Abstract:beta-glucosidase produced by Aspergillus niger AS 3.4309 was carried out under Solid State Fermentation (SSF). The effects of medium composition and incubation conditions on the enzyme activity were investigated and suitable solid medium components and optimum incubation conditions for fermentation process were therefore established. The maximum beta-glucosidase production (508 U g(-1) dry matter) occurred after 72 h of incubation at 28 degrees C in optimized solid medium containing wheat bran 100%, ammonium chloride 2%, MgSO4 center dot 7H(2)O 0.5% and KH2PO4, 1.5% at the optimal conditions including moisture content of solid substrate 70% (w/w) initial pH 6.0 and inoculum level at 1 x 10(7) spores g(-1) of solid substrate.
A Qualitative and Quantitative High-Throughput Assay for Screening of Gluconate High-Yield Strains by Aspergillus Niger.

Abstract:A novel two-step high-throughput strategy was developed for screening of gluconate high-yield strains by Aspergillus niger. The first step was fast qualitative assay according to the indicator color change, the second step was quantitative assay according to the absorbance of chelate formed with Cu(2+) at 810nm. The accuracy of high-throughput assay was comparable to that of high-performance liquid chromatography (HPLC). The correlation coefficient between CuSO4 assay and HPLC assays was exceeding 0.99 by statistical analysis. As a result, 3 high-yield mutants were screened out from 1000 viable single colonies, the mutants II-2-A1, IV-7-C6, and V-11-C5 were further validated in 5L of bioreactor. The average production rates were 15.5%, 32.8%, and 12.1% higher than that of the parental strain, respectively.

Abstract:The Aspergillus niger strain ZBY-7 was selected as the original strain of glucose-6-phosphate dehydrogenase production. After mutagenesis of the strain by means of UV irradiation and nitrosoguanidine, mutants of Aspergillus niger resistant to a certain metabolic inhibitor were obtained. Five of the mutants showed increased glucose-6-phosphate dehydrogenase production. The mutant resistant to antimycin A ( Aspergillus niger AM-23) produced the highest level of glucose-6-phosphate dehydrogenase (695.9% of that produced by the original strain).

Abstract:α-Glucosidase is a crucial enzyme for the production of isomaltooligosaccharide. In this study, a novel method comprising eosin Y (EY) and α- d -methylglucoside (AMG) in glass plates was tested for the primary screening of α-glucosidaseproducing strains. First, α-glucosidase-producing Aspergillus niger strains were selected on plates containing EY and AMG based on transparent zone formation resulting from the solubilization of EY by the hydrolyzed product. Conventional methods that use trypan blue (TB) and p -nitrophenyl-α- d -glucopyranoside (pPNP) as indicators were then compared with the new strategy. The results showed that EY-containing plates provide the advantages of low price and higher specificity for the screening of α-glucosidase-producing strains. We then evaluated the correlation between the hydrolytic activity of α-glucosidase and diffusion distance, and found that good linearity could be established within a 6–75 U/ml enzyme concentration range. Finally, the hydrolytic and transglycosylation activities of α-glucosidase obtained from the target isolates were determined by EY plate assay and 3,5-dinitrosalicylic acid-Saccharomyces cerevisiae assay, respectively. The results showed that the diameter of the transparent zone varied among isolates was positively correlated with α-glucosidase hydrolytic activity, while good linearity could also be established between α-glucosidase transglycosylation activity and non-fermentable reducing sugars content. With this strategy, 7 Aspergillus niger mutants with high yield of α-glucosidase from 200 obvious single colonies on the primary screen plate were obtained.

Abstract:The filamentous fungus Aspergillus niger is widely exploited by the fermentation industry for the production of enzymes, particularly glucoamylase. Although a variety of genetic techniques have been successfully used in wild-type A. niger, the transformation of industrially used strains with few conidia (e.g., A. niger N1) or that are even aconidial (e.g., A. niger O1) remains laborious. Herein, we developed genetic tools, including the protoplast-mediated transformation and Agrobacterium tumefaciens-mediated transformation of the A. niger strains N1 and O1 using green fluorescent protein as a reporter marker. Following the optimization of various factors for protoplast release from mycelium, the protoplast-mediated transformation efficiency reached 89.3% (25/28) for N1 and 82.1% (32/39) for O1. The A. tumefaciens-mediated transformation efficiency was 98.2% (55/56) for N1 and 43.8% (28/64) for O1. We also developed a marker-free CRISPR/Cas9 genome editing system using an AMA1-based plasmid to express the Cas9 protein and sgRNA. Out of 22 transformants, 9 albA deletion mutants were constructed in the A. niger N1 background using the protoplast-mediated transformation method and the marker-free CRISPR/Cas9 system developed here. The genome editing methods improved here will accelerate the elucidation of the mechanism of glucoamylase hyperproduction in these industrial fungi and will contribute to the use of efficient targeted mutation in other industrial strains of A. niger.
Impact of Overexpressing NADH Kinase on Glucoamylase Production in Aspergillus Niger.

