Abstract:Over recent years, keratin has gained great popularity due to its exceptional biocompatible and biodegradable nature. It has shown promising results in various industries like poultry, textile, agriculture, cosmetics, and pharmaceutical. Keratin is a multipurpose biopolymer that has been used in the production of fibrous composites, and with necessary modifications, it can be developed into gels, films, nanoparticles, and microparticles. Its stability against enzymatic degradation and unique biocompatibility has found their way into biomedical applications and regenerative medicine. This review discusses the structure of keratin, its classification and its properties. It also covers various methods by which keratin is extracted like chemical hydrolysis, enzymatic and microbial treatment, dissolution in ionic liquids, microwave irradiation, steam explosion technique, and thermal hydrolysis or superheated process. Special emphasis is placed on its utilisation in the form of hydrogels, films, fibres, sponges, and scaffolds in various biotechnological and industrial sectors. The present review can be noteworthy for the researchers working on natural protein and related usage.

Abstract:Keratin constitutes the major component of the feather, hair, hooves, horns, and wool represents a group of biological material having high cysteine content (7-13%) as compared to other structural proteins. Keratin -based biomaterials have been investigated extensively over the past few decades due to their intrinsic biological properties and excellent biocompatibility. Unlike other natural polymers such as starch, collagen, chitosan, the complex three-dimensional structure of keratin requires the use of harsh chemical conditions for their dissolution and extraction. The most commonly used methods for keratin extraction are oxidation, reduction, steam explosion, microbial method, microwave irradiation and use of ionic liquids. Keratin -based materials have been used extensively for various biomedical applications such as drug delivery, wound healing, tissue engineering. This review covers the structure, properties, history of keratin research, methods of extraction and some recent advancements related to the use of keratin derived biomaterials in the form of a 3-D scaffold, films, fibers, and hydrogels.

Abstract:Keratins are a family of fibrous proteins anticipated to possess wide-ranging biomedical applications due to their abundance, physicochemical properties and intrinsic biological activity. This review mainly focuses on the biomaterials derived from three major sources of keratins; namely human hair, wool and feather, that have effective applications in tissue engineering, wound healing and drug delivery. This article offers five viewpoints regarding keratin i) an introduction to keratin protein extraction and keratin-based scaffold fabrication methods ii) applications in nerve and bone tissue engineering iii) a review on the keratin dressings applied to different types of wounds to facilitate wound healing and thereby repair the skin iv) the utilization of keratinous materials as a carrier system for therapeutics with a controlled manner v) a discussion regarding the main challenges for using keratin in biomedical applications as well as its future prospects.

Abstract:This review aims to trace the developments of keratin extraction techniques over the last 70 years and possibilities for future research. Keratin is a fibrous structural protein naturally present in the appendages of animals such as hair, wool, feathers, hooves and hides. Currently millions of tons of these appendages are discarded as waste products by the meat, poultry, textile and leather industries. These keratin-rich wastes lack environmental-friendly disposal methods, and are often dumped in landfills or incinerated. Over the last few years, several studies have developed various methods to repurpose these wastes as a potential source for obtaining keratin. Keratin has recently been demonstrated to have applications within the biomedical field (as a scaffold material in tissue engineering, for drug delivery and wound healing), cosmetic products and environmental remediation. To meet the demand for keratin in the above-mentioned applications, research on extracting keratin from different natural sources including keratinous wastes and improving the extraction efficiency thereof is imperative. Hence, with a brief introduction to the structure and occurrence of keratin, the present review extensively focuses on current methods of its extraction from hair, feathers, wool, hides and hooves. Also, the review identifies gaps in current research and outlines future directions in this field.

Abstract:Advances in the extraction, purification, and characterization of keratin proteins from hair and wool fibers over the past century have led to the development of a keratin-based biomaterials platform. Like many naturally-derived biomolecules, keratins have intrinsic biological activity and biocompatibility. In addition, extracted keratins are capable of forming self-assembled structures that regulate cellular recognition and behavior. These qualities have led to the development of keratin biomaterials with applications in wound healing, drug delivery, tissue engineering, trauma and medical devices. This review discusses the history of keratin research and the advancement of keratin biomaterials for biomedical applications.

