Abstract:Congenital amusia is a lifelong neurodevelopmental disorder of fine-grained pitch processing. In this fMRI study, we examined the neural bases of congenial amusia in speakers of a tonal language – Cantonese. Previous studies on non-tonal language speakers suggest that the neural deficits of congenital amusia lie in the music-selective neural circuitry in the right inferior frontal gyrus (IFG). However, it is unclear whether this finding can generalize to congenital amusics in tonal languages. Tonal language experience has been reported to shape the neural processing of pitch, which raises the question of how tonal language experience affects the neural bases of congenital amusia. To investigate this question, we examined the neural circuitries sub-serving the processing of relative pitch interval in pitch-matched Cantonese level tone and musical stimuli in 11 Cantonese-speaking amusics and 11 musically intact controls. Cantonese-speaking amusics exhibited abnormal brain activities in a widely distributed neural network during the processing of lexical tone and musical stimuli. Whereas the controls exhibited significant activation in the right superior temporal gyrus (STG) in the lexical tone condition and in the cerebellum regardless of the lexical tone and music conditions, no activation was found in the amusics in those regions, which likely reflects a dysfunctional neural mechanism of relative pitch processing in the amusics. Furthermore, the amusics showed abnormally strong activation of the right middle frontal gyrus and precuneus when the pitch stimuli were repeated, which presumably reflect deficits of attending to repeated pitch stimuli or encoding them into working memory. No significant group difference was found in the right IFG in either the whole-brain analysis or region-of-interest analysis. These findings imply that the neural deficits in tonal language speakers might differ from those in non-tonal language speakers, and overlap partly with the neural circuitries of lexical tone processing (e.g. right STG).

Abstract:Congenital amusia is a neuro-developmental disorder of music perception and production, with the observed deficits contrasting with the sophisticated music processing reported for the general population. Musical deficits within amusia have been hypothesized to arise from altered pitch processing, with impairments in pitch discrimination and, notably, short-term memory. We here review research investigating its behavioral and neural correlates, in particular the impairments at encoding, retention, and recollection of pitch information, as well as how these impairments extend to the processing of pitch cues in speech and emotion. The impairments have been related to altered brain responses in a distributed fronto-temporal network, which can be observed also at rest. Neuroimaging studies revealed changes in connectivity patterns within this network and beyond, shedding light on the brain dynamics underlying auditory cognition. Interestingly, some studies revealed spared implicit pitch processing in congenital amusia, showing the power of implicit cognition in the music domain. Building on these findings, together with audiovisual integration and other beneficial mechanisms, we outline perspectives for training and rehabilitation and the future directions of this research domain.

Abstract:Congenital amusia is a neurodevelopmental disorder characterized by deficits of music perception and production, which are related to altered pitch processing. The present study used a wide variety of tasks to test potential patterns of processing impairment in individuals with congenital amusia (N = 18) in comparison to matched controls (N = 19), notably classical pitch processing tests (i.e., pitch change detection, pitch direction of change identification, and pitch short-term memory tasks) together with tasks assessing other aspects of pitch-related auditory cognition, such as emotion recognition in speech, sound segregation in tone sequences, and speech-in-noise perception. Additional behavioral measures were also collected, including text reading/copying tests, visual control tasks, and a subjective assessment of hearing abilities. As expected, amusics' performance was impaired for the three pitch-specific tasks compared to controls. This deficit of pitch perception had a self-perceived impact on amusics' quality of hearing. Moreover, participants with amusia were impaired in emotion recognition in vowels compared to controls, but no group difference was observed for emotion recognition in sentences, replicating previous data. Despite pitch processing deficits, participants with amusia did not differ from controls in sound segregation and speech-in-noise perception. Text reading and visual control tests did not reveal any impairments in participants with amusia compared to controls. However, the copying test revealed more numerous eye-movements and a smaller memory span. These results allow us to refine the pattern of pitch processing and memory deficits in congenital amusia, thus contributing further to understand pitch-related auditory cognition. Together with previous reports suggesting a comorbidity between congenital amusia and dyslexia, the findings call for further investigation of language-related abilities in this disorder even in the absence of neurodevelopmental language disorder diagnosis.

