Abstract:Organic fluorines usually cannot act as halogen bond donors, as a result of the extreme electronegativity and least polarizability of the fluorine atom. However, when the electronegative ability of the substituents bound to the carbon atom is very strong, organic fluorines do show positive electrostatic potentials (ESPs) along the CF bond and thus can form halogen bonds with electron donors. In this work, the effects of six different substituents, i.e., NO2, CF3, CN, COOH, CHO, and CCH, on the ESPs of the F atom were studied using the M06-2x method. When two or three substituents with very strong electron-withdrawing ability, such as NO2 and CN, are linked to the C atom, positive ESPs take place on the outermost portion of the F atom. However, the ESPs remain negative along the CF bond with the introduction of relatively weak electron-withdrawing substituents, irrespective of the number of the subsituents. In addition, some complexes between fluorinated molecules with positive ESPs on the F atom and NH3 were calculated, to characterize the structural and energetic features of these specific halogen bonds. Finally, some crystal structures extracted from the Cambridge Structural Database were selected to provide experimental evidence of these interactions involving the CF bond. The results presented in this work are very important for the modern definition of halogen bonding as well as for the development of pharmaceuticals and a wide range of functional material. Study on Halogen Bonding of Organofluorine Compounds in China

Abstract:This review summarizes studies on halogen bonding of fluorine-containing alkyl and aryl iodides in China. From 1987 to 1993, Chen et al. found that there existed donor-acceptor interaction between fluorinated organic (di)iodides, as Lewis acids, and organic Lewis bases such as amines and ethers, which represented early important advances for the research on the non-covalent force currently called as halogen bonding. From 2001 to now, several groups have used halogen bonding as driving force to conduct researches on crystal engineering. In this catogery, Zhu et al. investigated the one-dimensional self-assembly between perfluoro-alpha, pi-diioodalkanes and amines, ethers, and hexamethylphosphamide. Jin et al. studied the complexation between fluorinated aryl iodides and various N-heterocycles, whereas Zhang and Li et al. constructed supramolecular double and quadruple helices from one or two molecular components. Jin et al. conducted extensive studies on C-I center dot center dot center dot pi halogen bonding and its applications in crystal engineering. Wang and Wan et al. utilized halogen bonding to induce trianglular aromatic molecules to co-assemble into two-dimensional honeycomb arrays on surface, whereas Wang et al. utilized halogen bond to induce mono-layer and layer-by-layer self-assembly of two polymers or organic molecules. Zhao and Li et al. developed the applications of halogen bonding in solution-phase multi-site molecular recognition of foldamer receptors for ICF2-incorporated tri-armed guests. Hu, Gong, Liao et al. utilized halogen bonding to improve the material properties of a variety of organic aromatic molecules. Several groups have also used halogen bonding to increase the selectivity of a number of organic reactions. Representative examples are described, which highlight the utility of halogen bonding.
Halogen Cation Transfer in Halogen Bonding Catalysis

Abstract:The potential of fluorine to participate in halogen bonding (XB) has remained a subject of ongoing debate. The discovery of N-fluoropyridinium triflate as a unique "F-cation organocatalyst" for aziridine synthesis is considered to be the first reported example of F-halogen bonding catalysis. Nevertheless, the mechanistic details of this reaction have remained elusive, and compelling evidence supporting the F-halogen bond catalysis has been lacking. In this study, we present an in-depth computational investigation of the mechanism of this reaction to gain insight into the intriguing role of the F-cation organocatalyst. Our results, however, are inconsistent with the previous prevalent F-halogen bonding catalysis mechanism but instead, bring to light a new fluorine cation transfer mechanism. Control experiments provide further support for this alternative mechanism. Moreover, preliminary results suggest that other reactions traditionally attributed to ‘classic’ halogen bonding catalysis might also involve a halogen cation transfer process. These mechanistic insights have important far reaching implications for the rational design of halogen bonding catalytic systems.

Abstract:Four intramolecularly F center dot center dot center dot H-N hydrogen-bonded aromatic amide and hydrazide derivatives have been prepared. Their crystal structures are investigated and compared with those of their MeO center dot center dot center dot H-N hydrogen-bonded analogues. It is found that all the F center dot center dot center dot H-N hydrogen-bonded molecules form intermolecular CO center dot center dot center dot H-N hydrogen bonding and, for two of them, weak F center dot center dot center dot H-C interactions. In contrast, the MeO center dot center dot center dot H-N hydrogen-bonded molecules display only very weak intermolecular C-H center dot center dot center dot pi interactions. The hydrogen-bonded amide units in the fluorine-bearing molecules exhibit large torsion from the connecting benzene units. This has been attributed to the weaker ability of fluorine as a hydrogen-bonding acceptor, its smaller size (relative to the MeO group), and consequently the strengthened intermolecular NH center dot center dot center dot OC hydrogen bonding. The results also suggest that, although the weakness of fluorine as a hydrogen-bonding acceptor has been mainly attributed to its low polarizability and tightly contracted lone pairs, the great tendency of fluorine-bearing aromatic amides to form intermolecular CO center dot center dot center dot H-N hydrogen bonding may also play a role.

