Abstract:Covalently modifying electrochemiluminescence (ECL) luminophores to alter their energy levels or generate energy/electron transfer processes for improved performance is hindered by the complex design and fabrication processes. In this study, non-covalent bond self-assembly was employed to enhance the ECL property of gold nanoclusters with tryptophan (Try) and mercaptopropionic acid (MPA) as ligands (Try-MPA-gold nanoclusters). Specifically, through the molecular recognition of Try by cucurbit[7]uril, some non-radiative transition channels of the charge carriers on the surface of the Try-MPA-gold nanoclusters were restricted, resulting in a significant enhancement of the ECL intensity of the nanoclusters. Furthermore, rigid macrocyclic molecules acted on the surface of the nanoclusters through self-assembly, forming a passive barrier that improved the physical stability of the nanoclusters in the water-phase and indirectly improved their luminescent stability. As an application, cucurbit[7]uril-treated Try-MPA-gold nanoclusters (cucurbit[7]uril@Try-MPA-gold nanoclusters) were used as signal probes, and Zn-doped SnO2 nanoflowers (Zn-SnO2 NFs) with high electron mobility were used as electrode modification material to establish an ECL sensor for kanamycin (KANA) detection, utilizing split aptamers as capture probes. The advanced split aptamer sensor demonstrated excellent sensitivity analysis for KANA in complex food substrates with a recovery rate of 96.2 to 106.0%.

Abstract:The authors describe an enzyme-free aptamer-based assay for the determination of the model antibiotic kanamycin (Kana). The method is making use of (a) microfluidic chip electrophoresis; (b) a stirring bar carrying a gold-labeled aptamer probe, and (c) the hybridization chain reaction (HCR) for signal amplification. Firstly, a stirring bar (length: 1 cm; diameter: 0.2 mm) was modified with a large amount of duplex DNA and then hybridized with aptamer and its partially complementary chains (cDNA). In the presence of Kana, the binding between the Kana and aptamer unwinds the duplex structures and releases a corresponding amount of cDNA into the supernatant. The released cDNA triggers the HCR in the presence of H1 and H2 DNA hairpin to produce a large amount of duplex DNA chains with different lengths. At the same time, the amounts of H1 and H2 are reduced. The decreased signal of H1/H2 after several HCR cycles can be used to quantify kana in the 1 pg·mL−1 to 10 ng·mL−1, with a detection limit of 0.29 pg·mL−1. The signal is generated by reading the fluorescence, best at excitation/emission maxima of 470/525 nm. The whole detection process takes 3 min only. The assay was employed to the detection of Kana in spiked milk and fish samples. Results are consistent with those of an enzyme linked immunosorbent assay. Ratiometric Dual Signal-Enhancing-Based Electrochemical Biosensor for Ultrasensitive Kanamycin Detection

Abstract:Based on the signal amplification elements of planar VS2/AuNPs nanocomposites and CoFe2O4 nanozyme, we herein developed an electrochemical biosensor for sensitive kanamycin (Kana) quantification. A ratiometric sensing platform was presented by incorporating VS2/AuNPs nanocomposites as a support material with excellent conductivity and high specific surface area, as well as hairpin DNA (hDNA) with complementary hybridization of biotinylated Kana-aptamer. In addition, streptavidin-functionalized CoFe2O4 nanozyme with superior peroxidase-like catalytic activity were immobilized onto the aptasensor, hence the peroxidase-like catalytic reaction could yield amplified electrochemical signals. With the presence of Kana, the aptamer-biorecognition resulted in a quantitative decrease of nanozyme accumulation and an increase of methylene blue response. Under optimal conditions, the electrochemical signal ratio of the aptasensor revealed a linear relation along with the logarithmic concentration of Kana from 1 pM to 1 μM, with the limit of detection reaching to 0.5 pM. Abstract:Chitosan (CS)-stabilized platinum nanoparticles (CS/PtNPs) were employed to develop a novel aptamer-based dual-mode colorimetric and photothermal biosensor for selective detection of kanamycin (KAN). As a peroxidase-like catalyst, the CS/PtNPs showed outstanding catalytic activity for the oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide (H2O2). As a stabilizing agent, CS excelled at fixing the KAN binding aptamer on the surface of the CS/PtNPs, amplifying their catalytic activity and enhancing colloidal dispersion and stability. The oxidized TMB (TMBox) functioned as a signal for the colorimetric, photothermal aptasensor because of its observable absorbance of light in the visible and near-infrared (NIR) regions. When light from a NIR laser was absorbed by the TMBox in the reaction solution, heat was generated in inverse proportion to the KAN concentration. The developed colorimetric and photothermal modes of the aptasensor showed a linear detection range of 0.1-50 and 0.5-50 μM, with a limit of detection (LOD) of 0.04 and 0.41 μM, respectively. Moreover, the aptasensor successfully determined KAN concentrations in spiked milk samples, verifying the reliability and reproducibility in practical applications. The dual-mode aptasensor based on CS/PtNPs for KAN detection, utilizing both color change and heat generation signals through a single probe (TMBox), demonstrates rapid response, simplicity in operation, cost-effectiveness, and high sensitivity. In addition, unlike typical immunoassays, this aptamer-based peroxidase-like nanozyme activation and inhibition strategy required no washing process, which was very effective in terms of reducing the time required for an assay and sustaining a high sensitivity.