Abstract:To investigate the differences in event-related potential parameters related to children's intelligence, 18 intellectually gifted children and 18 average children participated in this study. The electroencephalograms were recorded the auditory sensory memory that elicited the mismatch negativity (MMN) and late discriminative negativity (LDN), as well as involuntary attention switch that elicited the P3a and early MMN were analyzed. The results indicated that children with high intelligence had comparatively larger MMN, LDN, early MMN, P3a amplitudes, and earlier peak latency in LDN than average children. The enhanced neural function of the intellectually gifted children might be due to more spatially and temporally coordinated neural network, faster neural processing speed and more efficient neural activation functions.

Abstract:Interhemispheric connectivity of the two cerebral hemispheres is crucial for a broad repertoire of cognitive functions including music and language. Congenital amusia has been reported as a neurodevelopment disorder characterized by impaired music perception and production. However, little is known about the characteristics of the interhemispheric functional connectivity (FC) in amusia. In the present study, we used a newly developed voxel-mirrored homotopic connectivity (VMHC) method to investigate the interhemispheric FC of the whole brain in amusia at resting-state. Thirty amusics and 29 matched participants underwent a resting-state functional magnetic resonance imaging (fMRI) scanning. An automated VMHC approach was used to analyze the fMRI data. Compared to the control group, amusics showed increased VMHC within the posterior part of the default mode network (DMN) mainly in the posterior superior temporal gyrus (pSTG) and posterior cingulate cortex (PCC). Correlation analyses revealed negative correlations between the VMHC value in pSTG/PCC and the music perception ability among amusics. Further ROC analyses showed that the VMHC value of pSTG/PCC showed a good sensibility/specificity to differentiate the amusics from the controls. These findings provide a new perspective for understanding the neural basis of congenital amusia and imply the immature state of DMN may be a credible neural marker of amusia.

Congenital amusia and academic performance among Colombian university students

Abstract:Background: Congenital amusia is a rare neurogenetic and neuropsychological condition which hinders the ability to recognize variations in all aspects of a musical piece. Although previous studies have determined the prevalence of congenital amusia in the general population, few have studied its presence among university students. Findings regarding the association between this condition and academic performance are equivocal, although evidence suggests that musical training improves scholastic achievement. Methods: We conducted a cross-sectional study on a sample of 383 university students, all pursuing health-related degrees, comparing their class rank with their performance on the BRAMS Online Test for amusia. Results: We found a prevalence of 0.52% for pitch-based amusia. When applying the Off-Scale test failure criterion for the definition of amusia in our sample, we found a prevalence of 4.4%. Logistic models showed an increase in risk of poor academic performance (lowest quartile) in subjects who failed the off-scale test (Odds Ratio: 7.14 95% CI 2.59-19.6) and who met any of the described definitions of amusia (Odds Ratio: 4.89 95% CI 2.24-10.7). Conclusions: Both musical training and self-report of musical ability significantly affected test results. Although musical education shows some effect over academic performance, further studies are required to determine if this is due to differential effects in subjects with and without amusia.
Reduced Sensitivity to Between-Category Information but Preserved Categorical Perception of Lexical Tones in Tone Language Speakers With Congenital Amusia

Abstract:Previous studies have shown that for congenital amusics, long-term tone language experience cannot compensate for lexical tone processing difficulties. However, it is still unknown whether such difficulties are merely caused by domain-transferred insensitivity in lower-level acoustic processing and/or by higher-level phonological processing of linguistic pitch as well. The current P300 study links and extends previous studies by uncovering the neurophysiological mechanisms underpinning lexical tone perception difficulties in Mandarin-speaking amusics. Both the behavioral index (<i>d'</i>) and P300 amplitude showed reduced within-category as well as between-category sensitivity among the Mandarin-speaking amusics regardless of the linguistic status of the signal. The results suggest that acoustic pitch processing difficulties in amusics are manifested profoundly and further persist into the higher-level phonological processing that involves the neural processing of different lexical tone categories. Our findings indicate that long-term tone language experience may not compensate for the reduced acoustic pitch processing in tone language speakers with amusia but rather may extend to the neural processing of the phonological information of lexical tones during the attentive stage. However, from both the behavioral and neural evidence, the peakedness scores of the <i>d'</i> and P300 amplitude were comparable between amusics and controls. It seems that the basic categorical perception (CP) pattern of native lexical tones is preserved in Mandarin-speaking amusics, indicating that they may have normal or near normal long-term categorical memory.