Abstract:Quantum chemical calculations have been performed to investigate the halogen bond of HArF with some brominated hydrocarbons at the MP2/aug-cc-pVTZ level. The C–Br bond in F3CBr–FArH complex is contracted, while it is elongated in other halogen-bonded complexes. However, the C–Br stretch vibration has a small red shift in all complexes. The strength of halogen bond becomes stronger in order of Csp3–Br<Csp2–Br<Csp–Br. The substitution position in CH2CHBr has a prominent effect on the strength of halogen bond. With the number of F atom in CH2CHBr, the halogen bond is stronger and a negative nonadditivity is present for F substitution. The average contribution of each F atom to the interaction energy of halogen bond is estimated to be −4kJ/mol. Additionally, two hydrogen-bonded complexes of BrCCH–FArH and HCCBr-pi-HArF have also been studied. These complexes have been analyzed with the electrostatic potentials and NBO theory.

Abstract:The information assembled in this review of the gas phase molecular structures of fluorocarbons and related compounds were derived from electron diffraction patterns and microwave spectra. We placed primary emphasis on the geometrical effects of fluorine substitution (bond distances, bond angles and internal rotational parameters). Clearly other aspects of molecular structure, such as dipole moments, average bond dissociation energies, photoelectron spectra, etc. merit codification. At first glance, interatomic distances and bond angles call attention to features which appear to be strictly localized within molecules. It is evident, however, that such a local bond concept is not consistent with the totality of structural data on the level of precision currently available. Furthermore, it is also clear that the level of accuracy now generally attainable in the computation of molecular geometries is considerably below experimentally derived quantities. Regularities in the molecular structures of organic fluorides have been uncovered. For some structural types there are strong similarities and comparable trends among the fluorides of elements other than carbon. The substitution or environmental effects on molecular structures raise interesting questions regarding the nature of chemical bonds, and the contrasts between fluorine and hydrogen substituents provide an incisive focal points for theoreticians.

Influence of C—F…H—X Interactions on Organic Reactions

Abstract:Although the debate on whether or not C―F bonds can function as H-bond acceptors lasted for tens of years, dating back to 1939 when Pauling pointed out in The Nature of the Chemical Bond that C-F bonds do not have significant power to act as proton acceptors in the formation of hydrogen bonds, more and more evidences support the existence of C― F⋯H―X interactions, and in particular, C―F···H―O and C―F···H―N interactions cannot be ignored. Because the sum of the van der Waals radii of hydrogen and fluorine atoms is reported to be around as 2.55 Å, C―F···H―X interactions may exist if the calculated distance of F···H is less than 2.50 Å. Strong C―F···H―X interactions may occur if the calculated distance is less than 2.30 Å and the F···H―X angle is greater than 120. In 2011, we observed strong fluorine effects on the Strecker reaction of ketimines: while Schreiner's thiourea could catalyze the Strecker reaction of acetophenone derived ketimine using TMSCN, it was unable to mediate the corresponding reaction of analogy α-CF3 or α-CF2H ketimines. Theoretical calculations revealed that the C―F···H―N interactions between the C―F bond of fluorinated ketimines and thiourea played the key role. This is the first report on the influence of such subtle interactions on organic reactions. Since then, reports from our and other groups revealed various types of C―F⋯H―X interactions that may be present in the reaction course, to strongly influence the reactivity and selectivity. Although successful examples are still limited, these achievements have suggested that C―F⋯H―X interactions may exist between the substrate and the catalyst; the substrate and the solvent; different reaction partners, or engender in the transition state with the reaction intermediate. Importantly, known examples demonstrate it possible to harness C―F⋯H―X interactions to tune reactivity and/or selectivity, which are useful for new reaction development, as well as for the design of new catalysts. To provide reference and inspiration for researchers engaged in organic synthesis, especially the organic fluorine chemistry, we summarize in this review the recent advances in the study of the influences of C―F⋯H―X interactions on organic reactions.
Exploring Favorable Supramolecular Interactions of Multifluorinated Aromatics in Dendronized Push‐Pull Chromophores for Electro‐Optics

