Abstract:Pd-catalyzed reactions of diazo compounds result in unique transformations that are distinct from those of traditional processes catalyzed by Rh-II or Cu-I catalysts. Pd carbene migratory insertion was deemed to be the key step in a series of Pd-catalyzed reactions of diazo compounds. The combination of this unique reaction and classic Pd-catalyzed cross-coupling provides new potential for the development of novel Pd-catalyzed transformations, in particular for C=C double bond formation.

Abstract:Diazo compounds had been widely employed as a class of environment friendly and highly efficient coupling reagents in transition-metal-catalyzed C-H bonds insertion reactions.Recently, C-H alkylation of various arenes and heteroarenes has been achieved enormous successes using diazo compounds as the alkylation reagents under a new catalytic model.The recent progress in diazo compounds mediated transition-metal-catalyzed C-H alkylation and tandem C-H alkylation/cyclization is reviewed, including reaction mechanism and synthetic applications.Furthermore, the prospects of this reaction are also discussed.

Abstract:Copper-catalyzed reaction of diazo compounds generates copper carbene intermediates that undergo diverse transformations. In recent years, the selectivity and efficiency of these important conversions have been further improved. In particular, breakthroughs have been made in catalytic asymmetric polar X-H bond insertion reactions. Moreover, novel transformations based on copper carbene, namely copper-catalyzed cross-couplings of diazo compounds, have emerged as powerful methods for carbon-carbon bond formations. This feature article summarizes the most recent developments in this area.

Abstract:The Pd/C-catalyzed reactions, including reduction reactions and cross-coupling reactions, play an irreplaceable role in modern organic synthesis. Compared to the homogeneous palladium catalyst system, the heterogeneous Pd/C catalyst system offers an alternative protocol that has particular advantages and applications. Herein, a review on Pd/C-catalyzed reactions is presented. Both the advances in Pd/C-catalyzed methodologies and the application of Pd/C-catalysis in total synthesis are covered in this review.

Abstract:Palladium-catalyzed C-H functionalization of acyldiazomethanes with aryl iodides has been developed. This reaction is featured by the retention of the diazo functionality in the transformation, thus constituting a novel method for the introduction of diazo functionality to organic molecules. Consistent with the experimental results, the density functional theory (DFT) calculation indicates that the formation of Pd-carbene species in the catalytic cycle through dinitrogen extrusion from the palladium ethyl diazoacetate (Pd-EDA) complex is less favorable. The reaction instead proceeds through Ag2CO3 assisted deprotonation and subsequently reductive elimination to afford the products with diazo functionality remained. This C-H functionalization transformation can be further combined with the recently evolved palladium-catalyzed cross-coupling reaction of diazo compounds with aryl iodides to develop a tandem coupling process for the synthesis of α,α-diaryl esters. DFT calculation supports the involvement of Pd-carbene as reactive intermediate in the catalytic cycle, which goes through facile carbene migratory insertion with a low energy barrier (3.8 kcal/mol).

The authors have demonstrated that by choosing a suitable Pd catalyst the sensitive diazo function is left intact and that subsequent coupling with alkenyl and aryl iodides is possible under basic conditions. A protocol for carbonylation with CO is also presented.

Abstract:In this Perspective, we describe recent advances on Pd-catalyzed oxidative cascade carbocyclizations. These cascade processes enable efficient construction of the molecular complexity and structural diversity of carbocyclic compounds via introducing diverse functionalities concomitant with multiple C-C bond-formations in one-pot operations. In many cases, these processes are facilitated by Pd-catalysts alone, while cocatalysis by combination of Pd catalyst and other catalysts are also discussed, since they represent a new entry to address the preparation of functionalized cyclic compounds with high efficiency and selectivity.