Abstract:Glucoamylase has a wide range of applications in the production of glucose, antibiotics, amino acids, and other fermentation industries. Fungal glucoamylase, in particular, has attracted much attention because of its wide application in different industries, among which Aspergillus niger is the most popular strain producing glucoamylase. The low availability of NADPH was found to be one of the limiting factors for the overproduction of glucoamylase. In this study, 3 NADH kinases (AN03, AN14, and AN17) and malic enzyme (maeA) were overexpressed in aconidial A. niger by CRISPR/Cas9 technology, significantly increasing the size of the NADPH pool, resulting in the activity of glucoamylase was improved by about 70%, 50%, 90%, and 70%, respectively; the total secreted protein was increased by about 25%, 22%, 52%, and 26%, respectively. Furthermore, the combination of the mitochondrial NADH kinase (AN17) and the malic enzyme (maeA) increased glucoamylase activity by a further 19%. This study provided an effective strategy for enhancing glucoamylase production of A. niger.

Abstract:Aspergillus niger is widely used in industrial enzyme production for its excellent protein expression and secretion capacity. The differences of physiological behaviors and metabolic flux distribution between Aspergillus niger mutant and wild strains under same cultivation conditions are investigated,so as to determine limited factors in glucoamylase production. Based on kinetic analysis,it is confirmed that the mutant strain gets a higher maximum specific growth rate( + 30%),a lower by-product productivity(- 90%) and a higher substrate uptake efficiency( + 30%),which implies significant differences in carbon distribution and substrate usage efficiency between these two strains. By applying Flux Balance Analysis( FBA),it is found that supplies of reducing power and ribose are main factors which effect cell growth. What's more,precursor amino acids is confirmed to be the main limited factor in glucoamylase production. These conclusions provide significances for subsequent bioprocess optimization and strain gene modification.

Abstract:A mutant of Aspergillus niger 3.2130,deficient in acid protease expression,have been isolated after in vivro UV-mutagenesis of conidiospores and selected by halo-screening on the gelatin/casein plate.According to the study of mutagenesis condition,its optimum condition was found.Acid protease activities of 3.2130-L4 was 3.7% of the parental strain and β-glucosidase activity was mainly normal.This strain may be used as the host for efficient expression of foreign proteins.

Abstract:Cellulase producing strain,Aspergillus glaucus XC9,was selected as the initial strain for mutation.Four mutant strains(E2-26,E5-65,U6-31,EU7-22)were obtained after it was treated multiplicatively with UV-ray,ethyl methane sulphonate(EMS)and LiCl.The CMC activity of mutants increased to 1.7,1.4,1.3 and 2.0 times comparing with the initial strain respectively.The spore colours of mutants also varied.The morphological and fermentative properties of mutant strain EU7-22 were studied in this paper.The producing capability of cellulase of mutant strain was remarkably enhanced.

Abstract:α-Galactosidases are broadly used in feed, food, chemical, pulp, and pharmaceutical industries. However, there lacks a satisfactory microbial cell factory that is able to produce α-galactosidases efficiently and cost-effectively to date, which prevents these important enzymes from greater application. In this study, the secretory expression of an Aspergillus niger α-galactosidase (AGA) in Pichia pastoris was systematically investigated. Through codon optimization, signal peptide replacement, comparative selection of host strain, and saturation mutagenesis of the P1' residue of Kex2 protease cleavage site for efficient signal peptide removal, a mutant P. pastoris KM71H (Muts) strain of AGA-I with the specific P1' site substitution (Glu to Ile) demonstrated remarkable extracellular α-galactosidase activity of 1299 U/ml upon a 72 h methanol induction in 2.0 L fermenter. The engineered yeast strain AGA-I demonstrated approximately 12-fold higher extracellular activity compared to the initial P. pastoris strain. To the best of our knowledge, this represents the highest yield and productivity of a secreted α-galactosidase in P. pastoris, thus holding great potential for industrial application.
Mutation-screening in Xylanase High-yield Strains by Low-energy Ion Implantation Technique

Abstract:With ion implantation,a high xylanase-producing strain Aspergillus niger N212 was selected.Based on an orthogonal experiment,an optimal fermentation condition was obtained for this high-yield strain.The suitable medium was composed of 8 % corncob,1.0 % wheat bran,0.1 % Tween 80,0.5 %(NH4)2SO4,0.5 % NaNO3,and the other component was the same as Mandels'.At present,under our experiment condition,xylanase activity of Aspergillus niger N212 reached to a level of 600 IU/mL.