Abstract:Multifluorinated aromatics serve as supramolecular synthons in the research of organic electro‐optic (EO) materials by exploiting π−π stacking interaction between the aromatic hydrocarbon and multifluorinated aromatic groups for performance improvement. However, non‐classical hydrogen bonding remains largely unexplored in fluorinated EO dendrimers. In this study, three Fréchet‐type generation 1 benzyl ether co‐dendrons were synthesized by replacing one benzyl group with 2,3,5,6‐tetrafluorobenzyl (p‐HF4Bz), pentafluorobenzyl (C6F5Bz), and 2,3,4,5‐tetrafluorobenzyl (o‐HF4Bz) groups, to afford the benzoic acid derivatives D1, D2, and D3, which were further bonded to the donor and π‐bridge moieties to afford three co‐dendronized push‐pull phenyltetraene chromophores EOD1, EOD2, and EOD3, respectively. The weak C−H⋅⋅⋅X (X = O, F) interactions in the crystal structure of D1 cumulatively add to the benzoic acid dimers to form an extended hydrogen‐bonded network, while D2 is crystallized into a centric one‐dimensional chain with strong intermolecular interactions. The poled films of EOD1 with PMMA exhibited the largest and most stable EO activity with optical homogeneity among the series. The results identify the effectiveness of weak but favorable hydrogen bonds enabled by the enhanced carbon acidity of p‐HF4Bz synthon in D1, over the interactions in D2 and D3, for the rational design of supramolecular EO dendrimers.

Experimental/Computational Study on the Impact of Fluorine on the Structure and Noncovalent Interactions in the Monohydrated Cluster of ortho-Fluorinated 2-Phenylethylamine

Abstract:Fluorine-containing medicinal compounds frequently allow modulation of physical-chemical properties. Here, we address the effect of fluorine, near the ethylamino side chain, on conformational flexibility and noncovalent interactions (NCIs) of the selected jet-cooled monohydrated cluster of 2-(2-fluoro-phenyl)-ethylamine (2-FPEA) by mass-selected resonance-enhanced two-photon ionization and ionization-loss stimulated Raman spectroscopies. Our results show that Raman spectral signatures of the 2-FPEA-H2O cluster match the scaled harmonic vibrational Raman frequencies, resulting from density functional theory calculations of the most stable 2-FPEA gauche conformer hydrogen-bonded (HB) to water, confirming the three-dimensional cluster structure. This predicted electronic structure, together with NCI analysis, allows visualization and assessment of the attractive and repulsive interactions. The comparison of the NCIs and revealed red (O-H and N-H stretches) and blue shifts (C-H stretches and CH2 out-of-plane bends) of the cluster to other class members confirm O-H···N, N-H···π, C-H···O, and C-H···F HB formation and their contribution to structure stabilization, uncovering the potential of the approach.
A Quantitative Insight into the ‘amphiphilicity’ of Covalently-Bonded Halogens from Ab Initio Calculations

Abstract:A series of complexes formed between bromine-containing molecules RBr and HF have been investigated at the MP2(full)/aug-cc-pvdz level of theory. This, together with a previous work [J.W. Zou, Y.J. Jiang, M. Guo, G.X. Hu, B. Zhang, H.C. Liu, Q.S. Yu, Chem. Eur. J. 11 (2005) 740] on the halogen-bonded RBr⋯NH3 complexes, offers a quantitative insight into the nucleophile ‘head on’ and electrophile ‘side on’ interactions of halogens. It has been shown that the stronger electron-withdrawing ability of the moiety bound to the bromine is, the weaker is the Br⋯H interaction to which it gives rise, which is contrary to the trend probed in the RBr⋯NH3 complexes. Several linear relationships have been established to provide further understanding of the ‘amphiphilicity’ of covalently-bonded halogens. The topological parameters obtained in terms of Bader's atoms in molecules (AIM) theory have also been applied for the analysis of these Br⋯H interactions.

Abstract:Intermolecular interactions, such as halogen bonding, π-π stacking, F…π interactions, and anion-π interactions, play pivotal roles in the fields of chemistry, materials science, and biological technology. In this study, N-aromatic L-proline and its fluorinated derivatives were chosen as exemplars. X-ray diffraction and NMR techniques were employed to investigate these interactions in both solid-state and solution phases respectively. Theoretical investigations encompassing geometry optimization along with QTAIM (Quantum Theory of Atoms in Molecule) and IGMH (Independent Gradient Model based on the Hirshfeld partition) analyses provided profound insights into the intermolecular interactions. Parameters obtained from X-ray diffraction analysis of single crystals provide compelling evidence for F…F interaction with intermolecular F…F distances measuring 2.879 Å and 3.125 Å in compounds 2 and 3 crystal structures respectively. QTAIM and IGMH analyses indicate that F…π contacts significantly contribute to the formation of these two fluorinated crystals while H-bonding and CH…π contacts predominantly shape the crystal structure of the non-fluorinated L-proline derivative.
General alkyl fluoride functionalization via short-lived carbocation-organozincate ion pairs