Distributional learning of musical pitch despite tone deafness in individuals with congenital amusia

Abstract:Congenital amusia is an innate and lifelong deficit of music processing. This study investigated whether adult listeners with amusia were still able to learn pitch-related musical chords based on stimulus frequency of statistical distribution, i.e., via distributional learning. Following a pretest-training-posttest design, 18 amusics and 19 typical, musically intact listeners were assigned to bimodal and unimodal conditions that differed in distribution of the stimuli. Participants' task was to discriminate chord minimal pairs, which were transposed to a novel microtonal scale. Accuracy rates for each test session were collected and compared between the two groups using generalized mixed-effects models. Results showed that amusics were less accurate than typical listeners at all comparisons, thus corroborating previous findings. Importantly, amusics-like typical listeners-demonstrated perceptual gains from pretest to posttest in the bimodal condition (but not the unimodal condition). The findings reveal that amusics' distributional learning of music remains largely preserved despite their deficient music processing. Implications of the results for statistical learning and intervention programs to mitigate amusia are discussed.

Abstract:Background: Congenital amusia (CA) is a rare disorder characterized by deficits in pitch perception, and many structural and functional magnetic resonance imaging studies have been conducted to better understand its neural bases. However, a structural magnetic resonance imaging analysis using a surface-based morphology method to identify regions with cortical features abnormalities at the vertex-based level has not yet been performed. Methods: Fifteen participants with CA and 13 healthy controls underwent structural magnetic resonance imaging. A surface-based morphology method was used to identify anatomical abnormalities. Then, the surface parameters' mean value of the identified clusters with statistically significant between-group differences were extracted and compared. Finally, Pearson's correlation analysis was used to assess the correlation between the Montreal Battery of Evaluation of Amusia (MBEA) scores and surface parameters. Results: The CA group had significantly lower MBEA scores than the healthy controls ( p = 0.000). The CA group exhibited a significant higher fractal dimension in the right caudal middle frontal gyrus and a lower sulcal depth in the right pars triangularis gyrus ( p < 0.05; false discovery rate-corrected at the cluster level) compared to healthy controls. There were negative correlations between the mean fractal dimension values in the right caudal middle frontal gyrus and MBEA score, including the mean MBEA score ( r = −0.5398, p = 0.0030), scale score ( r = −0.5712, p = 0.0015), contour score ( r = −0.4662, p = 0.0124), interval score ( r = −0.4564, p = 0.0146), rhythmic score ( r = −0.5133, p = 0.0052), meter score ( r = −0.3937, p = 0.0382), and memory score ( r = −0.3879, p = 0.0414). There was a significant positive correlation between the mean sulcal depth in the right pars triangularis gyrus and the MBEA score, including the mean score ( r = 0.5130, p = 0.0052), scale score ( r = 0.5328, p = 0.0035), interval score ( r = 0.4059, p = 0.0321), rhythmic score ( r = 0.5733, p = 0.0014), meter score ( r = 0.5061, p = 0.0060), and memory score ( r = 0.4001, p = 0.0349). Conclusion: Individuals with CA exhibit cortical morphological changes in the right hemisphere. These findings may indicate that the neural basis of speech perception and memory impairments in individuals with CA is associated with abnormalities in the right pars triangularis gyrus and middle frontal gyrus, and that these cortical abnormalities may be a neural marker of CA.

