Abstract:The CCCH type zinc finger proteins are a super family involved in many aspects of plant growth and development. In this study, we investigated the response of one CCCH type zinc finger protein AtZFP1 (At2g25900) to salt stress in Arabidopsis. The expression of AtZFP1 was upregulated by salt stress. Compared to transgenic strains, the germination rate, emerging rate of cotyledons and root length of wild plants were significantly lower under NaCl treatments, while the inhibitory effect was significantly severe in T-DNA insertion mutant strains. At germination stage, it was mainly osmotic stress when treated with NaCl. Relative to wild plants, overexpression strains maintained a higher K+, K+/Na+, chlorophyll and proline content, and lower Na+ and MDA content. Quantitative real-time PCR analysis revealed that the expression of stress related marker genes KIN1, RD29B and RD22 increased more significantly in transgenic strains by salt stress. Overexpression of AtZFP1 also enhanced oxidative and osmotic stress tolerance which was determined by measuring the expression of a set of antioxidant genes, osmotic stress genes and ion transport protein genes such as SOS1, AtP5CS1 and AtGSTU5. Overall, our results suggest that overexpression of AtZFP1 enhanced salt tolerance by maintaining ionic balance and limiting oxidative and osmotic stress.

Abstract:Salinity causes widespread crop loss and prompts plants to adapt through changes in gene expression. In this study, we aimed to investigate the function of the non-tandem CCCH zinc-finger (non-TZF) protein gene AtC3H3 in response to salt stress in Arabidopsis. AtC3H3, a gene from the non-TZF gene family known for its RNA-binding and RNase activities, was up-regulated under osmotic stress, such as high salt and drought. When overexpressed in Arabidopsis, AtC3H3 improved tolerance to salt stress, but not drought stress. The expression of well-known abscisic acid (ABA)-dependent salt stress-responsive genes, namely Responsive to Desiccation 29B (RD29B), RD22, and Responsive to ABA 18 (RAB18), and representative ABA-independent salt stress-responsive genes, namely Dehydration-Responsive Element Binding protein 2A (DREB2A) and DREB2B, was significantly higher in AtC3H3-overexpressing transgenic plants (AtC3H3 OXs) than in wild-type plants (WT) under NaCl treatment, indicating its significance in both ABA-dependent and -independent signal transduction pathways. mRNA-sequencing (mRNA-Seq) analysis using NaCl-treated WT and AtC3H3 OXs revealed no potential target mRNAs for the RNase function of AtC3H3, suggesting that the potential targets of AtC3H3 might be noncoding RNAs and not mRNAs. Through this study, we conclusively demonstrated that AtC3H3 plays a crucial role in salt stress tolerance by influencing the expression of salt stress-responsive genes. These findings offer new insights into plant stress response mechanisms and suggest potential strategies for improving crop resilience to salinity stress.

Abstract:In present study,the expression of AtHAK5 gene were found to be induced by some abiotic stress, such as low potassium,low calcium,NaCl and ABA by GUS activity based histochemical staining of the Arabidopsis thaliana transgenic plants containing a promoter AtHAK5::GUS fusion gene. These stress could up-regulate the ex-pression of AtHAK5 gene of Arabidopsis thaliana confirmed by Real time RT-PCR. The differences of growth pheno-type between wild-type Arabidopsis thaliana(Col-0)and the mutant(athak5)in the various abiotic stress treatments proved that AtHAK5 participated in the response to salt and ABA in the plant root.

