Abstract:Photoredox-catalyzed aminoarylation and thioamination of unactivated alkenes have been developed, providing novel synthetic routes to access synthetically challenging quaternary carbon-centered benzoindolizidinones and trifluoromethylthiolated piperidines using readily available starting materials. Notably, these transformations were enabled by merging amidyl radical generation from N-alkyl benzamides with oxidant incorporation. Density functional theory calculations were performed to understand the reaction mechanism and to rationalize the regioselectivities. Moreover, the newly developed catalytic aminoarylation provided a convenient synthetic route for natural product tylophorine and its gem-dimethyl analogues with greatly improved drug-like properties such as enhanced solubility and stability. Abstract:[GRAPHICS] Photochromic pyrans, including [3H]naphtho[2,1-b]pyrans, [2H]naphtho[1,2-b]pyrans, indeno-fused naphtho[1,2-b]pyrans, and heteroannulated pyrans, were synthesized in excellent yields through a facile one-pot procedure by reaction of propargyl alcohol and naphthol or phenol derivatives in the presence of 5 mol % PPTS and 2 equiv of (MeO)(3)CH. Symmetrical and nonsymmetrical bispyrans can also be prepared using the protocol.

Abstract:A facile approach toward 3-(o-aminophenyl)pyrroles was achieved by remodeling of N-sulfonyl-3-acyl indoles with N-phenylglycines under photoredox catalysis. This strategy enables the conversion of fused bicyclic systems into biaryl scaffolds, mechanically involving a Giese-type radical addition to C2=C3, C2–N cleavage of resulting indoline skeleton, and regenerating a new pyrrole motif. This novel methodology may provide an innovative strategy for the synthesis of potentially valuable 3-(o-aminophenyl)pyrroles and related alkaloids.

Abstract:In this study, ten phenylpropionyl phenylethylamines, including five previously undescribed ones ( 1a / b , 2a / b , and 3 ), five known analogues ( 4 – 8 ), and two established phenylpropanoids precursors ( 9 , 10 ) were isolated from the aerial parts of Chloranthus henryi Hemsl. Their structures, including absolute configurations, were determined by high-resolution mass spectrometry, enantio-separation, electronic circular dichroism calculation, and single crystal diffraction. Compounds 1a and 1b were the first examples of natural hetero- [2 + 2] cycloaddition products between phenylpropionyl phenylethylamine and phenylpropene. The plausible hetero- [2 + 2] biosynthesis pathway was confirmed by a photocatalytic biomimetic synthesis in eight steps, which also led to the production of three other potential natural homo -[2 + 2] adducts ( 1′a / b , 2′ , and 3′ ). Bioactivity screening indicated that these adducts bear medium inhibitory activity on nitric oxide generation, with IC 50 values of 6 – 35 μ M in RAW 264.7 macrophages.

Abstract:The synthesis, properties, and application of carbonyl-bridged phenanthrene, namely phenanthrenone are described. A new synthetic route involving palladium-catalyzed annulation between 4,5-dibromo-9-fluorenone and internal alkynes was developed to simplify the synthesis of phenanthrenone, which is a type of undeveloped building blocks. The reaction tolerates a variety of substituents at the alkynes and proceeds in moderate yields. Phenanthrenone derivatives possess the advantages of both fluorenone and phenanthrene backbones, such as extended conjugation, good planarity and rigidity, and tunable energy gap. These attractive features make phenanthrenone an ideal electron-deficient π-building block for developing novel polycyclic aromatic hydrocarbons. In addition, the phenanthrenone derivatives can be served as organic photoredox catalysts for organocatalytic atom transfer radical polymerization and oxidation of alcohol.

Abstract:A novel and convenient approach to the synthesis of polysubstituted isochromanones is described. Irradiation of 2-formyl phenylalkeno-derivatives with UV light in benzene solution afforded the corresponding products in up to 98% yield. Abstract:Photochemical approach to the synthesis of novel benzo[a]pyrano[3,2‐c]phenazin‐4‐ones was elaborated. Photochemical properties of terarylenes with quinoxalines bridge unit and allomaltol fragment was investigated. We have demonstrated that starting compounds with hydroxyl group in 3‐hydroxy‐4‐pyranone substituent does not undergo any photoinduced transformations. Wherein, conversion to methoxy derivatives allows one to realize photochemical 6π‐electrocyclization for considered quinoxalines. Based on data of x‐ray analysis the observed difference in reactivity is connected with the presence of hydrogen bond in hydroxyl derivatives. As a result of carried out research photochemical approach to novel benzo[a]pyrano[3,2‐c]phenazin‐4‐ones was implemented.