Abstract:Background: Colorimetric biosensors have important value for antibiotic residue testing. However, many previous methods were constructed based on the optical density change of certain unstable single-colored products with poor discrimination for visual measurements. Moreover, their low extinction coefficients usually result in low sensitivity of biosensors. In addition, many conventional signal amplification strategies often involve sophisticated nanomaterial preparation, inconvenient multi-step assay manipulation and limited signal amplification ability. Therefore, the development of new colorimetric biosensing strategies with excellent visual discrimination, high sensitivity and convenient manipulation is highly desirable. Results: We designed a target recycling accelerated cascade DNA walking amplification mechanism to trigger a telomerase extension-related enzymatic reaction, and developed a novel colorimetric biosensing strategy for kanamycin (Kana) assay. The target recycling was induced by an exonuclease III-assisted aptamer recognition reaction, which could also trigger the successive DNA walking at the streptavidin (SA)- and magnetic bead (MB)-based tracks. This not only caused the quantitative exposure of the telomeric substrate primers on MB surfaces but also released another strand to accelerate the SA-based DNA walking. By using the telomerase extension product to link numerous alkaline phosphatases and induce the plasmonic property change of gold nanobipyramids (Au NBPs), a colorimetric signal output strategy was constructed. This method could be applied for the high-resolution visual screening of Kana, and it also showed a very low detection limit of 17.6 fg mL-1 for assaying Kana over a wide, five-order-magnitude linear range. Significance: The quadruple nucleic acid recycling-enhanced telomerase extension resulted in the ultrahigh sensitivity of the method and also excluded the sophisticated manipulations involved in conventional biosensing strategies. The multiple enzyme catalysis-induced plasmonic property change of Au NBPs realized the stable and multicolor visual signal transduction. Together with its low cost, simple operation, high selectivity, excellent repeatability, and reliable performances, this method exhibits great potential for use in practical applications.

Abstract:The authors describe a colorimetric method for determination of kanamycin by using gold nanoparticles (AuNPs) as the element of signal-conversion and by applying hybridization chain reaction-assisted signal amplification. The assay is carried out by monitoring the absorbance change and color change adding salt to the reaction solution containing kanamycin (analyte), hairpin DNA probe, and AuNPs. Three hairpin DNA probes with sticky ends were absorbed on the AuNPs via their sticky ends. Cating with DNA prevents them from salt-induced aggregation (which leads to a color change from red to blue) in the complete absence of kanamycin. In contrast, in the presence of kanamycin, the aptamer hairpin DNA probe binds kanamycin, and the newly exposed section of DNA triggers a cascade of hybridization chain reactions with formation of numerous dsDNAs. On addition of salt, the AuNPs form blue aggregates due to the repulsion between dsDNA and AuNPs. Under optimal conditions, the ration of absorbance at 520 and 630 nm drops with the kanamycin concentration in the range from 1 to 40 μM, and the limit of detection is 0.68 μM. The assay can selectively distinguish kanamycin from other antibiotics. The method was applied to kanamycin detection in (spiked) milk samples and gave excellent recoveries.