Abstract:The family of formins, evolutionarily conserved multidomain proteins engaged in the control of actin and microtubule cytoskeleton organization, exhibits considerable diversity in plants. Angiosperms have two formin clades consisting of multiple paralogs, Class I and Class II, the former being often transmembrane proteins located at the plasmalemma or endomembranes. According to available transcriptome data, the Arabidopsis thaliana Class I transmembrane formin AtFH5 (At5g54650) exhibits a distinct pattern of transcript abundance in various seedling root tissues with massive increase of transcript level upon salinity stress. To examine a possible role of AtFH5 in NaCl stress response, we generated transgenic plants expressing green fluorescent protein (GFP)-tagged AtFH5 under its native promoter and characterized its tissue and intracellular localization under standard culture conditions and under NaCl stress. While we confirmed the induction of AtFH5 expression by salt treatment, the distribution of tagged protein, with maxima in the border-like cells of the root cap, in the phloem and at lateral root emergence sites, did not reflect previously reported transcript abundance, suggesting posttranscriptional regulation of gene expression. Subcellular localization studies employing also membrane trafficking inhibitors suggested that AtFH5 protein level may be modulated by endocytosis and autophagy. Notably, loss-of-function atfh5 mutants exhibited increased sensitivity to NaCl stress, indicating that AtFH5 contributes to the development of seedling salt tolerance. These findings highlight the functional importance of AtFH5 in abiotic stress responses.

Abstract:The NAC (NAM, ATAF1/2, CUC2) family of transcription factors (TFs) is a vital transcription factor family of plants. It controls multiple parts of plant development, tissue formation, and abiotic stress response. We cloned the FvNAC29 gene from Fragaria vesca (a diploid strawberry) for this research. There is a conserved NAM structural domain in the FvNAC29 protein. The highest homology between FvNAC29 and PaNAC1 was found by phylogenetic tree analysis. Subcellular localization revealed that FvNAC29 is localized onto the nucleus. Compared to other tissues, the expression level of FvNAC29 was higher in young leaves and roots. In addition, Arabidopsis plants overexpressing FvNAC29 had higher cold and high-salinity tolerance than the wild type (WT) and unloaded line with empty vector (UL). The proline and chlorophyll contents of transgenic Arabidopsis plants, along with the activities of the antioxidant enzymes like catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD) under 200 mM NaCl treatment or −8 °C treatment, were higher than those activities of the control. Meanwhile, malondialdehyde (MDA) and the reactive oxygen species (ROS) content were higher in the WT and UL lines. FvNAC29 improves transgenic plant resistance to cold and salt stress by regulating the expression levels of AtRD29a, AtCCA1, AtP5CS1, and AtSnRK2.4. It also improves the potential to tolerate cold stress by positively regulating the expression levels of AtCBF1, AtCBF4, AtCOR15a, and AtCOR47. These findings suggest that FvNAC29 may be related to the processes and the molecular mechanisms of F. vesca response to high-salinity stress and LT stress, providing a comprehensive understanding of the NAC TFs.

Abstract:Agropyron elongatum, a species in grass family, has a strong tolerance to salt stress. To study the molecular mechanism of Agropyron elongatum in salt tolerance, we isolated a homolog of Na+/H+ antiporters from the root tissues of Agropyron plants. Sequence analysis revealed that this gene encodes a putative vacuolar Na+/H+ antiporter and was designated as AeNHX1. The AeNHX1–GFP fusion protein was clearly targeted to the vacuolar membrane in a transient transfection assay. Northern analysis indicated that AeNHX1 was expressed in a root-specific manner. Expression of AeNHX1 in yeast Na+/H+ antiporter mutants showed function complementation. Further, overexpression of AeNHX1 promoted salt tolerance of Arabidopsis plants, and improved osmotic adjustment and photosynthesis which might be responsible for normal development of transgenic plants under salt stress. Similarly, AeNHX1 also functioned in transgenic Festuca plants. The results suggest that this gene might function in the roots of Agropyron plants, and its expression is involved in the improvement of salt tolerance.

Abstract:In plant cells, anion channels and transporters are essential for key functions.Members of the chloride channel (CLC) family located in intracellular organelles are required for anion accumulation, pH adjustment, and salt tolerance.Here, we cloned a maize (Zea mays L.) CLC gene, named ZmCLC-d, and found that its transcription was up-regulated under cold, drought, salt, and heat stresses, and after hydrogen peroxide (H 2 O 2 ) and abscisic acid (ABA) treatments.The overexpression of ZmCLC-d in Arabidopsis conferred tolerance to cold, drought, and salt stresses; this tolerance was primarily displayed by an increased germination rate, root length, plant survival rate, antioxidant enzyme (catalase, peroxidase, and superoxide dismutase) activities, and a reduced accumulation of Cl -in transgenic plants as compared with wild type (WT) plants.The accumulation of H 2 O 2 and superoxide anion in leaves of the ZmCLC-d-overexpressing plants is much less than that of the WT plants.The expressions of some stress related genes, such as CBF1, CBF2, CBF3, DREB2A, and RCI2A, increased to a greater extent in the ZmCLC-d-overexpressing plants than in the WT.Our results strongly suggest that ZmCLC-d played an important role in stress tolerance.