Abstract:[reaction: see text] Phenylene-linked bisnaphthopyrans were synthesized in good yields via the one-pot reaction of bis-propargyl alcohols with naphthols. Temperature-dependent photochromism in 1,4-phenylene-linked bispyrans leads to up to 60 nm bathochromic shift between the colored species formed at room temperature and at -20 degrees C. Better fatigue resistance and higher colorability was observed in 1,4-phenylene-linked bis-[2H]-naphtho[1,2-b]pyrans by comparison to the 1,3-phenylene linked bis-[2H]-naphtho[1,2-b]pyrans.

Abstract:Semi-one-pot synthesis of phenanthrenes from styrenes and arenes was developed through cross-dehydrogenative coupling. A sequence of Heck-type coupling and photo-cyclization were involved and a variety of functionalities were tolerated. Abstract:AbstractFacile syntheses of 2H‐chromenes or 9‐functionalized phenanthrenes under mild conditions in moderate to good yields have been developed. They each involve annulations of arynes with α,β‐unsaturated compounds bearing different electron‐withdrawing groups (EWGs). Depending on the natures of the different EWGs, the reactions proceed by different pathways: enals react with arynes through a tandem [2+2] cycloaddition/thermal electrocyclic ring‐opening/6e‐electrocyclization sequence to afford 2H‐chromenes, whereas acyl‐/ethoxycarbonyl‐/cyano‐substituted styrenes undergo Diels–Alder reactions with arynes followed by aromatization to afford 9‐functionalized phenanthrenes. The scope, limitations, regioselectivities and mechanisms have been studied and are discussed in detail.

Abstract:We report the photo-mediated 1,2-aryl migration of 2‑chloro-1-arylpropanones to 2-arylpropionic acids using HCOONa as an acid scavenger. This pragmatically focused study obviates the multiple-step sequence in the industrially employed, ZnO-promoted rearrangement strategy, and offers rapid access to various 2-arylpropionic acids under environmentally friendly conditions. Furthermore, the successful transfer of this batch photochemistry to a continuous flow platform led to improved scalability and enabled the gram-scale synthesis of loxoprofen.

Abstract:A concise and efficient photoinduced rearrangement of (E)-3-arylvinyl-4H-chromen-4-ones for the synthesis of benzoaryl-5-yl(2-hydroxyphenyl)methanones is described. Benzoaryl-5-yl-(2-hydroxyphenyl)methanones were obtained in 77-95% yields via the irradiation of (E)-3-arylvinyl-chromones in the 95% EtOH with a high-pressure mercury lamp at room temperature under Ar atmosphere. The reported method provides a novel procedure for the synthesis of α,α'-diaryl ketone derivatives without addition of any transition metals and oxidants or other additives. A plausible mechanism was proposed, and the rearrangement product was characterized by NMR, HRMS, and X-ray.

Abstract:Both sulfonyl and phosphorothioate are important privileged structural motifs which are widely presented in pharmaceuticals and agrochemicals. Herein, we describe an efficient approach to synthesizing sulfonyl-containing phosphorothioates by merging photoredox and copper catalysis at room temperature. This protocol is compatible with a wide range of substrates and can be applied to the late-stage modification of complex molecules. Control experiments are conducted to demonstrate the generation of the sulfonyl radical in the transformation.

Abstract:A unified strategy involving visible-light-induced iminyl-radical formation has been established for the construction of pyridines, quinolines, and phenanthridines from acyl oximes. With fac[Ir(ppy)(3)] as a photoredox catalyst, the acyl oximes were converted by 1e(-) reduction into iminyl radical intermediates, which then underwent intramolecular homolytic aromatic substitution (HAS) to give the N-containing arenes. These reactions proceeded with a broad range of substrates at room temperature in high yield. This strategy of visible-lightinduced iminyl-radical formation was successfully applied to a five-step concise synthesis of benzo[c]-phenanthridine alkaloids.