Abstract:Background Glucoamylase is one of the most industrially applied enzymes, produced by Aspergillus species, like Aspergillus niger . Compared to the traditional ways of process optimization, the metabolic engineering strategies to improve glucoamylase production are relatively scarce. Results In the previous study combined multi-omics integrative analysis and amino acid supplementation experiment, we predicted four amino acids (alanine, glutamate, glycine and aspartate) as the limited precursors for glucoamylase production in A. niger . To further verify this, five mutants namely OE-ala, OE-glu, OE-gly, OE-asp1 and OE-asp2, derived from the parental strain A. niger CBS 513.88, were constructed respectively for the overexpression of five genes responsible for the biosynthesis of the four kinds of amino acids ( An11g02620 , An04g00990 , An05g00410 , An04g06380 and An16g05570 ). Real-time quantitative PCR revealed that all these genes were successfully overexpressed at the mRNA level while the five mutants exhibited different performance in glucoamylase production in shake flask cultivation. Notably, the results demonstrated that mutant OE-asp2 which was constructed for reinforcing cytosolic aspartate synthetic pathway, exhibited significantly increased glucoamylase activity by 23.5% and 60.3% compared to CBS 513.88 in the cultivation of shake flask and the 5 L fermentor, respectively. Compared to A. niger CBS 513.88, mutant OE-asp2 has a higher intracellular amino acid pool, in particular, alanine, leucine, glycine and glutamine, while the pool of glutamate was decreased. Conclusion Our study combines the target prediction from multi-omics analysis with the experimental validation and proves the possibility of increasing glucoamylase production by enhancing limited amino acid biosynthesis. In short, this systematically conducted study will surely deepen the understanding of resources allocation in cell factory and provide new strategies for the rational design of enzyme production strains.

Abstract:Enzymatic saccharification is a key step in the green conversion of lignocellulose to biofuels and other products. A key deficiency in common biocatalytic systems, such as Trichoderma reesei, is the insufficient presence of beta-glucosidase (BGL). This study intended to develop an efficient process of BGL production as an enhancement to the T. reesei system. The authors investigated the process optimization of BGL by the mutant strain Aspergillus niger C112, which was previously developed in the authors' laboratory. The culture medium and process (carbon, nitrogen, temperature, and pH) were optimized for cost-effective BGL production, which led to a maximum BGL activity of 8.91 +/- 0.35 U/mL. In addition, the dynamics of the physio-chemical parameters (zeta potential and dissolved organic matter) of the process were studied and showed good correlations to the yield of BGL. Furthermore, a three-dimensional excitation-emission matrix fluorescence spectroscopy was successfully applied for analyzing the component, origin, and dynamics of dissolved organic matter, which contributed to a further understanding and optimization of BGL production.
Optimization by Response Surface Methodology of Solid-state Fermentation Medium for α-Galactosidase Production by Aspergillus niger ZJUQH

Abstract:In order to enhance the production of α-galactosidase by Aspergillus niger ZJUQH,response surface methodology was applied to optimize the solid culture medium.A fractional factorial design was used to investigate the main factors affecting α-galactosidase yield.The results showed that two factors played important roles in the production of α-galactosidase.Central composite design and response surface analysis were applied to establish a second-order regression equation model for the yield of α-galactosidase.At last,the optimal fementation condition were obtained by response surface analysis as follows: peptone 0.6 g,Na2HPO4.12H2O 0.08 g,ratio of solid to water 0.41 and inoculum quantity 1.5 mL/5g.Under the optimal conditions,the activity of α-galactosidase reached(77.21±2.01) U/g,which was increase by 57.41% compared with(49.05 ± 2.11) U/g before the oprimization,that is,peptone 0.4 g,Na2HPO4.12H2O 0.08 g,ratio of solid to water 0.33 and inoculation quantity 1.5 mL/5 g.
Screening of Aspergillus oryzae strain with a high production of aminoacylase by color-changing circle method