Abstract:Aptamer-based lateral flow analysis (Apt-LFAs) has promising applications in many fields. Nanozymes have demonstrated high potential in improving the performance of Apt-LFAs and have been increasingly utilized in recent studies. In this study, we developed a nanozyme-based Apt-LFA for the rapid and sensitive detection of kanamycin by using a novel dual-functionalized AuNPs@polyA-DNA/GpG-Cu2+ nanozyme as a nanoprobe. In the nanoprobe design, the polyA-cDNA strand can discriminate a kanamycin aptamer from the kanamycin/aptamer complex, and the GpG-Cu2+ complex can amplify the detection signal by catalyzing the chromogenic reaction. The nanozyme Apt-LFA can quantify kanamycin in the range of 1–250 ng/mL with an LOD of 0.08 ng/mL, which demonstrated a 4-fold sensitivity improvement and had a wider linear range than the conventional AuNP-based LFA. The Apt-LFA was successfully applied to the detection of kanamycin in honey with good recoveries. Our dual-functionalized AuNP nanoprobe is easily prepared and can be highly compatible with the conventional AuNP-DNA-based LFA platform; thus, it can be extended to the application of Apt-LFAs for other small molecules.

Abstract:The misuse of kanamycin in the breeding industry can pose a threat to human health through food exposure. Therefore, it is crucial to monitor kanamycin (Kana) levels in food. This study presents a novel colorimetric approach for detecting kanamycin based on the aggregation of gold nanoparticles (AuNPs) induced by kanamycin. To achieve this, a single-standard DNA (ssDNA) aptamer was employed to bind the surface of AuNPs and maintain their dispersion under high salt concentrations. Upon adding Kana, the aptamer selectively binds to it and separates from the gold surface, resulting in the aggregation of AuNPs. This leads to a color change in the solution (from red to purple to blue) which can be observed under salt conditions. The proposed sensor demonstrated a linear range of 0.5 - 3 nM and a limit of detection (LOD) of 0.11 nM under optimal conditions. Its practicability was tested by monitoring kanamycin in three food samples, namely milk, honey, and vitamin C effervescent tablets, which resulted in satisfactory spiked recoveries. The sensor's miniaturization, convenience, simplicity, and low cost make it a desirable choice for fast and highly sensitive detection of Kana.

Abstract:The analysis of residual kanamycin (KANA) has drawn great attentions based on its potential threat to human health and environment caused by its residual in aquatic and soil system. Herein, an ultrasensitive fluorescent aptasensing system was proposed based on the fluorescence change resulting from the conformation switch of kanamycin-aptamer (KANA-aptamer) with or without kanamycin in the sensing system, in which berberine was employed as the fluorescence reporter and KANA-aptamer as the recognition unit. Free berberine emits weak fluorescence at 535 nm in aqueous solution, and its fluorescence enhanced remarkably after it intercalated to KANA-aptamer. Once kanamycin was introduced into the sensing system, the randomly coiling structure of KANA-aptamer switched to a hairpin form, which made the dissociation of berberine from the KANA-aptamer, thus the fluorescence was quenched. Various experimental conditions including the concentration of berberine, KANA-aptamer, pH, ion strength and equilibrium time were studied in detail. Under the optimal experimental condition, a good correlation was obtained between the fluorescence of the sensing system and the concentration of kanamycin ranged from 5.0 nM to 71.0 nM with LOD at 2.3 nM. The presented fluorescent aptasensor also showed a delightful recovery of 96.50%–101.20% in human serum and milk. The excellent analytical performance of kanamycin shows a promising prospect in the practical application.

Abstract:In this work, a multiply-amplified peroxidase-like colorimetric strategy was proposed for the high-specific recognition and ultrasensitive detection of kanamycin (Kana). Based on two Kana-aptamer triggered sequential reactions, G-quadruplex (G4) and DNA (hairpins) modified Ni–Fe layered double oxides (LDOs) could be obtained simultaneously. Later, a three-dimensional G4/LDO frame networks, as a novel DNAzyme, with enhanced peroxidase-like catalytic activity was assembled through electrostatic interaction. This DNAzyme catalyzed 3,3′,5,5′-tetramethylbenzidine oxidation for the colorimetric detection of Kana. The enhancement principle was discussed and the charge transfer process during the catalytic reaction was investigated. Under the optimal experiment conditions, the proposed method exhibited high sensitivity, where the linear range is from 10 fM to 10 nM (r2 = 0.992), and the limit of detection is 3 fM (S/N = 3). The practicability of this assay was demonstrated by successfully application of residual Kana detection in genuine milk and urine samples.