Abstract:Purpose: Congenital amusia is a neurogenetic disorder of musical pitch processing. Its linguistic consequences have been examined separately for speech intonations and lexical tones. However, in a tonal language such as Chinese, the processing of intonations and lexical tones interacts with each other during online speech perception. Whether and how the musical pitch disorder might affect linguistic pitch processing during online speech perception remains unknown. Method: We investigated this question with intonation (question vs. statement) and lexical tone (rising Tone 2 vs. falling Tone 4) identification tasks using the same set of sentences, comparing behavioral and event-related potential measurements between Mandarin-speaking amusics and matched controls. We specifically focused on the amusics without behavioral lexical tone deficits (the majority, i.e., pure amusics). Results: Results showed that, despite relative to normal performance when tested in word lexical tone test, pure amusics demonstrated inferior recognition than controls during sentence tone and intonation identification. Compared to controls, pure amusics had larger N400 amplitudes in question stimuli during tone task and smaller P600 amplitudes in intonation task. Conclusion: These data indicate that musical pitch disorder affects both tone and intonation processing during sentence processing even for pure amusics, whose lexical tone processing was intact when tested with words.

In this study, we investigated the FC characteristics of amusia in Mandarin speakers in resting‐state functional magnetic resonance imaging data by voxel‐wise connectivity analyses with seeds in left and right Heschl's gyri (HG) and region of interest (ROI)‐to‐ROI connectivity analyses. Our findings indicate increased connectivity between right HG and bilateral posterior superior temporal gyrus, as determined by voxel‐wise connectivity analyses in amusia. Conversely, reduced connectivity was observed between bilateral HG and bilateral IFG (orbital part) as assessed through ROI‐to‐ROI connectivity analyses in amusia when compared to controls. Moreover, the music perception scores of amusia in Mandarin speakers were associated with diminished connectivity between the left HG and the right IFG. This study furnishes direct evidence for the link between music perception deficits and the aberrant frontotemporal connectivity of congenital amusia in tonal language speakers in resting state.

Abstract:Purpose: Individuals with congenital amusia exhibit degraded speech perception. This study examined whether adult Chinese Mandarin listeners with amusia were still able to extract the statistical regularities of Mandarin speech sounds, despite their degraded speech perception. Method: Using the gating paradigm with monosyllabic syllable-tone words, we tested 19 Mandarin-speaking amusics and 19 musically intact controls. Listeners heard increasingly longer fragments of the acoustic signal across eight duration-blocked gates. The stimuli varied in syllable token frequency and syllable-tone co-occurrence probability. The correct syllable-tone word, correct syllable-only, correct tone-only, and correct syllable-incorrect tone responses were compared respectively between the two groups using mixed-effects models. Results: Amusics were less accurate than controls in terms of the correct word, correct syllable-only, and correct tone-only responses. Amusics, however, showed consistent patterns of top-down processing, as indicated by more accurate responses to high-frequency syllables, high-probability tones, and tone errors all in manners similar to those of the control listeners. Conclusions: Amusics are able to learn syllable and tone statistical regularities from the language input. This extends previous work by showing that amusics can track phonological segment and pitch cues despite their degraded speech perception. The observed speech deficits in amusics are therefore not due to an abnormal statistical learning mechanism. These results support rehabilitation programs aimed at improving amusics' sensitivity to pitch.
Impaired socio-emotional processing in a developmental music disorder

Abstract:Some individuals show a congenital deficit for music processing despite normal peripheral auditory processing, cognitive functioning, and music exposure. This condition, termed congenital amusia, is typically approached regarding its profile of musical and pitch difficulties. Here, we examine whether amusia also affects socio-emotional processing, probing auditory and visual domains. Thirteen adults with amusia and 11 controls completed two experiments. In Experiment 1, participants judged emotions in emotional speech prosody, nonverbal vocalizations (e.g., crying), and (silent) facial expressions. Target emotions were: amusement, anger, disgust, fear, pleasure, relief, and sadness. Compared to controls, amusics were impaired for all stimulus types, and the magnitude of their impairment was similar for auditory and visual emotions. In Experiment 2, participants listened to spontaneous and posed laughs, and either inferred the authenticity of the speaker’s state, or judged how much laughs were contagious. Amusics showed decreased sensitivity to laughter authenticity, but normal contagion responses. Across the experiments, mixed-effects models revealed that the acoustic features of vocal signals predicted socio-emotional evaluations in both groups, but the profile of predictive acoustic features was different in amusia. These findings suggest that a developmental music disorder can affect socio-emotional cognition in subtle ways, an impairment not restricted to auditory information.