Abstract:The color-changing circle method was adopted to screening Aspergillus oryzae (Asp.) strains with a high yield of aminoacylase, which reduced the screening load and selection time. This method was validated by experiments and applied further to screening of Asp. strains induced by ultraviolet ray and diethyl sulfate, a mutation named W53 with a high yield of aminoacylase was obtained, whose growth characteristics and enzyme activity time course were investigated. The results show that the enzyme production rate and growing rate of Asp. Strains are synchronized and the cell biomass and the enzyme activity both reach their highest simultaneously. The enzyme production process of the mutation W53 is found consistent with that of original Asp. strain W19; however, the enzyme activity of mutation W53 can reach as high as 2 569 U/g when disposing TV-acetyl-DL-methionine (N-Ac-DL-Met), which is 14.9% higher than that of strain W19.

Abstract:With ion implantation (N+, energy 10 keV and dosage 1.56 × 1015 N+ cm-2), a high xylanase-producing strain Aspergillus niger N212 was selected. Based on an orthogonal experiment, an optimal fermentation condition was designed for this high-yield strain. The suitable medium was composed of 8% corncob; 1.0% wheat bran; 0.1% TWEEN20; 0.5% (NH4)2SO4; 0.5% NaNO3; 0.5% FeSO4, 7.5 × 10-4; MnSO4·H2O, 2.5 × 10-4; ZnSO4, 2.0 × 10-4; CoCl2, 3.0 × 10-4. At present, under our experiment condition, xylanase activity of Aspergillus niger N212 reached a level of 600 IU/ml, almost increased by 100% in xylanase production and the time of yielding xylanase was largely reduced to 12 h at 28 °C.

Abstract:From the 10 strains of Aspergillus,the strain A.niger FTA-008 was selected and it possessed highβ-glucosidase activity(2.89 U/mL).The period ofβ-glucosidase production by this strain was 3 days.A set of primers were synthesized based on the sequence ofβ-glucosidase gene(bgl1)(AJ132386)of A.niger.The cDNA fragment was amplified with RT-PCR using the total RNA of A.niger FTA-008 as template.The cloned gene fragment was 2 511 bp.Compared with sequence AJ132386, the identity of amino acid sequence was 96.8%.The cDNA was successfully expressed in Pichia pastoris and the maximum β-glucosidase activity(7.85 U/mg)was achieved in the BMMY medium containing 0.5%methanol and 240 r/min at pH 6.0 under 30°C for 108 h.Fig 3,Tab 1,Ref 13

Abstract:The purpose of this study was to obtain an engineered Aspergillus niger strain with high glucoamylase activity by overexpressing the glucoamylase gene glaA and α-amylase gene amyA in A. niger CICC2462. Three recombinant strains containing a single copy of amyA (1A), containing two copies of amyA (2A), and coexpressing amyA and glaA (AG), respectively, were constructed. The transcript levels of amyA in 1A and 2A were increased by 2.95 folds and 3.09 folds, respectively. The levels of amyA and glaA in AG were increased by 1.21 folds and 2.86 folds, but the maximum extracellular glucoamylase activities did not differ significantly. In addition, after 1% casein phosphopeptides (CPPs) was added to the fermentation medium, the maximum extracellular glucoamylase activities for strains 1A, 2A, and AG were 35,200, 37,300, and 40,710 U/ml, respectively, which were significantly higher than that of the parental strain CICC2462 (28,250 U/ml), while CPPs alone had no effect on the parental strain CICC2462. We demonstrate that overexpression of amyA and glaA substantially increases the expression and secretion of glucoamylase in A. niger, and CPPs effectively improves the yield of glucoamylase in recombinant A. niger strains overexpressing amyA and glaA. The newly developed strains and culture methods may have extensive industrial applications.

Abstract:This work mainly investigated the screening and breeding of α-galactosidase-producing microorganisms.The strain was isolated by preliminary screening with selection plate and re-screening with shaking flask culture from soy fermented products and other strains from traditional fermented foods.The strain selected can produce α-galactosidase of 22.27 U/mL at 96h cultivation.The isolated strain was tentatively identified by traditional identification methods as Aspergillus foetidus,and denominated as Aspergillus foetidus ZU-G.The Aspergillus foetidus ZU-G was further breed by 60Co-gamma irradiation and low energy N+ implanting methods.Experimental results demonstrated that α-galactosidase activity was increased by 20.7 %,23.4% contrasting with the original strain,and enzyme activities were about 26.88 U/mL and 33.18 U/mL after breeding,respectively.
