecular weight of SDFs extracted from raw black soybean hulls was 2.815 × 10<sup>5</sup> Da. By comparison, the average molecular weight of SDFs from ultrasound‐microwave comodified hulls and enzyme‐modified hulls decreased by 33.21% and 45.29%, respectively. The water‐holding capacity (WHC), water‐swelling capacity (WSC), and oil‐holding capacity (OHC) of the extracted SDFs modified by the ultrasound‐microwave method were 3.79 g/g, 1.39 mL/g, and 1.14 g/g, respectively, a 9.54%, 23.01%, and 17.53% increase from the values of raw SDF. The WHC, WSC, and OHC of SDFs modified via the enzyme method were 3.59 g/g, 1.25 mL/g, and 1.03 g/g, respectively, with a 3.76%, 10.62%, and 6.19% increase when compared to raw SDFs. The cholesterol‐binding capacity of SDFs modified via the ultrasound‐microwave and enzyme methods was 13.82 and 12.34 mg/g, respectively, with an increase of 47.98% and 32.20% when compared to raw SDFs. The changes in structure and physical and chemical properties were shown to be closely related to the significantly improved cholesterol‐binding capacity of the SDFs from modified black soybean hulls. This provides a theoretical basis for subsequent research and development of black soybean hulls products. </p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Practical Application</h3><p>At present, the black soybean hull, a byproduct of general grains, is usually abandoned, but black soybean hull is rich in dietary fiber. Enzymatic modification and ultrasound‐microwave comodification were used to treat black soybean hull to prepare small molecular weight, highly active soluble dietary fiber. This research is of great significance to the deep processing of black soybean hull and improvement of the economic benefits of black soybean byproducts.</p></section>

Abstract:Dietary fiber (DF) is an important active polysaccharide in Hericium erinaceus. Obesity can lead to a wide range of diseases. In this work, we investigated the in vitro lipid-lowering effect of soluble dietary fiber (SDF) from H. erinaceus, aiming to provide a basis for the subsequent development of lipid-lowering products. Ultrasound-assisted enzymatic extraction (UAEE) of SDF from H. erinaceus was performed. The optimal extraction parameters determined via single-factor experiments and response surface methodology (RSM) were as follows: Lywallzyme concentration, 1.0%; complex protease concentration, 1.2%; ultrasonication time, 35 min; and ultrasonication power, 150 W. In vitro lipid-lowering experiments revealed that the adsorption amount of cholesterol micelles by H. erinaceus SDF was 11.91 mg/g. The binding amount and binding rate of sodium taurocholate were 3.73 mg/g and 42.47%, respectively, and those of sodium glycocholate were 3.43 mg/g and 39.12%, respectively. The pancreatic lipase inhibition rate reached 52.11%, and the type of inhibition was competitive. Therefore, H. erinaceus SDF has good in vitro lipid-lowering ability.

Abstract:This paper deals with the physicochemical and functional properties of polysaccharides isolated from soybean hull (SHP) and soybean okara (SOP). In the investigation, first, crude SHP and SOP were fractioned and purified, and three purified fractions were obtained from SHP or SOP. Then, the physicochemical properties of the two types of crude polysaccharides and the fractions were analyzed by means of GC and GPC, and the functional properties of the samples were discussed by emulsifying and bile acid binding experiments. The results show that SHP fractions are acid heteropolysaccharides without protein, the relative molecular mass of which is 45~150 ku, while SOP fractions are glycosidoprotein with a relative molecular mass of 14~370 ku; and that, as a water-soluble polysaccharide with excellent functional properties, SOP is superior to SHP in terms of emulsifying performance and in-vitro bile acid binding ability.

Abstract:The preparation of dietary fiber based on soy hull with enzymic hydrolysis, acid hydrolysis and alkali dissolutionwas studied.The advantages and disadvantages of preparation method were compared, and then the optimum process param-eters were drawn. In enzymic hydrolysis, the optimum conditions were: 0.2 percent concentration of neutral protease,hydrolysis temperature 35℃,hydrolysis time 1h and the liquid to solid ratio 6:1.Under thes conditions, the product with 74.93 precent of crude fiber was obtained. Acid hydrolysis was not fit for the manufacture of dietary fiber acording to the qualityof final product. The optimum parameters of alkali dissolution were: 1.0 percent of sodium hydroxide, the liquid to solid ratio6:1, and cooking time 1h. Under these conditions, the final product of 90.42 percent of crude fiber was produced.