Abstract:Music is a universal human attribute. The study of amusia, a neurologic music processing deficit, has increasingly elaborated our view on the neural organization of the musical brain. However, lesions causing amusia occur in multiple brain locations and often also cause aphasia, leaving the distinct neural circuits for amusia unclear. Here, we utilized lesion network mapping to identify these circuits. A systematic literature search was carried out to identify all published case reports of lesion-induced amusia. The reproducibility and specificity of the identified amusia network were then tested in an independent prospective cohort of 97 stroke patients (46 female and 51 male) with repeated structural brain imaging, specifically assessed for both music perception and language abilities. Lesion locations in the case reports were heterogenous, but connected to common brain regions, including bilateral temporoparietal and insular cortices, precentral gyrus, and cingulum. In the prospective cohort, lesions causing amusia mapped to a common brain network, centering on the right superior temporal cortex and clearly distinct from the network causally associated with aphasia. Lesion-induced longitudinal structural effects in the amusia circuit were confirmed as reduction of both grey and white matter volume, which correlated with the severity of amusia. We demonstrate that despite the heterogeneity of lesion locations disrupting music processing, there is a common brain network, that is distinct from the language network. These results provide evidence for the distinct neural substrate of music processing, differentiating music-related functions from language, providing a testable target for non-invasive brain stimulation to treat amusia. Significance Statement Perspectives on the neural organization of language and music have been elaborated by studying the respective brain lesion induced deficits, aphasia and amusia, but without leading to an agreement regarding their shared and distinct neural circuitries. This study demonstrates that lesions disrupting music processing map to a common brain network, which is distinct from speech processing networks in the brain. Importantly, our data confirms, the lesion-induced longitudinal structural effects in the central nodes of the amusia network, resulting in both grey and white matter atrophy. The results lend insight into the neural substrate of music processing, add neural evidence for the differentiability of music-related functions from language, and provide first testable targets for non-invasive brain stimulation to treat amusia.

Abstract:Music is inherent in human life and is a significant topic of cognitive neuroscience. Previous studies focused on amusia suggested that two frontotemporal circuits engage in music processing. Structure-function coupling is an important feature of human brain, which is associated with cognition and allows for a more sensitive investigation of brain-behavior association. However, we still have limited knowledge about the relation between structure-function coupling, music processing and other regional neural profiles. We recruited 106 participants (43 subjects were diagnosed with congenital amusia) and measured their music perception by Montreal Battery of Evaluation of Amusia (MBEA). Then we utilized support vector regression algorithm and mediation analysis, and employed amplitude of low frequency fluctuation (ALFF), structural/functional degree centrality (DC) and structure-function coupling to explore their relation with global averaged MBEA score. We found structure-function coupling of widespread brain regions in both hemispheres, rather than ALFF or DC, contributed to predict MBEA score. Left middle frontal gyrus, bilateral inferior temporal gyrus and right insula were most predictive regions, and these regions were involved in memory and cognitive control according to meta-analysis. Further, coupling of left middle frontal gyrus, a region that is homologous to and is connected with typical music circuits, fully mediated the negative relation between ALFF and MBEA score. Our findings provide further understanding for the neural basis of music, and have implications for neural plasticity, neuromodulation therapy and cognitive causes of amusia.

Using newly developed technology and method, especially functional magnetic resonance imaging (fMRI), people have accumulated some knowledge about the musical ability in normal human, the speciality of the musician, the emotional effect of music. Further understanding the neural fundament of musical phenomenon could propel the cognition of the human brain in a lot of aspects.