Abstract:Fluorinated organic compounds are frequently used across the chemical and life sciences. Although a large, structurally diverse pool of alkyl fluorides is nowadays available, synthetic applications trail behind the widely accepted utility of other halides. We envisioned that C(sp2)-C(sp3) cross-coupling reactions of alkyl fluorides with fluorophilic organozinc compounds should be possible through a heterolytic mechanism that involves short-lived ion pairs and uses the stability of the Zn-F bond as the thermodynamic driving force. This would be mechanistically different from previously reported radical reactions and overcome long-standing limitations of organometallic cross-coupling methodology, including competing β-hydride elimination, homodimerization and hydrodefluorination. Here, we show a practical Csp3-F bond functionalization method that expands the currently restricted synthetic space of unactivated primary, secondary and tertiary C(sp3)-F bonds but also uses benzylic, propargylic and acyl fluorides. Many functional groups and sterically demanding substrates are tolerated, which allows practical carbon-carbon bond formation and late-stage functionalization.
Electron Topological and Energetic Study of the Intermolecular Halogen Bonding Interactions in Complexes H2o Center Dot Center Dot Center Dot M (M = F-2, Clf, and Cf4)

Abstract:The halogen bonding complexes H2O center dot center dot center dot M (M = F-2, ClF, and CF4) in comparison with the hydrogen bonding H2O center dot center dot center dot HF complex are studied by high-level ab initio calculations and electron topological atoms-in-molecules (AIM) analyses. The basis set superposition error corrections are important to predict if the structures are in good agreement with the experimental results. Both the CCSD(T)/aug-cc-pVTZ calculations and the AIM analyses indicate a interaction strength order: H2O center dot center dot center dot HF > H2O center dot center dot center dot ClF > H2O center dot center dot center dot F-2 > H2O center dot center dot center dot CF4, with the interaction energies -7.91, -4.16, -1.11, and -1.05 kcal/mol, respectively. The symmetry-adapted perturbation theory analyses have been carried out towards understanding of the nature of the halogen bonding interactions in the complexes H2O center dot center dot center dot M (M = F-2, ClF, and CF4), where the exchange energies are the predominant repulsive components. For the complexes involving polar monomers, the hydrogen bonding H2O center dot center dot center dot HF and the halogen bonding H2O center dot center dot center dot ClF, the largest attractive contributions are the electrostatic energies. However, in H2O center dot center dot center dot F-2 and H2O center dot center dot center dot CF4, the attractive dispersion energies become more important, and the induction energy in the former complex is a little higher than that in the latter. In contrary to the red-shifts of H-F, Cl-F, and F-F bond stretching vibrational frequencies in the complexes H2O center dot center dot center dot M (M = HF, ClF, and F-2), the blue-shifts are predicted for C-F bonds neighboring water in H2O center dot center dot center dot CF4.

Abstract:Using ab initio calculations, the authors' predicted for the first time that the halogen-bonded complex FBr delta+center dot delta+BrF and hydrogen-bonded complex FBr delta+center dot delta+HF formed by the interactions between two positively charged atoms of different polar molecules can be stable in gas phase. It shows that halogen bond or hydrogen bond not only exists between oppositely charged atoms but also between like-charged atoms. That the attraction arising from the special halogen bond or hydrogen bond can exceed the electrostatic repulsion between two contact positively charged atoms stabilizes the complex. Of course, from the point of view of physics they can consider the interactions in FBr delta+center dot delta+BrF and FBr delta+center dot delta+HF as mainly the sum of the long range molecular interactions, namely, electrostatic, induction, and dispersion with some short-range repulsion. They found that the intermolecular electron correlation contribution representing dispersion interaction plays a crucial role in the stabilities of seemingly repulsive complexes FBr delta+center dot delta+BrF and FBr delta+center dot delta+HF. Energetic Effects Between Halogen Bonds and Anion-π or Lone Pair-π Interactions: A Theoretical Study

Abstract:Energetic effects between halogen bonds and anion-π or lone pair-π interactions have been investigated by means of ab initio MP2 calculations. 1,4-diiodo-perfluorobenzene, a very effective building block for crystal engineering based on halogen bonding, is selected in this work both as electron-deficient π aromatic ring and as halogen bond donor. Additive and diminutive effects are observed when halogen bonds and anion-π/lone pair-π interactions coexist in the same complex, which can be ascribed to the same direction of charge transfer for the two interactions. These effects have been analyzed in detail by the structural, energetic, and AIM properties of the complexes. Finally, experimental evidence of the combination of the interactions has been obtained from the Cambridge Structural Database.
