Abstract:Dendrimer is one type of macromolecules with well-defined, highly-branched and nano-scaled architectures, composed of three distinct domains: core, branches and terminal groups. Its potential functions and applications are explored based on the unique physical and chemical features due to special molecular architecture. Using polyhedral oligomeric silsesquioxanes (POSS) as the core of dendrimers is an effective and facile way to simplify the tedious repetitive steps of preparation and separation. Since POSS allows eight branches to radiate from a silicon-oxygen right cubic core, the dendrimers can be constructed in a well-controlled globular, three-dimensional framework, and large numbers of peripheral groups can be obtained at relatively low generation numbers. As a kind of novel nano-hybrid supermolecules, POSS-based dendrimers have attracted considerable interest in materials science. In this review, we briefly introduce the synthetic approaches of some typical POSS-based dendrimers categorized by the different chemical composition of branching units, and then focus on the potential functions and applications of this nano-hybrid material in the field of catalysis, gene and drug delivery, liquid crystals, light harvesting and energy transfer. The specific and excellent properties imparted by the incorporation of POSS into dendrimers are also presented. Finally, the advances of POSS-based dendrimers are prospected.

Abstract:Dendrimers have successfully proved themselves as useful additives in different routes of drug administration because they can render drugs greater water-solubility, bioavailability, and biocompatibility. This review demonstrated the potential of dendrimers to be applied in these detailed routes with particular reference to intravenous, oral, transdermal, and ocular delivery systems. As a necessary introduction, the structures, synthesis, and properties of dendrimers were presented. Furthermore, the interaction mechanisms between dendrimers and drug molecules, including simple encapsulation, electrostatic interaction, and covalent conjugation, were elaborated. Abstract:Dendrimers are new artificial macromolecules which have the structure like a tree. They are hyperbranched and monodisperse three-dimensional molecules with defined molecular weights, large numbers of functional groups on the surface and well-established host-guest entrapment properties. Recently, dendrimers have successfully proved themselves as promising nanocarriers for drug delivery because they can render drug molecules a greater water-solubility, bioavailability, and biocompatibility. In this review, recent progress in the pharmaceutical applications of dendrimers as delivery systems for drugs, paticularly, the non-steroidal anti-inflammatory, anti-microbial/anti-viral and potent anticancer drugs is discussed. Three possible interaction mechanisms between dendrimers and drug molecules are presented. In addition, the pharmacodynamic and pharmacokinetic properties of both dendrimer/drug complex and dendrimer-drug conjugation after their administration to animals are evaluated.

In the paper the recent advances of dendrimer in gene transfer vehicles and drug delivery are separately reviewed. These advances illustrate the direction of the future development of dendrimers.

Abstract:Dendrimers, as a type of artificial polymers with unique structural features, have been extensively explored for their applications in biomedical fields, especially in drug delivery. However, one important concern about the most commonly used dendrimers exists - the nondegradability, which may cause side effects induced by the accumulation of synthetic polymers in cells or tissues. Therefore, biodegradable dendrimers incorporating biodegradability with merits of dendrimers such as well-defined architectures, copious internal cavities and surface functionalities, are much more promising for developing novel nontoxic drug carriers. Herein, we review the recent advances in design and synthesis of biodegradable dendrimers, as well as their applications in fabricating drug delivery systems, with the aim to provide researchers in the related fields a good understanding of biodegradable dendrimers for drug delivery.

Abstract:Dendrimers are nanosized, monodisperse, highly branched polymers with well-defined topological structure which have attracted much attention for drug delivery recently. To further improve the performance of dendrimers in drug delivery, various functional dendrimers are developed by decorating the dendrimers with targeting agents, imaging agents, or stimuli-sensitive moieties. They show good biocompatibility, visibility, tumor targeting and stimuli-sensitive properties for drug or gene delivery. This chapter will focus on the design of multifunctional nanocarriers based on the dendrimers. Therefore, the chapter will provide the ideas for designing the dendrimers based nanocarriers for controllable drug delivery and let more people know the development of dendrimers for drug delivery in recent years.

Abstract:Dendrimers, as a type of artificially synthesized polymers, have been increasingly attracting attention in many research fields, including the material and medical sciences, due to their unique characteristics that include their highly branched and well-defined molecular architecture, multivalency and tunable chemical compositions. Abstract:Dendrimers are polymers with well-defined 3D branched structures that are vastly utilized in various neurotheranostics and biomedical applications, particularly as nanocarrier vectors. Imaging agents can be loaded into dendrimers to improve the accuracy of diagnostic imaging processes. Likewise, combining pharmaceutical agents and anticancer drugs with dendrimers can enhance their solubility, biocompatibility, and efficiency. Practically, by modifying ligands on the surface of dendrimers, effective therapeutic and diagnostic platforms can be constructed and implemented for targeted delivery. Dendrimer-based nanocarriers also show great potential in gene delivery. Since enzymes can degrade genetic materials during their blood circulation, dendrimers exhibit promising packaging and delivery alternatives, particularly for central nervous system (CNS) treatments. The DNA and RNA encapsulated in dendrimers represented by polyamidoamine that are used for targeted brain delivery, via chemical-structural adjustments and appropriate generation, significantly improve the correlation between transfection efficiency and cytotoxicity. This article reports a comprehensive review of dendrimers' structures, synthesis processes, and biological applications. Recent progress in diagnostic imaging processes and therapeutic applications for cancers and other CNS diseases are presented. Potential challenges and future directions in the development of dendrimers, which provide the theoretical basis for their broader applications in healthcare, are also discussed.