Abstract:While the issue of individual variation has been widely studied in second language learning or processing, it is less well understood how perceptual and musical aptitude differences can explain individual variation in native speech processing. In the current study, we make use of tone merger in Hong Kong Cantonese, an ongoing sound change that concerns the merging of tones in perception, production or both in a portion of native speakers, to examine the possible relationship between tone merger and musical and pitch abilities. Although a previous study has reported the occurrence of tone merger independently of musical training, it has not been investigated before whether tone-merging individuals, especially those merging tones in perception, would have inferior musical perception and fine-grained pitch sensitivities, given the close relationship of speech and music. To this end, we tested three groups of tone-merging individuals with various tone perception and production profiles on musical perception and pitch threshold tasks, in comparison to a group of Cantonese speakers with congenital amusia, and another group of controls without tone merger or amusia. Additionally, the amusics were compared with tone-merging individuals on the details of their tone discrimination and production profiles. The results showed a clear dissociation of tone merger and amusia, with the tone-merging individuals exhibiting intact musical and pitch abilities; on the other hand, the amusics demonstrated widespread difficulties in tone discrimination yet intact tone production, in contrast to the highly selective confusion of a specific tone pair in production or discrimination in tone-merging individuals. These findings provide the first evidence that tone merger and amusia are distinct from each other, and further suggest that the cause of tone merger may lie elsewhere rather than being driven by musical or pitch deficits. We also discussed issues arising from the current findings regarding the neural mechanisms of tone merger and amusia.
Neural Correlates of Consonance, Dissonance, and the Hierarchy of Musical Pitch in the Human Brainstem

Abstract:Consonant and dissonant pitch relationships in music provide the foundation of melody and harmony, the building blocks of Western tonal music. We hypothesized that phase-locked neural activity within the brainstem may preserve information relevant to these important perceptual attributes of music. To this end, we measured brainstem frequency-following responses (FFRs) from nonmusicians in response to the dichotic presentation of nine musical intervals that varied in their degree of consonance and dissonance. Neural pitch salience was computed for each response using temporally based autocorrelation and harmonic pitch sieve analyses. Brainstem responses to consonant intervals were more robust and yielded stronger pitch salience than those to dissonant intervals. In addition, the ordering of neural pitch salience across musical intervals followed the hierarchical arrangement of pitch stipulated by Western music theory. Finally, pitch salience derived from neural data showed high correspondence with behavioral consonance judgments (r = 0.81). These results suggest that brainstem neural mechanisms mediating pitch processing show preferential encoding of consonant musical relationships and, furthermore, preserve the hierarchical pitch relationships found in music, even for individuals without formal musical training. We infer that the basic pitch relationships governing music may be rooted in low-level sensory processing and that an encoding scheme that favors consonant pitch relationships may be one reason why such intervals are preferred behaviorally.

Abstract:Congenital amusia is a neurogenetic disorder, affecting music pitch processing. It also transfers to the language domain and negatively influences the perception of linguistic components relying on pitch, such as lexical tones. It has been well established that unfavorable listening conditions impact lexical tone perception in amusics. For instance, both Mandarin- and Cantonese-speaking amusics were impaired in tone processing under simultaneously noisy conditions. Backward noise is one of the adverse listening conditions, but its interference mechanism is distinct from the simultaneous noise. Therefore, it warrants more studies to explore whether and how backward noise affects tone processing in amusics. In the current study, eighteen Mandarin-speaking amusics and 18 controls were tested on discrimination of Mandarin tones under two conditions: a quiet condition involving relatively low-level processing and a backward masking condition involving high-level processing (e.g., tone categorization) where a native multi-talker babble noise was added to target tones. The results revealed that amusics performed similarly to controls in quiet conditions, whereas their performance was found poorer in backward masking conditions. These findings shed light on how adverse listening environments influence amusics' lexical tone processing and provided further empirical evidence that amusics may be impaired in the high-level phonological processing of lexical tone.

Abstract:Music, as language, is a universal and specific trait to humans; it is a complex ability with characteristics that are unique compared to other cognitive abilities. Nevertheless, several issues are still open to debate, such as, for example, whether music is a faculty that is independent from the rest of the cognitive system, and whether musical skills are mediated by a single mechanism or by a combination of processes that are independent from one another. Moreover, the anatomical correlations of music have yet to be clarified. The goal of this review is to illustrate the current condition of the neuropsychology of music and to describe different approaches to the study of the musical functions. Hereby, we will describe the neuropsychological findings, suggesting that music is a special function carried out by different and dedicated processes that are probably subserved by different anatomical regions of the brain. Moreover, we will review the evidence obtained by working with brain-damaged patients suffering from music agnosia, a selective impairment in music recognition.