Abstract:Herein, we report the development of a simple, sensitive, inexpensive, and homogeneous detection method for the analysis of DNA hybridization based on the optical properties of hydroxylamine-amplified gold nanoparticles (Au NPs) in solution phase. The assay relies on a sandwich-type DNA hybridization in which DNA targets are first hybridized with capture DNA probes immobilized on the surface of magnetic beads and then sandwiched with Au NPs modified with biotinylated reporter DNA. Au NPs, after being anchored on the magnetic beads, are then dispersed in solution by the dehybridization and enlarged by using a mixture of HAuCl4 and NH2OH. The Au NP growth signal which is used for the quantitative analysis of sequence-specific DNA can be easily monitored by the naked eye directly or an UV-vis spectrophotometer. Surface plasmonic signature of the enlarged Au NPs and the kinetics of the Au NP growth in the homogenous phase containing of HAuCl4 and NH2OH have also been studied. As a result, such a homogeneous assay allows the detection of 30-base DNA targets down to the 100 amol level, which offers great promise for facilitating sensitive detection of other biorecognition events.

Abstract:We have developed a new nanoparticle (NP)-based chemiluminescent (CL) method for the sensitive detection of DNA hybridization. The "sandwich-type'' DNA technique comprises NH2-functionalized capture DNA immobilized on magnetic beads that can hybridize with one end of the target DNA. The other end of the target is recognized by a reporter DNA probe labeled with Au NPs on its 3-terminus. The hybridization events were monitored by a simple and direct luminol-AgNO3 CL reaction with a detection limit of 0.5 pM through a non-stripping Au NPs catalysis. Moreover, the CL reaction of non-stripping Au NPs-luminol-AgNO3 offers the advantages of being simpler and faster as compared to previous stripping Ag NPs and CuS methods. This simple and easy-to-use technique holds great promise for clinical applications of DNA hybridization.

Abstract:Simple assay format-based SERS methods for sensitive target substance analysis is of great significance for the development of on-site monitoring biosensors. Herein, taking the typical antibacterial kanamycin (KANA) as a subject, a simple, highly sensitive and specific SERS aptasensor was developed by manipulating DNA hydrogel network to fish plasmonic core-shell nanoparticles. A competitive binding mode of aptamer, ligation-rolling circle amplification (L-RCA), gap-containing nanoparticles (GCNPs) with embedded Raman reporters were integrated into the sensor. In the presence of KANA, the double stranded DNA (dsDNA) structure of the aptamer was disrupted, and the released primers were used to construct two kinds of circularized padlock probes (CPPs) which were partially complementary. DNA hydrogel network was formed through the intertwining and self-assembly of two RCA-generated single stranded DNA (ssDNA) chains, during which GCNPs and magnetic beads (MBs) were entangled and incorporated. Finally, KANA quantification was successfully achieved through the quantification of the DNA hydrogel. Overall, this novel SERS aptasensor realized a simple and ultrasensitive quantification of KANA down to 2.3 fM, plus excellent selectivity, and precision even for real food samples. In view of innovative fusion across L-RCA-based DNA hydrogel and SERS technique, the proposed method has promising potential for application in on-site detection and quantification of trace food contaminants.

Fluorescent and polarity-switchable photoelectrochemical dual-mode homogeneous sensing platform for ultrasensitive kanamycin detection based on EXO III-driven signal amplification

Abstract:A fluorescent and photoelectrochemical (PEC) dual-mode biosensor based on target biorecognition-triggered cyclic amplification was constructed for Kana detection. With the assistance of the catalyzed reaction of exonuclease III, a Kana-aptamer DNA duplex was designed for conducting the cyclic release of G-rich DNA sequence as well as output DNA S2. The released G-rich sequence triggers the fluorescence (FL) of thioflavin T (ThT), the intensity of which is positively correlated with the Kana concentration. The linear range is 0.2 to 30 nM, and the detection limit reaches 0.07 nM. Simultaneously, the released output DNA S2 was captured by Fe3O4@CdTe-probe ssDNA and then combined with methylene blue to realize the transduction of polarity-reversed PEC signal, leading to the sensitive detection of Kana with a linear range of 0.2 to 40 nM and a calculated detection limit of 0.2 nM. The outstanding performance endows the dual-mode biosensor a promising prospect for practical application.