Abstract:The Arabidopsis thaliana AtNHX1 gene encodes a vacuolar Na+/H+ antiporter that is important in salt tolerance. We report here the tissue distribution and regulation of AtNHX1 expression by salt stress and abscisic acid (ABA). The steady-state level of AtNHX1 transcript was up-regulated by treatment with NaCl, KCl or ABA. AtNHX1 promoter-GUS analysis in transgenic Arabidopsis showed that AtNHX1 was expressed in all tissues except the root tip. Strong GUS expression was detected in guard cells, suggesting that AtNHX1 may play a role in pH regulation and/or K+ homeostasis in the specialized cells. AtNHX1 promoter activity was substantially up-regulated by NaCl, KCl or ABA, demonstrating that salt and ABA regulation of AtNHX1 expression occurs at the transcriptional level. Strong induction of GUS activity in root hair cells was observed, which suggests a role of AtNHX1 in storing Na+ in the enlarged vacuoles in root hair cells. The up-regulation of AtNHX1 transcript levels by NaCl was reduced in abi1-1, aba2-1 and aba3-1, but not in abi2-1, sos1, sos2 or sos3 mutants. ABA-induced AtNHX1 expression was also decreased in abi1-1 but not in abi2-1. These results suggest that salt stress up-regulates AtNHX1 expression transcriptionally and the up-regulation is partially dependent on ABA biosynthesis and ABA signaling through ABI1.

Abstract:Soil salinity seriously affects plants distribution and yield, while salt stress induces SOS genes, and voltage-dependent anion channels (VDAC) and a mitochondrial porin, are induced too. In this paper, phenotypes of AtVDAC2 transgenic lines and wild type (RLD) were analyzed. It was found that AtVDAC2 over-expressing transgenic plants were more sensitive to NaCl, and produced more H2O2 in the NaCl treatment. Also, to find the inner reason, the salt overly sensitive gene 3 (SOS3) expression level was changed with the expression of AtVDAC2. So, it was conjectured that the signal of salt stress response was first sent to AtVDAC2, then AtVDAC2 expression improved, leading to the down-stream signals changes, such as accumulation of H2O2 and improved expression of SOS3. So, it was found that in the over-expression of transgenic lines with AtVDAC2 up-regulation, SOS3 expression increased significantly, and in the inhibited-expressing lines, it was vice versa. In summary, AtVDAC2 was involved in salt stress signaling pathway, and it regulated SOS3 gene expression.
Salt-Specific Gene Expression Reveals Elevated Auxin Levels in Arabidopsis thaliana Plants Grown Under Saline Conditions

Abstract:Soil salinization is increasing globally, driving a reduction in crop yields that threatens food security. Salinity stress reduces plant growth by exerting two stresses on plants: rapid shoot ion-independent effects which are largely osmotic and delayed ionic effects that are specific to salinity stress. In this study we set out to delineate the osmotic from the ionic effects of salinity stress. Arabidopsis thaliana plants were germinated and grown for two weeks in media supplemented with 50, 75, 100, or 125 mM NaCl (that imposes both an ionic and osmotic stress) or iso-osmolar concentrations (100, 150, 200, or 250 mM) of sorbitol, that imposes only an osmotic stress. A subsequent transcriptional analysis was performed to identify sets of genes that are differentially expressed in plants grown in (1) NaCl or (2) sorbitol compared to controls. A comparison of the gene sets identified genes that are differentially expressed under both challenge conditions (osmotic genes) and genes that are only differentially expressed in plants grown on NaCl (ionic genes, hereafter referred to as salt-specific genes). A pathway analysis of the osmotic and salt-specific gene lists revealed that distinct biological processes are modulated during growth under the two conditions. The list of salt-specific genes was enriched in the gene ontology (GO) term “response to auxin.” Quantification of the predominant auxin, indole-3-acetic acid (IAA) and IAA biosynthetic intermediates revealed that IAA levels are elevated in a salt-specific manner through increased IAA biosynthesis. Furthermore, the expression of NITRILASE 2 ( NIT2 ), which hydrolyses indole-3-acetonitile (IAN) into IAA, increased in a salt-specific manner. Overexpression of NIT2 resulted in increased IAA levels, improved Na:K ratios and enhanced survival and growth of Arabidopsis under saline conditions. Overall, our data suggest that auxin is involved in maintaining growth during the ionic stress imposed by saline conditions.