Abstract:Dendritic compounds (so-called dendrimers) with nano-scale represent unique symmetric and spherical structures. They are usually prepared through two fundamental methods based on molecular level which involved the convergent method and divergent method, although the synthetic methods have a tendency towards the diversity and functionalization. The outershell of dendrimers with rich peripheral reactive sites are easily modified by small functional molecules. Moreover, the higher generation dendrimers also possess more exposed functional groups on the surface prior to the previous one and they are much easily customized for much more applications. Consequently, based on previous researches, this review summarized wide applications of dendritic compounds in many fields which were investigated in detail, including gene vector, drug carrier, catalysis, sensor industry, photoelectric material, etc. Dendrimers provide promising tools for the cellular delivery of molecular cargos ranging in size from small molecules and peptides to proteins and DNA. More importantly, it is necessary to explore new synthetic methods and undiscovered applications of new dendrimers.

Abstract:Dendrimers have been recognized as promising materials for biomedical applications due to their unique properties, such as well-defined unimolecular structures, precise molecular weights and high degrees of branching and surface functionality. The dendrimers with a polyhedral oligomeric silsesquioxane core – nanoglobules – have many advantageous features over traditional dendrimers, including more functional groups at the same generation of dendrimers, well-defined 3D structures and relatively compact morphologies. Various nanoglobules of polyhedral oligomeric silsesquioxane-core dendrimers have been synthesized and investigated for biomedical applications. Nanoglobules have been used as carriers for developing drug-delivery systems, gene-delivery systems and imaging contrast agents with precisely defined structures and sizes. This article summarizes the recent developments in nanoglobules for biomedical applications.

Abstract:Dendrimers are highly branched, with abundant functional groups on surfaces to be modified. Dendrimer micelles are ideal nano‐anticancer drug carriers. This article reviews the classification, preparation, structure, surface modification of dendrimer micelles and their applications as various platforms for biomedical applications related to cancer therapy including drug delivery, gene transfection, nano‐contrast for imaging, and combined therapy. Dendrimers are a family of polymers with highly branched structure, well‐defined composition, and extensive functional groups, which have attracted great attention in biomedical applications. Micelles formed by dendrimers are ideal nanocarriers for delivering anticancer agents due to the explicit study of their characteristics of particle size, charge, and biological properties such as toxicity, blood circulation time, biodistribution, and cellular internalization. Here, the classification, preparation, and structure of dendrimer micelles are reviewed, and the specific functional groups modified on the surface of dendrimers for tumor active targeting, stimuli‐responsive drug release, reduced toxicity, and prolonged blood circulation time are discussed. In addition, their applications are summarized as various platforms for biomedical applications related to cancer therapy including drug delivery, gene transfection, nano‐contrast for imaging, and combined therapy. Other applications such as tissue engineering and biosensor are also involved. Finally, the possible challenges and perspectives of dendrimer micelles for their further applications are discussed.

Abstract:Dendrimers are nanoparticles with unique features including globular 3D shape and nanometer size. The availability of numerous terminal functional groups and modifiable surface engineering permit modification of dendrimer surface with several therapeutic agents, diagnostic moieties and targeting substances. The aim. To enlighten the readers regarding design, development, limitations, challenges and future directions regarding anticancer bio-dendrimers. Materials and methods. The data base was represented by such systems as Medline, Cochrane Central Register of Controlled Trials, Scopus, Web of Science Core Collection, PubMed. gov, Google-Academy. A search was carried out for the following keywords and combinations: Polypropylene imine (PPI); Poly-L-lysine (PLL); polyamidoamine (PAMAM); cancer; drug delivery; dendrimers. Results. High encapsulation of drug and effective passive targeting are also among their therapeutic uses. Herein, we have described latest developments in chemotherapeutic delivery of drugs by dendrimers. For the most part, the potential and efficacy of dendrimers are anticipated to have considerable progressive effect on drug targeting and delivery. Conclusion. The newest discoveries have shown that the dendritic nanocarriers have many unique features that endorse more research and development.