Abstract:Four kinds of soluble dietary fibers (raw soybean dregs SDF1,enzymatic hydrolysis SDF2,extruded SDF3,extrusion-enzymatic SDF4)were extracted from soybean dregs and their structural and functional properties were investigated using electron microscope,laser particle size analyzer,ultraviolet spectrophotometer,rotary viscomete and in vitro antioxidant Kits.The results showed that the volume average particle size of SDF4 was the smallest of(39.311 ± 0.657)μm.The surface structures of SDF2 and SDF4 particles were much looser with almost no edges.SDF4 exhibited the lowest viscosity,stronger cation-exchange and cholesterol-adsorption capacity.The FRAP,the inhibition capacity on ABTS+ · and ·OH radical of SDF4 was the largest of(0.169 ±0.004)mmol/g,(0.806 ± ± 0.028) mmol/g and (305.739 ± 4.965) U/mL,respectively.Extrusion-enzymatic hydrolysis had a significant impact on the structure and function of soluble dietary fiber(p ＜0.05).

Compared with the other DFs, water extraction SDF (W-SDF) and acid extraction SDF (AC-SDF) exhibited a more complete structure, higher thermal stability, and larger apparent viscosity, which in turn enhanced their functional properties. Notably, the oil holding capacity of W-SDF and AC-SDF reached very high levels at 19.89 ± 1.47 g/g and 12.62 ± 1.31 g/g, respectively. In vivo experiments in mice showed that W-SDF and AC-SDF effectively inhibited soybean oil absorption, promoted its release from the body and decreased serum triglyceride levels. In summary, W-SDF and AC-SDF from R. chingii fruits can effectively reduce the absorption of fats.

Abstract:Studies have suggested that food rich in dietary fiber may facilitate body weight loss, lower total and LDL-cholesterol levels, and reduce body fat. This study examined the effects of soy fiber (SF) on body weight, body composition, and blood lipids in overweight and obese participants.Thirty-nine overweight and obese college adults (19-39 years of age) were randomly assigned to consume control biscuits or biscuits supplemented with SF for their breakfast for 12 wk (approximately 100 g/day). There were significant differences in changes on body weight, BMI, and LDL-cholesterol (LDL-C) between the two groups after 12-wk intervention (p < 0.05). The changes of body weight, BMI, waist circumference, diastolic blood pressure, serum levels of total cholesterol, LDL-C, and glucose, body fat, and trunk fat of participants in SF group were observed significantly after 12 wk.SF had favorable effects on body weight, BMI, and fasting LDL-C levels in overweight and obese adults. These effects may be beneficial in antiobesity and the improvement of hyperlipidemia and hypertension (ClinicalTrials.gov registration number-NCT01802840).

Abstract:Black soybean hull was subjected to steam explosion (SE) treatment under different conditions to improve the yield and properties of soluble dietary fiber (SDF) extract. Optimal conditions for SE treatment were found to be moisture content of 15%, pressure level of 1.0 MPa, and treatment time of 80 s. Under these conditions, the yield of SDF increased from 10.20% to 17.49%. In addition, structural and functional properties of SDF from untreated and SE-treated black soybean hull were investigated. Soluble dietary fiber extracted from SE-treated hull exhibited lower molecular weight and improved functional properties, such as cholesterol-binding capacity, when compared to SDF extracted from untreated soybean hull. In addition, SDF extracted from SE-treated black soybean hull showed a rough surface structure, while a smooth surface structure was found for SDF extracted from the untreated hull. The obtained results indicate that SE treatment can be successfully used to enhance the yield and the property of yolk cholesterol absorption of SDF adsorption of yolk cholesterol functional properties of SDF from black soybean hull.