Abstract:Genes for V-H(+)-ATPase subunits were identified and cloned from the salt-tolerant wheat mutant RH8706-49. Sequences of these genes are highly conserved in plants. Overexpression of these genes in Arabidopsis thaliana improved its salt tolerance, and increased the activities of V-H(+)-ATPase and Na(+)/H(+) exchange, with the largest increase in plants carrying the c subunit of V-H(+)-ATPase. Results from quantitative RT-PCR analysis indicated that the mRNA level of each V-H(+)-ATPase subunit in the Arabidopsis increased under salt stress. Overall, our results suggest that each V-H(+)-ATPase subunit plays a key role in enhancing salt tolerance in plants.
Genome-wide analysis of ZAT gene family revealed GhZAT6 regulates salt stress tolerance in G. hirsutum

Abstract:High salt environments can induce stress in different plants. The genes containing the ZAT domain constitute a family that belongs to a branch of the C2H2 family, which plays a vital role in responding to abiotic stresses. In this study, we identified 169 ZAT genes from seven plant species, including 44 ZAT genes from G. hirsutum. Phylogenetic tree analysis divided ZAT genes in six groups with conserved gene structure, protein motifs. Two C2H2 domains and an EAR domain and even chromosomal distribution on At and Dt sub-genome chromosomes of G. hirsutum was observed. GhZAT6 was primarily expressed in the root tissue and responded to NaCl and ABA treatments. Subcellular localization found that GhZAT6 was located in the nucleus and demonstrated transactivation activity during a transactivation activity assay. Arabidopsis transgenic lines overexpressing the GhZAT6 gene showed salt tolerance and grew more vigorously than WT on MS medium supplemented with 100 mmol NaCl. Additionally, the silencing of the GhZAT6 gene in cotton plants showed more obvious leaf wilting than the control plants, which were subjected to 400 mmol NaCl treatment. Next, the expressions of GhAPX1, GhFSD1, GhFSD2, and GhSOS3 were significantly lower in the GhZAT6-silenced plants treated with NaCl than the control. Based on these findings, GhZAT6 may be involved in the ABA pathway and mediate salt stress tolerance by regulating ROS-related gene expression.

Abstract:Salt stress causes reduced plant growth and alters the plant development process, resulting in a threat to global crop production. The exploring of unique genes conferring to salt tolerance from plants that inhabit extreme environments remains urgent. Haloxylon ammodendron is a desert xero-halophyte shrub with a strong tolerance to drought and salt stresses. We previously reported that the drought tolerance of Arabidopsis thaliana was improved by the overexpression of HaASR1 from H. ammodendron. In this work, the effects of HaASR1 overexpression on the salt tolerance of Arabidopsis were investigated. HaASR1 overexpression significantly enhanced the growth of Arabidopsis lines under salinity and plant tissue water content through enhancing the osmotic adjustment ability, maintaining the membrane integrity, improving the chlorophyll content and leaf area, and thereby enhancing photosynthesis capacity. Taken together, the overexpression of HaASR1 from H. ammodendron improved the salt tolerance of the transgenic lines of Arabidopsis. These results indicated that HaASR1 from H. ammodendron has potential application values in increasing the salt tolerance of grass and crop plants by genetic engineering.