Abstract:Based on the well development of diazo compound transformations in organic synthesis, it is expected to extend its applications to the field of polymer synthesis. It is of vital importance to transform efficient organic reactions of diazo compounds into new polymerization methodology in order to afford polymers with new structures and functions. Currently, there is still great development potential in the application of diazo compounds in polymer synthesis. This review highlights the recent advancements of polymer synthesis chemistry with diazo compounds, including chain-growth polymerization, step-growth polymerization, end-group functionalization and post-polymerization modification. Transition-metal-catalyzed chain-growth polymerization of diazoacetates constructs C-C main chain from one carbon unit. Through the development of catalytic diazoacetate polymerization, the synthesis of high molecular weight polymers, stereocontrol polymerization and living/controlled polymerization can be realized successively. The stepwise polymerization of diazo compounds originated from their efficient organic conversions. The insertion reaction of diazo compounds into O-H and N-H bonds can be extended to stepwise polymerization to afford polyethers, polyesters and polyamines that are not easily accessible through other synthetic methods, while cross-coupling polymerization of diazo compounds via metal carbene insertion-migration mechanism can realize the synthesis of polymers with novel structures. Diazoacetates and diazomalonates can be employed to regulate the terminal structures of polymers synthesized through metal-catalyzed vinyl addition polymerization or ring-opening metathesis polymerization of olefins, and successively end-capped by metal carbene. The post-polymerization modifications based on diazo compounds can be applied to the introduction of polar functional groups into polyolefin. Compared to the copolymerization of polar olefins, the post-polymerization functionalization strategy avoids the depressed reactivity of varied monomers, and can be applicable to a broad scope of functional groups. The thermal and mechanical properties of polymers can be improved by this method. Finally, the future challenges in expanding the approaches on polymer chemistry of diazo compounds are prospected in this review.

Abstract:Over the past few decades, there have been major developments in transition metal-catalyzed asymmetric cyclization reactions, enabling the convenient access to a wide spectrum of structurally diverse chiral carbo- and hetero-cycles, common skeletons found in fine chemicals, natural products, pharmaceuticals, agrochemicals, and materials. In particular, a plethora of enantioselective cyclization reactions have been promoted by chiral palladium catalysts owing to their outstanding features. This review aims to collect the latest advancements in enantioselective palladium-catalyzed cyclization reactions over the past eleven years, and it is organized into thirteen sections depending on the different types of transformations involved.

Abstract:Palladium‐catalyzed decarboxylative cycloadditions have emerged as highly effective methods for constructing structurally diverse carbo‐ and heterocycles because of the formation of at least two carbon‐carbon or carbon‐heteroatom bonds in a single step. It is of great interest to chemists that this type of cycloaddition reactions possesses some special advantages such as high reactivity, exclusive regioselectivity, and good functional group compatibility. Based on these qualities, palladium‐catalyzed decarboxylative cycloadditions present strong ability in synthetic chemistry and have been flourished especially in the last five years. In this review, the achievements in palladium‐catalyzed decarboxylative cycloadditions involving cyclic carbonates, carbamates, and lactones for accessing oxacyclo‐, azacyclo‐ and carbocyclic compounds are addressed. Mechanistic insights and some synthetic applications toward the synthesis of natural products are discussed. The challenges and opportunities of this field are also outlined. Abstract:Transition-metal-catalyzed carbene transformations and cross-couplings represent two major reaction types in organometallic chemistry and organic synthesis. However, for a long period of time, these two important areas have evolved separately, with essentially no overlap or integration. Thus, an intriguing question has emerged: can cross-coupling and metal carbene transformations be merged into a single reaction cycle? Such a combination could facilitate the development of novel carbon-carbon bond-forming methodologies. Although this concept was first explored about 10 years ago, rapid developments inthis area have been achieved recently. Palladium catalysts can be used to couple diazo compounds with a wide variety of organic halides. Under oxidative coupling conditions, diazo compounds can also react with arylboronic acids and terminal alkynes. Both of these coupling reactions form carbon-carbon double bonds. As the key step in these catalytic processes, Pd carbene migratory insertion plays a vital role in merging the elementary steps of Pd intermediates, leading to novel carbon-carbon bond formations. Because the diazo substrates can be generated in situ from N-tosylhydrazones in the presence of base, the N-tosylhydrazones can be used as reaction partners, making this type of cross-coupling reaction practical in organic synthesis. N-Tosylhydrazones are easily derived from the corresponding aldehydes or ketones. The Pd-catalyzed cross-coupling of N-tosylhydrazones is considered a complementary reaction to the classic Shapiro reaction for converting carbonyl functionalities into carbon-carbon double bonds. It can also serve as an alternative approach for the Pd-catalyzed cross-coupling of carbonyl compounds, which is usually achieved via triflates. The combination of carbene formation and cross-coupling in a single catalytic cycle is not limited to Pd-catalyzed reactions. Recent studies of Cu-, Rh-, Ni-, and Co-catalyzed cross-coupling reactions with diazo compounds or N-tosylhydrazones show that these transformations also work with other transition metals, demonstrating the generality of the diazo compounds as new cross-coupling partners in transition-metal-catalyzed coupling reactions.