Effect of Different Extraction Methods on Physicochemical and Functional Properties of Soybean Dietary Fiber

Abstract:The residue from biological dissociation（BD） of soybeans can be considered as a rich source of dietary fibers due to its richness in dietary fibers. In order to investigate the effect of BD on the quality of soybean dietary fiber and hence to obtain high-quality soybean dietary fiber, the chemical composition, physicochemical and functional properties of the resulting soybean dietary fiber were determined and compared with those obtained from water extraction, chemical treatment, fermentation and water extraction following extrusion pretreatment. The results showed that the purity of dietary fiber prepared by BD was 82.58%, and approximately 60% of the total dietary fiber was soluble, suggesting that high-quality dietary fiber was obtained. The water-holding capacity, oil-holding capacity, expansibility and solubility of dietary fiber from BD were 6.87 g/g, 5.48 g/g, 8.22 m L/g and 5.07%, respectively, which were significantly higher than those of dietary fibers from other extraction methods. The results of functional properties showed that the functional properties of dietary fibers from different extraction methods were in the decreasing order of BD, extrusion, fermentation, chemical extraction, and water extraction. In addition, the adsorption capacity of BD dietary fiber toward Pb2+, As3+, and Cu2+ were 351.2, 304.1 and 214.1 μmol/g at p H 7.0, respectively. The glucose adsorption capacity, α-amylase-inhibitory activity and bile acid retardation index were 6.56–35.78 mmol/g, 18.42%, and 33.12%–35.52%, respectively, which were significantly higher than dietary fibers from other extraction methods. Therefore, the residue from biological dissociation of soybeans has good physicochemical and functional properties, indicating that it has great potential for application in the food industry as a new source of dietary fiber.

Abstract:In this study, the functional properties of the soybean hull soluble fractions and insoluble fiber in stabilizing oil-in-water emulsions were investigated by changing the soluble fraction (SF) content in the soybean hull suspensions. High-pressure homogenization (HPH) caused the release of soluble materials (Polysaccharides and proteins) and the deagglomeration of insoluble fibers (IF) from soybean hulls. The apparent viscosity of the soybean hull fiber suspension increased as the SF content of the suspension increased; The absolute value of ζ- potential increased from 18 to 28 mV. In addition, the IF individually stabilized emulsion had the largest emulsion particle size (32.10 μm), but decreased as the SF content in the suspension increased to 10.53 μm. The microstructure of the emulsions showed that surface-active SF adsorbed at the oil-water interface formed an interfacial film, and microfibrils in IF formed a three-dimensional network in the aqueous phase, which synergistically stabilized the oil-in-water emulsion. The findings of this study are important for understanding emulsion systems stabilized by agricultural by-products.

Abstract:Different methods can be used to change the fiber compositions of food, and they consequently affect the physicochemical properties and physiological activities. The present study compared the effects of a blanching treatment on the physicochemical properties of water-insoluble fiber enriched fraction (WIFF) from three varieties of vegetable soybean pod hulls (tea vegetable soybean pod hull, TVSPH; black vegetable soybean pod hull, BVSPH; 305 vegetable soybean pod hulls, 305VSPH) and evaluated their effects on intestinal health in hamsters. Blanching may increase the soluble dietary fiber (SDF) content of WIFF in the 305VSPH variety by solubilizing cell wall components and releasing water-soluble sugars. Thus, the WIFF in the 305VSPH variety after blanching may be composed of cellulose and pectic substances. The WIFF of the blanched 305VSPH (B-305VSPH) variety exhibited the highest physicochemical properties, such as a water-retention capacity (11.7 g/g), oil-holding capacity (9.34 g/g), swelling property (10.8 mL/g), solubility (12.2%), and cation-exchange capacity (221 meq/kg), of the three varieties examined. The supplementation of B-305VSPH WIFF in the diet resulted in significantly (p < 0.05) lower cecal and fecal ammonia; activities of fecal β-d-glucosidase, β-d-glucuronidase, mucinase, and urease; as well as higher cecal total short-chain fatty acids relative to other diets. In addition, microbial analysis suggested that fecal bifidobacteria growth was enhanced by the consumption of B-305VSPH WIFF. Therefore, B-305VSPH WIFF may be applicable as a potential functional ingredient in the food industry for the improvement of intestinal health.

Abstract:Soybeans and their food products exist in the market in various forms, ranging from crude oils and bean meals to nutritious products (e.g. soy milk powers). With the availability of technologies for mass production of soy products and for enrichment of soy components (e.g. phospholipids, saponins, isoflavones, oligosaccharides and edible fiber), the nutritional values of soy products have been enhanced remarkably, offering the potential for functional food development. Among different bioactive components in soybeans, one important component is isoflavones, which have been widely exploited for health implications. While there are studies supporting the health benefits of isoflavones, concerns on adverse effects have been raised in the literature. The objective of this article is to review the recent understanding of the biological activities, adverse effects, and use of isoflavones in functional food development.