Abstract:Haloxylon ammodendron is a typical desert xerophyte with worthy genetic resources, therefore, it is necessary to explore the resistance genes of H. ammodendron, and to improve the stress tolerance of other plant species through genetic engineering. In this study, a heat shock transcription factor gene, HaHSFA1, was firstly cloned from H. ammodendron and overexpressed in Arabidopsis thaliana. The sensitivity of transgenic plants to exogenous ABA, osmotic stress and salt stress were analyzed; the effects of overexpression of HaHSFA1 in Arabidopsis on salt tolerance were deeply investigated. The results indicated that overexpression of HaHSFA1 increased chlorophyll content in Arabidopsis, and the growth of plants was promoted. The leaf soluble sugar and proline contents in OE lines were significantly increased compared with those in WT. OE lines maintained lower osmotic potential and higher shoot water content. Overexpression of HaHSFA1 in Arabidopsis also enhanced antioxidant activity, which may be important for protecting plasma membrane integrity in cells. Overexpression of HaHSFA1 in Arabidopsis improved root activity and increased selective absorption and transport capacity for K+ over Na+, which was of great significance to reduce the toxic effect of Na+. This study indicated that overexpression of HaHSFA1 from H. ammodendron significantly elevates the salt tolerance of Arabidopsis, and HaHSFA1 has potential application values in improving abiotic stress tolerance in crops by genetic engineering.

Abstract:High-salinity stress represses plant growth by inhibiting various metabolic processes. In contrast to the well-studied mechanisms mediating tolerance to high levels of salt, the effects of low levels of salts have not been well studied. In this study, we examined the growth of Arabidopsis thaliana plants under different NaCl concentrations. Interestingly, both shoot and root biomass increased in the presence of 5 mM NaCl, whereas more than 10 mM NaCl decreased plant biomass. To clarify the biological mechanism by which a low level of NaCl stimulated plant growth, we analyzed element accumulation in plants grown under different NaCl concentrations. In addition to the Na and Cl contents, C, S, Zn, and Cu contents were increased under 5 mM NaCl in shoots; this was not observed at higher NaCl concentrations. Adverse effects of high salinity, such as decreased levels of nitrate, phosphate, sulfate, and some cations, did not occur in the presence of 5 mM NaCl. An increase in C was possibly attributed to increased photosynthesis supported by Cl, Zn, and Cu, which also increased in shoots after NaCl application. Salt stress-responsive gene expression was enhanced under 20 mM NaCl but not at lower doses. Among the S metabolites analyzed, cysteine (Cys) was increased by 5 mM NaCl, suggesting that S assimilation was promoted by this dose of NaCl. These results indicate the usefulness of NaCl for plant growth stimulation.

Abstract:Salt stress is one of the most serious abiotic stresses limiting crop growth and yield.Exploring the salt stress response gene can provide a new direction for solving salt damage.In order to reveal the Arabidopsis thaliana genes expression under salt stress,we explore the digital gene expression profiles (DGEP) of Arabidopsis thaliana (Columbia-0) leaves treated with water (control) or 200 mmol/L NaC1 for 2 h.By comparison of gene expression of the treatment and control,4 400 genes are identified to be differentially expressed,among which 1 513 genes are up-regulated,2 887 genes are down-regulated.Gene ontology (GO) reveals that these genes are involved in 22 GO terms such as structural constituent of ribosome,membrane,response to stimulus,response to stress,and proline metabolism.Thirty-two pathways are enriched by Kyoto encyclopedia of genes and genomes (KEGG),including basic metabolism,secondary metabolism and oxidation-reduction processes.Meanwhile,6 genes encoding late embryogenesis abundant (LEA) proteins are identified to express differently,which indicates that these LEA genes might be important in stress response process.
SLAH1 is Involved in the Long-Distance Transport of Cl− from Roots into Shoots in the Cl−-tolerant Xerophyte Pugionium Cornutum under Salt Stress