Abstract:The signal transduction of most target induced strand displacement-based assays relies on the conformational changes of aptamers, significantly limiting the general applications of these sensors. We report on a very simple and general sensor named signaling-probe displacement electrochemical aptamer-based sensor (SD-EAB), in which signal transduction is induced only by the affinity binding between an aptamer and its target and completely independent of the conformational state of the aptamer. A typical SD-EAB is comprised of a gold electrode immobilized with DNA duplexes formed between a thiolated capture probe (aptamer or its short complementary strand) and a redox tagged signaling probe (short complementary strand or aptamer). In the presence of target, the signaling probe is displaced and released from the electrode surface, leading to the decrease of current proportional to the logarithm of target concentrations. SD-EAB achieved the reagentless detection of kanamycin A with 7 orders of magnitude dynamic ranges (1nM–10mM). Amazingly, SD-EAB clearly differentiated kanamycin A from its structural analogues kanamycin B by showing opposite current change. In contrast, its counterpart, the label-free electrochemical impedance spectroscopy (EIS)-based sensor, was one thousand times less sensitive than SD-EAB and had a narrow dynamic range (1–100μM) due to its limited tolerance of nonspecific adsorption of kanamycin A.

Abstract:The plasmonic colorimetric biosensors are very favorable for the on-site testing and naked-eye screening of analytes from real samples, but how to realize their highly sensitive assays with simple manipulations is still a great challenge. Herein we designed a target-triggered dual cascade nucleic acid recycling strategy to amplify the assembly of a hyperbranched DNA nanostructure and thus developed a novel kanamycin colorimetric biosensing method. The first cycle arising from the aptamer recognition-triggered strand displacement reaction and its cascade cycle constructed on the catalytic reaction of two nucleases could release an output DNA to trigger the assembly of the DNA nanostructure. Based on the high capture of alkaline phosphatase at this DNA nanostructure to induce the localized surface plasmon resonance change of gold nanobipyramids (Au NBPs), an ultrasensitive colorimetric signal transduction strategy was constructed. Through the measurement of the shift of the characteristic absorption wavelength of Au NBPs, a very wide linear range from 10 fg mL to 1 ng mL and a very low detection limit of 1.4 fg mL were obtained. Meanwhile, the obvious multicolor change of Au NBPs could be used for the visual semi-quantitative analysis of Kana residues. The whole homogeneous assay process well simplified the manipulation and also ensured the excellent repeatability. These excellent performances determine the great potential of the method for future applications.

Abstract:By the employment of a homogeneous biorecognition reaction to induce the assembled formation of DNA nanostructures at an electrode, herein we develop a novel biosensing method for the ultrasensitive electrochemical detection of kanamycin (Kana) antibiotic. A DNA complex consisting of Kana-aptamer and a hairpin DNA with an exposed 3'-end was first designed for conducting the homogeneous reaction with Kana in the presence of exonuclease I (Exo I). It resulted in the production of a hairpin DNA with a blunt terminus, which could be used for triggering the assembled formation of a layer of DNA nanostructures with orderly distribution and abundant biotin sites at a gold electrode. Then, high-content methylene blue and horseradish peroxidase (HRP)-functionalized gold nanotags would be captured onto the electrode to realize the electrocatalytic signal transduction. Due to the Exo I and HRP-assisted dual signal amplification, a very low detection limit of 9.1 fg mL-1 was obtained for the Kana assay along with a very wide linear range over five-order of magnitude. Considering the excellent performance of the method, it exhibits a promising prospect for practical applications.

Abstract:The authors describe an aptamer-based liquid crystal (LC) assay for kanamycin (KNM) that displays visible signal changes. The KNM aptamer was immobilized, along with the N,N-dimethyl-N-(3-(trimethoxysilyl)propyl)-1-octadecanaminiuchloride (DMOAP) on a glass slide, and this results in a homeotropic orientation of the LCs film. In the presence of KNM, its interaction with the aptamers will result in the formation of G-quadruplexes. These will destroy the orientated arrangement of the LCs on the surface to result in a color change from pink to green that can be visually observed by using crossed polarizers. The method possesses high specificity and the detection limit is as low as 1 nM. Unlike current laboratory-based KNM assays, this method does not require instrumental read-out which is highly beneficial in terms of on-site food analysis.Graphical abstractSchematic presentation of an aptamer-based liquid crystal assay for detection of kanamycin.