Abstract:Adsorption capacity of dietary fiber complexes for fat,cholesterol and bile acid in vitro.An in vitro study was carried out to test adsorption capacity of wheat bran,fruit fiber,soybean fiber and the product,which is made,by their combinations and fructose for fat,cholesterol and bile acid.The results show that wheat bran has highest adsorption capacity for lard and peanut oil;the maximum absorption is 5.64g/g and 2.68g/g.And fruit fiber for cholesterol and bile acid,and the dietary fiber complexes consistently ranked second for adsorbing lard,peanut oil,cholesterol and bile acid.The absorptions are 5.37g/g,2.35g/g,9.23g/g and 45.37mg/mL.It can be concluded that the complexes in combination of water-soluble and water-insoluble dietary fibers would play synergic function of all types of dietary fibers.
Insoluble dietary fiber from soy hulls regulates the gut microbiota in vitro and increases the abundance of bifidobacteriales and lactobacillales

Abstract:We evaluated soy hull dietary fibers (SHDF) extracted from different raw materials, in terms of their chemical composition, physicochemical properties, structure, and ability to regulate fecal microflora, in order to investigate the properties and functions of SHDF. The structures of insoluble dietary fiber from soy hull with oxalic acid extraction (IDFO) and insoluble dietary fiber from soy hull with citric acid extraction (IDFC) were characterized by scanning electron microscopy, Fourier transform infrared spectroscopy, and X-ray diffraction. Compared with IDFO, IDFC had larger crystalline regions, and a higher water retention capacity (4.92 g/g), water swelling capacity (4.77 mL/g), oil adsorption capacity (1.60%), α-amylase activity inhibition ratio (12.72%), glucose adsorption capacity (1.59-13.42%), and bile acid retardation index (5.18-26.61%). Given that the gut microbiota plays a pivotal role in health homeostasis, we performed a detailed investigation of the effects of dietary fiber on fecal microbiota through 16S rDNA high-throughput sequencing. As revealed by Venn, principal component analysis, and 3D-principal co-ordinates analysis analysis, the structure of the fecal microbiota community was markedly altered by intake of IDFO and IDFC. In particular, the abundance of Bifidobacteriales and Lactobacillales significantly increased to varying degrees as a result of IDFO and IDFC intake. Altogether, this study demonstrates a prebiotic effect of SHDF on the fecal microbiota in vitro and provides a basis for the development of SHDF as a novel gut microbiota modulator for health promotion.
In Vitro Binding Capacities and Physicochemical Properties of Soluble Fiber Prepared by Microfluidization Pretreatment and Cellulase Hydrolysis of Peach Pomace

Abstract:The effects of microfluidization coupled with cellulase hydrolysis (MC), solely cellulase hydrolysis (C) and solely water extraction (W) on the in vitro binding capacities of soluble peach fibers (SPFs) to sodium cholate and cholesterol were investigated. The yield and efficiency of soluble fiber production were significantly improved by the MC treatment. Pectic polysaccharides and beta-glucan appeared to be the major components of the three SPFs as indicated by the saccharide composition analysis and FTIR. As opposed to C-SPF and W-SPF, MC-SPF exhibited a greater amount of pectic polysaccharides with a highly branched structure. Moreover, MC-SPF and C-SPF were characterized by a lower molecular weight profile and more significant shear thinning properties compared to W-SPF. The binding capacities of MC-SPF and C-SPF to sodium cholate and cholesterol were much stronger than that of W-SPF, which could be probably attributed to their more sponge-like porous network. Abstract:Soluble dietary fiber can dissolve in water and swelling,and it can be degraded in the large intestine of fiber. The physiological functions of soluble dietary fiber including:excreting toxins from the body,reducing blood fat,lowering blood glucose and so on. It may play an important role in preventing diabetes,cardiovascular disease and cancer. Especially,reducing blood lipid by soluble dietary fiber has made some breakthroughs. Epidemiological studies have shown that soluble dietary fiber such as oat beta glucan has a cholesterol-lowering effect. Animal experiments found that dietary fiber composites have obvious lipid-lowering effect in the hypercholesterolemia mice. Intervention study in population found that soluble dietary fiber and soy protein can reduce cholesterol and triglyceride in serum etc. In recent years,some studies found that soluble dietary fiber can regulate lipid and cholesterol-related gene expressions. This paper summarizes the recent progress of soluble dietary fiber in the field of anti-hypolipidemia and the possible molecular mechanisms are involved in.