Abstract:Efficient palladium-catalyzed cross-coupling reactions of alpha-diazocarbonyl compounds and arylboronic acids or aryl halides have been developed. The reaction proceeds smoothly for a range of diazo compounds, boronic acids, and halides. The coupling reaction conditions tolerate various substituents on the aromatic rings of the substrates, such as chloro, fluoro, acyl, oxo, ester, and nitro groups. This coupling reaction constitutes a novel access to alpha-aryl-substituted alpha,beta-unsaturated carbonyl compounds. Mechanistically, palladium-carbene is supposed to be the key intermediate; its formation is followed by migratory insertion of an aryl group to the carbenic carbon of the palladium-carbene complex and subsequent beta-hydride elimination. Kinetic isotope effect (KIE) data measured for intra- and intermolecular competition experiments suggest that beta-hydride elimination is not involved in the rate-determining step.

Abstract:We report the recent progress in the preparation of functional poly(disubstituted acetylene)s (PDSAs) through post-polymerization modification routes. The metathesis polymerization of disubstituted acetylene monomers activated by Mo/W-Sn complex catalysts, which do not tolerate highly polar functionalities, was assumed to be a key step in the polymer synthetic procedures. We and other groups have explored several approaches to prepare PDSAs with latent reactive functionalities, which are inactive to Mo/W-Sn complex catalysts but can be used as highly reactive sites for post-polymerization modification. Click chemistry, Michael-type addition reactions, the use of activated esters and other strategies are demonstrated by recently published examples. These works indicate that post-polymerization modification is an efficient route to the synthesis of various functional PDSAs.

Abstract:The carbonylation of carbenes through catalytic cycles is highly desirable due to the importance of ketene-mediated reactions in organic synthesis. In this investigation, a highly efficient and mild catalytic approach toward ketene intermediates has been developed based on Pd-catalyzed carbonylation of diazo compounds with CO. When alpha-diazocarbonyl compounds or N-tosylhydrazone salts are heated in the presence of a palladium catalyst under atmospheric pressure of CO, ketene intermediates are formed in situ, where they undergo further reactions with various nucleophiles such as alcohols, amines, or imines. The Pd-catalyzed tandem carbonylation-Staudinger cycloaddition gives beta-lactam derivatives in good yields with excellent trans diastereoselectivity. The results from DFT calculation on the reaction mechanism suggest that Pd is involved in the [2 + 2] cycloaddition process and affects the diastereoselectivity of the beta-lactam products by assisting isomerization of the addition intermediate. On the other hand, the acylketenes generated from the Pd-catalyzed carbonylation of alpha-diazoketones react with imines in a formal [4 + 2] cycloaddition manner to afford 1,3-dioxin-4-one derivatives. This straightforward carbonylation provides a new approach toward highly efficient catalytic generation of ketene species under mild conditions.

Abstract:This review reports a series of mechanistic studies on Pd-catalyzed C−C cross-coupling reactions via density functional theory (DFT) calculations. A brief introduction of fundamental steps involved in these reactions is given, including oxidative addition, transmetallation and reductive elimination. We aim to provide an important review of recent progress on theoretical studies of palladium-catalyzed carbon–carbon cross-coupling reactions, including the C−C bond formation via C−H bond activation, decarboxylation, Pd(II)/Pd(IV) catalytic cycle and double palladiums catalysis.

Abstract:Addition reactions involving conjugate addition and 1,2-addition of various nucleophiles to unsaturated bonds is one of the most important areas in modern organic chemistry, allowing the direct formation of carbon-carbon or carbon-heteroatom bonds under mild conditions. In comparison with other transition metals, palladium complexes provides high catalytic efficiency, also being inexpensive when compared with rhodium. The Pd-catalyzed asymmetric addition reactions have become a common tool for the construction of optically active compounds as well as biologically active molecules and natural products. In the past decades, many novel and efficient Pd catalyst systems based on various chiral ligands have been developed and high stereoinduction could be achieved with both nucleophilic and electrophilic reactions partners. The asymmetric conjugate additions of different types of nucleophiles, such as boronic acids, diphenylphosphine, aryltriethylsiloxanes and amines, to electron-deficient olefins provided a useful strategy to construct carbon stereocenters with highly chemo-, regio- and stereoselectivity. The palladium-catalyzed enantioselective 1,2-addition to various electrophiles, including aldehydes, ketones, aldimines, ketimines, olefins, allenes and cumulenes, have emerged as a powerful approach for the construction of chiral alcohols, amines, allylic compounds, etc. In this chapter, the recent achievements and developments in palladium-catalyzed enantioselective addition processes and synthetic applications are summarized.