Abstract:Since the first dendrimer was reported in 1978 by Fritz Vögtle, dendrimer research has grown exponentially, from synthesis to application in the past four decades. The distinct structure characteristics of dendrimers include nanoscopic size, multi-functionalized surface, high branching, cavernous interior, and so on, making dendrimers themselves ideal drug delivery vehicles. This mini review article provides a brief overview of dendrimer's history and properties and the latest developments of dendrimers as drug delivery systems. This review focuses on the latest progress in the applications of dendrimers as drug and gene carriers, including 1) active drug release strategies to dissociate drug/gene from dendrimer in response to stimuli; 2) size-adaptive and charge reversal dendrimer delivery systems that can better take advantage of the size and surface properties of dendrimer; 3) bulk and micro/nano dendrimer gel delivery systems. The recent advances in dendrimer formulations may lead to the generation of new drug and gene products and enable the development of novel combination therapies.

Abstract:Dendrimers are hyper-branched organic compounds characterized via a three-dimensional structure possessing functional groups on the surface. These terminals groups can be simply modified to enhance the functionality of dendrimers and produce biocompatible and versatile products. They are a promising agent for nanomedicine applications because of their unique properties, including nanoscale size, globular shape, and high reactivity, solubility in water, internal cavities, and comfortable synthesis methods. The use of dendrimers as drug delivery systems have received great attention from researchers. Dendrimers can be applied as carriers for different therapeutic agents. They can reduce the toxicity of drugs and increase their efficacy. This review provides a general outline of the structure and types of dendrimers, the synthesis of dendrimers, and applications in the nanomedicine field with emphasis on drug delivery.

Abstract:Dendrimers have shown great promise as carriers in drug delivery due to their unique structures and superior properties. However, the precise control of payload release from a dendrimer matrix still presents a great challenge. Stimuli-responsive dendrimers that release payloads in response to a specific trigger could offer distinct clinical advantages over those dendrimers that release payloads passively. These smart polymers are designed to specifically release their payloads at targeted regions or at constant release profiles for specific therapies. They represent an attractive alternative to targeted dendrimers and enable dendrimer-based therapeutics to be more effective, more convenient, and much safer. The wide range of stimuli, either endogenous( acid, enzyme, and redox potentials) or exogenous ( light, ultrasound, and temperature change), allows great flexibility in the design of stimuli-responsive dendrimers. In this review article, we will highlight recent advances and opportunities in the development of stimuli-responsive dendrimers for the treatment of various diseases, with emphasis on cancer. Specifically, the applications of stimuli-responsive dendrimers in drug delivery as well as their mechanisms are intensively reviewed.

Abstract:Abstract Dendrimers are nano-sized, radially symmetric molecules with well-defined, homogeneous, and monodisperse structure that has a typically symmetric core, an inner shell, and an outer shell. Their three traditional macromolecular architectural classes are broadly recognized to generate rather polydisperse products of different molecular weights. A variety of dendrimers exist, and each has biological properties such as polyvalency, self-assembling, electrostatic interactions, chemical stability, low cytotoxicity, and solubility. These varied characteristics make dendrimers a good choice in the medical field, and this review covers their diverse applications.

Abstract:Dendrimers are a new class of artificial macromolecules with well-defined hyperbranched structures which enable bio-active molecules such as drugs to be presented in a highly multi-valent fashion. Covalent conjugation of drugs to the surface of dendrimers can be easily achieved either by direct chemical reactions between dendrimers and drug molecules including esterification and amidation or through cleavable linkers, depending on the functional groups on the surface of dendrimers. The pharmacological properties of these dendrimer-based prodrugs such as biocompatibility, biodistribution, biostability, bioadhesion and biopermeability can be modulated by further modifying dendrimers with specific functional molecules to fit a specific medicinal purpose. In this mini-review, recent advances on the use of dendrimers as prodrug nano-scaffolds were briefly demonstrated. The design and synthesis of dendrimer-based prodrugs as well as screening their intrinsic properties in biological systems were fully discussed.

Abstract:Dendrimers, distinguished by their mono-dispersed globular or spheroid macromolecular structures with repeated branches, exhibit unique and desirable characteristics that set them apart from other polymers. These characteristics include their nanoscopic size, precise control over molecular structure, low polydispersity index, water solubility, exceptional physicochemical stability, self-assembly capabilities, polyvalency, chemical stability, electrostatic interactions, solubility, and minimal cytotoxicity. Dendrimers are prepared from monomers using either divergent or convergent step-growth polymerization. Dendrimers have the ability to reduce the toxicity of medications while enhancing their effectiveness. This review will delve into various types of dendrimers such as poly (amidoamine) dendrimers, peptide dendrimers, smart dendrimers, polyethylene glycol dendrimers, poly (propylene imine) dendrimers, polyester dendrimers, Janus dendrimers, and etc. in the field of drug delivery and gene delivery systems that utilize them. Dendrimer-drug conjugates offer numerous benefits over conventional drug carriers, such as enhanced drug uptake by the tumor, reduced systemic toxicity issues, and increased antitumor activity of the administrated medications. To achieve this objective, the authors utilized the Science Direct, PubMed, and Scopus databases to acquire the information, evaluating references individually, extracting essential details, and assessing the quality of the included studies.