Abstract:Over the past few decades, naturally derived biomaterials have been extensively used in tissue engineering and regenerative medicine owing to their biological function, structural support, excellent biocompatibility, and favorable biodegradability characteristics. Traditionally, keratin has been extracted from wool, feathers, horns, and other animal sources for industrial use, and it has also been used as a biomaterial to develop scaffolds, hydrogels, and other forms for biomedical applications. Recently, keratin extracted from human hair has emerged as a fascinating biomaterial, which as a human-derived protein, exhibits excellent biocompatibility, no immune reaction upon transplantation, good cellular interaction activity, and biodegradability. Recent development of well-defined and proficient processes for human hair keratin extraction has led to the fabrication of various types of keratin-based biomaterials, which have been employed in successful approaches for tissue regeneration. The use of human hair keratin-based biomaterials for translational biomedical applications requires better understanding of the molecular properties and biological function of keratin. This review provides a critical summary of molecular characteristics, cellular interactions, various extraction strategies, and recent advances in biomedical applications of human hair keratin that could be used to improve tissue regeneration for regenerative medicine.

Abstract:Keratin, a bio-sourced polymer of high sulfur content, reckons as the major component of the feather, hair, hooves, horns, and wool. Keratin-based formulations (KFs) have drawn great academic interest due to their intrinsic biofunctionality and high biocompatibility. KFs have been widely utilized in different fields, e. g., drug delivery, wound healing, tissue engineering, etc. Besides pure KFs, the blend systems of keratin and other (bio)polymers, as novel keratinous formulations, have started to trigger great interest, in recent years. In this study, an overview of the structure, features, extraction methods, and recent advances related to the application of KFs for various areas is provided. Furthermore, an overview of different KFs (e. g., 3-D scaffolds, films, fibers, and even hydrogels) and keratin blended with natural/synthetic polymers are discussed.

Abstract:Keratin is a structural fibrous protein and the core constituent of animal by-products from livestock such as wool, feathers, hooves, horns, and pig bristles. This natural polymer is also the main component of human hair and is present at an important percentage in human and animal skin. Significant amounts of keratin-rich animal tissues are discarded worldwide each year, ca. 12 M tons, and the share used for keratin extraction and added-value applications is still very low. An important stream of new potential raw materials, represented by animal by-products and human hair, is thus being lost, while a large-scale valorization could contribute to a circular bioeconomy and to the reduction in the environmental fingerprint of those tissues. Fortunately, scientific research has made much important progress in the last 10–15 years in the better understanding of the complex keratin architecture and its variability among different animal tissues, in the development of tailored extraction processes, and in the screening of new potential applications. Hence, this review aims at a discussion of the recent findings in the characterization of keratin and keratin-rich animal by-product structures, as well as in keratin recovery by conventional and emerging techniques and advances in valorization in several fields.

Abstract:Keratins are naturally derived proteins that can be fabricated into several biomaterials morphologies including films, sponges and hydrogels. As a physical matrix, keratin biomaterials have several advantages of both natural and synthetic materials that are useful in tissue engineering and controlled released applications. Like other naturally derived protein biomaterials, such as collagen, keratin possess amino acid sequences, similar to the ones found on extracellular matrix (ECM), that may interact with integrins showing their ability to support cellular attachment, proliferation and migration. The ability of developing biomaterials that mimic ECM has the potential to control several biological processes and this is the case for keratin which has been used in a variety of biomedical applications due to its biocompatibility and biodegradability. This review describes the progress to date towards the use of keratin in the field of wound healing, tissue engineering and drug delivery applications, with highlight to reports of particular relevance to the development of the underlying biomaterials science in this area.

