Mutagenicity Test of ABS and AS and Its Inhibitory Effect on Mitomycin C Induced SCE Frequency in Chinese Hamster Cells

Abstract:Alkylbenzene sulfonate (ABS) and alkyl sulfonate (AS) are the major active components of synthetic detergent. The potential mutagenicity of ABS and AS were tested with Salmonella/microsome test, there was no significant or consistant difference between ABS (5~30μg/ml) or AS (0.01~0.5μg/ml) and controls. There was also no difference whether the metabolic activation system of Sg-mix was added or not.Treatment or ABS (1~10μg/ml) of AS (0.001~0.05μg/ml) alone caused no significant increase of SCE frequency in Chinese hamster Wg3-h cells as compared to the controls. Treatment of Wg3-h cells with various doses of Mitomycin C (MMC) from 0.005 to 0.05μg/ml, all caused significant dose-dependent increase of SCE frequencies. Such increase was significantly inhibited by concurrent addition of a fixed dose of ABS (5 μg/ml)or AS (0.01μg/ml). Futhermore, Wg3-h cells exposed to MMC at a fixed dose of 0.025 μg/ml plus various lower ABS doses from 0.01~0.005μg/ml or exposed to ABS at a fixed dose of 0. 005μg/ml plus various MMC doses from 0.05~0.005μg/ml all showed a dose-dependent inhibitory effect on MMC-induced SCE. These results indicate that at certain conditions, ABS and/or AS have the ability to suppress the SCE frequencies induced by MMC, which suggests that ABS and/or AS might have some effect on the protection of environmental harmful factors to human.
