Abstract:Biological ion channels are key molecules for cellular regulation and communication. To mimic the structure and functions of nature ion channels, a new class of light-regulated transmembrane ion channels was reported based on tri(macrocycle) hydraphile and azobenzene photoswitch (hydraphile 1). The liposome-based proton transport assays showed that hydraphile I exhibited excellent transmembrane activity (10, and Y-max arrived 0.7 at 401 mu mo1/L. The successful isomerization of azobenzene moiety was confirmed and qualified by UV and NMR spectra. Upon alternative irradiation of 365 nm UV light and 450 nm visible light, the transmembrane activity of hydraphile I was regulated between 0.35 and 0.5, reversibly. All the obtained results have demonstrated the promise of developing excellent synthetic ion channels with ion gating properties based on simple molecular design. 2015 Chinese Chemical Society and Institute of Materia Medica, Chinese Academy of Medical Sciences. Abstract:The controllable ion transport in the photoreceptors of rod cells is essentially important for the light detection and information transduction in visual systems. Herein, inspired by the photochromism-regulated ion transport in rod cells with stacking structure, layered ion channels have been developed with a visual photochromic function induced by the alternate irradiation with visible and UV light. The layered structure is formed by stacking spiropyran-modified montmorillonite 2D nanosheets on the surface of an alumina nanoporous membrane. The visual photochromism resulting from the photoisomerization of spiropyran chromophores reversibly regulates the ion transport through layered ion channels. Furthermore, the cooperation of photochromism and pH value achieves multiple switchable states of layered ion channels for the controllable ion transport mimicking the biological process of the visual cycle. The ion transport properties of these states are explained quantitatively by a theoretical calculation based on the Poisson and Nernst-Plank (PNP) equations.

Abstract:It goes both ways: A thiol-reactive cross-linker based on a bridged azobenzene derivative permits photoreversible control of peptide conformation on irradiation with violet (407 nm) and green (500-550 nm) light (see picture) through isomerization of the cross-linker. The large separation of the absorbance bands of the cis (yellow) and trans (red) isomers enables complete bidirectional photoswitching.

Abstract:Inspired by the critical role of ion channel proteins in the regulation of cellular activities, here we developed a new type of synthetic ion channel by simple benzocrown ether-based derivatives M1 and M2, where M1 had a dodecyl tail and M2 had a diethylene glycol-conjugated dodecyl tail. Being amphiphilic in nature, the two small molecules were assumed to form crown ether channels through supramolecular interactions in bilayer lipid membranes (BLMs). The efficient ion transport was investigated by both a fluorescence-based vesicle assay and a planar bilayer conductance measurement, and M2 with diethylene glycol substitution exhibited more efficient activity comparable to amphotericin B. Moreover, the presence of a photosensitive o-nitrobenzyl group provided the light-regulation to deactivate ion transport by destroying the channel assembly of the molecules in BLMs, which provides new opportunities for developing intelligent light-regulated systems for biomedical applications based on synthetic small molecules.

Abstract:A versatile design for fabricating guard cell‐inspired photoswitchable ion channels is presented by infiltrating azobenzene‐cross‐linked polymer (AAZO‐PDAC) into nanoporous anodic aluminum oxide (AAO) membranes. By adding ionic liquids, the ion conductivity of the light‐driven ion nanochannels can be controlled with good repeatability and fast responses (within seconds) in multiple cycles. Stimuli‐responsive ion nanochannels have attracted considerable attention in various fields because of their remote controllability of ionic transportation. For photoresponsive ion nanochannels, however, achieving precise regulation of ion conductivity is still challenging, primarily due to the difficulty of programmable structural changes in confined environments. Moreover, the relationship between noncontact photo‐stimulation in nanoscale and light‐induced ion conductivity has not been well understood. In this work, a versatile design for fabricating guard cell‐inspired photoswitchable ion channels is presented by infiltrating azobenzene‐cross‐linked polymer (AAZO‐PDAC) into nanoporous anodic aluminum oxide (AAO) membranes. The azobenzene‐cross‐linked polymer is formed by azobenzene chromophore (AAZO)‐cross‐linked poly(diallyldimethylammonium chloride) (PDAC) with electrostatic interactions. Under UV irradiation, the trans‐AAZO isomerizes to the cis‐AAZO, causing the volume compression of the polymer network, whereas, in darkness, the cis‐AAZO reverts to the trans‐AAZO, leading to the recovery of the structure. Consequently, the resultant nanopore sizes can be manipulated by the photomechanical effect of the AAZO‐PDAC polymers. By adding ionic liquids, the ion conductivity of the light‐driven ion nanochannels can be controlled with good repeatability and fast responses (within seconds) in multiple cycles. The ion channels have promising potential in the applications of biomimetic materials, sensors, and biomedical sciences.