Abstract:In tissue engineering and regenerative medicine applications, the utilization of bioactive materials has become a routine tool. The goal of tissue engineering is to create new organs and tissues by combining cell biology, materials science, reactor engineering, and clinical research. As part of the growth pattern for primary cells in an organ, backing material is frequently used as a supporting material. A porous three-dimensional (3D) scaffold can provide cells with optimal conditions for proliferating, migrating, differentiating, and functioning as a framework. Optimizing the scaffolds' structure and altering their surface may improve cell adhesion and proliferation. A keratin-based biomaterials platform has been developed as a result of discoveries made over the past century in the extraction, purification, and characterization of keratin proteins from hair and wool fibers. Biocompatibility, biodegradability, intrinsic biological activity, and cellular binding motifs make keratin an attractive biomaterial for tissue engineering scaffolds. Scaffolds for tissue engineering have been developed from extracted keratin proteins because of their capacity to self-assemble and polymerize into intricate 3D structures. In this review article, applications of keratin-based scaffolds in different tissues including bone, skin, nerve, and vascular are explained based on common methods of fabrication such as electrospinning, freeze-drying process, and sponge replication method.

Abstract:Keratin is a kind of natural polymer that is abundant in feathers, wool, and hair. Being one of the natural biomolecules, keratin has excellent biological activity, biocompatibility, biodegradability, favorable material mechanical properties, and natural abundance, which exhibit significant biological and biomedical application potentials. At present, the strategies commonly used for preparing keratin from hair, feathers, wool, etc. include physical, chemical, and enzymatic methods. The present article mainly reviews the structure, classification, preparation methods, and the main biological applications of keratin, and these applications cover wound healing, hemostasis, targeted release of tissue engineering drugs, and so on. Extraction and characterization of keratin from bovine hoof: A potential material for biomedical applications

Abstract:Abstract Keratin from the hoof is a less explored source for making valuable products. In this paper we present the extraction of pure keratin from bovine hooves and characterized them to better address the possible exploitation of this bio-resource as an alternative material for tissue engineering applications. The keratin protein from the pulverized hooves was extracted by reduction, which was observed to be pure, and two polypeptide chains of molecular weight in the range of 45–50 and 55–60 KDa were determined using SDS-PAGE assay. FTIR analysis complementing circular dichroism (CD) data, established that hoof keratin predominantly adopted α-helical conformation with admixture of β-sheet. The keratin was shown to have appreciably high denaturation temperature (215°C) as indicated by differential scanning calorimetric (DSC) analysis. Thermogravimetric analysis (TGA) also showed the retention of 50% of the original weight of the sample even at a temperature of 346°C. The keratin from the hoof had been observed to be biocompatible when analyzed with MTT assay using fibroblast cells, showing more than 90% cell viability. Hence, hoof keratin would be useful for high value biomedical applications.

Abstract:Keratin is a durable, fibrous protein which is mainly present in higher vertebrates (mammals, birds and reptiles) and humans epithelial cells. Food industry especially the meat market, slaughter house and wool industry produces million of tons of keratin containing biomass. These industries are constantly growing and the major producers include USA, Brazil and China, account for more than 40 million tons per year. These proteins constitute keratin by-products have from 15 to 18% nitrogen, 2-5% sulphur, 3.20% mineral elements and 1.27% fat and 90% of proteins. The organic waste rich in keratin can be utilized as a natural source using chemical and mechanical methods. The natural keratin obtained by biomass does not contain any harmful chemical and can be used directly to produce variety of cosmetics, creams, shampoos, hair conditioners and biomedical products. The natural protein is more compatible to use or apply on human skin and hairs. The monomeric units of natural keratin can penetrate in the skin and hair cuticle and able to nourish the skin without any side effects. In the present review various strategies for the purification and separation of keratin from the organic waste have been described and use of natural keratin in cosmetics and pharmaceutical industry has also been explored.