Abstract:Different from most crops and forages, the xerophyte Pugionium cornutum can absorb high quantities of Cl− and then efficiently transport them into shoots to enhance osmotic adjustment ability under salt stress. However, the molecular mechanism underlying long-distance Cl− transport remains unclear. SLAH1 is crucial for root-to-shoot Cl− transport in Arabidopsis only under non-saline conditions. This study aimed to evaluate the function of PcSLAH1 from P. cornutum in long-distance Cl− transport under salt stress. PcSLAH1 was cloned, its tissue and subcellular locations and expression patterns in response to salt treatments were analyzed. The effect of PcSLAH1 on ion accumulation and the expression of key genes involved in Na+ and Cl− transport were investigated by ectopically overexpressing in Arabidopsis driven by a root stelar-specific promoter. PcSLAH1 was specifically expressed at root stelar cells and located on the plasma membrane. Opposing with AtSLAH1 in Arabidopsis, the relative expression level of PcSLAH1 in roots was significantly induced by high Cl− treatments. The root stelar-specific expression of PcSLAH1 in Arabidopsis resulted in a significant increase in shoot Cl− content under NaCl or KCl treatment. PcSLAH1 could coordinate with CLCg and NHX1 to regulate shoot Cl− and Na+ homeostasis under salt treatment. In addition, the root stelar-specific expression of PcSLAH1 conferred a stronger long-distance transport ability of Cl− than AtSLAH1. PcSLAH1 functions in facilitating the long-distance transport of Cl− and modulating shoot Na+ and Cl− homeostasis under salt stress. The up-regulated expression of PcSLAH1 is conducive to salt tolerance of P. cornutum.
Comprehensive transcriptional profiling of NaCl-stressed Arabidopsis roots reveals novel classes of responsive genes

Abstract:BackgroundRoots are an attractive system for genomic and post-genomic studies of NaCl responses, due to their primary importance to agriculture, and because of their relative structural and biochemical simplicity. Excellent genomic resources have been established for the study of Arabidopsis roots, however, a comprehensive microarray analysis of the root transcriptome following NaCl exposure is required to further understand plant responses to abiotic stress and facilitate future, systems-based analyses of the underlying regulatory networks.ResultsWe used microarrays of 70-mer oligonucleotide probes representing 23,686 Arabidopsis genes to identify root transcripts that changed in relative abundance following 6 h, 24 h, or 48 h of hydroponic exposure to 150 mM NaCl. Enrichment analysis identified groups of structurally or functionally related genes whose members were statistically over-represented among up- or down-regulated transcripts. Our results are consistent with generally observed stress response themes, and highlight potentially important roles for underappreciated gene families, including: several groups of transporters (e.g. MATE, LeOPT1-like); signalling molecules (e.g. PERK kinases, MLO-like receptors), carbohydrate active enzymes (e.g. XTH18), transcription factors (e.g. members of ZIM, WRKY, NAC), and other proteins (e.g. 4CL-like, COMT-like, LOB-Class 1). We verified the NaCl-inducible expression of selected transcription factors and other genes by qRT-PCR.ConclusionMicorarray profiling of NaCl-treated Arabidopsis roots revealed dynamic changes in transcript abundance for at least 20% of the genome, including hundreds of transcription factors, kinases/phosphatases, hormone-related genes, and effectors of homeostasis, all of which highlight the complexity of this stress response. Our identification of these transcriptional responses, and groups of evolutionarily related genes with either similar or divergent transcriptional responses to stress, will facilitate mapping of regulatory networks and extend our ability to improve salt tolerance in plants.

Expression Analysis of LeNHX1 Gene in Mycorrhizal Tomato under Salt Stress.

Abstract:The plant growth, stem sap flow, Na+ and Cl− content, and the expression of vacuolar Na+/H+ antiporter gene (LeNHX1) in the leaves and roots of tomato under different NaCl stresses (0.5% and 1%) were studied to analyze the effect of arbuscular mycorrhizal fungi (AMF) on Na+ and Cl− accumulation and ion exchange. The results showed that arbuscular mycorrhizal (AM) plant growth and stem sap flow increased and salt tolerance improved, whereas Na+ and Cl− accumulated. Na+ significantly decreased, and no significant decline was detected in Cl− content after AMF inoculation compared with the non-AM plants. The LeNHX1 gene expression was induced in the AM and non-AM plants by NaCl stress. However, AMF did not improve the LeNHX1 level, and low expression was observed in the AM tomato. Hence, the mechanism that reduced the Na+ damage to tomato induced by AMF has little relation to LeNHX1, which can export Na+ from the cytosol to the vacuole across the tonoplast.