Acylhydrazone-based reversibly photoswitchable ion pair transporter with OFF-ON cotransport activity

Abstract:The cellular membrane transport of physiologically important cations and anions is omnipresent and regulates different physiological functions. Whereas a notable number of cation-anion transporters are being developed to transport salts across the membrane, developing an artificial cation-anion symporter with stimulus-responsive activities is an immense obstacle. Herein, for the first time, we report reversibly photoswitchable acylhydrazone-based transporter 2 that has distinctive OFF-ON cation-anion co-transport abilities. The substituent was modified in 1a-1c and 2, to change the to-and-fro movement of the transporter to enhance the ion transport efficiency. Ion transport experiments across the lipid bilayer membrane demonstrate that 1a has the highest transport activity among the series with irreversible photoisomerization properties, whereas 2 has a unique reversible photoisomerization property. A detailed transport study indicated that the E-conformer of compound 2 facilitates Na+/Cl- transport via the symport process by following the carrier mode of ion transport. 23Na NMR and chloride selective electrode assays confirmed the OFF and ON state of ion transport of compound 2 with photoirradiation. An assembly of [(2 E )2 + NaCl] was subjected to geometry optimization to understand the responsible ion binding motif. Geometry optimization followed by the natural bond orbital analysis of 1a Z and 2 Z demonstrated that 1a Z forms comparatively stronger intramolecular H-bonding than 2 Z , making it accessible for reversible photoisomerization.

Abstract:The spatiotemporal regulation of light-gated ion channels is a powerful tool to study physiological pathways and develop personalized theranostic modalities. So far, most existing light-gated channels are limited by their action spectra in the ultraviolet (UV) or visible region. Simple and innovative strategies for the specific attachment of photoswitches on the cell surface without modifying or genetically encoding channel structures, and more importantly, that enable the remote activation of ion-channel functions within near-infrared (NIR) spectral window in living systems, remain a challenging concern. Herein, metabolic glycan biosynthesis is used to achieve site-specific covalent attachment of near-infrared-light-mediated lanthanide-doped upconversion nanocrystals (UCNs) to the cell surface through copper-free click cyclization. Upon irradiation with 808 nm light, the converted emission at 480 nm could activate a light-gated ion channel, channelrhodopsins-2 (ChR2), and thus remotely control the cation influx. This unique strategy provides valuable insights on the specific regulation membrane-associated activities in vivo.

Abstract:Supramolecular synthetic transporters are crucial to understand and activate the passage across lipid membranes of hydrophilic effector molecules. Herein, we introduce photoswitchable calixarenes for the light-controlled transport activation of cationic peptide cargos across model lipid bilayers and inside living cells. Our approach was based on rationally designed p-sulfonatocalix[4]arene receptors equipped with a hydrophobic azobenzene arm, which recognize cationic peptide sequences at the nM range. Activation of membrane peptide transport is confirmed, in synthetic vesicles and living cells, for calixarene activators featuring the azobenzene arm in the E configuration. Therefore, this method allows the modulation of the transmembrane transport of peptide cargos upon Z-E photoisomerization of functionalized calixarenes using 500 nm visible light. These results showcase the potential of photoswitchable counterion activators for the light-triggered delivery of hydrophilic biomolecules and pave the way for potential applications in remotely controlled membrane transport and photopharmacology applications of hydrophilic functional biomolecules.

Abstract:Three new artificial transmembrane channel molecules have been designed and synthesized by attaching positively charged Arg-incorporated tripeptide chains to pillar[5]arene. Fluorescent and patch-clamp experiments revealed that voltage can drive the molecules to insert into and leave from a lipid bilayer and thus switch on and off the transport of K(+) ions. One of the molecules was found to display antimicrobial activity toward Bacillus subtilis with half maximal inhibitory concentration (IC50 ) of 10 μM which is comparable to that of natural channel-forming peptide alamethicin.

Abstract:Stimuli-responsive synthetic ionophores allow for spatial and temporal control over ion transport, with promise for applications in targeted therapy. Relay transporters have emerged as a new class of ion transporters - these are anchored carriers that sit in both leaflets of the bilayer and mediate transport across the membrane by passing ions between them. The relays are themselves membrane impermeable, and so must be incorporated into the membrane during vesicle preparation. Here we show that relay transporters can be delivered to both sides of the membrane of vesicles using a synthetic flippase. By incorporating an aryl azopyrazole photo-switch into the movable arm of the relay transporters the ion transport activity can be very efficiently and reversibly switched between off and on states. This control is achieved by extension and contraction of their movable arms via photo-isomerization of the central aryl azopyrazole moiety, hence modulating the ability of the relays to pass ions across the membrane.

Abstract:Natural redox-regulated channel proteins often utilize disulfide bonds as redox sensors for adaptive regulation of channel conformations in response to diverse physiological environments. In this study, we developed novel synthetic ion channels capable of reversibly switching their ion-transport capabilities by incorporating multiple disulfide bonds into artificial systems. X-ray structural analysis and electrophysiological experiments demonstrated that these disulfide-bridged molecules possess well-defined tubular cavities and can be efficiently inserted into lipid bilayers to form artificial ion channels. More importantly, the disulfide bonds in these molecules serve as redox-tunable switches to regulate the formation and disruption of ion-permeation pathways, thereby achieving a transition in the transmembrane transport process between the ON and OFF states.

Abstract:Inspired by the light-gated ion channels in cell membranes that play important roles in many biological activities, herein, we developed an artificial light-gated ion channel membrane out of conjugated microporous polymers. Through bottom-up design of the monomer molecular structure and by the electropolymerization method, the membrane pore size and thickness were precisely controlled on the molecular level. The obtained membrane exhibited uniform pore size and highly sensitive light-switchable response. The photoisomerization of the polymer chain resulted in a reversible "on and off" light control over the pore size and subsequently led to light-gated ion transport across the membrane for a series of ions including hydrogen, potassium, sodium, lithium, calcium, magnesium, and aluminum ions.