Abstract:In the present review, we have focused on the utilization of keratin from poultry bio-mass. Keratin is a tough, fibrous protein that is mostly found. There are millions of tonnes of keratin-containing biomass produced by the food sector, particularly the meat market, slaughterhouses and wool industry. These proteins which make up the byproduct of keratin contain between 15-18% of Nitrogen, 2-5% of sulphur, 1.20% of Fat and 90% of Protein. It is an eco-friendly and economical method for the production of uniaxially and randomly oriented polymeric products. The chemical and mechanical techniques can be utilized right away to make a wide range of cosmetics, lotions, shampoos, hair conditioners and biomedical items. The natural protein is better suited for usage on or application to human hair and skin. In the present study, various sources of keratin, purification and separation of keratin from organic waste have industrial application and their future perspectives have all been discussed.

Abstract:Keratin is a natural protein with a high content of cysteine residues (7–13%) and is widely found in hair, wool, horns, hooves, and nails. Keratin possesses abundant cell-binding motifs such as leucine-aspartate-valine (LDV), glutamate-aspartate-serine (EDS), and arginine-glycine-aspartate (RGD), which benefit cell attachment and proliferation. It has been confirmed that keratin plays important roles in every stage of wound healing, including hemostasis, inflammation, proliferation, and remodeling, making keratin-based materials good candidates for wound dressings. In combination with synthetic and natural polymers, keratin-based wound dressings in the forms of films, hydrogels, and nanofibers can be achieved with improved mechanical properties. This review focuses on the recent development of keratin-based wound dressings. Firstly, the physicochemical and biological properties of keratin, are systematically discussed. Secondly, the role of keratin in wound healing is proposed. Thirdly, the applications of keratin-based wound dressings are summarized, in terms of the forms and functionalization. Finally, the current challenges and future development of keratin-based wound dressings are presented.

Abstract:Keratin is widely recognized as a high-quality renewable protein resource for biomedical applications. A large amount of rabbit hair waste is produced in textile industries, because it has high medullary layer content, but poor spinnability. Therefore, it is of great significance to extract keratin from waste rabbit hair for recycling. In this research, an ultrasonic-assisted reducing agent-based extraction method was developed and applied to extract keratin from rabbit hair. The results showed that the ultrasonic treatment had a certain destructive effect on the structure of the fiber, and when combined with reducing agent, it could effectively promote the dissolution of rabbit hair, and extract keratin with high molecular weight between 31 and 94 kDa. The structure and properties of keratin were studied. Compared to the rabbit hair, the cystine content of keratin was significantly reduced, and the secondary structure changed from α-helix to β-sheet. The keratin products show excellent biocompatibility and antioxidant capacity. In addition, large keratin particles can be formed by assembly with a balance between intermolecular hydrophobic attraction as the concentration of urea in keratin solution decreased during dialysis.

Abstract:Orthopedic implants and bone regeneration are widely recognized as the most effective treatments for bone tissue disorders. Despite this, there exists a substantial disparity between the growing demand for bone implants and their insufficient supply, emphasizing the need for meticulous attention in the creation of engineered bone constructs. Bioactive materials have garnered significant attention for their ability to mimic tissues in terms of mechanical, chemical, and physical properties. While numerous natural and synthetic polymers have been discovered, keratin stands out in tissue engineering due to its natural self-assembly tendency, biocompatibility, biodegradability, non-toxicity, and ability to prevent bacterial biofilm formation. This review presents the rationale for using keratin and its composites in bone tissue engineering, as well as the techniques for fabricating and enhancing the properties of keratin-based biomaterials for treating orthopedic deformities. The focus of this study is on the potential of keratin, along with emerging technologies, in the development of orthopedic implants. Various extraction methods for keratin and the development of keratin-based biomaterials for bone implants are explored, with the aim of achieving more precise and eco-friendly methods for site-specific tissue replacement. The combination of keratin with different components has the potential to revolutionize biomaterials development for bone tissue regeneration.Graphical Abstract
